国内6种地衣芽孢杆菌饲料添加剂比较试验

摘要:
为了解国内地衣芽孢杆菌添加剂中菌种的数量及性能,对6家不同公司生产的地表芽孢杆菌制剂进行计数,并研究其生物学特性的差异.分别对6种地衣芽孢杆菌制剂计数后筛选出其中的地衣芽孢杆菌,对其进行高温处理、酸处理和猪胆盐处理后计算存活率并比较其耐受性,通过测定常见病原病的抑菌圈直径,比较其抑菌特性.结果表明:6种地表芽孢杆菌均能耐受90 ℃高温,但存活率差异较大(59.84 %~72.52%);经过pH 2.0酸处理后存活率也各不相同(39.51%~52.85%),经过1.0%猪胆盐溶液处理后存活率也存在差异(13.24 %~25.39%);6种菌株均具有抑菌特性,抑菌效果差异不明显.研究发现,国内生产的地衣芽孢杆菌添加剂品质参差不齐,只有2种产品的数量与其标明数量相符,且仅2种产品的地衣芽孢杆菌生物学特性相对较好.

摘要:
Alpha-ketoglutarate (AKG) is an intermediate of the Krebs cycle which bridges amino acid metabolism with glucose oxidation in animals. Of particular interest is the conversion of AKG into glutamate by mitochondrial glutamate dehydrogenase in the gastrointestinal tract where glutamate has multiple physiological functions (including regulation of cell function, neurotransmission, and gastric emptying). Additionally, AKG stimulates the initiation of catabolism of branched-chain amino acids (BCAA) via BCAA transaminase in enterocytes. Oxidation of AKG also provides large amounts of ATP and modulates cellular redox state in the small intestine. Translating the basic research into practice, results of recent studies indicate that dietary supplementation with AKG alleviates oxidative stress and injury in intestinal mucosal cells, while improving intestinal mucosal integrity and absorption of nutrients in endotoxin-challenged pigs. The beneficial effects of AKG are associated with increased activation of the mTOR signaling pathway and net protein synthesis. Thus, AKG is a novel and promising supplement in diets to improve intestinal health in animals and possibly humans.

摘要:
试验研究了乙酰半胱氨酸(NAC)对脂多糖(LPS)多次刺激造成的免疫应激仔猪炎性介质的影响,旨在探讨NAC是否有缓解仔猪免疫应激的作用.选取来源一致、体重相近的18头健康仔猪[杜洛克×长白×大白,体重(11.58±0.26)kg随机分成3个处理(对照组、LPS组和NAC组),每个处理6个重复,每个重复1头猪.对照组和LPS组饲喂基础日粮,试验组(NAC组)为基础日粮+500 mg/kg NAC,LPS组和NAC组仔猪分别于试验的第10、13、20 d腹膜注射100μg/kg BW的LPS,对照组注射等量的灭菌生理盐水.试验第10、20 d在注射LPS 3 h后前腔静脉采血,测定血浆TNF-α、COR、IL-6和PGE<,2>含量.试验结果显示:日粮中添加NAC显著缓解了LPS刺激导致的血浆(第10 d)和血浆(第20 d)中TNF-α、COR、IL-6和PGE<,2>含量的升高(P<0.05).由此可见,在本试验条件下,LPS刺激引起仔猪血浆中炎性介质升高,日粮中添加500 mg/kg NAC缓解LPS导致的血浆中炎性介质的升高,缓解免疫应激,最终可能缓解LPS刺激引起的生长抑制.

摘要:
[目的]研究α-酮戊二酸(AKG)对经脂多糖(LPS)多次刺激后断奶仔猪肌肉能量代谢的影响及其分子机制.[方法]采用2×2因子试验设计,LPS刺激(有、无)和AKG添加量(0、1%)为2个主效应,共4个处理组.选取24头(6.79±0.32)kg的断奶仔猪(杜洛克×长白×大白),随机分配到4个处理组,每个处理设6个重复(猪).LPS刺激组仔猪在试验的第10、13和15天腹腔注射100 μg·kg(-1)BW LPS,无LPS刺激组注射等量的生理盐水.第16天仔猪麻醉后进行屠宰,取腓肠肌样品,反相高效液相色谱法检测腓肠肌细胞内腺苷酸水平,Western blotting检测腓肠肌AMP激活蛋白激酶(AMPK)和乙酰CoA羧化酶(ACC)表达及其磷酸化.[结果](1)日粮添加1%AKG和LPS刺激对仔猪腓肠肌AMP含量和AMP/ATP的影响存在交互作用(P<0.05).仔猪日粮中添加1% AKG能显著提高腓肠肌ADP水平(P<0.05),缓解了LPS刺激引起的AMP水平和AMP/ATP升高、ADP和能荷(EC)降低(P<0.05).(2)LPS刺激和日粮中添加AKG对AMPK表达量和AMPK磷酸化水平(pAMPK/AMPK)均无影响.(3)日粮中添加1% AKG抑制了ACC的磷酸化;LPS刺激使ACC表达量升高(P<0.05),并促进ACC磷酸化(P<0.05).[结论]LPS刺激增加肌肉能量损耗,日粮中添加1% AKG有利于肌肉维持正常的能量代谢;日粮添加AKG通过调控ACC的活性与表达影响肌肉能量代谢;AKG可能通过ACC信号影响肌肉能量代谢,进而改善LPS刺激下机体能量代谢状态.

摘要:
The present study determined whether α-ketoglutarate (AKG) might affect the expression of AMP-activated protein kinase (AMPK) and energy status in the intestinal mucosa of piglets challenged with Escherichia coli lipopolysaccharide (LPS). A total of eighteen piglets (weaned at 21 d of age) were allocated to one of three treatments: (1) non-challenged (control); (2) LPS-challenged (LPS); (3) LPS+1 % AKG (LPS+AKG). Piglets in the control and LPS groups were fed a maize-and soyabean meal-based diet, and the LPS+AKG group was fed the basal diet supplemented with 1 % AKG. On days 10, 12, 14 and 16 of the trial, piglets in the LPS and LPS+AKG groups were challenged with LPS (80 Î1/4g/kg body weight), whereas piglets in the control group received the same volume of sterile saline. Pigs were euthanised 24 h after the last administration of LPS or saline to obtain intestinal mucosae for biochemical analysis. Compared with the control group, LPS administration decreased (P < 0•05) the oxidation of AKG, oleic acid, glutamine and glucose in enterocytes, decreased concentrations of ATP in the duodenal and jejunal mucosae and decreased adenylate energy charge (AMP:ATP ratio) in the jejunal and ileal mucosae. Additionally, LPS treatment reduced (P < 0•05) mucosal concentrations of phosphorylated AMPK in the jejunum and ileum as well as acetyl-CoA carboxylase in all segments of the small intestine. The adverse effects of LPS were reversed by AKG. Collectively, these results indicate that dietary supplementation with 1 % AKG beneficially modulates the AMPK signalling pathway to improve energy status in the small intestine of LPS-challenged piglets. © 2011 The Authors.

摘要:
This study determined effects of dietary supplementation with L-arginine (Arg) or N-carbamylglu-tamate (NCG) on intestinal health and growth in early-weaned pigs. Eighty-four Landrace × Yorkshire pigs (average body weight of 5.56 ± 0.07 kg; weaned at 21 days of age) were fed for 7 days one of the three iso-nitrogenous diets: (1) a corn-and soybean meal-based diet (CSM), (2) CSM + 0.08% NCG (0.08%), and (3) CSM + 0.6% Arg. There were four pens of pigs per diet (7 pigs/pen). At the end of a 7-day feeding period, six piglets were randomly selected from each treatment for tissue collections. Compared with the control group, Arg or NCG supplementation increased (P < 0.05): (1) Arg concentrations in plasma, (2) small-intestinal growth, (3) villus height in duodenum, jejunum and ileum, (4) crypt depth in jejunum and ileum, (5) goblet cell counts in intestinal mucosae, and (6) whole-body weight gain in pigs. Realtime polymerase chain reaction and western blotting analyses revealed that both mRNA and protein levels for heat shock protein-70 (HSP70) were higher (P< 0.05) in the intestinal mucosae of Arg-or NCG-supplemented pigs than in the control group. Furthermore, the incidence of diarrhea in the NCG group was 18% lower (P < 0.01) than that in the control group. Collectively, these results indicate that dietary supplementation with 0.6% Arg or 0.08% NCG enhances intestinal HSP70 gene expression, intestinal growth and integrity, and the availability of dietary nutrients for whole-body weight gain in postweaning pigs fed a CSM-based diet. Thus, Arg or NCG is a functional ingredient in the weaning diet to improve nutrition, health, and growth performance of these neonates. © Springer-Verlag 2010.

试验研究了脯氨酸(pmline,PRO)对脂多糖(LPS)多次刺激的仔猪生长性能、血液生化指标的影响.选取来源一致、体重相近[(9.13±0.85)kgl的健康断奶仔猪36头,采用2×3因子试验设计.设免疫应激和PRO添加量2个因子,其中免疫应激设2个水平:无应激(不注射LPS,注射相同剂量的生理盐水)和LPS刺激(腹膜注射100 μg/kgBW LPS);PRO添加量设3个水平:0.05%和1.0%.共6个处理组,每个处理6个重复,每重复1头仔猪,试验期20 d.试验前10 d,未进行LPS处理,于试验的第10、13和20 d对应激组仔猪腹膜注射100μg/kgBW LPS,无应激组仔猪注射相应剂量的生理盐水.试验第10、20 d空腹称重,并于注射LPS 3 h后前腔静脉采血,观测有关指标.试验结果显示:①LPS刺激降低了仔猪生长性能、提高了血浆中尿素氮,对过氧化氢酶(CAT)、丙二醛(MDA)和超氧化物歧化酶(SOD)均无影响.②日粮添加PRO可以显著降低血浆BUN和MDA,提高血浆SOD,以0.5%的效果较好.在本次试验条件下,LPS刺激显著降低了仔猪的生长性能,提高了血浆中尿素氮水平,日粮添加0.5%的PRO可提高机体抗氧化能力和降低血浆中尿素氮水平;添加脯氨酸对LPS刺激导致的生长抑制没有缓解作用.

Neonates are at increased risk for inflammatory bowel disease, but effective prevention and treatments are currently limited. This study was conducted with the lipopolysaccharide (LPS)-challenged piglet model to determine the effects of dietary supplementation with α-ketoglutarate (AKG) on the intestinal morphology and function. Eighteen 24-day-old pigs (weaned at 21 days of age) were assigned randomly to control, LPS, and LPS + AKG groups. The piglets in the control and LPS groups were fed a corn-and soybean meal-based diet, whereas the LPS + AKG group was fed the basal diet supplemented with 1% AKG. On days 10, 12, 14, and 16, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16, D-xylose was orally administrated to all pigs at the dose of 0.1 g/kg BW, 2 h after LPS or saline injection, and blood samples were collected 3 h thereafter. Twenty-four hours post-administration of LPS or saline, pigs were killed to obtain intestinal mucosae for analysis. Compared with the control group, LPS challenge reduced (P < 0.05) protein levels, the ratio of villus height to crypt depth, and the ratio of phosphorylated mTOR to total mTOR in duodenal, jejunal, and ileal mucosa. These adverse effects of LPS were attenuated (P > 0.05) by AKG supplementation. Moreover, AKG prevented the LPS-induced increase in intestinal HSP70 expression. Collectively, these novel results indicate that dietary supplementation with 1% AKG activates the mTOR signaling, alleviates the mucosal damage, and improves the absorptive function of the small intestine in LPS-challenged piglets. The findings not only help understand the mode of AKGs actions in the neonatal gut but also have important implications for infant nutrition under inflammatory conditions. © Springer-Verlag 2010.

摘要:
L-type amino acid transporter-1 (LAT1) transports large, branched-chain, aromatic and neutral amino acids. About 64 Duroc Landrace Yorkshire pigs were used to study the effects of dietary crude protein (CP), energy and arginine on LAT1 expression in forebrain. The results showed that LAT1 expression in forebrain was sensitive to different levels of CP, energy and arginine. On the basis of Western blot analysis, a lower level of LAT1 presented in the brain tissues of pigs fed the low dietary CP diet (P < 0.05), a higher level were found in pigs fed the higher CP diet, with moderate to intense staining seen in pigs fed the diet plus 1% arginine. In contrast, pigs fed the control-energy diet had weak LAT1 expression, and those fed the diet supplemented with 1% arginine showed lowest LAT1 expression (P < 0.05). These results showed that LAT1 was highly expressed in the forebrain, and expression of LAT1 was affected by dietary protein, energy and arginine differently. Copyright © The Animal Consortium 2010.

α-酮戊二酸对脂多糖应激断奶仔猪生长抑制的缓解作用

摘要:
试验用大肠杆菌脂多糖(LPS)多次刺激断奶仔猪建立应激模型,研究α-酮戊二酸(AKG)对断奶仔猪的生长性能和血浆激素水平的影响,探讨AKG能否缓解仔猪应激反应.选用24头健康断奶仔猪(杜洛克×长白×大白,体重(7.25±0.23)kg),随机分成3个组(空白对照组、应激对照组和AKG组),每组8个重复,每个重复1头猪.2个对照组饲喂基础日粮+1%淀粉,AKG组饲喂基础日粮+1%AKG,在试验第10天、第12天、第14天和第16天分别在应激对照组和AKG组仔猪的腹膜注射80 μg/kg LPS.试验结果显示:(1)试验第1~9天,未注射LPS时,AKG组平均日增重分别比空白对照组和应激对照组提高30.0%(P<0.05)和31.1%(P<0.05),料重比降低12.4%(P<0.05)和12.8%(P<0.05);试验第10~16天(注射LPS),应激对照组与空白对照组相比平均日增重降低28.8%(P<0.05),料重比升高16.8%(P<0.05),但AKG组的平均日增重和料重比与空白对照组相比无显著性差异(P>0.05);(2)与空白对照组相比,试验第10天,首次LPS刺激后,应激对照组的血浆胰岛素样生长因子(IGF-I)水平降低22.7%(P<0.05),而AKG组无显著变化(P>0.05);试验第16天,多次LPS刺激后,与空白对照组比较,应激对照组的IGF-I和胰岛素(INS)水平分别降低21.2%(P<0.05)和25.7%(P<0.05),而AKG组无显著变化(P>0.05).结果表明,日粮中添加1%AKG能显著缓解LPS应激对杜×长×大断奶仔猪生长的负影响.

摘要:
酵母是一种重要的单细胞微生物,与人们的日常生活和工业应用密切相关。酵母的生产和发展已经有200多年的历史。在美国,用酵母产品饲喂动物的历史已超过100年。这些产品包括在农场里制作的酵母发酵饲料、酿酒厂产生的酵母副产品以及专门为饲喂动物而生产的商业性酵母产品(龙凌,2001)。酵母在饲料中最初作为单细胞蛋白产品替代饲料中部分鱼粉,不仅解决了部分地区蛋白资源紧缺的问题,也大大降低了饲料成本。随着酵母工业的发展和对酵母产品的研究不断加深,酵母及其相关产品在饲料工业中有了更广泛的应用,也展现出了广阔的应用前景。

A study on oxytetracycline-human serum albumin interaction based on flow-injection chemiluminescence analysis combined with ultrafiltration sampling

摘要:
Binding properties of oxytetracycline (OTC) and human serum albumin (HSA) have been studied in details. This investigation was based on flow injection chemiluminescence (FI-CL) analysis and ultrafiltration (UF) sampling technique. Chemiluminescence (CL) emission was observed during oxidation or OTC with KMnO4 in acidic medium in the presence of formaldehyde as a sensitizer. Coupled with flow injection technique, OTC can be determined in the range of 8.0 x 10(-7)-1.2 x 10(-4) mol L-1. Oxytetracycline and HSA were mixed in different molar ratios and incubated at 37+/-0.5 degrees C. An ultrafiltration probe was introduced to the sample containing unbound drug and the concentration of this free drug was determined by the CL method. The obtained data were analysed applying the Scatchard plot. The binding constant (K) and the number of binding sites (n) were 3.19 x 10(4) (mol L-1) and 1.10, respectively.
