Aspartate attenuates intestinal injury and inhibits TLR4 and NODs/NF-κB and p38 signaling in weaned pigs after LPS challenge

摘要:
Purpose: This study was conducted to investigate whether aspartate (Asp) could alleviate Escherichia coli lipopolysaccharide (LPS)-induced intestinal injury by modulating intestine inflammatory response. Methods: Twenty-four weaned piglets were divided into four treatments: (1) non-challenged control; (2) LPS-challenged control; (3) LPS + 0.5 % Asp; and (4) LPS + 1.0 % Asp. After feeding with control, 0.5 or 1.0 % Asp-supplemented diets for 21 days, pigs were injected intraperitoneally with saline or LPS. At 4 h postinjection, blood and intestine samples were obtained. Results: Asp supplementation to LPS-challenged pigs improved intestinal morphology, indicated by higher jejunal and ileal villus height/crypt depth ratio and lower ileal crypt depth linearly or quadratically. Asp also improved intestinal barrier function, indicated by increased jejunal and ileal diamine oxidase activities as well as enhanced protein expression of jejunal claudin-1 linearly or quadratically. In addition, Asp decreased plasma, jejunal and ileal tumor necrosis factor-α concentration and ileal caspase-3 protein expression linearly and quadratically. Moreover, Asp down-regulated the mRNA expression of toll-like receptor 4 (TLR4) and nucleotide-binding oligomerization domain protein (NOD) signaling-related genes, nuclear factor-κB (NF-κB) p65 and p38, decreased phosphorylation of jejunal p38, and increased phosphorylation of ileal extracellular signal-related kinase 1/2 linearly or quadratically. Finally, Asp increased mRNA expressions of TLR4 and NOD signaling negative regulators including radioprotective 105, suppressor of cytokine signaling 1, toll-interacting protein, Erbb2 interacting protein and centaurin β1 linearly or quadratically. Conclusions: These results indicate that Asp supplementation is associated with inhibition of TLR4 and NODs/NF-κB and p38 signaling pathways and concomitant improvement of intestinal integrity under an inflammatory condition. © 2016, Springer-Verlag Berlin Heidelberg.

摘要:
旨在研究表达I型耐热肠毒素(STa)的重组大肠杆菌对7日龄仔猪肠道吸收、屏障、转运及抗氧化功能的影响。选取24头7日龄仔猪,分成4个处理组,分别为对照组(基础日粮)、STa组(基础日粮+ 2×10~9CFU重组菌 LMG194-STa)、LMG194组(基础日粮+ 2×10~9 CFU大肠杆菌LMG194)和K88组(基础日粮+ 2×10~9 CFU大肠杆菌K88)。使用人工乳饲养,第5天进行攻毒,第7天屠宰取样。测量回肠免疫相关基因IFN-γ、IL-4与VNN1及回肠黏膜损伤基因Villin与MMP3的相对转录量,空肠黏膜上皮连接蛋白Occludin与Claudin-1相对表达量,以及十二指肠、回肠氧化相关酶谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)的活力与氧化相关产物丙二醛(MDA)的含量。试验结果表明,与对照组相比,STa组显著降低了回肠IFN-γ和IL-4基因相对转录量,VNN1相对转录量显著上调(P<0.05);STa组显著降低了基因Villin与MMP3相对转录量(P<0.05)及蛋白Occludin与Claudin-1相对表达量(P<0.05);同时,STa组十二指肠、回肠GSH-Px和回肠MPO活力显著降低(P<0.05);STa组十二指肠丙二醛(MDA)含量显著升高(P<0.05)。结果显示,表达I型耐热肠毒素的重组大肠杆菌可导致7日龄仔猪肠道免疫、屏障及抗氧化功能下降。

Clostridium perfringens is the causative pathogen of avian necrotic enteritis. Lactobacillus spp. are well-characterized probiotics with anti-microbial and immune-modulatory activities. In the present study, we investigated the effects of L. acidophilus and L. fermentum on the growth, α-toxin production and inflammatory responses of C. perfringens. In in vitro culture experiments, both lactobacilli inhibited the growth of C. perfringens (P < 0.01), accompanied with a decrease in pH (P < 0.01). Supernatants from lactobacilli cultures also suppressed the growth of C. perfringens during 24 h of incubation (P < 0.01), but this inhibitory effect disappeared after 48 h. Both lactobacilli decreased the α-toxin production of C. perfringens (P < 0.01) without influencing its biomass, and even degraded the established α-toxin (P < 0.01). Lower environmental pH reduced the α-toxin production as well (P < 0.01). Preincubation with L. acidophilus decreased the attachment of C. perfringens to cells (P < 0.01) with the cell cytotoxicity being unaffected. Both lactobacilli pretreatment reduced the up-regulation of proinflammatory factors, peptidoglycan (PGN) receptors and nuclear factor kappa B (NF-κB) p65 in C. perfringens-challenged chicken intestinal epithelial cells (P < 0.05). In conclusion, L. acidophilus and L. fermentum inhibited the pathological effects of C. perfringens in vitro conditions. © 2017 Guo, Liu, Zhang, Li, Li, Ding and Guo.

Supplementation of branched-chain amino acids (BCAA) has been demonstrated to promote skeletal muscle mass gain, but the mechanisms underlying this observation are still unknown. Since the regulation of muscle mass depends on a dynamic equilibrium (fasted losses-fed gains) in protein turnover, the aim of this study was to investigate the effects of BCAA supplementation on muscle protein synthesis and degradation in fed/fasted states and the related mechanisms. Fourteen 26-(Experiment 1) and 28-day-old (Experiment 2) piglets were fed reduced-protein diets without or with supplemental BCAA. After a four-week acclimation period, skeletal muscle mass and components of anabolic and catabolic signaling in muscle samples after overnight fasting were determined in Experiment 1. Pigs in Experiment 2 were implanted with carotid arterial, jugular venous, femoral arterial and venous catheters, and fed once hourly along with the intravenous infusion of NaH 13 CO 3 for 2 h, followed by a 6-h infusion of [1- 13 C]leucine. Muscle leucine kinetics were measured using arteriovenous difference technique. The mass of most muscles was increased by BCAA supplementation. During feeding, BCAA supplementation increased leucine uptake, protein synthesis, protein degradation and net transamination. The greater increase in protein synthesis than in protein degradation resulted in elevated protein deposition. Protein synthesis was strongly and positively correlated with the intramuscular net production of α-ketoisocaproate (KIC) and protein degradation. Moreover, BCAA supplementation enhanced the fasted-state phosphorylation of protein translation initiation factors and inhibited the protein-degradation signaling of ubiquitin-proteasome and autophagy-lysosome systems. In conclusion, supplementation of BCAA to reduced-protein diet increases fed-state protein synthesis and inhibits fasted-state protein degradation, both of which could contribute to the elevation of skeletal muscle mass in piglets. The effect of BCAA supplementation on muscle protein synthesis is associated with the increase in protein degradation and KIC production in the fed state. © 2016 by the author; licensee MDPI, Basel, Switzerland.

This study determined whether N-acetylcysteine (NAC) could improve intestinal function through phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR), epithelial growth factor receptor (EGFR), toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-κB), adenosine 5′-monophosphate-activated protein kinase (AMPK), and type I interferon (IFN) signaling pathways in a piglet model of lipopolysaccharides (LPS) challenge. Thirty-two piglets (24-day-old) were randomly allocated to one of four treatments, with eight replicates per treatment and one piglet per replicate. The experiment consisted of four treatments in a 2 × 2 factorial arrangement with two diets (supplemented with 0 or 500 mg NAC/kg diet) and saline or LPS administration. On day 20 of the trial, piglets in the LPS and LPS + NAC groups were intraperitoneally injected with 0 (saline) or 100 μg LPS/kg BW. Blood samples were obtained at 3 h and intestinal mucosae were collected at 6 h post LPS or saline injection. The growth performance was not affected by dietary NAC. LPS induced intestinal dysfunction, as indicated by: (1) reductions in the small-intestinal glutathione concentrations and plasma d-xylose levels; (2) elevations in plasma diamine oxidase activity, mucosal MMP3 mRNA levels and caspase-3 protein abundance; (3) reduced the activities of the small-intestinal mucosal maltase, sucrase and lactase. The adverse effects of LPS on porcine intestinal function and redox status were mitigated by NAC supplementation through the activation of multiple signaling pathways involving PI3K/Akt/mTOR, EGFR, TLR4/NF-κB, AMPK, and type I IFN. Our findings provide novel mechanisms for beneficial effects of NAC in protecting the intestine from inflammation in animals. © 2017, Springer-Verlag Wien.

Inflammatory bowel disease (IBD), which includes both ulcerative colitis and Crohn's disease, is a chronic relapsing inflammation of the gastrointestinal tract, and is difficult to treat. The pathophysiology of IBD is multifactorial and not completely understood, but genetic components, dysregulated immune responses, oxidative stress, and inflammatory mediators are known to be involved. Animal models of IBD can be chemically induced, and are used to study etiology and to evaluate potential treatments of IBD. Currently available IBD treatments can decrease the duration of active disease but because of their adverse effects, the search for novel therapeutic strategies that can restore intestinal homeostasis continues. This review summarizes and discusses what is currently known of the effects of amino acids on the reduction of inflammation, oxidative stress, and cell death in the gut when IBD is present. Recent studies in animal models have identified dietary amino acids that improve IBD, but amino acid supplementation may not be adequate to replace conventional therapy. The animal models used in dietary amino acid research in IBD are described.

Clostridium perfringens encodes at least two different quorum sensing (QS) systems, the Agr-like and LuxS, and recent studies have highlighted their importance in the regulation of toxin production and virulence. The role of QS in the pathogenesis of necrotic enteritis (NE) in poultry and the regulation of NetB, the key toxin involved, has not yet been investigated. We have generated isogenic agrB-null and complemented strains from parent strain CP1 and demonstrated that the virulence of the agrB-null mutant was strongly attenuated in a chicken NE model system and restored by complementation. The production of NetB, a key NE-associated toxin, was dramatically reduced in the agrB mutant at both the transcriptional and protein levels, though not in a luxS mutant. Transwell assays confirmed that the Agrlike QS system controls NetB production through a diffusible signal. Global gene expression analysis of the agrB mutant identified additional genes modulated by Agrlike QS, including operons related to phospholipid metabolism and adherence, which may also play a role in NE pathogenesis. This study provides the first evidence that the Agr-like QS system is critical for NE pathogenesis and identifies a number of Agr-regulated genes, most notably netB, that are potentially involved in mediating its effects. The Agr-like QS system thus may serve as a target for developing novel interventions to prevent NE in chickens. © 2017 American Society for Microbiology. All Rights Reserved.

Pork is one of the most economical sources of animal protein for human consumption. Meat quality is an important economic trait for the swine industry, which is primarily determined by prenatal muscle development and postnatal growth. Identification of the molecular mechanisms underlying skeletal muscle development is a key priority. MicroRNAs (miRNAs) are a class of small noncoding RNAs that have emerged as key regulators of skeletal muscle development. A number of muscle-related miRNAs have been identified by functional gain and loss experiments in mouse model. However, determining miRNA-mRNA interactions involved in pig skeletal muscle still remains a significant challenge. For a comprehensive understanding of miRNA-mediated mechanisms underlying muscle development, miRNAome analyses of pig skeletal muscle have been performed by deep sequencing. Additionally, porcine miRNA single nucleotide polymorphisms have been implicated in muscle fiber types and meat quality. The present review provides an overview of current knowledge on recently identified miRNAs involved in myogenesis, muscle fiber type and muscle protein metabolism. Undoubtedly, further systematic understanding of the functions of miRNAs in pig skeletal muscle development will be helpful to expand the knowledge of basic skeletal muscle biology and be beneficial for the genetic improvement of meat quality traits.

杏鲍菇菌糠固态发酵工艺条件的优化及在生长猪上的应用

本试验旨在研究杏鲍菇菌糠最佳发酵工艺条件及其在生长猪日粮中的适宜添加量。以采摘3茬的杏鲍菇菌糠为原料,选取黑曲霉、枯草芽孢杆菌和嗜酸乳杆菌3株菌种,采用单因素试验和正交试验设计,考察接种量、发酵时间、料水比和混菌比例对杏鲍菇菌糠发酵后营养价值的影响。动物试验选取平均体重为20 kg的杜×长×大三元杂交生长猪24头,随机分为4组,每组6个重复,每个重复1头猪;对照组饲喂基础日粮,试验组日粮分别用5%、10%、15%发酵菌糠替代麸皮,试验期为21 d。结果表明:最佳条件为菌种接种量12%、发酵时间84 h、料水比1:1.5、混菌比例2:1:1、发酵温度37℃,在此条件下粗蛋白提高39.9%、粗纤维降低18.9%;与对照组相比,添加5%和10%发酵菌糠对生长猪的生长性能无显著影响(P>0.05),5%菌糠组和10%菌糠组的表观消化率明显优于对照组(P>0.05)。综合考虑,通过优化发酵条件,提高了杏鲍菇菌糠的营养价值,且在生长猪饲料中杏鲍菇菌糠的适宜添加量为10%。

摘要:
本实验旨在研究RNA干扰(RNAi)RACK1基因对C2C12成肌细胞中肌分化标志基因MHC和MyoG表达的影响。将人工合成的靶向RAKC1基因的3条si RNA(1,2,3)转染C2C12细胞,用RT-qPCR方法检测并筛选干扰效率最高的1条siRNA。将筛选出的si RNA转染C2C12成肌细胞并诱导细胞分化,通过RT-qPCR、Western blot和免疫荧光方法在mRNA及蛋白水平检测RACK1基因沉默后对MHC和MyoG表达的影响。此外,Western blot方法检测RACK1基因沉默后对PI3K/Akt和Erk/MAPK通路激活的影响。结果表明:RACK1-si RNA-2干扰效率最高,转染48 h后抑制率可达70%。随后,C2C12细胞转染si RNA-2,诱导分化48h后RTqPCR表明,MHC和MyoG的m RNA表达均显著性下调(P〈0.05);诱导分化72 h后,Western blot和免疫荧光结果表明MHC和MyoG的蛋白表达明显低于对照组,但AKT和Erk磷酸化水平未见明显变化。上述结果表明,干扰RACK1能显著抑制MHC和MyoG的表达,提示RACK1正向调控C2C12成肌细胞的分化。

本试验旨在研究二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)对体外培养的C2C12成肌细胞增殖和凋亡的影响.用不同终浓度DHA和EPA(0、6 25、12 5、25、50、100 μmol/L)分别作用C2C12细胞12、24、48 h后,用CCK-8法检测细胞的增殖情况;用不同终浓度DHA和EPA(0、50、100 μmol/L)分别作用C2C12细胞48 h后,用TUNEL法检测细胞凋亡情况.结果表明:终浓度为50、100 μmol/L的EPA处理细胞48 h,C2C12细胞增殖能力受到显著抑制,但不同浓度DHA处理组细胞的增殖能力无明显变化.此外,EPA处理C2C12细胞后,TUNEL法显示凋亡细胞数增加,且100μmol/L的EPA处理组细胞凋亡最明显.然而,与对照相比,DHA处理组细胞无明显凋亡.上述结果表明,EPA能抑制C2C12成肌细胞增殖并促进其凋亡,而DHA对细胞增殖和凋亡无明显影响.

胸膜肺炎放线杆菌(Actinobacillus pleuropneumoniae,APP)是一种能够引起猪传染性胸膜肺炎的细菌病原体,它给养猪业带来巨大的经济损失。外膜蛋白W(Omp W)在细菌的生命活动中起着重要作用,大量研究表明Omp W与细菌的感染、耐药性以及分子调节等都有着紧密联系,因而阐明ompW基因对胸膜肺炎放线杆菌调节机制对于该疾病的防控具有重要意义。以胸膜肺炎放线杆菌ompW基因为研究对象,成功构建自杀性质粒p EM-Omp W,为构建胸膜肺炎放线杆菌Omp W突变株以及探究ompW基因对胸膜肺炎放线杆菌分子调节机制提供了基础。

摘要:
利用自制触球及迷宫法评估了百里香酚、香芹酚、丁香酚和肉桂醛4种香精对(4.45± 0.58) g黄颡鱼(Pelteobargrus fulridraco)幼鱼的诱食剂量效应反应,试验结果表明:百里香酚、香芹酚和肉桂醛都对黄颡鱼有明显的诱食作用,其最适诱食浓度分别为(0.025~ 0.05)、0.025、0.007 g/mL;百里香酚诱食效果最佳(P < 0.05),但浓度过高,也会出现诱食抑制;而丁香酚对黄颡鱼幼鱼均无明显的诱食作用(P >0.05)。随后在基础饲料中分别添加75、150、250、500和1000 mg/kg的百里香酚饲喂黄颡鱼幼鱼,8周的养殖结果表明黄颡鱼增重率(Y)与饲料中百里香酚添加量(X)之间可用二次方程式表示Y=-0.000 7 X~2+ 0.796 3 X+ 107.08(R~2= 0.777 2),当黄颡鱼获得最大增重时,饲料中百里香酚的添加量为568.8 mg/kg。

鄂西北地区规模猪场猪瘟疫苗使用情况及免疫效果

为了解鄂西北地区规模化猪场所用猪瘟疫苗的类别及免疫效果,对鄂西北地区38家规模化猪场猪瘟疫苗的使用情况进行调研,采集829份血清进行了猪瘟抗体水平检测.结果表明:38家规模猪场中有26家(68.4%)使用政府供应疫苗,12家(31.6%)使用商品疫苗;猪瘟细胞苗和脾淋苗在猪场的使用比例分别为71.1%和28.9%;猪场母猪猪瘟疫苗使用剂量高于推荐剂量的1.2 ~6倍,生长育肥猪使用的剂量为推荐剂量的1~4倍,猪场使用政府供应的疫苗时剂量高于商品疫苗;20家猪场群体免疫合格(52.6%),18家猪场群体免疫不合格(47.4%);受检的829份血清中,阴性血清158份(19.1%),阳性血清609份(73.5%).说明鄂西北地区猪瘟疫苗以政府供应猪瘟细胞苗为主,不同养殖场免疫效果差别大,需要规范免疫程序,加强免疫抗体水平检测,以降低该地区猪瘟疫病风险.

摘要:
This study was conducted to evaluate the effects of biofloc on the species and quantity of plankton in pond water. There were four treatments in the experiment: the control, no supplementation; treatment I, pond water supplemented with glucose; treatment II, pond water supplemented with both glucose and Bacillus subtilis; and treatment III, pond water supplemented with Bacillus subtilis only. Water quality, plankton, and biofloc formation were monitored every five days. Results showed that biofloc formation was enhanced by adding glucose to the pond water. The addition of glucose together with Bacillus subtilis shortened the formation time of mature bioflocs by about five days than the supplementation with glucose only. In ponds supplemented with glucose and those supplemented with glucose + bacillus, the ammonia nitrogen, and nitrite nitrogen content in the water column decreased, and the number of cyanobacteria and green algae were significantly inhibited, while the number of zooplankton such as Rotatoria increased during the experimental period. © 2017, Israeli Journal of Aquaculture - Bamidgeh. 摘要:
This study was designed to evaluate the effect of the replacement of fish oil (FO) by soybean oil (SO) on growth performance, liver lipid peroxidation, and biochemical composition in juvenile Chinese sucker, Myxocyprinus asiaticus. Fish (13.7 ± 0.2 g) in triplicate were fed five experimental diets in which 0% (FO as control), 40% (SO40), 60% (SO60), 80% (SO40), and 100% (SO100) FO were replaced by SO. The body weight gain of fish fed SO40, SO60, or SO80 diet was similar to FO group, but diets that have 100% soybean oil as dietary lipid significantly reduced fish growth (P < 0.05). Although the level of SO resulted in increasing crude lipid content of the liver, the level of SO did not significantly alter the hepatosomatic index (HSI). Indicators of peroxidation, such as vitamin E (V E ) and thiobarbituric acid-reactive substance (TBARS) contents, were changed as increasing dietary SO. It was shown that the inclusion of SO in the diets increased V E concentrations, but reduced TBARS in the liver and total cholesterol (T-CHO) in the plasma. Linoleic acid (LA) and linolenic acid (LNA) significantly increased in fish liver fed diets that contained SO, but eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and the ratio n-3/n-6 were significantly reduced by the inclusion of dietary SO (P < 0.05). Our results indicated that the inclusion of SO increased the hepatic V E content and reduced lipid peroxidation in fish. However, diet containing 100% SO as dietary lipid could reduce growth performance. Thus, we recommended that 40–80% SO can be used as dietary lipid to replace FO for juvenile Chinese sucker. © 2017, Springer Science+Business Media Dordrecht.

试验研究不同水平的发酵木薯酒精糟对生长猪生产性能的影响.选择200头初始体重约(26.35±0.43) kg的生长猪,随机分为5个处理组,每组4个重复,每个重复10头猪.5个处理组分别为:A组为对照组,饲喂基础日粮;B组、C组和D组分别为发酵木薯酒精糟用量为2%、6%和15%的配合日粮组,E组则为用发酵木薯酒精糟直接替代6%基础日粮的混合日粮组.结果表明,B组和C组的日增重较A组分别提高了6.45%(P>0.05)和4.84%(P>0.05),各试验组料肉比均低于对照组,分别降低了7.48%、9.06%、5.12%和0.39%.日粮中添加发酵木薯酒精糟后有效改善了营养物质消化率,B组、C组、D组及E组干物质消化率较对照组分别提高了8.62%(P<0.05)、7.50%(P>0.05)、4.75%(P>0.05)和5.26%(P>0.05);粗蛋白消化率较对照组分别提高了11.43%(P<0.05)、9.50%(P<0.05)、9.61%(P<0.05)和10.61%(P<0.05);粗脂肪消化率较对照组分别提高了15.66%(P<0.05)、15.86%(P<0.05)、12.98%(P<0.05)和14.10% (P<0.05);钙的消化率则较对照组分别提高了10.46%(P<0.05)、6.50%(P>0.05)、6.93%(P>0.05)和10.60%(P<0.05).B组、C组、D组及E组较A组的养殖效益均有所提高,每千克饲料的增重成本分别降低了0.683、0.833、0.523元和0.238元,其中使用处理C组饲料的效益最好.本试验结果说明,生长猪日粮中发酵木薯酒精糟的适宜添加量为2%-6%,能有效提高猪的生产性能和养殖效益.

摘要:
The morphological revisions of Macrocytopharynxa pyriformis (Nie, 1932) Li et al., 2002; collected from the rectum of Fejervarya limnocharis (=Rana limnocharis), are presented in this paper: (1) two surfaces of the organism are not - identical left side narrower and convex, right broader and flat or slightly concave; (2) infundibulum is large and well-developed with no "fold" or "plicature" present in the middle or posterior portion; (3) micronucleus is tiny and ovoid shaped and always embedded in the middle concavity of macronucleus, which can be well revealed by ammoniacal silver staining. Our phylogenetic analysis based on SSU-rDNA showed that M. pyriformis fell into the Nyctotheroides Glade, within which four definite Nyctotheroides species were involved - N. cordiformis, N. deslierresae, N. parvus and N. hubeiensis. In combination with their morphological features, we discussed the reliability of using karyophore organelles or kinetal suture patterns as the generic taxonomic criteria. Besides, we considered that the genus Macrocytopharynxa is a junior synonym of Nyctotheroides and we transfer its type species to Nyctotheroides as Nyctotheroides pyriformis n. comb. The phylogenetic pattern of the family Nyctotheridae was also indicated in our work, but it will be necessary to analyze more species from fishes and reptiles before coming to a sound conclusion. 摘要:
microRNA分子在调节宿主细胞应对细菌感染的过程中起重要作用。为研究miR-194b-3p对马链球菌兽疫亚种(SeZ)粘附猪肾上皮细胞(PK15)能力的影响,本实验将miR-194b-3pagomir和antagomir分别转染PK15细胞,检测SeZ对PK15细胞粘附能力的变化。结果表明:在PK15细胞中转染miR-194b-3p agomir,SeZ C55138粘附率显著下降;转染miR-194b-3p antagomir, SeZ C55138粘附率显著上升,确定了miR-194b-3p能够显著抑制SeZC55138对PK15细胞的粘附。同时对其机制进行研究,显示PK15细胞表面CD44是miR-194b-3p的一个调控靶基因, miR-194b-3p通过调节CD44的表达参与影响了上述粘附过程。本实验为深入理解SeZ与PK15细胞的互作机制提供了线索。
