2,4-dinitrotoluene (Dnt) perturbs yolk absorption, liver development and lipid metabolism/oxygen transport gene expression in zebrafish embryos and larvae

2,4-dinitrotoluene (2,4-DNT) is a common environmental pollutant, and was classified as a group 2B human carcinogenic compound by the International Agency for Research on Cancer. This study determined the toxic effects of 2,4-DNT exposure on zebrafish at the embryo-larvae stage, in terms of organ morphogenesis and the expression pattern of selected target genes related to lipid metabolism and oxygen transportation. The results showed that the 120-h post-fertilization LC50 of 2,4-DNT was 9.59 mg/L with a 95% confidence interval of 8.89–10.44 mg/L. The larvae treated with 2,4-DNT showed toxic symptoms including smaller body, less skin pigment production, yolk malabsorption, and disordered liver development. Further studies on the expression of genes related to lipid transport and metabolism, and respiration indicated that they were significantly affected by 2,4-DNT. It is concluded that 2,4-DNT exposure perturbed liver development and yolk absorption in early-life zebrafish, and disturbed the lipid metabolism /oxygen transport gene expression. © 2019 by the authors. Yeast cell wall (YCW) preparations are important nutritional additives in the aquaculture industry. Head-kidney macrophages play important roles in innate immune functions in fish. However, the mechanisms underlying their stimulation are not yet characterized. The purpose of this study was to investigate the effects of YCW on immunological functions of channel catfish (Ictalurus punctatus) head-kidney macrophages. Our results showed that YCW preparations not only improved the proliferation and phagocytosis of macrophages but also induced the secretion of reactive oxygen species (ROS), nitric oxide (NO), inducible nitric oxide synthase (iNOS), and cytokines, such as tumor necrosis factor-alpha (TNF-alpha) and interleukin 1 beta (IL-1 beta). At a concentration of 500.0 mu g/mL, YCW preparations stimulated macrophage proliferation and phagocytosis to 1.98 and 1.8 times, respectively, when compared with the negative group. Equally, YCW preparations raised the production of NO by inducing iNOS and ROS in a concentration-dependent manner. Furthermore, these preparations enhanced TNF-alpha and IL-1 beta mRNA levels to multiple digits from 1.2 to 3.0 times, respectively. In conclusion, the stimulatory effects of YCW on the phagocytic activity of the fish macrophages were associated with respiratory bursts coupled with nitric oxide production.

Morphological studies of Sicuophora (Syn. Wichtermania) multigranularis Xiao et al., 2002, from the rectum of the frog, Quasipaa spinosa, performed using silver impregnation and scanning electron microscopy, confirmed the following newly recognized features: (1) only one apical suture on the right surface; (2) two naked regions at the posterior end of both the left and the right side of the body. Phylogenetic analysis based on the SSU-rRNA gene showed that S. multigranularis is a sister to a clade comprising all other Clevelandellida, strongly supporting the validity of the genus Sicuophora. This is also the first molecular data obtained for the genus Sicuophora. Because of the lack of molecular data, it will be necessary to obtain more genetic data from the family Sicuophoridae to discuss the question of the taxonomic status of the genus Sicuophora. Balantidium grimi n. sp. is described from the rectum of the frog Quasipaa spinosa (Amphibia, Dicroglossidae) from Lishui, Zhejiang Province, China. The new species is described by both light microscopy (LM) and scanning electron microscopy (SEM), and a molecular phylogenetic analysis is also presented. This species has unique morphological features in that the body shape is somewhat flattened and the vestibulum is "V"-shaped, occupying nearly 3/8 to 4/7 of the body length. Only one contractile vacuole, situated at the posterior body, was observed. The phylogenetic analysis based on SSU-rDNA indicates that B. grimi groups together with B. duodeni and B. entozoon. In addition, the genus Balantidium is clearly polyphyletic.

摘要:
研究外源核苷酸——5’-腺苷酸、5’-尿苷酸二钠、5’-胞苷酸二钠、5’-鸟苷酸二钠和5’-肌苷酸二钠对斑点叉尾鮰幼鱼生长性能、体成分和胃肠道形态的影响。结果表明:基础饲料中添加外源核苷酸没有显著提高斑点叉尾鮰幼鱼的增重率和特定生长率,但显著改善了鱼体的干物质和粗蛋白质含量,同时对胃肠道形态的发育具有显著地促进作用,其中以0.10%核苷酸混合物添加水平较佳。

本研究旨在探索不同品种鸡蛋蛋黄的营养效果,首先测定了3种市售鸡蛋蛋黄的营养成分,进而通过高效液相色谱和质谱法分析了其脂肪酸含量,最后以斑马鱼幼鱼为受试生物,分析了3种蛋黄的饲喂效果.结果表明,不同品种鸡蛋蛋黄的粗蛋白含量不同,蛋黄3的粗蛋白含量显著高于试验蛋黄1(P<0.05),而各组蛋黄的粗脂肪含量差异不显著,高效液相色谱和质谱分析显示其脂肪酸含量存在差异,蛋黄3的二十碳以上不饱和脂肪酸含量较高;通过饲喂斑马鱼幼鱼显示其营养效果存在差异,与蛋黄1、2相比,蛋黄3能显著提高斑马鱼幼鱼的生长性能(P<0.05).总之,不同品种鸡蛋蛋黄的蛋白质含量和不饱和脂肪酸含量存在差异,这直接影响了蛋黄的营养效果.

A study was conducted to investigate the effect of dietary yeast polysaccharides on some hematologic parameters and intestinal morphology of channel catfish. Channel catfish were fed diets containing yeast polysaccharides at 0 (control), 0.1, 0.2, or 0.3 % for 7 weeks. Each diet was provided to 10 channel catfish specimens (5.82 ± 0.13 g initial weight) replicated 3 times in individual 250 L fiberglass tanks. Some hematologic parameters, leukocyte phagocytic activity, and intestinal morphology were monitored. After 7 weeks of trial, 0.2 % yeast polysaccharides resulted in significantly higher (P < 0.05) monocyte numbers. Furthermore, fish fed 0.2 % yeast polysaccharide diet had higher (P < 0.05) phagocytic rate of leukocyte. And 0.3 % yeast polysaccharide enhanced (P < 0.05) phagocytic index of leukocyte. Histological evaluation showed yeast polysaccharide supplementation increased the height of intestine fold (0.1, 0.2 and 0.3 %) and the thick of muscular layers (0.2 %) in intestine (P < 0.05). In addition, 0.1 and 0.3 % yeast polysaccharide supplementation improved the number of goblet cells (P < 0.05). The results of this trial indicate that yeast polysaccharides supplementation could affect blood monocytes, improve leukocytes phagocytic activity, and the development of intestine in channel catfish. © 2012 Springer Science+Business Media B.V.

研究稀土壳聚糖螯合盐对鲫鱼非特异性免疫力的影响。结果表明:饲喂20天后,3个试验组血清溶菌酶活性较对照组均有不同程度提高,且0.24%组血清溶菌酶显著高于对照组;但在第40、60天时,3个试验组血清溶菌酶活性与对照组相比差异均不显著。在第40天时,3个试验组血清超氧化物歧化酶活性均高于对照组;在第20、60天时,各组血清超氧化物歧化酶活性无明显变化规律,差异均不显著。第20天时,0.24%组鲫鱼脾脏指数、头肾指数均显著高于对照组,0.08%组、0.16%组脾脏指数、头肾指数也均高于对照组;但在第40、60天时,各组脾脏指数、头肾指数无明显变化规律,差异均不显著。

摘要:
研究在日粮中添加不同水平的酵母多糖对斑点叉尾鮰免疫功能的影响.结果表明:O.2%、0.3%酵母多糖组斑点叉尾细的头肾指数比对照组分别提高15.09%和7.50%,但试验组间差异不显著;0.3%酵母多糖组脾脏指数比对照组提高9.84%,但试验组间差异不显著.酵母多糖组斑点叉尾鮰血液中白细胞总数比对照组呈不同程度的降低,但各组间差异不显著.0.2%酵母多糖组斑点叉尾鮰白细胞吞噬率显著高于对照组和0.1%酵母多糖组,而0.2%、0.3%酵母多糖组间差异不显著;0.3%酵母多糖组斑点叉尾鮰吞噬指数比对照组显著提高29.77%.
