摘要:
本研究以从断奶仔猪肠道分离的益生菌为试验菌株,将大肠杆菌K88和益生菌接种到体外培养的猪小肠上皮细胞(intestinal porcine epithelial cell-1,IPEC-1)中,测定培养2.5h上清液中的乳酸脱氢酶(lactate dehydrogenase,LDH)活性,同时将益生菌和大肠杆菌K88体外混合培养2.5h,进行平板计数,统计大肠杆菌K88菌数的变化,筛选出可以抑制大肠杆菌K88的益生菌。试验结果显示,干酪乳杆菌和凝结芽孢杆菌能显著降低IPEC-1培养上清液中的LDH活性(P<0.05),降低大肠杆菌K88对细胞的损伤;凝结芽孢杆菌能降低DMEM培养基中大肠杆菌K88的生长速度,结合模拟制粒过程及胃肠道环境的耐高温、耐酸及耐胆盐研究进行综合分析,该凝结芽胞杆菌对大肠杆菌K88有较好的抑制作用且具有良好的耐高温、耐酸及耐胆盐性能,具有作为微生态制剂菌株的应用潜力。本试验建立了能够抑制大肠杆菌K88的益生菌体外筛选技术模型。

摘要:
Background: Skeletal muscle undergoes rapid loss in response to inflammation. a-Ketoglutarate (AKG) has been reported to enhance muscle growth in piglets, but the underlying mechanisms are largely unknown. Objectives: This study tested the hypothesis that dietary AKG supplementation activates mechanistic target of rapamycin (mTOR) signaling and improves skeletal muscle energy metabolism in lipopolysaccharide (LPS)-challenged piglets. Methods: Forty-eightmale piglets (Duroc × Landrace × Yorkshire)wereweaned at 21 d of age to a corn- and soybeanmeal- based diet. After a 3-d period of adaptation, piglets with a mean weight of 7.21 kg were randomly assigned to control, LPS (intraperitoneal administration of 80 μg LPS/kg body weight on days 10, 12, 14, and 16), or LPS plus 1% dietary AKG (LPS +AKG) groups. On day 16, blood sampleswere collected from8 piglets/group 3 h after LPS administration.On day 17, piglets were killed to obtain gastrocnemius muscle from 8 piglets/group for biochemical analysis. Results: Compared with the control group, LPS administration increased (P < 0.05) plasma concentrations of globulin (by 14%) and tumor necrosis factor α (by 59%) and the intramuscular ratio of AMP to ATP (by 93%) and abundance of phosphorylated acetyl- coenzyme A carboxylase (ACC) β protein (by 64%). Comparedwith the control group, LPS administration reduced (P < 0.05) weight gain (by 15%); plasma concentrations of glutamine (by 20%), glucose (by 23%), insulin, insulin-like growth factor I, and epidermal growth factor; intramuscular concentrations of glutamine (by 27%), ATP (by 12%), ADP (by 22%), and total adenine nucleotides; and intramuscular ratios of phosphorylated mTOR to total mTOR (by 38%) and of phosphorylated 70-kDa ribosomal protein S6 kinase (p70S6K) to total p70S6K (by 39%). These adverse effects of LPS were ameliorated (P < 0.05) by AKG supplementation. Conclusions: Dietary AKG supplementation activated mTOR signaling, inhibited ACC-β, and improved energy status in skeletal muscle of LPS-challenged piglets. These results provide a biochemical basis for the use of AKG to enhance piglet growth under inflammatory or practical postweaning conditions. © 2016 American Society for Nutrition.

摘要:
This study was carried out to investigate the effects of N-acetylcysteine (NAC) on growth performance and intestinal function of heat-stressed broilers. A total of two hundred 1-d-old Cobb male chicks were allocated to 1 of 4 treatments, with 5 replicated pens per treatment and 10 birds per pen. The experiment consisted of 4 treatments in a 2 × 2 factorial arrangements with two diets (basal diet or 1 g/kg NAC diet) and two temperatures (thermoneutral or heat stress). From day 8–35 of age, broilers were raised at thermoneutral (26 ± 1 °C) or exposed to cyclic heat stress (36 ± 1 °C from 0800 to 1800 and 26 ± 1 °C from 1800 to 0800). The results showed that heat stress reduced ADFI, ADG, plasma concentrations of triiodothyronine (T3) and thyroxine (T4), intestinal villus height (VH), ratio of VH to crypt depth (CD), intestinal mucosal ATP level, adenylate energy charge (AEC), activities of alkaline phosphatase (AKP), antioxidative and digestive enzymes, and mRNA level for Bcl-2, whereas increased the feed/gain, mortality rate, plasma corticosterone level, intestinal CD, intestinal mucosal AMP and malondialdehyde (MDA) levels, and mRNA levels of heat shock protein (HSP70), caspase-3, AMP-activated protein kinase (AMPK), heme-oxigenase (HMOX), and xanthine oxidoreductase (XOR). Dietary supplementation with NAC decreased the feed/gain, mortality rate, plasma corticosterone level, MDA concentration and intestinal mucosal mRNA levels of HSP70, AMPK and HMOX, while elevated the ratio of VH to CD, ATP, catalase (CAT) and trypsine activity in the small intestine of heat-stressed broilers. Taken together, these results suggest that dietary supplementation of 1 g/kg NAC could improve growth performance and intestinal function of broilers exposed to heat stress. © 2016 Elsevier B.V.

摘要:
Abstract There is growing interest in glutamate as a functional amino acid in nutrition and health. This study was conducted to determine whether glutamate precursor α-ketoglutarate (AKG) could alleviate lipopolysaccharide (LPS)-induced liver injury in young pigs. Twenty-four piglets were randomly assigned to the control, LPS, or LPS + AKG group. Piglets in the control and LPS groups were fed a basal diet, whereas piglets in the NAC group were fed the basal diet supplemented with 1 % AKG. On days 10, 12, 14, and 16 of the trial, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16 of the trial, blood samples were collected 3 h after LPS or saline injection. Twenty-four hours post-administration of LPS or saline (on day 17 of the trial), piglets were killed to obtain liver for analysis. Dietary AKG supplementation alleviated LPS-induced histomorphological abnormalities and mitigated LPS-induced increases in aspartate aminotransferase (AST) activity and AST/ALT ratio (P < 0.05). Compared with the LPS group, dietary supplementation with AKG decreased plasma glutamate concentration, while increasing hepatic concentrations of glutamate, glutamine, leucine, asparagine, lysine, alanine, serine, threonine, valine, and phenylalanine (P < 0.05). LPS challenge dramatically increased concentrations of malondialdehyde and decreased glutathione peroxidase activity in the liver. Additionally, LPS challenge enhanced concentrations of AMP and total protein, as well as RNA/DNA and total protein/DNA ratios, while decreasing hepatic ADP concentrations. These adverse effects of LPS challenge were ameliorated by AKG supplementation. Collectively, dietary AKG supplementation provides a new means to ameliorate LPS-induced liver injury by increasing anti-oxidative capacity and improving energy metabolism in young pigs. © 2015 Springer-Verlag Wien.

为深入研究嗜酸乳杆菌的作用机理,以温敏型穿梭载体pSET4s为基本骨架,将绿色荧光蛋白(GFP)和嗜酸乳杆菌上、下游同源臂插入该载体,构建GFP标记的穿梭表达载体pSET4s-P1-GFP-P2。利用电转化方法将穿梭表达载体pSET4s-P1-GFP-P2转化入嗜酸乳杆菌,通过抗性和温度双重筛选获得能稳定表达GFP的重组嗜酸乳杆菌,将Western blotting和荧光显微镜观察验证的重组菌命名为ΔMG6243(GFP)。利用荧光显微镜观察和平板计数的方法对重组菌进行遗传稳定性和生物学特性研究。结果显示,重组菌在蓝光激发下可见明显绿色荧光,连传10代仍可见明显荧光,与野生菌相比,其生长特性、酸碱盐耐受性均无显著差别。结果表明,本试验已成功构建稳定表达GFP基因的重组嗜酸乳杆菌,利用温敏型穿梭载体pSET4s实现了外源基因在嗜酸乳杆菌中非抗性、整合型表达,为嗜酸乳杆菌基因重组菌的构建奠定了基础,同时GFP的标记作用也为嗜酸乳杆菌在动物体内分布、定植规律和作用机理创造了可行的条件。

Neonates (including human infants) require l-glutamine (Gln) for optimal intestinal health. This study tested the hypothesis that Gln enhances enterocyte growth via both mammalian target of rapamycin (mTOR) and AMP-activated kinase (AMPK) signaling pathways. Intestinal porcine epithelial cells (IPEC-1) were cultured for 3 days in Gln-free Dulbecco's modified Eagle medium containing 0 or 2 mM Gln. To determine the role of mTOR and AMPK on cell growth, additional experiments were conducted where medium contained 2 mM Gln and 10 nM rapamycin (Rap, an inhibitor of mTOR) or 1 μM compound C (an inhibitor of AMPK). IPEC-1 cell growth increased with increasing concentrations of Gln from 0 to 2 mM. Compared with 0 mM Gln, 2 mM Gln increased (P < 0.05) the amounts of phosphorylated 4E-binding protein 1 (4E-BP1) and ribosomal protein S6 kinase (p70S6 kinase) proteins but did not affect abundances of total or phosphorylated AMPK protein. Gln also increased mRNA levels for Bcl-2, mTOR, p70S6 kinase, 4E-BP1, COX7C, ASCT2, ODC, SGLT-1, CFTR, Na+/K+-ATPase, HSP70, and ZO-1. Similarly, cells cultured with Rap and Gln exhibited higher (P < 0.05) abundances of phosphorylated 4E-BP1 and p70S6 kinase proteins than the Rap-only group, whereas abundances of phosphorylated mTOR and 4E-BP1 proteins were increased when AMPK was inhibited by compound C. Conversely, the amount of phosphorylated AMPK increased when mTOR was inhibited by Rap, suggesting a negative cross-talk between mTOR and AMPK. Collectively, these results indicate that Gln stimulates enterocyte growth by activating the mTOR signaling pathway independently of AMPK. © 2014 Springer-Verlag Wien.

摘要:
Cinnamon is a traditional herb used for treatment of many human diseases. The most important chemical compounds of the essential oil are cinnamaldehyde and eugenol. Oleum cinnamomi (OCM, cinnamon oil) is increasingly used as a feed additive to animal diets. Beneficial effects of OCM in protecting tissues from inflammation and injury by endogenous and exogenous agents (such as hydrogen peroxide and lipopolysaccharide (LPS)) may result, in part, from its action on regulating amino acid metabolism in cells to favor the synthesis of glutathione (a major low-molecular-weight antioxidant) from cysteine, glycine and glutamate. In support of this notion, results of recent studies indicate that supplementing OCM (50 mg/kg diet) to a corn- and soybean meal-based diet for piglets weaned at 21 days of age enhances intestinal anti-oxidative capacity and reduces the incidence of diarrhea. Additionally, dietary supplementation with OCM ameliorates LPS-induced mucosal barrier dysfunction and mucosal damage in the small intestine. OCM holds great promise for protecting the gut from injury under conditions of inflammation, infections, and oxidative stress. © 2015, Frontiers in Bioscience. All rights reserved.

摘要:
The integrity of the intestinal epithelium ensures its normal physiological function. Consequently, damage to the mucosal epithelium can impair the absorption of nutrients, thereby reducing the growth performance and compromising the health of animals. N-acetylcysteine (NAC) is pharmaceutically available either intravenously, orally, or by inhalation for reducing endothelial dysfunction, inflammation, fibrosis, invasion, cartilage erosion, acetaminophen detoxification, and transplant prolongation. NAC is rapidly metabolized by the small intestine to produce glutathione and can not be detected in animals without supplementation. The physiologic functions and therapeutic effects of NAC are largely associated with maintaining intracellular concentrations of reduced glutathione. Results from recent studies indicate that NAC reduces inflammation, alleviates oxidative stress, improves energy status, and ameliorates tissue damage in the intestine of lipopolysaccharide-challenged piglets. Moreover, dietary supplementation with NAC ameliorates acetic acid-induced colitis in a porcine model. The effects of NAC are associated with some intestinal cell signaling pathways, such as EGFR, TLR4, apoptosis and tight junction signaling. The current review focuses on the protective effects of NAC on intestinal health and the molecular mechanisms of its action. © 2015, Frontiers in Bioscience. All rights reserved.

This study was carried out to investigate the effects of tributyrin (TB) on the growth performance, pro-inflammatory cytokines, intestinal morphology, energy status, disaccharidase activity, and antioxidative capacity of broilers challenged with lipopolysaccharide (LPS). A total of 160 one-day-old Cobb broilers were allocated to 1 of 4 treatments, with 4 replicated pens per treatment and 10 birds per pen. The experiment consisted of a 2×2 factorial arrangements of treatments with TB supplementation (0 or 500 mg/kg) and LPS challenge (0 or 500 μg/kg body weight [BW]). On days 22, 24, and 26 of the trial, broilers received an intraperitoneal administration of 500 μ g/kg BW LPS or saline. Dietary TB showed no effect on growth performance. However, LPS challenge decreased the average daily gain of broilers from day 22 to day 26 of the trial. Dietary TB supplementation inhibited the increase of interleukin-1β (in the jejunum and ileum), interleukin-6 (in the duodenum and jejunum), and prostaglandin E2 (in the duodenum) of LPS-challenged broilers. Similar inhibitory effects of TB in the activities of total nitric oxide synthase (in the ileum) and inducible nitric oxide synthase (in the jejunum) were also observed in birds challenged with LPS. Additionally, TB supplementation mitigated the decrease of ileal adenosine triphosphate, adenosine diphosphate and total adenine nucleotide and the reduction of jejunal catalase activity induced by LPS. Taken together, these results suggest that the TB supplementation was able to reduce the release of pro-inflammatory cytokines and improve the energy status and anti-oxidative capacity in the small intestine of LPS-challenged broilers. 为考查N-乙酰半胱氨酸(NAC)是否可以改善肉鸭肝脏的能量代谢状况及组织抗氧化能力,试验选用90只1 d樱桃谷肉鸭,随机分成3个处理组:对照组(基础日粮)、500 mg/kg NAC组(基础日粮+500 mg/kg NAC)、1 000 mg/kg NAC组(基础日粮+1 000 mg/kg NAC),每组3个重复,每个重复10只鸭。试验期为3周(1~21日龄)。结果表明:与对照组相比,日粮添加500 mg/kg NAC显著提高了血浆、肝脏的过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的活力及肝脏谷胱甘肽过氧化物酶(GSH-Px)的酶活力、三磷酸腺苷(ATP)、二磷酸腺苷(ADP)的含量和能荷(EC)值(P<0.05),显著降低了血浆丙二醛(MDA)和肝脏过氧化氢(H2O2)的含量及肝脏AMP/ATP值和肝脏低氧诱导因子1(HIF-1)、黄嘌呤氧化还原酶(XOR)的m RNA水平(P<0.05);1 000 mg/kg NAC组显著提高了血浆和肝脏CAT、SOD的活力及肝脏GSH-Px的酶活力、ATP的含量、EC值和AMPK-α1的m RNA水平(P<0.05),显著降低了血浆和肝脏H2O2的含量及肝脏MDA的含量、HIF-1、XOR的m RNA水平。表明NAC可以提高肉鸭的组织抗氧化能力、改善肝脏的能量代谢,这可能与NAC对AMPK-α1、HIF-1及XOR的基因表达调控有关。

本试验旨在筛选出抑菌效果最佳的植物精油并通过动物饲养实验研究该植物精油对断奶仔猪生长性能,血液生化指标的影响,为开发绿色饲料添加剂建立工作基础和提供理论依据.试验通过微量肉汤稀释法和刃天青指示剂测定肉桂油、牛至油、百里香油、茶树油的最小抑菌浓度(MIC),选出抑菌能力最强的植物精油,然后挑选16头体重相近,健康的21日龄"杜×长×大"三元杂交仔猪,分为2个处理组(对照组和肉桂油组),每个处理8个重复.对照组饲喂基础饲粮,肉桂油组饲喂含50 mg/kg肉桂油的基础饲粮.试验期为21d,于试验第21d,前腔静脉采血,分离血浆.采完血之后屠宰,取肝脏组织待测.(结果)结果表明:肉桂油对大肠杆菌和沙门氏菌的抑菌能力最强(MIC为800 μL/L).与对照组相比,添加50 mg/kg肉桂油能显著提高仔猪血液的总蛋白(P<0.05)和白蛋白(P<0.05)水平,红细胞比容提高了11.74% (P=0.082),谷胱甘肽过氧化物酶的活力提高了13.66% (P=0.085).由此可见:肉桂油有较强的抑菌能力,在饲粮中添加50 mg/kg的肉桂油可在一定程度上改善断奶仔猪营养状况,并有提高仔猪抗氧化能力的趋势.

试验旨在研究α-酮戊二酸对热应激肉鸡生长性能和肌肉品质的影响.通过给高温应激肉鸡饲喂含0.5% α-酮戊二酸(α-ketoglutarate,AKG)的日粮35 d,研究AKG对热应激肉鸡生长性能、肌肉品质、抗氧化功能和能量代谢的影响.结果表明:①与对照组相比,热应激处理显著降低了肉鸡(1~35日龄)的生长性能(P<0.05)和组织(胸腺、肝脏、胸肌和腹脂)的相对重量(P<0.05),同时也降低了肉鸡胸肌的a值、抗氧化酶(过氧化氢酶CAT和谷胱甘肽过氧化物酶GSH-Px)的活力、三磷酸腺苷(ATP)和二磷酸腺苷(ADP)含量及能荷(EC)值,但是提高了肉鸡胸肌一磷酸腺苷(AMP)的含量(P<0.05).②日粮中添加0.5%的AKG降低了热应激肉鸡的料重比(1~35日龄)、胸肌AMP含量(P<0.05),提高了热应激肉鸡血液抗氧化酶(CAT、SOD和GSH-Px)的活力(P<0.05),胸肌ATP、ADP和腺苷酸池(TAN)的含量及EC值(P<0.05).因此,日粮中添加0.5%的AKG可以降低热应激肉鸡的料重比,提高血液的抗氧化能力,改善胸肌的能量代谢.
