摘要:
试验研究罗格列酮(ROS)对脂多糖(LPS)刺激的断奶仔猪肠黏膜结构和抗氧化能力的影响.试验包括3个处理组,对照组注射生理盐水,LPS组注射100 μg/kg体重的LPS,ROS组在注射LPS之前的30 min注射3 mg/kg体重的RPS.注射LPS 3 h后取样.结果表明:ROS提高了十二指肠和回肠DNA的含量(p<O.05);ROS缓解了LPS导致的十二指肠、空肠和回肠隐窝深度的增加(p<0.10)和绒毛高度/隐窝深度的降低(p<0.01);ROS降低了空肠丙二醛含量(P<0.10),提高了十二指肠和空肠SOD活性(P<0.05).结果表明,ROS可能是通过提高肠道抗氧化功能而对LPS刺激的断奶仔猪小肠形态结构的损伤起到一定保护作用.

摘要:
A study was conducted to investigate the effects of l-arginine (Arg) on performance and immune function in cyclophosphamide (CY) immunosuppressed weaned pigs. The weaned pigs were allotted randomly into one of three treatments, including: (1) non-challenged control; (2) CY-challenged group; and (3) CY + 0.5% Arg. On days 14 and 21 of the trial, pigs were injected with CY or sterile saline. Blood samples were obtained on days 21 and 28 of the trial for further analysis. On day 28, delayed-type hypersensitivity reaction was evaluated. Arg alleviated the decrease of average daily gain (P < 0.05) induced by CY challenge from days 21 to 28. Arg mitigated the CY-induced decrease of total white blood cell numbers (P < 0.05) on day 28 and improved the lymphocyte percentage on day 21 (P < 0.05). Arg increased the delayed-type hypersensitivity reaction (P < 0.05), and attenuated the decrease of bovine serum albumin antibody level caused by CY treatment (P < 0.05) on day 28. In addition, Arg elevated the levels of serum interleukin-2 and interferon-γ (P < 0.05) on day 28, and mitigated the decrease of serum interferon-γ level on day 21 (P < 0.05). These results indicate that Arg supplementation has beneficial effects in attenuating the immunosuppressive effects of CY challenge, therefore improving growth performance of young pigs. © 2008 Springer-Verlag.

摘要:
Our previous study demonstrated mRNA and protein expression of peroxisome proliferator-activated receptor-γ (PPAR-γ) in the immune system of weaned pigs. In this report, to test the hypothesis that activation of PPAR-γ in immune system modulates inflammatory response, and adrenal and somatotropic responses associated with immune challenge, we administered intraperitoneally PPAR-γ agonist and/or antagonist in weaned pigs subjected to Escherichia coli lipopolysaccharide (LPS) challenge. Unexpectedly, we found that a single injection of the PPAR-γ agonist rosiglitazone (given at 3 mg/kg body weight 30 min before LPS injection) failed to block pro-inflammatory cytokine production induced by LPS injection. Rather, plasma levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6), mRNA abundance of TNF-α in thymus, spleen, mesenteric lymph node and peripheral white blood cells, mRNA abundance of IL-6 in thymus, protein levels of TNF-α in spleen and mesenteric lymph node, and protein levels of IL-6 in spleen and mesenteric lymph node, were elevated beyond the levels in control pigs injected with LPS. Furthermore, rosiglitazone potentiated the increase of plasma cortisol and prostaglandin E2 concentrations, and the decrease of plasma insulin-like growth factor-1 concentration induced by LPS injection. Co-administration of the PPAR-γ antagonist bisphenol A diglycidyl ether (given 30 mg/kg body weight) 30 min prior to treatment with rosiglitazone antagonized the effect of the PPAR-γ agonist, indicating a PPAR-γ-dependent effect. Our data indicate that ligand-induced activation of PPAR-γ does not ameliorate but enhances pro-inflammatory cytokine production, and further potentiates the adrenal and somatotropic changes in weaned pigs subjected to E. coli LPS challenge, which suggests that PPAR-γ activation may not be useful, but potentially harmful, in the treatment of immune challenge in livestock. Our results raise doubts about the prevalently accepted anti-inflammatory role for PPAR-γ activation. © SAGE Publications 2009.

摘要:
研究在日粮中添加不同水平的酵母多糖对斑点叉尾鮰免疫功能的影响.结果表明:O.2%、0.3%酵母多糖组斑点叉尾细的头肾指数比对照组分别提高15.09%和7.50%,但试验组间差异不显著;0.3%酵母多糖组脾脏指数比对照组提高9.84%,但试验组间差异不显著.酵母多糖组斑点叉尾鮰血液中白细胞总数比对照组呈不同程度的降低,但各组间差异不显著.0.2%酵母多糖组斑点叉尾鮰白细胞吞噬率显著高于对照组和0.1%酵母多糖组,而0.2%、0.3%酵母多糖组间差异不显著;0.3%酵母多糖组斑点叉尾鮰吞噬指数比对照组显著提高29.77%.

试验用大肠杆菌脂多糖(LPS)多次刺激断奶仔猪建立应激模型,研究α-酮戊二酸(AKG)对断奶仔猪的生长性能和血浆激素水平的影响,探讨AKG能否缓解仔猪应激反应.选用24头健康断奶仔猪(杜洛克×长白×大白,体重(7.25±0.23)kg),随机分成3个组(空白对照组、应激对照组和AKG组),每组8个重复,每个重复1头猪.2个对照组饲喂基础日粮+1%淀粉,AKG组饲喂基础日粮+1%AKG,在试验第10天、第12天、第14天和第16天分别在应激对照组和AKG组仔猪的腹膜注射80 μg/kg LPS.试验结果显示:(1)试验第1~9天,未注射LPS时,AKG组平均日增重分别比空白对照组和应激对照组提高30.0%(P<0.05)和31.1%(P<0.05),料重比降低12.4%(P<0.05)和12.8%(P<0.05);试验第10~16天(注射LPS),应激对照组与空白对照组相比平均日增重降低28.8%(P<0.05),料重比升高16.8%(P<0.05),但AKG组的平均日增重和料重比与空白对照组相比无显著性差异(P>0.05);(2)与空白对照组相比,试验第10天,首次LPS刺激后,应激对照组的血浆胰岛素样生长因子(IGF-I)水平降低22.7%(P<0.05),而AKG组无显著变化(P>0.05);试验第16天,多次LPS刺激后,与空白对照组比较,应激对照组的IGF-I和胰岛素(INS)水平分别降低21.2%(P<0.05)和25.7%(P<0.05),而AKG组无显著变化(P>0.05).结果表明,日粮中添加1%AKG能显著缓解LPS应激对杜×长×大断奶仔猪生长的负影响.

This study evaluated whether arginine (Arg) supplementation could attenuate gut injury induced by Escherichia coli lipopolysaccharide (LPS) challenge through an anti-inflammatory role in weaned pigs. Pigs were allotted to four treatments including: (1) non-challenged control; (2) LPS-challenged control; (3) LPS+0.5% Arg; (4) LPS+1.0% Arg. On day 16, pigs were injected with LPS or sterile saline. At 6 h post-injection, pigs were killed for evaluation of small intestinal morphology and intestinal gene expression. Within 48 h of challenge, 0.5% Arg alleviated the weight loss induced by LPS challenge (P<=0.025). In all three intestinal segments, 0.5 or 1.0% Arg mitigated intestinal morphology impairment (e.g. lower villus height and higher crypt depth) induced by LPS challenge (P<0.05), and alleviated the decrease of crypt cell proliferation and the increase of villus cell apoptosis after LPS challenge (P< 0.01). The 0.5% Arg prevented the elevation of jejunal IL-6 mRNA abundance (P=0.082), and jejunal (P=0.030) and ileal (P=0.039) TNF-γ mRNA abundance induced by LPS challenge. The 1.0% Arg alleviated the elevation of jejunal IL-6 mRNA abundance (P=0.053) and jejunal TNF-γ mRNA abundance (P=0.003) induced by LPS challenge. The 0.5% Arg increased PPARγ mRNA abundance in all three intestinal segments (P< 0.10), and 1.0% Arg increased duodenal PPARγ mRNA abundance (P=0.094). These results indicate that Arg supplementation has beneficial effects in alleviating gut mucosal injury induced by LPS challenge. Additionally, it is possible that the protective effects of Arg on the intestine are associated with decreasing the expression of intestinal pro-inflammatory cytokines through activating PPARγ expression. © The Authors 2008.

A study was conducted with 48 weaned barrows ($28{\pm}3d$, $8.45{\pm}0.14kg$) to determine the effect of Achyranthes bidentata polysaccharide (ABPS) supplementation on pig performance, immunological, adrenal and somatotropic responses following Escherichia coli lipopolysaccharide (LPS) challenge. The experiment was a $2{\times}2$ factorial design; the main factors included diet (supplementation with 0 or 500 mg/kg ABPS) and immunological challenge (LPS or saline). On d 14 and 21 of the trial, pigs were given an intraperitoneal injection with either $100{\mu}g/kg$ BW of LPS or an equivalent amount of sterile saline. Blood samples were obtained 3 h after injection for analysis of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), prostaglandin $E_2$ ($PGE_2$), cortisol, growth hormone (GH), insulin-like growth factor (IGF)-I and immunoglobulin G (IgG). On d 2 after LPS challenge, peripheral blood lymphocyte proliferation (PBLP) was measured. LPS administration decreased average daily feed intake (ADFI) (p<0.05), had a tendency to decrease average daily gain (ADG) (p<0.10) during both the first and second challenge periods and increased (p<0.05) feed:gain ratio only during the first challenge period. ABPS tended to improve ADG (p<0.10) during the first challenge period, and improved ADG (p<0.05) and tended to improve ADFI (p<0.10) during the second challenge period. ABPS did not affect feed:gain ratio. An interaction (p<0.05) between LPS challenge and diet was observed for the plasma concentrations of TNF-${\alpha}$, $PGE_2$ and cortisol after both LPS challenges such that, among LPS-treated pigs, pigs fed the ABPS diet were lower for these indices than those receiving the control diet. In contrast, pigs fed the ABPS diet had higher IGF-I (p<0.05) compared with those fed the control diet. No effect of diet, LPS challenge or both on GH and IgG was observed after both LPS administrations. LPS challenge increased PBLP when these cells were incubated with $8{\mu}g/ml$ of LPS during both the challenge periods, and did likewise when incubated with $8{\mu}g/ml$ of concanavalin A only after the first challenge. ABPS had no effect on PBLP. These data demonstrate that ABPS alters the release of pro-inflammatory cytokines following an immunological challenge, which might enable pigs to achieve better performance.

[目的]研究免疫应激对断奶仔猪脾脏、胸腺和外周血白细胞过氧化物酶体增殖物活化受体γ(PPARγ)蛋白表达水平的影响.[方法]选取12头健康断奶仔猪,分成2组,每组6个重复.试验组注射100μg-kg~(-1) BW 的脂多糖(LPS)建立免疫应激模型,对照组注射等量生理盐水.注射后3 h,采血分离血浆和白细胞待测,然后立即屠宰仔猪,取脾脏和胸腺,通过Western blot测定脾脏、胸腺和白细胞中PPAR γ蛋白表达水平.[结果]LPS刺激仔猪,导致血浆白细胞介素-6、肿瘤坏死因子-α、皮质醇和前列腺素E2含量显著升高(P<0.05),胰岛素、类胰岛素生长因子-1含量显著降低(P<0.05);LPS刺激导致血浆葡萄糖、总蛋白、白蛋白和球蛋白含量显著降低(P<0.05);LPS刺激导致脾脏(P<0.05)、胸腺(P<0.1)和白细胞(P<0.05)PPAR v蛋白表达水平升高.[结论]脂多糖刺激可提高仔猪脾脏、胸腺和外周血自细胞PPARγ蛋白的表达,显示PPARγ可能参与仔猪免疫应激的调控,可能成为缓解仔猪免疫应激的一个新靶点.

本研究通过试验探讨不同水平的α-酮戊二酸(AKG)对小肠组织学形态和功能的影响。试验1和试验2各选取32头仔猪,按体重随机分为4个处理,分别饲喂添加0、0.5%、1.0%和2.0%AKG的试验日粮;于试验第14天,测定血浆D-木糖浓度、二胺氧化酶(DAO)活性、内毒素含量。试验3选用12头仔猪,按体重随机分为2个处理,分别为基础日粮组和添加1.0%AKG的试验日粮组;于试验第14天,观察小肠黏膜形态。试验结果表明:试验1和试验2中,D-木糖含量均随AKG水平的增高而上升(P<0.05),当AKG水平为1.0%时达到最高(二次,P<0.05);试验1中血浆中的DAO活性随着AKG水平增高而降低(P<0.01),当AKG水平为1.0%时最低(二次,P<0.05);试验3中1.0%AKG组各肠段绒毛高度与隐窝深度比值(VH/CD)较对照组显著下降(P<0.05)。由试验结果得出,日粮中添加AKG可在一定程度上改善仔猪的小肠组织学形态和吸收功能,以1.0%比例添加效果较好。

摘要:
Peroxisome proliferator-activated receptor gamma (PPAR gamma), a member of the nuclear hormone receptor superfamily, has been implicated in regulation of immunity and inflammation in rodents and humans. The objective of the current study was to investigate whether the expression of PPAR gamma was altered in the immune system of weaned pigs after Escherichia coli lipopolysaccharide (LPS) injection. PPAR gamma expression was investigated in the thymus, spleen, mesenteric lymph node and peripheral white blood cells of weaned pigs (8.54 +/- 0.24 kg BW) after LPS injection (100 mu g/kg BW, n = 6) and controls (sterile saline, n = 6), by using real-time polymerase chain reaction, Western blot analysis, and immunohistochemistry. Plasma pro-inflammatory cytokines and hormones were also assessed. LPS triggered PPAR gamma mRNA and protein expression in the thymus (P < 0.05, 4.24-fold; P < 0.10, 1.46-fold), spleen (P < 0.10, 2.75-fold; P < 0.05, 1.84-fold), mesenteric lymph node (P < 0.05, 4.32-fold; P < 0.05, 1.96-fold) and peripheral white blood cells (P < 0.001, 24.44-fold; P < 0.001, 1.58-fold). The LPS-injected pigs showed an increase in PPAR gamma staining in splenic corpuscle and periarterial lymphatic sheath of white pulp (P < 0.05) and red pulp (P < 0.001) of spleen, and in medullas of thymus lobule of thymus (P < 0.05), and in thymus-dependent area of mesenteric lymph node (P < 0.05) compared to the control pigs. Concurrent with up-regulation of PPAR gamma expression, LPS induced increases in plasma interleukin-6 (P < 0.001), tumor necrosis factor-alpha (P < 0.001), cortisol (P < 0.001), prostaglandin E-2 (P < 0.01) and 15-deoxy-Delta(12,14)-prostaglandin J(2) (15d-PGJ(2)) (P < 0.05), and decreases in plasma insulin (P < 0.10) and insulin-like growth factor-1 (P < 0.001). These results suggest that induction of PPAR gamma expression in immune system may be associated with the release of the natural PPAR gamma activating ligand 15d-PGJ(2), and play an important role in host response to immunological stress. Additionally, it is possible that PPAR gamma would be a new therapeutic target in treatment of immunological stress of livestock. (c) 2008 Elsevier B.V. All rights reserved.

摘要:
研究脂多糖(Lipopolysaccharide,LPS)对断奶仔猪外周血白细胞、胸腺、脾脏和肠道淋巴结过氧化物酶体增殖物活化受体(PPARγ)mRNA表达水平的影响,探讨免疫应激是否影响免疫细胞和免疫器官PPARγmRNA的表达。选取12头健康的(21±1)d断奶仔猪,分成2个处理,每个处理6个重复。试验组注射100μg/kg BW的LPS,对照组注射等量的生理盐水。于注射LPS后1.5和3 h,采血分离血浆、白细胞;注射LPS后3 h屠宰仔猪,取胸腺、脾脏和肠道淋巴结进行相关检测。结果表明:(1)LPS刺激后1.5和3 h,肿瘤坏死因子TNF-α和皮质醇含量均急剧上升(P〈0.001);LPS刺激后3 h,胰岛素含量显著下降(P〈0.001),胰高血糖素含量显著上升(P〈0.001)。此外,LPS刺激也导致血液生化指标发生了显著变化。这表明LPS刺激导致机体处于急性免疫应激状态。(2)LPS刺激导致外周血白细胞(P〈0.001)、胸腺(P〈0.05)和肠道淋巴结(P〈0.05)的PPARγmRNA表达量显著升高,表明免疫应激诱导了免疫细胞和免疫器官中PPARγmRNA的表达。提示PPARγ可能参与仔猪免疫应激的调控,可能成为缓解免疫应激的一个新靶点。

本文旨在研究精胺(spermine)对断奶仔猪小肠组织形态与功能的影响.试验选取来源一致、体重相近的21日龄断奶仔猪30头,随机分为5组,每组6个重复,分别口腔灌服0、300、600、900和1 200μmol/d的精胺,连续灌服3 d.试验期为14 d.试验结束末,屠宰全部仔猪,取小肠组织样品,分析小肠黏膜形态、蛋白质含量、DNA和RNA含量以及双糖酶活性.于试验第7天和第14天灌服D-木糖,60 min后采血,测定血清中D-木糖含量.结果表明:(1)灌服1 200 μmol/d的精胺可提高小肠绒毛高度、降低隐窝深度(P<0.05);(2)1 200 μmol/d的精胺组空肠麦芽糖酶活性显著高于对照组(P<0.05);(3)断奶后第7天,精胺各组与对照组血清D-木糖含量无显著差异(P>0.05);断奶后第14天,1 200 μmol/d精胺组血清D-木糖含量显著高于对照组(P<0.05).口腔灌服1 200 μmol/d剂量的精胺可改善断奶仔猪小肠形态结构与吸收功能,作用效果具有后续性.
