One of the most important technologies for heterologous gene expression is gene/pathway assembly before transformation into the heterologous host. Normally, biosynthetic PKS and NRPS gene clusters are large, ranging from 10–120 kb, and the targeted genes must be placed under the control of appropriate promoters to ensure the successful expression, which make the demands for facile DNA assembly technologies with higher efficiency, fidelity, and modularity. High throughput genome sequencing has revealed a multitude of potential secondary metabolites biosynthetic pathways that remain cryptic. Pathway reconstruction coupled with genetic engineering via heterologous expression enables discovery of novel compounds, elucidation of biosynthetic pathways, and optimization of product yields.
