Pro-inflammatory cytokines play a key role in many models of hepatic damage. In addition, asparagine (Asn) plays an important role in immune function. We aimed to investigate whether Asn could attenuate lipopolysaccharide (LPS)-induced liver damage. Forty-eight castrated barrows were allotted to four groups including: (1) non-challenged control; (2) LPS-challenged control; (3) LPS+0·5 % Asn; and (4) LPS+1·0 % Asn. After 19 d feeding with control, 0·5 or 1·0 % Asn diets, pigs were injected with LPS or saline. Blood and liver samples were obtained at 4 h (early stage) and 24 h (late stage) post-injection. Asn alleviated liver injury, indicated by reduced serum aspartate aminotransferase and alkaline phosphatase activities linearly and quadratically; it increased claudin-1 protein expression linearly and quadratically at 24 h, and less severe liver morphological impairment at 4 or 24 h. In addition, Asn decreased mRNA expression of TNF-α and heat shock protein 70 (HSP70) linearly and quadratically at 4 h; it increased TNF-α mRNA expression, and HSP70 protein expression linearly and quadratically at 24 h. Moreover, Asn increased inducible NO synthase activity linearly and quadratically. Finally, Asn down-regulated the mRNA expression of Toll-like receptor 4 (TLR4) signalling molecules (TLR4, IL-1 receptor-associated kinase 1 (IRAK1), TNF-α receptor-associated factor 6), nucleotide-binding oligomerisation domain protein (NOD) signalling molecules (NOD1, NOD2 and their adaptor molecule receptor-interacting serine/threonine-protein kinase 2 (RIPK2)), and NF-κB p65 linearly or quadratically at 4 h. Oppositely, Asn up-regulated mRNA expressions of TLR4 and NOD signalling molecules (TLR4, myeloid differentiation factor 88, IRAK1, NOD2 and RIPK2), and their negative regulators (radioprotective 105, single Ig IL-1R-related molecule, Erbb2 interacting protein and centaurin β1) linearly or quadratically at 24 h. These results indicate that, in early and late stages of LPS challenge, Asn improves liver integrity and exerts different regulatory effects on mRNA expression of TLR4 and NOD signalling molecules. © The Authors 2015.

Abstract There is growing interest in glutamate as a functional amino acid in nutrition and health. This study was conducted to determine whether glutamate precursor α-ketoglutarate (AKG) could alleviate lipopolysaccharide (LPS)-induced liver injury in young pigs. Twenty-four piglets were randomly assigned to the control, LPS, or LPS + AKG group. Piglets in the control and LPS groups were fed a basal diet, whereas piglets in the NAC group were fed the basal diet supplemented with 1 % AKG. On days 10, 12, 14, and 16 of the trial, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16 of the trial, blood samples were collected 3 h after LPS or saline injection. Twenty-four hours post-administration of LPS or saline (on day 17 of the trial), piglets were killed to obtain liver for analysis. Dietary AKG supplementation alleviated LPS-induced histomorphological abnormalities and mitigated LPS-induced increases in aspartate aminotransferase (AST) activity and AST/ALT ratio (P < 0.05). Compared with the LPS group, dietary supplementation with AKG decreased plasma glutamate concentration, while increasing hepatic concentrations of glutamate, glutamine, leucine, asparagine, lysine, alanine, serine, threonine, valine, and phenylalanine (P < 0.05). LPS challenge dramatically increased concentrations of malondialdehyde and decreased glutathione peroxidase activity in the liver. Additionally, LPS challenge enhanced concentrations of AMP and total protein, as well as RNA/DNA and total protein/DNA ratios, while decreasing hepatic ADP concentrations. These adverse effects of LPS challenge were ameliorated by AKG supplementation. Collectively, dietary AKG supplementation provides a new means to ameliorate LPS-induced liver injury by increasing anti-oxidative capacity and improving energy metabolism in young pigs. © 2015 Springer-Verlag Wien.

Purpose: This study was conducted to investigate whether aspartate (Asp) could improve liver energy status in the lipopolysaccharide (LPS)-challenged pigs. Methods: Twenty-four weaned pigs were assigned to four treatments: (1) nonchallenged control (control diet and saline-treated); (2) LPS-challenged control (the same control diet and LPS-challenged); (3) LPS + 0.5 % Asp treatment (0.5 % Asp diet and LPS-challenged); and (4) LPS + 1.0 % Asp treatment (a 1.0 % Asp diet and LPS-challenged). On d 19, the pigs were injected intraperitoneally with Escherichia coli LPS at 100 μg/kg body weight, and the same volume of 0.9 % NaCl solution, respectively. All pigs were slaughtered at 24 h after LPS or saline injection, and the liver was collected for further analysis. Results: Dietary supplementation with Asp improved liver energy status evidenced by the increased ATP concentration and adenylate energy charges, and the decreased AMP concentration and AMP/ATP ratio (p < 0.05). Asp supplementation increased the mRNA expression of key enzymes in hepatic glycolysis and tricarboxylic acid (TCA) cycle, including pyruvate kinase and citrate synthase (p < 0.05), and had a tendency to increase hepatic pyruvate dehydrogenase and isocitrate dehydrogenase β mRNA expression (p < 0.10). In addition, Asp increased the mRNA expressions of hepatic AMP-activated protein kinase (AMPK) α1, AMPKα2, silent information regulator (Sirt1), and proliferator-activated receptor-γ coactivator 1α (PGC1α) (p < 0.05). Moreover, Asp increased AMPKα phosphorylation (p < 0.05). Conclusions: These results indicated that dietary supplementation of Asp could improve energy status in LPS-injured liver, which might result from motivating the metabolism pathway of TCA cycle and glycolysis and stimulating the AMPK signaling pathway. © 2014, Springer-Verlag Berlin Heidelberg.

本试验旨在研究天冬氨酸(ASP)对脂多糖(LPS)刺激仔猪结肠黏膜结构和屏障功能的影响。试验选用24头仔猪,4个处理组分别为对照组、LPS组、0.5%ASP组、1.0%ASP组。试验第19天,分别给LPS组、0.5%ASP组、1.0%ASP组试验猪注射100μg/kg体重的LPS,对照组注射等量的生理盐水。注射后24 h屠宰,取结肠样品待测。结果表明:对照组仔猪结肠形态结构完整;注射LPS后,结肠上皮部分脱落;ASP组轻度脱落,与LPS组相比,0.5%ASP组结肠隐窝深度显著升高(P<0.05);0.5%和1.0%ASP可显著缓解LPS刺激导致的结肠杯状细胞数目的增多(P<0.05);0.5%ASP具有提高结肠中性粒细胞密度的趋势(P<0.10)。0.5%和1.0%ASP可显著缓解LPS刺激导致的结肠和血浆二胺氧化酶活性的降低(P<0.05)。以上研究结果表明,日粮添加ASP可有效阻止LPS对结肠结构和屏障功能的损伤。

摘要:
Cinnamon is a traditional herb used for treatment of many human diseases. The most important chemical compounds of the essential oil are cinnamaldehyde and eugenol. Oleum cinnamomi (OCM, cinnamon oil) is increasingly used as a feed additive to animal diets. Beneficial effects of OCM in protecting tissues from inflammation and injury by endogenous and exogenous agents (such as hydrogen peroxide and lipopolysaccharide (LPS)) may result, in part, from its action on regulating amino acid metabolism in cells to favor the synthesis of glutathione (a major low-molecular-weight antioxidant) from cysteine, glycine and glutamate. In support of this notion, results of recent studies indicate that supplementing OCM (50 mg/kg diet) to a corn- and soybean meal-based diet for piglets weaned at 21 days of age enhances intestinal anti-oxidative capacity and reduces the incidence of diarrhea. Additionally, dietary supplementation with OCM ameliorates LPS-induced mucosal barrier dysfunction and mucosal damage in the small intestine. OCM holds great promise for protecting the gut from injury under conditions of inflammation, infections, and oxidative stress. © 2015, Frontiers in Bioscience. All rights reserved.

The intestine requires a high amount of energy to maintain its health and function; thus, energy deficits in intestinal mucosa may lead to intestinal damage. Asparagine (Asn) is a precursor for many other amino acids such as aspartate, glutamine and glutamate, which can be used to supply energy to enterocytes. In the present study, we hypothesise that dietary supplementation of Asn could alleviate bacterial lipopolysaccharide (LPS)-induced intestinal injury via improvement of intestinal energy status. A total of twenty-four weaned piglets were assigned to one of four treatments: (1) non-challenged control; (2) LPS+0 % Asn; (3) LPS+0.5 % Asn; (4) LPS+1.0 % Asn. On day 19, piglets were injected with LPS or saline. At 24 h post-injection, piglets were slaughtered and intestinal samples were collected. Asn supplementation improved intestinal morphology, indicated by higher villus height and villus height:crypt depth ratio, and lower crypt depth. Asn supplementation also increased the ratios of RNA:DNA and protein:DNA as well as disaccharidase activities in intestinal mucosa. In addition, Asn supplementation attenuated bacterial LPS-induced intestinal energy deficits, indicated by increased ATP and adenylate energy charge levels, and decreased AMP:ATP ratio. Moreover, Asn administration increased the activities of key enzymes involved in the tricarboxylic acid cycle, including citrate synthase, isocitrate dehydrogenase and α-ketoglutarate dehydrogenase complex. Finally, Asn administration decreased the mRNA abundance of intestinal AMP-activated protein kinase-α1 (AMPKα1), AMPKα2, silent information regulator 1 (SIRT1) and PPARγ coactivator-1α (PGC1α), and reduced intestinal AMPKα phosphorylation. Collectively, these results indicate that Asn supplementation alleviates bacterial LPS-induced intestinal injury by modulating the AMPK signalling pathway and improving energy status. 本试验旨在研究脂多糖(Lipopolysaccharide,LPS)刺激对仔猪核苷酸结合寡聚化结构域蛋白(NODs)信号通路关键基因表达的影响.试验选取12头杜×长×大断奶仔猪,分成2个处理组,每个处理组6个重复.试验组注射100 μg/kg体重的LPS,对照组注射等量的生理盐水.注射LPS或生理盐水4h后,屠宰仔猪,取空肠、回肠、肝脏、背最长肌和腓肠肌,应用实时荧光定量PCR技术测定NODs信号通路关键基因mRNA表达水平.包括NOD1、NOD2、受体相互作用蛋白激酶2 (RIPK2)、核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、矢车菊苷β1(ACAP1)和Erbb2相互作用蛋白(ERBB2IP)mRNA表达水平.结果表明:与对照组相比,LPS刺激显著提高了肝脏NOD1、NOD2、RIPK2、NF-κB和TNF-α,背最长肌ⅣOD2、RIPK2和TNF-α,腓肠肌NOD2、RIPK2、NF-κB和TNF-α,空肠RIPK2和TNF-α的mRNA表达量(P<0.05),LPS刺激有提高回肠RIPK2和NF-κB mRNA表达量的趋势(P<0.10);同时,LPS刺激显著降低组织中的负调控因子,如空肠、回肠和肝脏ACAP1,空肠ERBB2IP的mRNA的表达量(P<0.05).因此,综上表明,LPS激活仔猪肠道、肝脏和肌肉组织中NODs信号通路关键基因的表达.

摘要:
The study was performed to evaluate the effects of enzyme supplementation on performance and digestive parameters of broilers fed corn-soybean diets from 1 to 21 d of age. A total of 480 one-day-old Cobb broilers were allocated to 1 of 4 treatments, with 6 replicate pens per treatment and 20 birds per pen. The experiment consisted of a 2 × 2 factorial arrangement of treatments with 2 dietary ME levels (high ME, energy 1, 12.13 MJ/kg or low ME, energy 2, 11.92 MJ/kg) and 2 levels of supplemental enzyme (including xylanase, 1,800 IU/g, β-glucanase, 500 IU/g, and α-amylase, 800 U/g; 0 or 0.1% of diet). Enzyme supplementation had no effect on average daily weight gain, feed intake, and feed:gain. However, enzyme supplementation decreased the relative weight of the pancreas (d 7 and 21) in broilers fed the high ME diet. Low dietary ME level increased pancreatic lipase (d 7, P = 0.015), trypsin (d 14, P = 0.01; d 21, P = 0.014), amylase (d 21, P = 0.027), and pepsin (d 7, P = 0.001; d 21, P = 0.042) activity, but reduced pancreatic lipase (d 14, P = 0.03; d 21, P = 0.004) and amylase (d 14, P = 0.027) activity. Enzyme supplementation resulted in an increase in pancreatic amylase (d 7, P = 0.023), trypsin (d 7, P = 0.02; d 21, P = 0.004), lipase (d 21, P = 0.001), pepsin (d 7, P = 0.001; d 14, P = 0.004; d 21, P = 0.001), and maltase (d 14, P = 0.011, in ileum) activity. Moreover, broilers fed low dietary ME and enzyme supplementation diets had an increase in pancreatic lipase (d 21, P = 0.001) and pepsin (d 7, P = 0.001) activity. Low ME diets reduced jejunum villus height and jejunum and ileum crypt depth (d 7, 21). However, enzyme supplementation, especially enzyme supplementation in low ME diets, increased jejunum and ileum villus height and villus surface area. This suggested enzyme supplemented with low ME diet might be more effective to improve the activity of digestive enzymes and the absorptive capacity of the small intestine.

Tributyrin (TBU) is a good dietary source of butyrate and has beneficial effects on the maintenance of normal intestinal morphology. The present study tested the hypothesis that dietary TBU supplementation could alleviate intestinal injury in the acetic acid (ACA)-induced porcine model of colitis. A total of eighteen piglets (25Â d old) were randomly allocated to one of three treatment groups (control, ACA and TBU). The control and ACA groups were fed a basal diet and the TBU group was fed the basal diet supplemented with 0·1Â % TBU. On day 15 of the trial, under anaesthesia, a soft catheter was inserted into the rectum of piglets (20-25Â cm from the anus), followed by administration of either saline (control group) or ACA (10Â ml of 10Â % ACA solution for ACA and TBU groups). On day 22 of the trial, after venous blood samples were collected, piglets were killed to obtain mid-ileum and mid-colon mucosae. Compared with the control group, the ACA group exhibited an increase (P<Â 0·05) in lymphocyte counts, creatinine, PGE2, and malondialdehyde concentrations and diamine oxidase and inducible NO synthase activities in the plasma and lymphocyte density in the colon and a decrease in insulin concentrations and glutathione peroxidase activity, ileal villus height:crypt depth ratios and goblet cell numbers in the colon. These adverse effects of ACA were attenuated by TBU supplementation. Moreover, TBU prevented the ACA-induced increase in caspase-3 levels while enhancing claudin-1 protein and epidermal growth factor receptor (EGFR) mRNA expression in the colonic mucosa. Collectively, these results indicate that dietary supplementation with 0·1Â % TBU alleviates ACA-induced intestinal injury possibly by inhibiting apoptosis, promoting tight-junction formation and activating EGFR signalling. © 2014 The Authors.

Pro-inflammatory cytokines play a critical role in many models of liver injury. In addition, aspartate (Asp) plays an important role in many biological and physiological processes including liver physiology. We hypothesized that Asp could alleviate lipopolysaccharide (LPS)-induced liver injury. Forty-eight weanling pigs were assigned to four treatments including: (1) non-challenged control; (2) LPS challenged control; (3) LPS+0.5% Asp; (4) LPS+1.0% Asp. After 20-d feeding with control (0% Asp), 0.5% or 1.0% Asp supplemented diets, pigs were injected with saline or LPS. At 4 (early phase) and 24 h (late phase) post-injection, blood and liver samples were obtained. Asp attenuated liver injury indicated by reduced serum aspartate aminotransferase activity and increased ratio of serum alanine aminotransferase and aspartate aminotransferase at 24 h, and less severe histological liver damage induced by LPS challenge at 4 or 24 h. In addition, Asp supplementation to LPS challenged pigs decreased mRNA expressions of tumor necrosis factor (TNF)-alpha and cyclooxygenase-2 linearly and quadratically at 4 h, and increased mRNA expressions of these pro-inflammatory mediators linearly and quadratically at 24 h. Finally, Asp decreased mRNA expression of toll-like receptor 4 (TLR4) signaling related genes (TLR4, myeloid differentiation factor 88, IL-1 receptor-associated kinase 1, TNF-alpha receptor-associated factor (6), nucleotide-binding oligomerization domain protein (NOD) signaling related genes (NOD1, NOD2 and receptor-interacting serine/threonine-protein kinase 2) and nuclear factor-kappa B p65 linearly or quadratically at 4 h. However, Asp increased mRNA expressions of these signaling molecules linearly or quadratically at 24 h. These results indicate that, at early and late phases of LPS challenge, Asp exerts opposite regulatory effects on mRNA expression of hepatic pro-inflammatory cytokines and TLR4 and NOD signalling related genes, and improves liver integrity. 本试验旨在研究天冬氨酸(ASP)对脂多糖(LPS)刺激断奶仔猪肠道结构和黏膜抗氧化能力的影响.选取(21±1)d日龄断奶、平均体重(7.37±0.04)kg的杜×长×大三元杂交仔猪24头,随机分为对照组(基础日粮)、LPS组(基础日粮+LPS)、0.5% ASP组(基础日粮+0.5%ASP+LPS)、1.0% ASP组(基础日粮+1.0%ASP+LPS)4组,试验期24 d.试验第24天,分别给3个试验组的猪注射100 μg/kg体重的LPS,对照组注射等量的生理盐水.注射后24 h屠宰,取小肠样品待测.结果表明:0.5%或1.0% ASP可显著缓解LPS刺激导致的空肠绒毛高度降低、隐窝深度升高和绒毛高度/隐窝深度的降低,增加回肠的绒毛高度、绒毛高度/隐窝深度(P<0.05);0.5%ASP对空肠超氧化物歧化酶、回肠谷胱甘肽过氧化物酶活性有提高的趋势(P<0.10);0.5%或1.0% ASP显著降低了空肠和回肠丙二醛含量(P<0.05).以上研究结果显示,ASP有提高仔猪肠黏膜抗氧化相关酶的酶活趋势,降低过氧化物的产生,从而改善肠道形态结构.

The intestine has a high requirement for ATP to support its integrity, function and health, and thus, energy deficits in the intestinal mucosa may play a critical role in intestinal injury. Aspartate (Asp) is one of the major sources of ATP in mammalian enterocytes via mitochondrial oxidation. We hypothesized that dietary supplementation of Asp could attenuate lipopolysaccharide (LPS)-induced intestinal damage via modulation of intestinal energy status. Twenty-four weanling piglets were allotted to one of four treatments: (1) nonchallenged control, (2) LPS-challenged control, (3) LPS+0.5% Asp treatment, and (4) LPS+1.0% Asp treatment. On day 19, pigs were injected with saline or LPS. At 24 h postinjection, pigs were killed and intestinal samples were obtained. Asp attenuated LPS-induced intestinal damage indicated by greater villus height and villus height/crypt depth ratio as well as higher RNA/DNA and protein/DNA ratios. Asp improved intestinal function indicated by increased intestinal mucosal disaccharidase activities. Asp also improved intestinal energy status indicated by increased ATP, ADP and total adenine nucleotide contents, adenylate energy charge and decreased AMP/ATP ratio. In addition, Asp increased the activities of tricarboxylic acid cycle key enzymes including citrate synthase, isocitrate dehydrogenase and alpha-oxoglutarate dehydrogenase complex. Moreover, Asp down-regulated mRNA expression of intestinal AMP-activated protein kinase alpha 1 (AMPK alpha 1), AMPK alpha 2, silent information regulator 1 (SIRT1) and peroxisome proliferator activated receptor gamma coactivator-1 alpha (PGC1 alpha) and decreased intestinal AMPK alpha phosphorylation. These results indicate that Asp may alleviate LPS-induced intestinal damage and improve intestinal energy status. (C) 2014 Elsevier Inc. All rights reserved.

本试验旨在研究脂多糖(LPS)刺激对仔猪下丘脑-垂体-肾上腺(HPA)轴Toll样受体4(TLR4)信号通路关键基因表达的影响。选取12头断奶仔猪,分成2个组,每个组6个重复。试验组注射100μg/kg体重的LPS,对照组注射等量的生理盐水。注射LPS或生理盐水4 h后采血,测定血浆中与应激相关激素的含量;采血后屠宰仔猪,取下丘脑、垂体、肾上腺,测定TLR4信号通路关键基因的mRNA表达水平,包括TLR4、髓样分化因子88(My D88)、白细胞介素-1受体相关激酶1(IRAK1)、肿瘤坏死因子受体相关因子6(TRAF6)和核转录因子-κB(NF-κB)mRNA表达水平。结果表明:与对照组相比,1)LPS刺激导致血浆肿瘤坏死因子-α、皮质醇和促肾上腺皮质激素含量显著上升(P<0.05)。2)LPS刺激导致TLR4信号通路关键基因TLR4在下丘脑、垂体、肾上腺中mRNA表达水平显著升高(P<0.05);My D88在垂体、肾上腺中mRNA表达水平显著升高(P<0.05),在下丘脑中mRNA表达水平有升高的趋势(P<0.10);IRAK1在垂体、肾上腺中mRNA表达水平有升高的趋势(P<0.10);TRAF6在下丘脑、垂体、肾上腺中mRNA表达水平显著升高(P<0.05);NF-κB在垂体中mRNA表达水平显著升高(P<0.05)。试验结果表明LPS刺激激活了HPA轴,诱导了HPA轴的TLR4信号通路关键基因的表达。

试验旨在研究天冬酰胺(Asn)对脂多糖(LPS)刺激仔猪肠道结构及抗氧化能力的影响。选用24头断奶仔猪,分为对照组、LPS组、0.5%Asn组(0.5%Asn+LPS)和1.0%Asn组(1.0%Asn+LPS)。第24天,各组分别注射LPS或等量生理盐水24 h后屠宰。结果表明:LPS刺激导致空肠绒毛高度显著降低,日粮中添加0.5%或1.0%Asn显著降低了空肠、回肠的隐窝深度,提高了空肠、回肠的绒毛高度和绒毛高度/隐窝深度的比值(P<0.05);LPS刺激也导致空肠超氧化物歧化酶(SOD)活性和氧化型谷胱甘肽(GSSG)/还原性谷胱甘肽(GSH)比值显著升高(P<0.05);日粮中添加0.5%或1.0%Asn显著提高了空肠、回肠GSH的含量(P<0.05),降低了空肠、回肠GSSG/GSH的比值(P<0.01)和丙二醛(MDA)的含量(P<0.05)。结果显示,Asn缓解了LPS刺激引起的肠道结构损伤和抗氧化能力下降。

This trail was conducted to study the effect of L-proline on the growth performance, and blood parameter in the weaned lipopolysaccharide (LPS)-challenged pigs. Thirty six pigs (9.13±0.85 kg) were assigned randomly to dietary treatments in a 2×3 factorial arrangement in a 20-d growth assay. Factors were intraperitoneal injection with saline or LPS, and three dietary L-proline supplement levels (0%, 0.5%, or 1.0%). On d 10, blood samples were collected at 3 h after LPS (100 μg LPS/kg body weight [BW]) or saline injection. On d 20 of the trial, all pigs were orally administrated D-xylose (0.1 g/kg BW) at 2 h, and blood samples were collected at 3 h after LPS or saline injection. As a result, dietary supplementation with 0.5% proline had a tendency to increase average daily gain (ADG) in piglets during d 10 to 20 (p = 0.088). Without LPS challenge, dietary supplementation with 1.0% proline had no effect on growth hormone (GH) concentrations on d 10 (p>0.05), but decreased it after LPS challenge (p<0.05). There was LPS challenge× proline interaction for GH concentrations on d 10 (p<0.05). Dietary supplementation with 1.0% proline decreased glucagon concentration on d 10 after LPS challenge (p<0.05). In addition, dietary supplementation with proline increased superoxide dismutase (SOD) activity significantly on d 10 and 20 (p<0.05), and 1.0% proline increased heat shock proteins-70 concentration on d 10 (p<0.05). Moreover, proline supplementation increased diamine oxidase (DAO) concentrations after LPS challenge (p<0.05). There was LPS challenge×proline interaction for DAO (p<0.05). Furthermore, dietary supplementation with 1.0% proline increased the D-xylose level when no LPS challenge (p<0.05). These results indicate that proline supplementation could improve growth performance, increase SOD activities, and has a positive effect on the gastrointestinal tract digestibility in early weaned pigs. © 2014 by Asian-Australasian Journal of Animal Sciences.

本试验旨在研究Toll样受体4(TLR4)信号通路关键基因在断奶仔猪不同组织的分布情况。选择12头杜×长×大断奶仔猪,屠宰,取脾脏、胸腺、肠道淋巴结、下丘脑、垂体、肾上腺、肝脏、腓肠肌、皮下脂肪、空肠和回肠组织。应用实时荧光定量PCR技术测定TLR4信号通路关键基因,包括TLR4、髓样分化因子(MyD88)、IL-1受体相关激酶1(IRAK1)、肿瘤坏死因子受体相关因子6(TRAF6)、核因子-κB(NF-κB)和肿瘤坏死因子(TNF)-α在各组织的mRNA表达水平。结果表明:TLR4信号通路关键基因在所检测的11个组织中均有表达,并且组织分布规律基本一致。总体看来,主要在免疫组织(脾脏、胸腺、淋巴结)表达量较高,在皮下脂肪和肠道(空肠、回肠)表达量居中,在其他组织中表达量较低。TLR4信号通路关键基因在不同组织中表达差异较大,可能与仔猪各组织对病原的识别和抵抗能力有关。
