摘要:
试验旨在研究复合植物精油(OC T)对脂多糖(L PS)诱导的断奶仔猪的免疫应激的影响.试验选取28日龄左右的杜×长×大三元杂交仔猪,随机分成3个处理组:对照组(基础日粮,注射生理盐水),L PS组(基础日粮,注射LPS),OCT组(基础日粮+50 mg/kg OCT,注射LPS).每组8个重复,每个重复1头猪.于试验第21天注射L PS(100μg/kg体重)或等体积的生理盐水,注射3 h后前腔静脉采血进行白细胞分类计数;6 h后屠宰,取胸腺、脾脏,-80℃保存,用于测量胸腺、脾脏免疫相关基因白介素-4(IL-4)、白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子(TNF-α)、核因子(NF-κB)以及转录相关基因转录激活因子3(STAT3)的基因相对表达量.结果表明:①与对照组相比,L PS刺激降低了仔猪血液中白细胞、中性粒细胞、淋巴细胞、单核细胞及嗜酸性粒细胞数量(P<0.05);与LPS组相比,日粮中添加OCT提高了白细胞的数量(P<0.05);②与对照组相比,LPS刺激上调了脾脏、胸腺IL-6、STAT3 mRNA水平(P<0.05),下调了脾脏IL-4、TNF-α及胸腺IL-4、TNF-α、NF-κB mRNA水平(P<0.05);与LPS组相比,添加OCT下调了脾脏、胸腺IL-6、STAT3 mRNA水平(P<0.05);上调了胸腺IL-4、IL-10、TNF-α、NF-κB的mRNA水平(P<0.05).综合上述结果,LPS诱导发生了仔猪免疫应激,OCT在一定程度具有缓解作用.

摘要:
为研究N-乙酰半胱氨酸(NAC)对乙酸(ACA)刺激仔猪肠黏膜屏障功能以及肠道黏膜生长的影响,试验选取体重相似且体况健康的仔猪18头,随机分为对照组、ACA组和NAC组,其中前两组饲喂基础饲粮,NAC组在基础饲粮中添加0.05%NAC,每组6个重复,每个重复1头猪。试验第15天对ACA组和NAC组仔猪直肠灌注10 m L10%的乙酸,对照组灌注相应剂量的生理盐水。第21天灌服D-木糖溶液后采集前腔静脉血样,深度麻醉后屠宰,取小肠黏膜样品待测。结果表明:与对照组相比,ACA组仔猪血浆二胺氧化酶(DAO)活性、十二指肠和回肠的隐窝深度分别提高了94.80%、14.31%、8.14%(P<0.05);十二指肠DAO活性降低了17.95%(P<0.05),空肠绒毛高度、十二指肠和回肠绒毛高度/隐窝深度比值,十二指肠和空肠绒毛表面积,及回肠RNA/DNA比值和TP/DNA比值分别降低了14.03%、26.72%、14.97%、12.40%、37.11%、38.91%、26.35%(P<0.05)。饲粮中添加NAC可以缓解以上这些负面影响。此外,相较于ACA组,NAC血浆中D-木糖和表皮生长因子(EGF)含量以及回肠中EGF m RNA水平提高了17.29%、26.20%、34.03%(P<0.05)。综上所述,NAC能够改善ACA刺激造成的仔猪小肠黏膜屏障功能受损,NAC可能是通过提高EGF含量以及EGF m RNA水平,促进细胞生长,修复肠道损伤。

摘要:
The present study was to determine the efficacy of dietary supplementation with oleum cinnamomi (OCM) on growth performance and intestinal functions in piglets. Sixteen piglets (24-day-old) were randomly assigned to the control or OCM groups. Piglets in the control group were fed a basal diet, whereas piglets in the OCM group were fed the basal diet supplemented with 50 mg/kg OCM. On day 20 of the trial, blood samples and intestinal tissues were obtained from piglets. Compared with the control group, dietary OCM supplementation increased (p < 0.05) average daily feed intake, plasma insulin levels, villus width and villous surface area in the duodenum and jejunum, DNA levels and RNA/DNA ratios in the ileum, the abundance of Enterococcus genus and Lactobacillus genus in caecum digesta, mRNA levels for epithelial growth factor receptor (EGFR), Ras, extracellular signal-regulated kinase 1/2 (Erk1/2), b-cell lymphoma-extra large (Bcl-xL), villin, junctional adhesion molecule A (JAM-A), myxovirus resistance (MX) 1, MX2 and regenerating islet-derived protein 3 gamma (REG3G), and protein abundances of Ras and claudin-1, but decreased (p < 0.05) diarrhoea incidence; the abundances of Enterobacteriaceae family, Enterococcus genus, Lactobacillus genus, Bifidobacterium genus, and Clostrium coccoides in the colon digesta, and AMP-activated protein kinase (AMPK) mRNA levels and caspase-3 protein abundance in the jejunal mucosa of piglets. Taken together, these data indicate that dietary OCM supplementation modulates intestinal microbiota and improves intestinal function in weanling pigs. OCM is an effective feed additive and alternative to feed antibiotics for improving intestinal health in swine.

摘要:
Enterotoxigenic Escherichia coli strains induce human and animal intestinal dysfunction and injury, and cause diarrhea in weanling pigs. A recombinant E.coli strain LMG194-STa which expressed single toxin STa of ETEC was constructed by directly inserting the STa gene of ETEC into the expression vector pBAD202 and then transferring the recombinant plasmid pBAD-STa into the E.coli LMG19 host strain. Diarrhea and intestinal injury in piglets were induced by oral administration of recombinant strain LMG194-Sta. In vitro, the recombinant strain LMG194-Sta had the same toxicity to the IPEC cell as wild type strain K88, and higher toxicity than the host strain LMG194. In vivo, LMG194-STa caused severe diarrhea in piglets as K88, the diarrhea rate of LMG194-STa and K88 groups was higher than that in the LMG194 and control groups. Both K88 and LMG194-STa induced intestinal inflammation, injury, and atrophy, while adversely affecting the expression of genes for cytokines, transporters and ion channels, and nutrient metabolism. Thus, we established a porcine model of recombinant E. coli strain LMG194-STa-induced diarrhea for future nutritional and mechanistic studies of intestinal dysfunction. © 2018 Frontiers in Bioscience. 摘要:
α-Ketoglutarate (AKG) is an extensively used dietary supplement in human and animal nutrition. The aim of the present study was to investigate effects of dietary AKG supplementation on the energy status and anti-oxidative capacity in liver and intestinal mucosa of Cherry Valley ducks. A total of 80 1-day-old ducks were randomly assigned into four groups, in which ducks were fed basal diets supplemented with 0% (control), 0.5%, 1.0% and 1.5% AKG, respectively. Graded doses of AKG supplementation linearly decreased the ratio of adenosine monophosphate (AMP) to adenosine triphosphate (ATP) in the liver, but increased ATP content and adenylate energy charge (AEC) in a quadratic and linear manner, respectively (P < 0.05). Increasing dietary AKG supplemental levels produced linear positive responses in ATP content and AEC, and negative responses in AMP concentration, the ratio of AMP to ATP and total adenine nucleotide in the ileal mucosa (P < 0.05). All levels of dietary AKG reduced the production of jejunal hydrogen peroxide and hepatic malondialdehyde (P < 0.05). Hepatic and ileal messenger RNA expression of AMP kinase α-1 and hypoxia-inducible factor-1α were linearly up-regulated as dietary AKG supplemental levels increased (P < 0.05). In conclusion, dietary AKG supplementation linearly or quadratically enhanced hepatic and intestinal energy storage and anti-oxidative capacity of Cherry Valley ducks.

Porcine epidemic diarrhea virus (PEDV) infects the intestine of young pigs, but effective measures for prevention and treatment are lacking. N-Acetylcysteine (NAC) has been shown to reduce endotoxin-induced intestinal dysfunction. This study was conducted with the PEDV-infected neonatal piglet model to determine the effect of NAC supplementation on intestinal function. Thirty-two 7-day-old piglets were randomly allocated to one of four treatments in a 2 × 2 factorial design consisting of two liquid diets (0 or 50 mg/kg BW NAC supplementation) and oral administration of 0 or 104.5 TCID50 (50% tissue culture infectious dose) PEDV. On day 7 of the trial, half of the pigs (n = 8) in each dietary treatment received either sterile saline or PEDV (Yunnan province strain) solution at 104.5 TCID50 per pig. On day 10 of the trial, d-xylose (0.1 g/kg BW) was orally administrated to all pigs. One hour later, jugular vein blood samples were collected, and then all pigs were killed to obtain the small intestine. PEDV infection increased diarrhea incidence, while reducing ADG. PEDV infection also decreased plasma d-xylose concentration, small intestinal villus height, mucosal I-FABP and villin mRNA levels but increased mucosal MX1 and GCNT3 mRNA levels (P < 0.05). Dietary NAC supplementation ameliorated the PEDV-induced abnormal changes in all the measured variables. Moreover, NAC reduced oxidative stress, as indicated by decreases in plasma and mucosal H2O2 levels. Collectively, these novel results indicate that dietary supplementation with NAC alleviates intestinal mucosal damage and improves the absorptive function of the small intestine in PEDV-infected piglets. © 2017, Springer-Verlag Wien.

为了研究谷氨酰胺对断奶仔猪生长性能、血液生化指标及血浆游离氨基酸含量的影响,试验将16头体重相近的21日龄健康断奶仔猪随机分为2组(对照组和谷氨酰胺组),试验期为21 d,研究在日粮中添加1%谷氨酰胺对仔猪生长性能、血液生化指标及血浆游离氨基酸含量的影响.结果表明:①日粮中添加谷氨酰胺有降低仔猪0~14 d和0~21 d腹泻率的趋势,显著降低了15 ~ 21 d仔猪料重比(P<0.05).②日粮中添加谷氨酰胺降低了仔猪血液中白细胞数、淋巴细胞绝对值、单核细胞绝对值(P<0.05).③添加谷氨酰胺可提高血浆尿素、丝氨酸、半胱氨酸、瓜氨酸、蛋氨酸、酪氨酸、β-丙氨酸、γ-氨基丁酸和色氨酸含量(P<0.05),降低α-氨基丁酸、组氨酸和脯氨酸含量(P<0.05).说明日粮中添加1%谷氨酰胺可提高仔猪饲料利用率并降低腹泻率,并可改变仔猪血液氨基酸组成,提示谷氨酰胺具有影响部分氨基酸的吸收与转运和改善肠道完整性的作用.

This study aims to determine whether Lactobacillus casei (L. casei) could relieve liver injury in piglets challenged with lipopolysaccharide (LPS). Piglets were randomly allocated into one of the three groups: control, LPS, and L. casei. The control and LPS groups were fed a corn- and soybean meal-based diet, whereas the L. casei group was fed the basal diet supplemented with 6 × 106 cfu/g L. casei. On Day 31 of the trial, piglets in the LPS and L. casei groups receivedintraperitoneal administration of LPS (100 μg/kg body weight), while the control group received the same volume of saline. Blood and liver samples were collected for analysis. Results showed that L. casei supplementation decreased the feed/gain ratio (p = 0.027) and diarrhea incidence (p < 0.001), and attenuated LPS-induced liver histomorphological abnormalities. Compared with the control group, LPS challenge dramatically increased glutamyl transpeptidase activity (p = 0.001) in plasma as well as the concentrations of Interleukin 6 (IL-6) (p = 0.048), Tumor necrosis factor-alpha (TNF-α) (p = 0.041), and Malondialdehyde (MDA) (p = 0.001) in the liver, while decreasing the hepatic SOD activity. LPS also increased (p < 0.05) the mRNA levels for IL-6, IL-8, TNF-α, Toll-like receptors 4 (TLR4), Nuclear factor k B (NF- k B) and Heat shock protein 70 (HSP70) in the liver. The adverse effects of LPS challenge were ameliorated by L. casei supplementation. In conclusion, dietary L. casei alleviates LPS-induced liver injury via reducing pro-inflammatory cytokines and increasing anti-oxidative capacity. © 2017 by the authors. β-Conglycinin (β-CG) is well known for inducing intestinal allergies and dysfunction in neonates and young pigs. However, the underlying mechanisms are largely unknown. In this study, to clarify the role of autophagy in β-CG-induced cytotoxicity, we investigated the effects of β-CG on cell viability and autophagy activity in porcine enterocytes (IPEC-1 cells). The results indicated that the cell viability was decreased with the increasing levels of β-CG. β-CG treatment enhanced the eGFP-LC3 puncta per cells and LC3-II/LC3-I, and the latter was further increased in IPEC-1 cells cultured with bafilomycin A1. We conclude that β-CG enhances autophagy activity in enterocytes. © 2016, Springer-Verlag Wien.

本试验旨在研究L-亮氨酸(Leu)对猪流行性腹泻病毒(PEDV)感染幼龄仔猪血液指标和空肠免疫相关基因m RNA表达量的影响,探索Leu对PEDV感染仔猪应激的缓解作用。选用30头体重相近的7日龄PEDV阴性仔猪,随机分为3个处理组:对照组、PEDV组和PEDV+Leu组,每个处理10个重复,每个重复一头猪。试验期10 d(7日龄~16日龄)。从第0天开始对PEDV+Leu组按2.8 g/kg体重饲喂Leu,而对照组和PEDV组均按4.1 g/kg体重饲喂L-丙氨酸。第7天早上对PEDV组和PEDV+Leu组采用口服的方式接种106.5TCID50的PEDV,同时对照组采用口服的方式接种等剂量的生理盐水,3 d后(16日龄)取血液和肠道样品。结果表明:与对照组相比,PEDV组仔猪血液中总胆红素(TBIL)水平、血小板分布宽度(PDW)及空肠黏膜CXCL11、REG3G m RNA表达量分别降低了20.92%、9.76%、56.00%、36.00%(P〈0.05),血液中白细胞(WBC)和淋巴细胞(LYM)数量、磷酸丝氨酸、酪氨酸和色氨酸浓度,及空肠粘膜GSTO2和HSPH1 m RNA水平分别提高了61.68%、27.66%、30.39%、50.54%、64.00%、75.00%、89.00%(P〈0.05);与PEDV组相比,PEDV+Leu组仔猪血液中组氨酸、天冬酰胺、尿素氮(BUN)浓度分别升高了55.43%、32.58%、83.33%(P〈0.05),而二胺氧化酶(DAO)含量、单核细胞(MONO)、单核细胞比(Monocyte Ratio,MONOR)、磷酸丝氨酸、谷氨酸、异亮氨酸、精氨酸、苏氨酸及空肠黏膜MX1和GSTO2 m RNA水平分别降低了28.80%、47.13%、43.87%、10.53%、42.12%、29.48%、29.82%、33.50%、32.11%、28.00%(P〈0.05)。因此,日粮中添加Leu提高了PEDV感染仔猪的免疫功能,缓解了PEDV感染仔猪的应激。

摘要:
This study determined whether N-acetylcysteine (NAC) could improve intestinal function through phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR), epithelial growth factor receptor (EGFR), toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-κB), adenosine 5′-monophosphate-activated protein kinase (AMPK), and type I interferon (IFN) signaling pathways in a piglet model of lipopolysaccharides (LPS) challenge. Thirty-two piglets (24-day-old) were randomly allocated to one of four treatments, with eight replicates per treatment and one piglet per replicate. The experiment consisted of four treatments in a 2 × 2 factorial arrangement with two diets (supplemented with 0 or 500 mg NAC/kg diet) and saline or LPS administration. On day 20 of the trial, piglets in the LPS and LPS + NAC groups were intraperitoneally injected with 0 (saline) or 100 μg LPS/kg BW. Blood samples were obtained at 3 h and intestinal mucosae were collected at 6 h post LPS or saline injection. The growth performance was not affected by dietary NAC. LPS induced intestinal dysfunction, as indicated by: (1) reductions in the small-intestinal glutathione concentrations and plasma d-xylose levels; (2) elevations in plasma diamine oxidase activity, mucosal MMP3 mRNA levels and caspase-3 protein abundance; (3) reduced the activities of the small-intestinal mucosal maltase, sucrase and lactase. The adverse effects of LPS on porcine intestinal function and redox status were mitigated by NAC supplementation through the activation of multiple signaling pathways involving PI3K/Akt/mTOR, EGFR, TLR4/NF-κB, AMPK, and type I IFN. Our findings provide novel mechanisms for beneficial effects of NAC in protecting the intestine from inflammation in animals. © 2017, Springer-Verlag Wien.

摘要:
试验研究了大豆抗原蛋白对仔猪生长性能及肠道功能的影响。试验选用21日龄(杜×长×大)仔猪12头,随机分成2个处理组:对照组(饲喂含4%酪蛋白的基础日粮)和大豆分离蛋白组(饲喂含4%伴大豆分离蛋白的基础日粮)。试验为期7 d,采血后屠宰,取十二指肠、空肠、回肠和结肠等组织样品,测定仔猪生长性能、血液生化指标、肠道损伤和肠道功能相关基因等数据。结果表明:1日粮中添加大豆分离蛋白降低了仔猪的日增重,提高了仔猪料重比和腹泻率。2大豆分离蛋白提高了仔猪十二指肠、空肠和结肠的隐窝深度,降低了十二指肠的绒毛宽度、回肠的绒毛高度和绒毛面积。3在日粮中添加大豆分离蛋白降低了十二指肠和回肠AQP 3、Villin、MMP3及空肠AQP10的基因表达水平;提高了空肠Bcl-2、CFTR、Pep T1、SGLT-1、NHE3、AQP 3、AQP 4、AQP 8和Occludin的相对表达量。因此,大豆抗原蛋白调控肠道黏膜损伤与修复和转运通道基因的表达,改变了肠道的正常组织形态,进而降低了仔猪的生长性能。

摘要:
Dietary supplementation with N-acetylcysteine (NAC) has been reported to improve intestinal health and treat gastrointestinal diseases. However, the underlying mechanisms are not fully understood. According to previous reports, NAC was thought to exert its effect through glutathione synthesis. This study tested the hypothesis that NAC enhances enterocyte growth and protein synthesis independently of cellular glutathione synthesis. Intestinal porcine epithelial cells were cultured for 3 days in Dulbecco's modified Eagle medium containing 0 or 100 μM NAC. To determine a possible role for GSH (the reduced form of glutathione) in mediating the effect of NAC on cell growth and protein synthesis, additional experiments were conducted using culture medium containing 100 μM GSH, 100 μM GSH ethyl ester (GSHee), diethylmaleate (a GSH-depletion agent; 10 μM), or a GSH-synthesis inhibitor (buthionine sulfoximine, BSO; 20 μM). NAC increased cell proliferation, GSH concentration, and protein synthesis, while inhibiting proteolysis. GSHee enhanced cell proliferation and GSH concentration without affecting protein synthesis but inhibited proteolysis. Conversely, BSO or diethylmaleate reduced cell proliferation and GSH concentration without affecting protein synthesis, while promoting protein degradation. At the signaling level, NAC augmented the protein abundance of total mTOR, phosphorylated mTOR, and phosphorylated 70S6 kinase as well as mRNA levels for mTOR and p70S6 kinase in IPEC-1 cells. Collectively, these results indicate that NAC upregulates expression of mTOR signaling proteins to stimulate protein synthesis in enterocytes independently of GSH generation. Our findings provide a hitherto unrecognized biochemical mechanism for beneficial effects of NAC in intestinal cells. © 2015 Springer-Verlag Wien.

摘要:
Bowel diseases are common in human and animals and are characterized by intestinal dysfunction and injury. A well-established porcine model of intestinal injury can be induced by lipopolysaccharide (LPS), an endotoxin released from the cell wall of pathogenic bacteria. LPS affects the expression of genes associated with intestinal immune response, mucosal growth, energy metabolism, absorption, mucosal barrier function, and antiviral function. Transcriptional analysis of intestinal genes reveals that the duodenum, jejunum, ileum and colon respond to LPS challenge in a similar pattern. Moreover, the jejunum and ileum exhibit greater responses to LPS challenge than the duodenum and colon with regard to gene expression. Additionally, over 85% of genes are co-expressed along the small and large intestines and there is a clear distinction in gene expression patterns amongst the different intestinal segments in pigs. These findings have important implications for underlying molecular mechanisms responsible for endotoxin-induced intestinal injury and dysfunction.
