摘要:
Three water-soluble hetero-polysaccharides, designated LP1–3, were isolated from litchi pulp. Their structures, solution properties and immunomodulatory activities were evaluated. LP1 contained (1 → 4,6)-β-D-Glc and (1 → 4)-α-L-Gal, while LP2 contained (1 → 3)-α-L-Ara and (l → 2)-β-D-Gal, and LP3 contained α-L-Ara and (l → 4)-β-Rha. Their molecular weights ranged from 105,880 to 986,470 g/mol. LP1 had a spherical conformation with hyper-branched structure and LP2 was semi-flexible chain, while the polysaccharide chains of LP3 were cross linked to form network-like conformation in solution. In addition, all fractions strongly stimulated mesenteric lymph node cell proliferation, IFN-γand IL-6 secretion in the dose range of 25–100 μg/mL compared with untreated control group (p < 0.05). LP1 exhibited the strongest stimulation of mesenteric lymph node cell proliferation and cytokine secretion, which may be attributed to its unique chemical structure and chain conformation. This is the first report on the solution properties and intestinal immunity activities of polysaccharides from litchi pulp.<br/> ©2016 Elsevier Ltd

Inulin is a type of fructose polymer that is commonly present in plants as a storage carbohydrate. Enzymatic hydrolysis of inulin via exo-inulinase to produce fructose is an efficient, green and state-of-the-art technique. To achieve the high-level secretory expression of inulinase and to realize enzymatic preparation of fructose syrup from inulin, an Aspergillus exo-inulinase gene inu was codon-optimized and co-expressed with the endoplasmic reticulum secretion protein in Pichia cells. After inducible expression in a 500-L pilot scale bioreactor, the inulinase activity of the recombinant strains reached 10,480 U/mL of cultivation broth. Next, according to the determined enzymatic characteristics of inulinase INU, we optimized the parameters for inulinase to hydrolyse inulin. Under the optimal condition of the enzyme/inulin ratio of 5000 U/g, 15% substrate and an incubation temperature of 50 °C for 4 h, the hydrolysis ratio of inulin reached 100%. The hydrolysis products of inulin contain two components, 95% fructose, and 5% glucose. This study has fulfilled the scaled-up production of inulinase and facilitated its industrial application for enzymatic preparation of fructose from inulin.

摘要:
The multi-porous ZnFe2O4 (ZFO) nanotubes are fabricated by the direct annealing process of ZFO precursor nanofibers, which is prepared by a facile electrospinning method. The crystal structure, composition, morphology, magnetic property, specific surface area and pore size distribution of multi-porous ZFO nanotubes are systematically characterized. The results indicated the multi-porous ZFO nanotubes with high purity phase possess obvious multi-porous and tubular microstructure, the mean inner diameter, specific surface area and pore volume are about 150 nm, 57.42 m(2) g(-1) and 0.16 cm(3) g(-1), respectively. Moreover, the multi-porous ZFO nanotubes exhibit a better photocatalytic activity than that of ZFO nanoparticles for photocatalytic degradation of RhB solution. (C) 2016 Elsevier B.V. In this study, a series of novel amide derivatives and sulfamide derivatives as potential E. coli PDHc E1 inhibitors were designed and synthesized by optimizing the linker between triazole and benzene ring moieties based on the structure of lead compound I as thiamin diphosphate (ThDP) analogs. Their inhibitory activity against E. coli PDHc E1 were examined in vitro and their inhibitory activity against microbial diseases were further evaluated. Most of these compounds exhibit good inhibitory activity against E. coli PHDc E1 (IC50 1.99 to 25.66 μM) and obvious antibacterial activity. 5a, 5c and 9i showed 90–100% antibacterial activity against Xanthomonas oryzae pv. oryzae (Xoo), Acidovorax avenae subsp. avenae (Aaa) and cyanobacteria. Sulfamide derivatives 9 showed more potent inhibitory activity against E. coli PDHc E1 (IC50 < 14 μM) than that of amide derivatives 5 or lead compound I. Especially 9d (IC50 = 2.95 μM) and 9k (IC50 = 1.99 μM) exhibited not only the most powerful inhibitory potency against E. coli PDHc E1, but also 9k showed 99% antibacterial activity against Aaa at 500 μg mL−1 and almost the best inhibition of 97% against cyanobacteria at 20 μg mL−1. Furthermore, the binding mode of 5d and 9d to E. coli PDHc E1 was analyzed by a molecular docking method. The possible interactions of 9d with the important residues of E. coli PDHc E1 were further verified via site-directed mutagenesis enzymatic assays, and fluorescence spectral analysis. Both theoretical and experimental results revealed that 9d could display a more powerful interaction than that of 5d or I by forming a hydrogen bond between a sulfamide linkage and residues Lsy392, Tyr599 and His106 at active site of E. coli PDHc E1. 9k, 9d and 9i with both potent enzyme inhibition and significant antibacterial activity, could be used as novel lead compounds for further optimization. These results proved that a series of compounds with potential antibacterial activity could be obtained by the biorational design of E. coli PDHc E1 inhibitors.

摘要:
In recent decades, the polysaccharides from the medicinal plants have attracted a lot of attention due to their significant bioactivities, such as anti-tumor activity, antioxidant activity, anticoagulant activity, antidiabetic activity, radioprotection effect, anti-viral activity, hypolipidemic and immunomodulatory activities, which make them suitable for medicinal applications. Previous studies have also shown that medicinal plant polysaccharides are non-toxic and show no side effects. Based on these encouraging observations, most researches have been focusing on the isolation and identification of polysaccharides, as well as their bioactivities. A large number of bioactive polysaccharides with different structural features and biological effects from medicinal plants have been purified and characterized. This review provides a comprehensive summary of the most recent developments in physiochemical, structural features and biological activities of bioactive polysaccharides from a number of important medicinal plants, such as polysaccharides from Astragalus membranaceus, Dendrobium plants, Bupleurum, Cactus fruits, Acanthopanax senticosus, Angelica sinensis (Oliv.) Diels, Aloe barbadensis Miller, and Dimocarpus longan Lour. Moreover, the paper has also been focused on the applications of bioactive polysaccharides for medicinal applications. Recent studies have provided evidence that polysaccharides from medicinal plants can play a vital role in bioactivities. The contents and data will serve as a useful reference material for further investigation, production, and application of these polysaccharides in functional foods and therapeutic agents. © Taylor & Francis Group, LLC.

Inulin constitutes an important food ingredient, widely used for its fiber content, and its ability to substitute fat and sugar ingredients. Traditionally, industrial inulin production from chicory roots requires high extraction temperature (70-80°C) and long extraction time (1-2 h). This conventional extraction is generally accompanied with the presence of a large amount of impurities in the extracted juice, mainly due to the application of high temperature, requiring thus further purification steps. To overcome these issues, developing novel extraction technologies, consuming less energy, faster, and providing high yield and purity, is of paramount importance to meet the requirements of a green extraction concept. In this review, the feasibility of using conventional and new promising technologies (enzyme assisted extraction, ultrasounds, microwaves, supercritical fluid extraction, and pulsed electric fields) to recover inulin from plant food materials and by-products from an environmental and economical point of view will be discussed. Industrial relevance: Inulin is widely used in food industries mainly due to its ability to substitute fat and sugar ingredients. However, the current industrial recovery process of this molecule is mainly carried out by diffusion in hot water (70-80°C), followed by a relatively complex purification process, due to the presence of a large amount of impurities generated by the application of high temperatures. The need for obtaining greener, sustainable, and viable processes has led food scientists to develop new processes in full correspondence with the green extraction concept based on the use of non-conventional technologies (i.e. pulsed electric fields, ultrasounds, microwaves, etc). The submitted review discusses the potential of some of these new promising technologies to allow the industrial sustainability and green recovery of inulin, which have as benefits: energy- and time-saving along with higher yields and milder temperatures, reducing thus the subsequent purification steps. © 2015 Elsevier Ltd. In this study, grass carp peptides were prepared by enzymatic hydrolysis of grass carp protein using the combination of Alcalase and Neutrase, and angiotensin-I converting enzyme (ACE) inhibitory activity in vitro, antihypertensive activity in vivo, antioxidant activities, and physicochemical properties of peptides achieved from grass carp protein were characterised after ultrafiltration and desalted processes using mixed ion exchange resins. The purified peptides exhibited strong ACE inhibitory activity (IC<inf>50</inf>= 105 μg mL<sup>-1</sup>), antihypertensive activity with the maximal drop for systolic blood pressure (SBP) of 43 mmHg at a dosage of 100 mg per kg body weight in spontaneously hypertensive rat (SHR), and antioxidant activities indicated by thiobarbituric acid-reactive substance values in a liposome-oxidising system, radical-scavenging activity and chelation of metal ions (Fe<sup>2+</sup>). The molecular weight of peptides was <1000 Da. Compared to grass carp protein, the peptides separated from enzymatic hydrolysates possessed similar amino acid compositions, but contained higher concentrations of essential amino acids. Moreover, the peptides exhibited excellent solubility at a wide range of pH values from 2 to 10, and lower apparent viscosity than the protein. The peptides separated from enzymatic hydrolysates might be used as a promising ingredient in antihypertensive functional foods and nutraceuticals. Changes in systolic blood pressure 6 (SBP) by administering the purified grass carp peptides (50 mg kg<sup>-1</sup>body weight and 100 mg kg<sup>-1</sup>body weight) in SHR.<br/> ©2016 Institute of Food Science and Technology.

The microbial flora of naturally fermented Suan yu was investigated by phenotype combined with gene analysis at different stages of fermentation. Results shows that lactic acid bacteria (LAB) and yeast were gradually increased during fermentation and the counts were ripening to 7.93–8.56 and 5.14–5.75 log cfu/g, respectively, at the end of the fermentation. Staphylococci gradually increased during the first fermentation stage and then slowly decreased until the end of ripening (4.13–4.92 log cfu/g). Majority of the LAB isolated from Suan yu were Lactobacillus plantarum and Pediococcus pentosaceus, followed by Leuconostoc and Lactobacillus paralimentarius species. The yeast mainly consisted of Saccharomyces cerevisiae and Hansenula anomala. The predominant Staphylococcus isolated from Suan yu comprised Staphylococcus saprophyticus and Staphylococcus xylosus. The main microorganism in early fermentation included Leuconostoc mesenteroides, Pseudomonas and Enterobacteriaceae and the followed subsequent stage included L. plantarum, P. pentosaceus, S. xylosus, S. saprophyticus, S. cerevisiae and H. anomala. Practical Applications: In this study, the specific microorganisms and their microbial dynamics were investigated during Suan yu fermentation. Results showed that lactic acid bacteria, coagulase-negative Staphylococci and yeat were predominant microorganisms during the ripening fermentation. Therefore, these predominant organisms are responsible for the Suan yu unique flavor, taste, color and ensure their safety. Furthermore, selecting the most appropriate autochthonous microflora as starter formulation can effectively preserve the typical characteristics of the Suan yu. ©2015 Wiley Periodicals, Inc.

The potential of stirring assisted dead-end ultrafiltration (UF) to recover polyphenols from purple sweet potato (PSP) juices was evaluated. For this purpose, the effects of rotation speed (200-600 rpm), transmembrane pressure (TMP) (0.2-0.4 MPa), and membrane molecular weight cut-off (30-100 kDa) were investigated with a lab-scale filtration module. The results showed that the highest protein removal (86%) and polyphenol selectivity (12.2) were both obtained under higher rotation speed (600 rpm). Moreover, filtration flux was more important at 600 rpm due to the anti-fouling effect of shear rate generated by the rotation. The results also revealed that under TMP of 0.4 MPa, the cake resistance was the dominating fouling resistance. In addition, it was shown that the results obtained after fitting the data obtained for the filtration with 100 kDa membranes under the different TMP could be modeled by an exponential model. The optimum UF conditions in this study were found under rotation speed of 600 rpm, using a 100 kDa membrane and at TMP of 0.3 MPa. Six main anthocyanins were identified by HPLC-DAD-ESI-MS<sup>2</sup>analysis in the unfiltered PSP juice and the permeate obtained under UF at the optimum conditions, being the intensities of the peaks lower in the filtrate obtained using UF. This study opens the doors of rotation-assisted UF as a promising anti-fouling technique, thus avoiding subsequent purification steps.<br/> ©2016 Elsevier B.V. All rights reserved.

摘要:
以鲜切草鱼鱼腩为原料,选取17种平皿体系条件下对假单胞菌抑菌效果好的保鲜剂对草鱼鱼腩进行保鲜处理,通过感官初筛选出对产品色泽、味道、组织形态、组织弹性影响小的保鲜剂;研究不同初筛得到的保鲜剂处理后冷鲜草鱼肉贮藏期间品质变化,并根据微生物生长曲线构建微生物生长一级模型,通过模型参数的变化定量反映抑菌剂对微生物生长的抑制作用,综合考虑保鲜剂对冷鲜鱼腩品质以及抑菌特性影响,选出草鱼鱼腩处理的最佳保鲜剂;并结合气调包装(50% CO_2+50% N_2)研究保鲜处理后草鱼鱼腩的货架期。结果表明:没食子酸、柠檬酸、鞣酸、磷酸三钾、ε-聚赖氨酸、植酸处理后鱼腩颜色正常,保持原有鱼香味,组织形态和弹性无不好影响;ε-聚赖氨酸处理后品质优于其他组,并在草鱼鱼腩基质上有较好抑菌特性,确定ε-聚赖氨酸为草鱼鱼腩最佳保鲜剂;经ε-聚赖氨酸处理的鲜切鱼腩结合气调包装后置于4 ℃贮藏时的货架期可达13 d,能较长时间保持鲜切草鱼鱼腩的品质。

摘要:
依据摩擦学定量构效关系理论(QSTR),采用比较分子力场分析(CoMFA)和比较分子相似性指数分析(CoMSIA)这两种方法研究了含氮杂环类润滑油添加剂的抗磨损性能的摩擦学三维定量构效关系(3D-QSTR),并建立了相应的3D-QSTR模型.结果表明:仅利用静电场构建CoMFA或CoMSIA模型时,模型预测能力最好, r~2, q~2均大于0.5.根据CoMFA或CoMSIA模型等高线图分析得出:分子静电场对含氮杂环类润滑油添加剂的抗磨损性能影响最大,在特定区域的引入带负电荷或带正电荷的基团将有助于抗磨性能的提高.

摘要:
Three litchi polysaccharide fractions (LPFs), LP-4, LP-6 and LP-8, were obtained by fractional precipitation using 40%, 60% and 80% ethanol, respectively. The physicochemical properties, chemical antioxidant, and cellular antioxidant and antiproliferative activities of the three polysaccharide fractions were compared. LP-6 contained the highest contents of uronic acid and binding protein among the three fractions, whereas LP-8 contained the least. Amino acid composition analysis of the binding protein revealed that LP-6 contained the most acidic and aromatic amino acids. However, LP-8 contained more galactose and mannose than LP-4 and LP-6. LP-6 exhibited the highest chemical antioxidant activities, with an oxygen radical absorbance capacity of 28.14 μmol TE per g DW. LP-8 exhibited higher cellular antioxidant activity and a greater inhibitory effect on the proliferation of A549, HepG2 and MGC-803 cells at a concentration of 100-800 μg mL-1 than LP-4 and LP-6. In summary, the different LPFs exhibited different antioxidant and antiproliferative activities with differential physicochemical properties. © The Royal Society of Chemistry.

摘要:
A new type of portable sensor is proposed to detect bacterial concentration based on the change in dielectric loss angle δ. The performance of the sensor is tested in four types of drinks, namely, milk, orange juice, peach juice, and tomato juice. Notably, the sensor can detect δwhen the bacterial concentration reaches a critical threshold value (approximately 10<sup>6</sup>cfu mL<sup>-1</sup>) by applying a 1 V peak-to-peak 200 Hz sinusoidal test signal of 0.25 mA at intervals of 10 min. This work introduces a portable and low-system for rapid detection. The system is a useful tool for microbial screening in industrial and commercial environments.<br/> ©2015 The Royal Society of Chemistry.

摘要:
对寇氏隐甲藻突变株产DHA的发酵培养基进行响应面优化。首先用Plackett-Burman试验筛选出影响显著的3个因素,再利用最陡爬坡试验和响应面试验对培养基进行优化并建立二次多元回归模型,最后用优化后的发酵培养基在50 L发酵罐上进行放大试验。结果表明在最佳培养基葡萄糖121.41 g/L,谷氨酸钠11.54 g/L,硫酸镁7.25 g/L时,DHA的产量为5.65 g/L发酵液,比优化前提高了10.35%。在50 L发酵罐上发酵培养,DHA产量为9.50 g/L发酵液,为摇瓶培养时的1.68倍。

摘要:
为解决菜籽蛋白原料及制品中真蛋白含量难以测定的问题,建立了一种基于分级和定量2步完成的高选择性菜籽蛋白含量测定方法。通过酸化丙酮溶液和沸水萃取实施分级处理,可将样品中的非蛋白氮分离;结合应用可溶性蛋白的染色法测定和不溶性蛋白的凯氏定氮法测定,可获得样品的真蛋白含量。该方法变异系数不大于2.83%,回收率95%~96%,并可排除本源性非蛋白氮(包括芥子碱和硫甙)及外源性非蛋白氮(包括硝酸盐、铵盐、三聚氰胺和尿素)的干扰。该方法具备良好的抗干扰能力且不受蛋白质溶解性的影响,可推广到更多的植物蛋白制品及原料的真蛋白含量测定中,并可为相关行业的掺杂辨识或品质评价提供可靠手段。

摘要:
龙眼是一种颇受欢迎的亚热带水果,其干制果肉作为传统中药材亦被人们所熟知,而多糖是主要的生物活性成分。基于多糖理化特征和免疫调节活性的分析评价,探析新鲜和干制龙眼果肉的营养性差异。通过气质联用、离子交换层析、凝胶过滤层析、粘度测定、紫外和红外光谱分析发现,新鲜果肉多糖(LPF)和干制果肉多糖(LPD)均为多糖-蛋白络合物,但两者的结合蛋白含量、中性与酸性多糖含量比、单糖组成、分子量分布和特征粘度均存在显著差异。体外细胞试验证实,LPD在50–400μg/m L剂量范围内对脾淋巴细胞增殖和巨噬细胞吞噬的增强作用明显强于LPF。LPD和LPF均能显著刺激Con A诱导的脾淋巴细胞增殖和巨噬细胞NO生成(P<0.05),但对LPS诱导的脾淋巴细胞增殖刺激作用较弱。龙眼果肉的干制处理可能通过改变水溶性多糖的理化特征而增强其体外免疫调节活性。

摘要:
Membrane dead-end ultrafiltration of sugar beet juice (obtained from pulsed electric field assisted nonthermal pressing) was studied as an alternative method to the conventional liming-carbonation purification. A dead-end filtration module, Amicon 8200, was first used to investigate the purification efficiency of ultrafiltration with various polyethersulfone membranes (10kDa, 50kDa, and 150kDa). The purity of sugar beet juice was increased from 93.6 ± 0.5 to 96.4 ± 0.8% with a 10-kDa membrane. In order to improve the filtration flux, which declined tremendously during dead-end filtration due to membrane fouling, a rotating disk module (RDM) was then applied, and the investigation of the dynamic filtration behavior and filtrate quality of sugar beet juice was carried out. Compared with Amicon stirred module, the RDM had a much higher permeate flux and filtrate purity. The effect of rotating speed and molecular weight cutoff (MWCO) of membranes were studied. The permeate flux increased with rotating speed and pore size of membrane. The permeate flux increased significantly when the rotating speed was raised from 500 to 1,000rpm, while it slightly increased at higher rotating speed (2,000rpm). The purification efficiency of dynamic RDM filtration was about twice than that of filtration performed with Amicon stirring module. © 2014, Springer Science+Business Media New York.

摘要:
This study investigated the protective properties of lotus seedpod oligomeric procyanidins (LSOPC) against nonalcoholic fatty liver disease (NAFLD) and its underlying mechanism. Sprague-Dawley (SD) male rats were fed a basic diet, a high-fat diet (HFD), or HFD plus 0.2 or 0.5% (w/w) LSOPC for 12 weeks. Administration of LSOPC markedly reduced serum and hepatic biochemical parameters and protein expression of advanced glycation endproducts (AGEs). Additionally, 0.5% (w/w) LSOPC treatment remarkably reversed the increasing tendency of receptor of advanced glycation endproduct (RAGE) to normal level. Furthermore, dietary LSOPC significantly decreased the protein levels of mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-κB) and down-regulated genes involved in pro-inflammatory cytokines and adhesion molecules. Taken together, these findings demonstrate that LSOPC may protect obese rats with long-term HFD-induced NAFLD against RAGE-MAPK-NF-κB signaling suppression.<br/> ©2015 American Chemical Society.

As for the improper water and land use structure and difficulty in bringing the system into maximum performance, this research takes the Sanjiang Branch of Heilongjiang Land Reclamation Bureau as an example and uses the concept of complex adaptive system to build an allocation model of the complex adaptive system of agricultural land and water resources of the Sanjiang Plain and to make evolution analysis of optimized allocation of agricultural land and water resources under different groundwater exploitation plans. This research shows that: with the increased exploitation of groundwater, the agricultural production value and the grain yield have increased; the ecological area has decreased within a certain range; with the increased restriction in groundwater exploitation year by year, the efficiently-used land area will decrease, resulting in a lot of land not being effectively utilized and seriously hindering the coupling between the regional water and land resources and the local economy development.

Longan is one of the most popular subtropical fruits in Southeast Asia, because of its flavor and benefits to health. As one of the important active ingredients of longan pulp, polysaccharide LPIIa was obtained by hot water extraction, ion-exchange chromatography and gel filtration chromatography. Its physicochemical characterization and immunostimulatory effects on macrophages were then investigated. Structural analyses indicated that LPIIa was a 44.7 kDa heteropolysaccharide mainly composed of →6)-Glc-(1→, →5)-Ara-(1→, →4)-Man-(1→and →6)-Gal-(1→. It enhanced macrophage phagocytosis and nitric oxide production in the dose range of 100-400 μg mL<sup>-1</sup>. Moreover, it significantly increased the inducible nitric oxide synthase activity, tumor necrosis factor-αand interleukin-6 secretion of macrophages at 200 μg mL<sup>-1</sup>. However, these effects were obviously weakened after toll-like receptor 4 (TLR4) or TLR2 was blocked. Likewise, the specific inhibitors of p38 mitogen-activated protein kinase (MAPK), protein kinase C, phosphatidylinositol 3-kinase, protein tyrosine kinase and nuclear factor κB (NF-κB) selectively depressed the immunostimulatory activities of LPIIa on macrophages. LPIIa stimulated macrophage activation partly via TLR4 and TLR2, followed by p38 MAPK- and NF-κB-dependent signaling pathways. The results suggested that LPIIa possessed potent immunomodulatory activity by stimulating macrophages and could be used as an immunotherapeutic adjuvant. ©2015 The Royal Society of Chemistry.
