This study was performed to determine if pyruvate, which acts as a critical intermediate in energy metabolism, can substitute the role of glutamate as a metabolic fuel and effectively reduce nitrogen excretion in pigs. First, the experiment in vitro was carried out to investigate the effects of culturing porcine small intestinal epithelial cell line with pyruvate on the oxidation. Then, barrows weighing 40 kg were used in the experiment investigating the changes of nitrogen balance in response to addition of pyruvate to low-protein diets. Last, barrows (40 kg), which were surgically fitted with permanent catheters in the mesenteric vein, portal vein, hepatic vein, and carotid artery, were used to investigate the effects of supplementing low-protein diets with calcium pyruvate on the net portal fluxes of amino acids (AAs) and the consumption of AAs in the liver. The results showed that culturing cells with sodium pyruvate significantly reduced the number of glutamate oxidation (P < 0.05). Addition of calcium pyruvate to low-protein diets significantly reduced urinary nitrogen excretion from 13.2 g/d (18.0% crude protein, CP) to 10.3 g/d (15.0% CP) or 7.80 g/d (13.5% CP) and total nitrogen excretion from 22.5 g/d (18.0% CP) to 17.8 g/d (15.0% CP) or 14.2 g/d (13.5% CP) (P < 0.05), without obviously negative effects on the nitrogen retention (P > 0.05). Addition of calcium pyruvate to low-protein diets significantly decreased essential AA consumption rate in the liver (P < 0.05). This diet modification reduced the net portal fluxes of NH3, glycine, and alanine, as well as urea production rate in the liver (P < 0.05). The results indicated that pyruvate is an effective substitute for glutamate as a supplement in low-protein diets, reducing porcine nitrogen excretion and nitrogen consumption. © The Author(s) 2018. 对采自湖北省洪湖地区的泽蛙(Fejervarya limnocharis)肠道内寄生的泽蛙原蛙片虫进行了形态学再描述,包括活体形态、固定染色标本形态及扫描电镜下超微结构。样本形态特征参数值与Nie(倪达书)首次发现并命名时所记述标本参数值颇为相符(包括体长、体宽,核长、核宽)。此外,本文对2个重要的分类特征进行了补充和修正:(1)缝线位于虫体顶端,贯穿背腹侧;所有体动基列均从缝线两侧发出。(2)胞核分裂时在两新核间产生的连接细丝,在虫体整个生活周期一直存在。并针对这2点特征与其他已知的原蛙片虫属种类进行了比较和讨论:认为缝线结构是较为稳定的分类特征之一,体动基列均由此处生发并与虫体纵轴平行延伸;推测泽蛙原蛙片虫子代虫体中的2个胞核均来自于母体中同一个核,而其2核间的连接细丝可能是胞核分裂进化历程中比较原始的残迹。

Reducing dietary crude protein (CP) intake effectively decreases nitrogen excretion in growing-finishing pigs but at the expense of poor growth when dietary CP content is reduced by ≥3%. In this study, we investigated the main disadvantages of low-protein diets supplemented with lysine, methionine, threonine, and tryptophan in pigs. First, changes in the nitrogen balance in response to differences in dietary CP content (18%, 15%, and 13.5%) were investigated in barrows (40 kg). Then, barrows (40 kg) surgically fitted with catheters in the mesenteric vein, portal vein, hepatic vein, and carotid artery were used to investigate changes in amino acid (AA) metabolism in the portal-drained viscera and liver in response to differences in dietary CP content. The results showed that low-protein diets reduced fecal and urinary nitrogen excretion (P < 0.05) meanwhile resulted in significant decreases in nitrogen retention (P < 0.05). Moreover, a reduction in the dietary CP content from 18% to 13.5% resulted in decreases in the net portal fluxes of NH3, glycine, and alanine as well as in the urea production in the liver (P < 0.05), whereas their values as a percentage of nitrogen intake did not decline (P > 0.05). The net portal fluxes of nonessential AA (NEAA) were reduced in the low-protein diet groups (P < 0.05), while essential AA consumption in the liver increased (P < 0.05). Thus, low-protein diets result in reductions in both nitrogen excretion and retention, and NEAA deficiency may be a major disadvantage of low-protein diets. © 2018 American Chemical Society.

Morphological studies of Sicuophora (Syn. Wichtermania) multigranularis Xiao et al., 2002, from the rectum of the frog, Quasipaa spinosa, performed using silver impregnation and scanning electron microscopy, confirmed the following newly recognized features: (1) only one apical suture on the right surface; (2) two naked regions at the posterior end of both the left and the right side of the body. Phylogenetic analysis based on the SSU-rRNA gene showed that S. multigranularis is a sister to a clade comprising all other Clevelandellida, strongly supporting the validity of the genus Sicuophora. This is also the first molecular data obtained for the genus Sicuophora. Because of the lack of molecular data, it will be necessary to obtain more genetic data from the family Sicuophoridae to discuss the question of the taxonomic status of the genus Sicuophora. The extensive metabolism of amino acids (AA) as fuel is an important reason for the low use efficiency of protein in pigs. In this study, we investigated whether regulation of the pyruvate dehydrogenase kinase (PDK)/pyruvate dehydrogenase alpha 1 (PDHA1) pathway affected AA consumption by porcine intestinal epithelial (IPEC-J2) cells and intestinal bacteria in pigs. The effects of knockdown of PDHA1 and PDK1 with small interfering RNA (siRNA) on nutrient consumption by IPEC-J2 cells were evaluated. IPEC-J2 cells were then cultured with sodium dichloroacetate (DCA) to quantify AA and glucose consumption and nutrient oxidative metabolism. The results showed that knockdown of PDHA1 using siRNA decreased glucose consumption but increased total AA (TAA) and glutamate (Glu) consumption by IPEC-J2 cells (P < 0.05). Opposite effects were observed using siRNA targeting PDK1 (P < 0.05). Additionally, culturing IPEC-J2 cells in the presence of 5 mM DCA markedly increased the phosphorylation of PDHA1 and PDH phosphatase 1, but inhibited PDK1 phosphorylation (P < 0.05). DCA treatment also reduced TAA and Glu consumption and increased glucose depletion (P < 0.05). These results indicated that PDH was the regulatory target for shifting from AA metabolism to glucose metabolism and that culturing cells with DCA decreased the consumption of AAs by increasing the depletion of glucose through PDH activation. © 2018 American Chemical Society.

Balantidium grimi n. sp. is described from the rectum of the frog Quasipaa spinosa (Amphibia, Dicroglossidae) from Lishui, Zhejiang Province, China. The new species is described by both light microscopy (LM) and scanning electron microscopy (SEM), and a molecular phylogenetic analysis is also presented. This species has unique morphological features in that the body shape is somewhat flattened and the vestibulum is "V"-shaped, occupying nearly 3/8 to 4/7 of the body length. Only one contractile vacuole, situated at the posterior body, was observed. The phylogenetic analysis based on SSU-rDNA indicates that B. grimi groups together with B. duodeni and B. entozoon. In addition, the genus Balantidium is clearly polyphyletic.

In this paper, we present transcriptome data for Balantidium ctenopharyngodoni Chen, 1955 collected from the hindgut of grass carp (Ctenopharyngodon idella). We evaluated sequence quality and de novo assembled a preliminary transcriptome, including 43.3 megabits and 119,141 transcripts. Then we obtained a final transcriptome, including 17.7 megabits and 35,560 transcripts, by removing contaminative and redundant sequences. Phylogenomic analysis based on a supermatrix with 132 genes comprising 53,873 amino acid residues and phylogenetic analysis based on SSU rDNA of 27 species were carried out herein to reveal the evolutionary relationships among six ciliate groups: Colpodea, Oligohymenophorea, Litostomatea, Spirotrichea, Heterotrichea and Protocruziida. The topologies of both phylogenomic and phylogenetic trees are discussed in this paper. In addition, our results suggest that single-cell sequencing is a sound method of obtaining sufficient omics data for phylogenomic analysis, which is a good choice for uncultivable ciliates. The transcriptome data for Balantidium ctenopharyngodoni are the first omics data within the subclass Trichostomatia, and provide a good basis for ciliate phylogenomic analysis, as well as related omics analysis.

Cepedea longa Bezzenberger, 1904, collected from Fejervarya limnocharis (Amphibia, Anura, Ranidae) from Honghu Lake, Hubei Province, China in May-July 2016, is described at both light and transmission electron microscope levels. This is the first electron microscopic study of this species. Cepedea longa possesses a developed fibrillar skeletal system, composed of longitudinal fibrillar bands and transversal fibrils as well as numerous thin microfibrils dispersed in the endoplasm, which may play an important role in morphogenesis and offer some resilience to deformations of the cell. Longitudinal microfibrils are polarizing elements of kineties, bordering the somatic kineties on the left side and possibly responsible for kinetosome alignment. Two types of vesicles exist in the somatic cortex: globular endocytotic vesicles and flattened exocytotic vesicles. As to the nuclei of C. longa, a thick microfibrillar layer was observed to attach to the cytoplasmic face of the nuclear envelope. This fact suggests no necessary connection between the presence of this microfibrillar layer and the number of nuclei. In addition, some unknown tightly-packed microtubular structures in the nucleoplasm were observed for the first time in opalinids; neither their nature nor physiological significance is known. The morphological revisions of Macrocytopharynxa pyriformis (Nie, 1932) Li et al., 2002; collected from the rectum of Fejervarya limnocharis (=Rana limnocharis), are presented in this paper: (1) two surfaces of the organism are not - identical left side narrower and convex, right broader and flat or slightly concave; (2) infundibulum is large and well-developed with no "fold" or "plicature" present in the middle or posterior portion; (3) micronucleus is tiny and ovoid shaped and always embedded in the middle concavity of macronucleus, which can be well revealed by ammoniacal silver staining. Our phylogenetic analysis based on SSU-rDNA showed that M. pyriformis fell into the Nyctotheroides Glade, within which four definite Nyctotheroides species were involved - N. cordiformis, N. deslierresae, N. parvus and N. hubeiensis. In combination with their morphological features, we discussed the reliability of using karyophore organelles or kinetal suture patterns as the generic taxonomic criteria. Besides, we considered that the genus Macrocytopharynxa is a junior synonym of Nyctotheroides and we transfer its type species to Nyctotheroides as Nyctotheroides pyriformis n. comb. The phylogenetic pattern of the family Nyctotheridae was also indicated in our work, but it will be necessary to analyze more species from fishes and reptiles before coming to a sound conclusion. This paper gave detailed description of the fine structure of Apiosoma piscicola Blanchard 1885. These parasites were collected from the fry of Tachysurus fulvidraco during parasite surveys of fish at Jiangxia Fish Hatchery in Wuhan, Hubei province in May 2011. Its pellicle, peristomial apparatus, nuclei, trochal band and scopula were described, respectively. The relationship between these peritrich ciliates and their hosts was well discussed herein. The scopula of A. piscicola did not adhere directly to the surface of the host epithelial cells, but by means of a secreted adhesive pad of material according to the present study. Besides, no ultrastructural damage to host epithelial cells could be detected in our work. We concluded that these sessilids should be ectocommensals rather than ectoparasites, but not always harmless. In addition, their great variability of body shape and non-strict host specificity were also discussed. © 2016, Indian Society for Parasitology.

The complete mitochondrial DNA sequence of Triplophysa venusta was determined using a PCR-based method in our present study. The genome is totally 16 574 bp in length and contains 13 protein-coding genes, 2 ribosomal RNA, 22 transfer RNA genes, and 1 non-coding control region. The overall base composition of T. venusta is A 27.8%, T 26.9%, C 26.9%, and G 18.4%, with an A + T bias of 54.7%. This complete mitogenome of T. venusta provides a basic data for studies on species identification, molecular systematics, and conservation genetics. The phylogenetic trees consistently showed that the Family Cobitidae was divided into three major clades: the subfamily Botiidae, the subfamily Nemaeheilidae, and the subfamily Cobitinae. Cobitinae and Nomaeheilidae formed a main Loach clade, with Botiidae clustered at the basal. © 2015 Taylor & Francis.

The morphology of Nyctotheroides hubeiensis (Acta Hydrobiol. Sin. 1998, 22(suppl.):187), collected from the rectum of Phelophylax nigromaculatus, is presented in this paper based on detailed morphological information and molecular data. Our phylogenetic analysis showed that N.hubeiensis fell into the Nyctotheroides clade, which was strongly supported as monophyletic and clustered as basal to the genera Nyctotherus and Clevelandella. Also, the monophyly of the Order Clevelandellida and the affinity of parasitic nyctotherids and free-living metopids were indicated in our work. The origin of clevelandellid ciliates as well as their possible evolutionary history was also discussed here; however, the analysis of more species from other vertebrate hosts (fish, reptiles) should be made before a well-supported conclusion can be drawn. © 2016 The Author(s) Journal of Eukaryotic Microbiology © 2016 International Society of Protistologists

A redescription of Protoopalina pingi Nie, 1935 is presented in this paper to complete Nie's description at both light and scanning electron microscope levels. These organisms were collected from the recta of the frogs Hylarana guentheri Boulenger, 1882 and Pelophylax nigromaculatus Hallowell, 1861 from Jialing River, Sichuan Province and Honghu Lake, Hubei Province, respectively, in China. This is the first record of its occurrence in H. guentheri and P. nigromaculatus. The body of P. pingi is elongated and somewhat spindle-like in shape, slightly narrowed and bluntly rounded at the anterior extremity, while the posterior end is tapering or sharply pointed. The body surface is thickly flagellated, with the caudal tip being barren. The falx, located at the margin of the anterior end, is composed of a narrow band of kinetosomes. Four round or oval-shaped nuclei, usually arranged in a straight line, are situated in the middle region of the body. Comparisons are made between P. pingi and its congeners. © 2014 W. We obtained sequences of the small subunit ribosomal RNA (rRNA) for two new isolates of Balantidium from fishes, Balantidium polyvacuolum and Balantidium ctenopharingodoni. This is the first introduction of molecular data of Balantidium species from fish hosts in the phylogenetic analyses of the ciliate subclass Trichostomatia. Despite the fact that these species share morphological characteristics common to other species of Balantidium, the phylogenetic analysis of their sequences has shown that they are to be placed in a different branch closely related to the so-called Australian clade. Thus, our results indicate that the genus Balantidium is polyphyletic and possibly should be represented by two different genera; however, the analysis of more species from other poikilothermic hosts (amphibians, reptiles) should be made before a revised taxonomical proposal could be made. © 2014, Springer-Verlag Berlin Heidelberg.

A detailed description of the fine structure of Balantidium ctenopharyngodoni Chen, 1955 with an emphasis on its vestibulum is given in the present paper. As to the vestibular kinetids, special attention is paid to the characters of T1, T2 microtubules and nematodesmata. Serving as the major skeleton to the vestibular cortex, the T1, T2 and Pc microtubules are described herein and their support function is also discussed. Moreover, the well-developed nematodesmata of the vestibular kinetids that form a large basket-like complex are described in detail.

A new trichostome ciliate, Balantidium honghuensis n. sp., was isolated from the recta of Rana nigromaculata and R. limnocharis during parasite surveys in Honghu Lake, Hubei Province, central China in summer of 2010 and 2011. Its detailed morphometric characters based on LM and SEM studies were described herein. The organism is oval in shape and thickly ciliated. The vestibulum is ""V"" shaped and occupies about 1/3 to 2/5 of the body length. The vestibular and nearby regions possess strong peripheral fibers which form a marked axial fiber about the cytopharynx. More than 10 contractile vacuoles are distributed along the periphery of the latter body. Comparisions were made between this new species and B. sinensis Nie, 1935. They were discriminated from each other in terms of general body forms, body size, and vestibulum shapes. Besides, special attention was paid to its high-speed daughter swarmers which we believed to be the infective stage of B. honghuensis. Possible infection routes of anura amphibian balantidia were discussed. © 2013, Korean Society for Parasitology and Tropical Medicine.

累枝虫(Epistylis sp.)隶属于缘毛亚纲,固着目,累枝虫科;是一类广泛附生于多种水生植物、无脊椎动物和脊椎动物的纤毛虫。在一定情况下,可以严重危害养殖的鱼苗、鱼种、鳖及各种虾、蟹的卵、幼体和成体。其中尤以对虾、蟹的幼体和鱼苗的危害最大,可引起大批死亡,残存的商品价值也大大降低。珠蚌累枝虫(Epistylis unioi)是龚循矩(1986)在圆顶珠蚌的鳃叶上发现并命名的。本文以寄生在黄颡鱼鱼苗的珠蚌累枝虫为主要研究对象,利用蛋白银染色法和扫描电镜技术,对这一虫种进行了再描述。珠蚌累枝虫在黄颡鱼苗体表的发现是其宿主的一个新纪录。结果表明:黄颡鱼鱼苗为大量的珠蚌累枝虫感染,鱼体表现为生命力减弱,游动缓慢,浮头、呼吸急促;置于解剖镜下观察发现大量的虫体存在于鱼体体表、鳍、须和鳃上。虫体细长,略呈花瓶状,活体状态下虫体完全伸展时体长56-88(68)μm,体宽22-38(30)μm,口围唇的直径27-45(35)μm。蛋白银染色状态下,体长34-53(41)μm,体宽16-26(22)μm。虫体表面可见多条横向排列的条纹,在口围与横纤毛带间有37-45(41)条,横纤毛带与帚胚间有26-33(29)条,相邻条纹间距约为0.5μm。横向条纹上可见许多排列稀疏的气孔。单毛基列和复毛基列均起自于细胞顶端口围唇的内侧,两者相伴而行,进入口区之前围绕口缘1.5圈。P1-3分别由三条毛基索组成,其中P1、P2远长于P3;P1的下端一直延伸至胞口处,而P2和P3末端交汇,位于P1的下端点之上。伸缩泡一个,位于虫体顶端口围盘稍下处。大核呈马蹄形,横位,位于虫体的前部。群体较小,呈不对称二叉式分支。此外,我们还发现了虫体的游泳体,在横纤毛带上生出密集的纤毛。扫描电镜显示,在虫体的体柄与寄主皮肤接触的部位,可见明显损伤。

The peritrich Epistylis unioi Gong, 1986 was collected from fry of Pelteobagrus fulvidraco during parasite surveys at Honghu Lake Fish Hatchery, Hubei Province, central China in May 2010 and redescribed. Some revisions were done based on live, silver-impregnated, and SEM specimens. The zooid is elongated and somewhat vase-like in shape, measuring 56-88 × 22-38 μm in vivo. A single contractile vacuole is apically located slightly below the peristome disc. The macronucleus is horseshoe-shaped, always transversely situated at the foreside of the body. Haplokinety (H) and polykinety (Po) complete one and one-half circuits on the peristome before entering the infundibulum, with a distal kinetal fragment present at the distal end. Silverline system consists of 37-45 pellicular striations between peristome and aboral trochal band (TB), and of 26-33 between TB and scopula. Colony is asymmetrically and dichotomously branched, usually with only two levels of branches. In addition, the telotrochs of E. unioi were also observed and its structures were described herein. Besides, obvious skin lesions caused by the ringlike base of E. unioi were detected and the relationship between these epizooites and their hosts was briefly discussed as well.
