Simultaneous determination of total fatty acid esters of chloropropanols in edible oils by gas chromatography–mass spectrometry with solid-supported liquid–liquid extraction

摘要:
This study aimed to establish a novel robust method for the simultaneous determination of total fatty acid esters of 4 chloropropanols including fatty acid esters of 3-monochloropropane-1,2-diol (3-MCPD esters), 2-monochloropropane-1,3-diol (2-MCPD esters), 1,3-dichloropropan-2-ol (1,3-DCP esters) and 2,3-dichloropropan-1-ol (2,3-DCP esters) in edible oils. In this method, sodium methylate in methanol was used as the reagent for the ester cleavage reaction of chloropropanols esters. The reaction products were extracted by a sodium sulfate solution, and then purified by solid-supported liquid-liquid extraction (SLE) using diatomaceous earth (Hydromatrix™) as the sorbent. Finally, the extracts were derivatized with heptafluorobutyrylim idazole (HFBI) and analyzed by gas chromatography-mass spectrometry (GC-MS). Quantification was achieved using deuterated chloropropanols as their respective internal standards, including 3-MCPD-d<inf>5</inf>, 2-MCPD-d<inf>5</inf>, 1,3-DCP-d<inf>5</inf> and 2,3-DCP-d<inf>5</inf>. A good linear relationship between peak area and concentrations was obtained within the range of 0.025-2.000mgL<sup>-1</sup> with a correlation coefficients not less than 0.999 for all the chloropropanols esters. The limits of detection (LODs) of esters of 3-MCPD, 2-MCPD, 1,3-DCP and 2,3-DCP (calculated as corresponding chloropropanols) were 30, 30, 100 and 30μgkg<sup>-1</sup>, respectively. The average recoveries of the 3-MCPD esters and the 4 chloropropanols spiked at 0.1, 0.5 and 2mgkg<sup>-1</sup> into blank oil matrix were typically in a range from 70.7% to 113.3%. The robust method validation data including calibration, LOD/LOQ, accuracy and repeatability and proficiency test results (Z-score: -0.5) of the official Food Analysis Performance Assessment Scheme (FAPAS) indicated that the present quantitative method could be successfully applied to the determination of total chloropropanols esters in various edible oils. ©2013 .

摘要:
The germicidal efficacy of six sanitizers against food-borne bacteria and spores (Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis spores) and the effect of these sanitizers in simulation of cleaning and sterilizing in place were evaluated. The most effective sanitizer solution was peracetic acid (PAA), which was capable of reducing E. coli populations by more than 5 log CFU/g at 60 mg/L. The rest of the sanitizers resulted in a population reduction of less than 5 log CFU/g at 150 mg/L in phenol coefficient test. The effect of acid-anionic sanitizer (ABF) and PAA against E. coli and S. aureus was affected by both pH and temperature. The synergism of sanitizers on germicidal efficacy has also been examined in this study. It was observed that the combination of PAA and ABF presented high efficacy against spores of B. subtilis. 0.2 % PAA in combination with 0.1 % ABF reduced 7.6 log spores. The simulation of cleaning-in-place (CIP) and sterilization-in-place (SIP) revealed that most of organisms were eliminated during CIP. B. subtilis spores holding strong attachment and heat resistance could be eliminated with the combination of 0.2 % PAA and 0.1 % ABF. These findings showed that germicidal efficacy against bacteria and spores was affected by the type of sanitizer, concentration, and environmental conditions, which provides the guidelines in lowering the concentration of sanitizers through synergistic activity and further may be valuable references for sanitation to compete the cycle of good hygiene practices in a proper CIP/SIP process. © 2013 Springer-Verlag Berlin Heidelberg.

The storage modulus (G') and fracture properties of the non-treated and NEM-treated emulsion gels were investigated in the absence and presence of unadsorbed soy protein aggregates (USPA). In the absence of USPA, a decrease in the G' of emulsion gels was observed after NEM treatment. However, in the presence of USPA, the addition of NEM only slightly decreased the G' (p < 0.05). For both non-treated and NEM-treated emulsions, a ∼63-folds higher G' value was obtained after the USPA addition. These results revealed the presence of sulphydryl group - disulfide bond interchange reactions at ambient temperature and under acidic conditions. In the absence of USPA, the sulphydryl group - disulfide bond interchange reactions was the main interactions responsible for the higher G' values of non-treated emulsion gels, but for the emulsions with USPA presented, the large quantity of non-covalent interactions (e.g. hydrophobic group & hydrogen bonds) is the main interactions responsible for the aggregation and gelation of emulsion droplets. In the presence of USPA, the sulphydryl group - disulfide bond interchange reactions formed in the non-treated emulsion gels did not increase the final G' but increased the stability of network. A power law relation was observed between the USPA concentration and the final G', as well as between the oil volume fraction and the fracture stress/strain. © 2012 Elsevier Ltd.

摘要:
An ultrasonic bleaching process for rapeseed oil was studied by monitoring the removal of oil-containing pigments spectrophotometrically at 446 nm in varied bleaching conditions. When low-power ultrasound was used, bleaching was resulted from pigment adsorption by adsorbents, among which bentonite decreased the A<inf>446</inf>the most from 1.82 to 0.65. Ultrasound enhanced the absorption rate rather than the total absorption capacity. When high-power ultrasound was used, bleaching took place even in the absence of adsorbent as evidenced by the decease of A<inf>446</inf>to 0.74, possibly due to the effects of combining heat and sonochemical degradation of pigments. The overall bleaching effects were enhanced with the increase of ultrasound power, sonication time and treatment temperature. The content of primary oxidation products in oils increased after sonication, whereas the content of secondary oxidation products remained constant. Due to its multiple mechanisms, ultrasonic bleaching may be a good alternative to the adsorption-based bleaching method. ©2013 Elsevier Ltd. All rights reserved.<br/>

In this study, angiotensin-I converting enzyme (ACE) inhibitory tripeptides from rice protein hydrolysate were purified and their properties were further characterized. Val-Asn-Pro (VNP) and Val-Trp-Pro (VWP) were successfully purified from rice protein hydrolysate through multi-step isolation and purification comprising of ultrafiltration and DA201-C macroporous adsorption resin, followed by a two-step reverse phase high performance liquid chromatography (RP-HPLC). The sequences of tripeptides were separately assayed by using automated protein/peptide sequencer. In addition, the stability of the tripeptides against ACE and simulated gastrointestinal proteases, ACE-inhibitory kinetics, and their antihypertensive effects in vivo were evaluated. Our results showed that VNP (IC50 value of 6.4 mu M) and VWP (IC50 value of 4.5 mu M) were both competitive ACE inhibitors, and stability against ACE and gastrointestinal proteases of pepsin and chymotrypsin. Furthermore, single oral administration of the tripeptides VNP and VWP at 5 mg/kg body weight in spontaneously hypertensive rats (SHRs) significantly decreased the systolic blood pressure (SBP) of 29 and 38 mmHg (P < 0.05), respectively, and the antihypertensive effects of tripeptides could last for 8 h minimally. The identified unique characterization of VNP and VWP may enable the application of rice protein hydrolysate as functional foods or pharmaceuticals against hypertension. (C) 2013 Elsevier Ltd. Lipids were extracted from organs of the starfish Asterias amurensis associated with different treatments (raw-control, boiling and heating), and then analyzed for lipid content, lipid oxidation index, lipid classes and fatty acid composition. Results showed that boiling softened the hard starfish shells, thus facilitating the collection of starfish organs. As compared with raw organs, the boiled organs had lower water content and higher lipid content, possibly due to the loss of water-holding capacity caused by protein denaturation. Both boiling and heating increased the peroxide value (PV), thiobarbituric acid (TBA) value and carbon value (CV) of lipids. Despite slight increases in the content of complex lipids, associated lipid composition had no substantial variations upon boiling and heating. For simple lipids, the content of 1, 2-diglyceride decreased in boiled and heated organs, with free fatty acids observed on thin layer chromatography (TLC). However, neither boiling nor heating significantly changed the fatty acid compositions of simple or complex lipids in starfish organs, suggesting that these two treatments had no significant effects on complex lipids in starfish organs. Together, our results indicated that boiling of starfish soon after capture facilitated the handling and extraction of useful complex lipids consisting of abundant glucosylceramide and eicosapentaenoic acid (EPA)-bounded phospholipids.

Aschersonia placenta had been recognized as an important fungal pathogen of whiteflies. In recent years, natural occurrence of Aschersonia in whitefly populations was observed in many citrus orchards in the southern regions of China. We analyzed 60 A. placenta isolates obtained from Chinese citrus orchards, using inter-simple sequence repeats to examine the genetic diversity and to determine whether intraspecific variation is correlated with geographic origin. One hundred and fourteen fragments were generated from these isolates; 97% were polymorphic. The Nei's gene diversity (H) was estimated to be 0.1748 within the populations (range 0.0974-0.2179) and 0.3057 at the species level. Analysis of molecular variance showed that the genetic variation was found mainly within populations (74.9%). The coefficient of gene differentiation (G ST = 0.4315) indicated that 56.85% of the genetic diversity resided within populations. The Mantel test revealed no significant correlation between the genetic distance and the corresponding geographical distance (r = 0.142 and P = 0.887); the unweighted pair-group method using arithmetic average clustering gave similar results. © FUNPEC-RP.

亚硝胺是三大强致癌食品污染物之一,普遍存在于腌制和热加工食品中。本研究建立了一种基于液质联用技术结合多级反应监测模式扫描(MRM)检测N-亚硝胺的新型固相萃取-液相色谱方法,可以快速直接检测腌菜中9种N-亚硝胺。实验优化了液相色谱分离与检测条件,同时考察了4种固相萃取单柱(碱性氧化铝Al_2O_3、C_(18)、硅胶、硅藻土)和2种双填料的混合固相萃取柱(Al_2O_3-C_(18),Al_2O_3-硅胶)吸附解析的净化效果。结果表明,采用Al_2O_3-C_(18)混合柱纯化,液相色谱检测,外标法定量,对9种亚硝胺的回收率达75.7%~116.0%,检出限为0.20~0.49 mg/L。采用本方法检测不同产地的6类腌菜产品中亚硝胺含量,二甲基亚硝胺和二乙基亚硝胺的检出率相对较高,分别为37.5%和25.0%,其中二乙基亚硝胺在雪里蕻腌菜中最高含量达59.1 mg/L,亚硝基吗啉在所有样品中未检出。

The solution properties of four fractions (LPI-IV) from crude longan pulp polysaccharides (LP3) were analyzed by size-exclusion chromatography combined with laser light scattering, viscometry, complex formation with Congo red, and atomic force microscopy. Their radii of gyration <S(2)>(z)((1/2)) were 43.3, 62.6, 43.2 and 77.3 nm, exponents of <S(2)>(z)((1/2)) = k M(w)(v) were 0.04, 0.50, 0.52 and 0.02, and intrinsic viscosities ([eta]) were 9.945, 25.38, 308.2 and 452.1 mL/g, respectively. Moreover, the dependence of [eta] on M(w) was established to be [eta] = 5.3 x 10(-)(2)M(w)(0).(6)(1) (mL/g). LPI had both a sphere-like conformation and a triple-helix structure, and LPII-IV existed as flexible chains. LP3, LPI, LPII and LPIII all exhibited direct inhibitory effects on A549, HeLa and HepG2 cells in a positive dose-dependent manner in the range of 50-400 microg/mL. The activities of LPIII, especially the inhibition of HepG2 cell proliferation, were stronger than those of others, which may be partly related to its flexible conformation. The present results support the cancer therapeutic potential of longan polysaccharides.

A simple and rapid spectrophotometric method is described for the determination of trace zinc(II) in zinc-enriched yeast using 2-(5- bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP) as a chromogenic reagent in the presence of surfactant OP. The analytical parameters such pH, reagent and coexistent ions were investigated. The results showed that the maximum absorption wavelength of the complex Zn-5-Br-PADAP was identified at 556 nm. The apparent molar absorptivity was 1.16 × 105 L mol-1 cm-1. Beer's law was obeyed in the concentration range of 0̃12 μg/25 mL for zinc. The interference of various ions was studied by sodium citrate and thiourea as masking agents. The proposed method has been applied to the determination of Zn in yeast-enriched samples with satisfactory and results agreed with those provided by atomic absorption spectrometry.

摘要:
A purified olive extract (POE) rich in phenolic and oleosidic compounds was prepared from olive mill wastewaters by adsorption onto an amphoteric polymer resin with the yield of 2.2% (w/v). In addition to complex and simple phenolic compounds found in the POE, the highly hydrophilic elenolic acid and DEDA (dialdehydic form of decarboxymethyl elenolic acid, a non-phenolic secoiridoids) were also present in high concentration (45%, w/w). Among the phenolic constituents, the major compounds are hydroxytyrosol and its glucoside, DEDA linked to hydroxytyrosol (Hy-DEDA), p-coumaric acid and caffeic acid, verbascoside and some flavonoids. The high content (23%, w/w in gallic acid equivalent) and structural diversity of these phenolic compounds confer superior antioxidant potential of POE with an IC50 less than 8.5 μg/ml determined by DPPH assay. The stability of bioactive components during storage of the extract and changes in antioxidant properties were studied under different conditions. Determination of kinetics of degradation revealed that air/oxygen is the determinant factor which influences the stability of POE under low temperature storage conditions, while exposure to sunlight did not have significant effect on their stability. Whilst an increase in storage temperature decreased the total content of phenolic compounds (20-24% reduction) and non-phenolic secoiridoid aglycons, a high antioxidant capacity was still observed particularly in the nitrogen-protected POE samples (93-95% of the initial value). This was believed to be due to the transformation of hydroxytyrosol-containing complex phenols, especially the secoiridoid ester Hy-DEDA, into free phenolic monomer. This contributed significantly to the maintenance of a high antioxidant potency of the whole extract during storage. The high antioxidant, radical scavenging activity and stability of the extract suggest potential applications in the food and pharmaceutical industries. © 2011 Elsevier Ltd. All rights reserved.

Peptides inhibiting angiotensin-I converting enzyme (ACE, EC. 3.4.15.1) are possible cures of hypertension. Food-derived ACE-inhibitory peptides are particularly attractive because of reduced side effects. Previously, we reported ACE-inhibitory activity of grass carp protein hydrolysates. In this work, we report steps for purifying the ACE-inhibitory peptide from the hydrolysate and its biochemical properties. Following steps of ultrafiltration, macroporous adsorption resin, and two steps of reversed phase high performance liquid chromatography (RE-HPLC), a single Val-Ala-Pro (VAP) tripeptide was identified. The tripeptide with excellent ACE-inhibitory activity (IC50 value of 0.00534 mg/mL) was a competitive ACE inhibitor and stable against both ACE and gastrointestinal enzymes of pepsin and chymotrypsin. This is the first report of food-derived VAP. The identified unique biochemical properties of VAP may enable the application of grass carp protein hydrolysates as a functional food for treatments of hypertension. The developed purification conditions also allow the production of VAP for pharmaceutical applications. (C) 2011 Elsevier Inc. All rights reserved.

Dietary fiber (DF) has attracted increasing interest from nutritionists. The yield of DF by traditional extraction methods, however, is very low. This paper aims to increase the yield of DF by solid state fermentation using the edible and medicinal fungus Schizophyllum commune. Sweet potato residue (SPR) was selected as raw material for producing DF. Results showed that SPR was a good feedstock for DF production by solid state fermentation. Optimized conditions of solid state fermentation of SPR for DF were obtained as follows: material particle size = 1.8 mm to 2.5 mm, water moisture at 65%, natural lighting radiation, and temperature at 27 °C. Under the optimal conditions, the yield and DF content in fermented SPR were more than 80% and 70%, respectively. The increased DF yield was mainly attributable to increased cellulose and hemicelluloses conversion. Swelling capacity, water-holding capacity, oil-holding capacity, and glucose absorption capacity of the fermented SPR were also determined, and the data indicated that the fermented SPR could be considered as a new good DF. Therefore, this work showed us a novel bioconversion method to produce high-quality DF, and the yield of DF increased 4-fold compared with traditional extraction methods.

Acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC) were extracted with the yield of 15. 5 and 26. 5% on the basis of dry weight from the skin of black carp (Mylopharyngodon piceus). ASC and PSC were similar in amino compositions and subunit compositions, but they were slightly different in the secondary structure. Both of ASC and PSC, either rehydrated in distilled water or in acetic acid, had a lower denaturation temperature than pig skin collagen (PPSC), but their denaturation temperature were higher than that of collagens from other freshwater fish. Intrinsic viscosity of different collagens decreased in the order of PPSC>PSC>ASC. Both of ASC and PSC showed only partial degradation in collagenase solution, but they revealed more sensitive to collagenase compared with PPSC. Black carp skin collagens did not induce a significant cytotoxic effect according to the results of in vitro cytotoxicity test. © 2012 The Korean Society of Food Science and Technology and Springer Science+Business Media Dordrecht.

Furazolidone has been banned from use in food animals because of its carcinogenicity and mutagenicity, but its continued misuse is widespread in aquacultures. Therefore, there is an urgent need for a simple, reliable, and rapid method for the detection of its marker residue, 3-amino-2-oxazolidinone (AOZ), in aquatic products. In this regard, we modified a simplified indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) to address this need. A good linearity was achieved over a concentration range of 0.05-12.15μgL-1, and the IC50 value was 0.96μgL-1. The sample preparation was simple and effective included water bath treatments, acid hydrolysis combined with overnight derivatization of AOZ by benzaldehyde. The limit of detection and the limit of quantification were 0.15 and 0.3μgkg-1. The recoveries of AOZ in all tissues were between 78.0-95.3% at the levels of 0.3, 1.0, and 2.0μgkg-1. The inter-assay variability was less than 19.1%. The modified ic-ELISA was applied in quantification of AOZ elimination in carp. The results showed that AOZ was quite difficult to eliminate. Good correlations of the results obtained by ELISA and LC-MS/MS were observed in incurred carp muscle (r=0.9923) and carp plasma (r=0.9915) at the levels of 2.5-571.8μgkg-1 (μgL-1). Better results were obtained by modified ic-ELISA when compared with commercial ELISA kit. Therefore, the present assay is considered a rapid, accurate, reliable, and inexpensive method for the detection of furazolidone-residues in the edible tissues of aquatic animals. © 2010 Elsevier B.V.

摘要:
Immobilized artificial membrane chromatography stationary phase was prepared by coating soybean phosphatidylcholine (PC) on zirconia-magnesia micro-particles. The stability and chromatographic properties were investigated and compared with the PC-coated silica chromatography stationary phase prepared by the same method. PC-coated zirconia-magnesia chromatography stationary phase was more stable than the silica especially on resisting organic solvents. Hydrophobic action was the main factor for the retention of analyte on the new artificial membrane chromatography stationary phase, and electrostatic interaction had some contribution to retention. In addition, the special interaction between analyte and matrix affected retention greatly. Basic solutes were appropriate to be analyzed on PC-coated zirconia-magnesia stationary phase and acidic solutes were appropriate to be done on the silica one. Hence, the two different matrices artificial membrane stationary phases were perfectly complementary. ©2010 WILEY-VCH Verlag GmbH &Co. KGaA, Weinheim.

Lycium barbarum and Ganoderma lucidum, highly valued Chinese medicinal fruit and mushroom, have attracted more and more attention because of the antitumour activities they have shown, and polysaccharides are considered their most important functional constituent. Forming new compounds of L. barbarum polysaccharides, isolated from dried L. barbarum fruits, and G. lucidum polysaccharides, derived from a submerged culture of G. lucidum, has been investigated in this paper. Our data illustrate that two polysaccharide compounds, fermented polysaccharides and mixed polysaccharides, at the appropriate ratio and concentration have stronger free radical scavenging ability than the single polysaccharide. Ion exchange chromatography and high-performance liquid chromatography (HPLC) analyses indicated that the components of the new polysaccharide compounds had changed compared to those of the single polysaccharide. In addition, similar fractions were shared with the two polysaccharide compounds. Hence, the findings demonstrate that these new polysaccharide compounds might have stronger bioactivity than a single polysaccharide and could be obtained more easily by fermentation.

With an aim to evaluate dye decolorization by white rot fungus on natural living conditions, reproducing by solidstate fermentation, the process of triphenylmethane dyes decolorization using the white rot fungus P. ostreatus BP, cultivated on rice straw solid-state medium, has been demonstrated. Three typical dyes, including malachite green, bromophenol blue, and crystal violet, were almost completely decolorized by the fungus after 9 days of incubation. During the process of dye decolorization, the activities of enzyme secreted by the fungus, and the contents of soluble components, such as phenolic compounds, protein, and sugar, changed regularly. The fungus could produce ligninolytic, cellulolytic, and hemicellulolytic enzymes and laccase was the most dominant enzyme in solid-state medium. Laccase, laccase isoenzyme, and the laccase mediator could explain the decolorization of malachite green, bromophenol blue, and crystal violet by the fungus in solid-state medium, respectively. It is worth noting that the presence of the water-soluble phenolic compounds could stimulate the growth of fungus, enhance the production of laccase, and accelerate dye decolorization. © The Korean Society and Biotechnology.

The amino acid compositions, secondary structure, and self-assembly of oat protein isolate (OPI), which was purified from the high-protein Chinese oat, have been investigated by using a combination of amino acid analysis, Fourier transform infrared spectroscopy (FTIR), and tapping mode atomic force microscopy (TP-AFM). OPI, with molecular weights ranging from 14.0 kDa to 66.0 kDa, was rich in essential amino acids and contained 24.7% glutamic acid and 8.1% leucine. The amino acid contents of OPI are 4.5-8.7 times higher than those of oat flour. The secondary structures of OPI have been quantified by the deconvolution of the amide I band of the FTIR spectrum of OPI, which were found to contain approximately 7% β-tum, 19% α-helix, and 74% β-sheet. Tapping mode AFM results further suggest that the oat protein isolate has two major types of shapes, ellipsoidal and disk-like. At protein concentrations below 0.5 mg/mL, most of the OPI molecules are in the isolated form. However, when the concentration of OPI reaches 1.0 mg/mL, some of the OPI molecules self-assembled into large and heterogeneous protein aggregates. © 2009 American Chemical Society.

Biodegradation of crystal violet by white rot fungus Pleurotus ostreatus BP in rice straw solid-state medium mostly depended on low molecular mass fraction (LMMF) and the hypothetical metabolic pathways of crystal violet by LMMF was proposed by the biodegradation of crystal violet and by the deduced six metabolites formed.
