Puerarin enhances intestinal function in piglets infected with porcine epidemic diarrhea virus

<jats:title>Abstract</jats:title><jats:p>Puerarin has been reported to be an excellent antioxidant, anti-inflammatory and antimicrobial agent, but the potential effect of puerarin on porcine epidemic diarrhea virus (PEDV) is unclear. This study aimed to determine whether puerarin could alleviate intestinal injury in piglets infected with PEDV. A PEDV (Yunnan province strain) infection model was applied to 7-day-old piglets at 10<jats:sup>4.5</jats:sup> TCID<jats:sub>50</jats:sub> (50% tissue culture infectious dose). Piglets were orally administered with puerarin at the dosage of 0.5mg/kg body weight from day 5 to day 9. On day 9 of the trial, piglets were inoculated orally with PEDV. Three days later, jugular vein blood and intestinal samples were collected. Results showed puerarin reduced morbidity of piglets infected with PEDV. In addition, puerarin reduced the activities of aspartate aminotransferase and alkaline phosphatase, the ratio of serum aspartate aminotransferase to serum alanine aminotransferase, the number of white blood cells and neutrophils, and the plasma concentrations of interleukin-6, interleukin-8 and tumor necrosis factor-α, as well as protein abundances of heat shock protein-70 in PEDV-infected piglets. Moreover, puerarin increased D-xylose concentration but decreased intestinal fatty acid-binding protein concentration and diamine oxidase activity in the plasma of piglets infected with PEDV. Puerarin increased the activities of total superoxide dismutase, glutathione peroxidase and catalase, while decreasing the activities of myeloperoxidase and concentration of hydrogen peroxide in both the intestine and plasma of PEDV-infected piglets. Puerarin decreased mRNA levels of glutathione S-transferase omega 2 but increased the levels of nuclear factor erythroid 2-related factor 2<jats:italic>.</jats:italic> Furthermore, puerarin increased the abundance of total eubacteria (16S rRNA)<jats:italic>, Enterococcus</jats:italic> genus<jats:italic>, Lactobacillus</jats:italic> genus and <jats:italic>Enterobacteriaceae</jats:italic> family in the intestine, but reduced the abundance of <jats:italic>Clostridium coccoides</jats:italic> in the caecum. These data indicate puerarin improved intestinal function in piglets infected by PEDV and may be a promising supplement for the prevention of PEDV infection.</jats:p>

摘要:
本试验旨在研究日粮中添加复合氨基酸对断奶仔猪免疫功能以及肠道能量和氨基酸水平的影响.将16头体重(4.83±0.16)kg的(21±3)日龄断奶仔猪随机分为对照组和试验组,每组8个重复,每个重复1头猪.对照组饲喂基础日粮+0.99%的丙氨酸(与试验组等氮),试验组饲喂基础日粮+1.00%的复合氨基酸(谷氨酸:谷氨酰胺:甘氨酸:精氨酸:N-乙酰半胱氨酸=10:4:4:2:1).于试验的第21天采集样品.结果表明:与对照组相比,试验组降低了断奶仔猪的腹泻率、血液白细胞和单核细胞的数量以及回肠黏病毒抗性蛋白-1 (MX1 )和干扰素诱导蛋白-1(IFIT1 )基因的相对表达量(P<0.05);有降低15~21 d耗料增重比的趋势(P<0.1);提高了血液血红蛋白含量、回肠干扰素(INF)-β和IFN-γ基因的相对表达量、空肠腺苷酸池以及空肠瓜氨酸和精氨酸的含量(P<0.05);有提高空肠三磷酸腺苷和单磷酸腺苷的趋势(P<0.1).由此可见,复合氨基酸可以有效减少断奶仔猪腹泻,这可能与其提高仔猪免疫力、增加肠道能量储备和精氨酸水平有关.

摘要:
为进一步探明复合功能性氨基酸的应用效果与机理,在前期研究的基础上,本试验研究了日粮中添加复合氨基酸对断奶仔猪肝脏能量状态、游离氨基酸及相关基因表达的影响.将16头体重相近的(21±3)日龄断奶仔猪随机分为对照组和复合氨基酸组,每组8个重复.分别饲喂添加0.99%丙氨酸(与试验组等氮)和1.00%复合氨基酸(谷氨酸∶谷氨酰胺∶精氨酸∶甘氨酸∶N-乙酰半胱氨酸=5∶2∶2∶1∶0.5)的基础日粮.于试验第21天前腔静脉采血,屠宰取肝脏样品,待测.结果 表明,与对照组相比,复合氨基酸显著降低了仔猪血浆ALT和AST活性(P<0.05);显著提高了仔猪肝脏总蛋白含量、RNA/DNA和总蛋白/DNA(P<0.05),显著降低了仔猪肝脏MDA和H2O2含量(P<0.05);显著提高了肝脏精氨酸和二磷酸腺苷(ADP)含量(P<0.05);显著上调了肝脏氨基酸转运载体b0,+ AT和y+LAT2基因、谷胱甘肽硫转移酶m2基因(GSTO2)、干扰素γ基因(IFN-γ)、脂蛋白酯酶基因(LPL)的表达(P<0.05),显著下调了脂肪酸合成酶基因(FASN)的表达(P<0.05).以上结果表明,日粮中添加1.00%复合氨基酸可以改善肝功能,提高抗氧化能力,可以调控仔猪肝脏能量状态及游离氨基酸代谢,并可能影响肝脏的脂肪代谢和免疫功能.

Vitamin A (VA) is an important nutrient for weaning piglets. It plays a significant role in the normal formation, development, and maintenance of epithelial cells. Previous studies have shown that VA supplements could improve the host's intestinal barrier function. Therefore, we hypothesized that VA supplements can affect intestinal function in weaned piglets by regulating intestinal stem cells. Thirty-two 21-d-old weaned [(Yorkshire × Landrace) × Duroc] piglets with an average weight of 8.34 ± 0.13 kg were randomly divided into 4 treatment groups, with 1) 2 mg/kg (control), 2) 4 mg/kg, 3) 8 mg/kg, and 4) 16 mg/kg doses of VA, respectively. The experiment lasted for 14 d. Weaned piglets were given ad libitum access to food and water during the test. The ADG (linear, P = 0.020) and G:F (linear, P = 0.005) of the piglets were found to increase significantly from days 8 to 14. The Lgr5+ gene expression (P = 0.012) in the jejunum mucosa of the 16 mg/kg VA group was increased. The jejunum villus height (P = 0.027) and villi surface area (P = 0.035) were significantly increased in the 4 mg/kg VA treatment group. The crypt depth increased significantly in the 4 and 8 mg/kg VA treatment groups (quadratic, P = 0.043), and the ratios of villus height to crypt depth significantly increased in the 16 mg/kg VA group (quadratic, P = 0.015). The maltase (P = 0.032), sucrose (P = 0.041), and alkaline phosphatase activity (linear, P = 0.024) were significantly increased when further supplemented with 4 mg/kg VA. Slc2a2 mRNA abundance was significantly increased in the 2 mg/kg VA group (linear, P = 0.024). Moreover, the budding rates, buddings number per organoid, and Chromogranin A and Muc2 expression of piglet intestinal organoids were significantly reduced (P < 0.05) by VA and its metabolites (retinoic acid). Compared with the control group, the expression of Spp1 and Trop2 increased. These results indicated that VA may increase the stemness of intestinal stem cell in vitro. This study suggested that VA could affect growth performance and intestinal function by regulating intestinal stem cells in the jejunum of weaned piglets. © 2020 The Author(s) 2020. 摘要:
Glycerol-lactate esters are energy supplements for exercise, but effects of trilactic glyceride (TLG) on intestinal function and hepatic metabolism are unknown. We found that dietary supplementation with 0.5% TLG to weanling piglets decreased plasma concentrations of low-density lipoprotein and gamma-glutamyl transferase but increased those of D-xylose and high-density lipoprotein. TLG supplementation enhanced mRNA levels for fatty acid synthase (FASN) and SLC27A2 in white adipose tissue; insulin receptor in duodenum; aquaporin-8 in ileum, jejunum and colon; aquaporin-10 in duodenum and ileum; nuclear factor like-2 in jejunum and colon; glutathione S-transferase and phosphoenolpyruvate carboxykinase-1 in intestines; and abundances of claudin-1 and occludin proteins. TLG supplementation decreased mRNA levels for: hepatic hormone-sensitive lipase E, lipoprotein lipase, FASN, insulin-like growth factor-binding protein-3, and SLC27A2; and intestinal lipoprotein lipase, FASN and NADPH oxidase. Furthermore, TLG supplementation enhanced abundances of genus Bifidobacterium, while reducing abundances of family Enterobacteriaceae in ileum, colon and cecum; jejunal caspase-3 protein and diarrhea rate. In conclusion, dietary supplementation with TLG modulated lipid metabolism and alleviated diarrhea by improving intestinal function and regulating intestinal microflora in piglets. © 2020 Frontiers in Bioscience. <jats:title>Abstract</jats:title><jats:p>The effect of holly polyphenols (HP) on intestinal inflammation and microbiota composition was evaluated in a piglet model of lipopolysaccharide (LPS)-induced intestinal injury. A total of twenty-four piglets were used in a 2 × 2 factorial design including diet type and LPS challenge. After 16 d of feeding with a basal diet supplemented with or without 250 mg/kg HP, pigs were challenged with LPS (100 μg/kg body weight) or an equal volume of saline for 4 h, followed by analysis of disaccharidase activities, gene expression levels of several representative tight junction proteins and inflammatory mediators, the SCFA concentrations and microbiota composition in intestinal contents as well as proinflammatory cytokine levels in plasma. Our results indicated that HP enhanced intestinal disaccharidase activities and reduced plasma proinflammatory cytokines including TNF-<jats:italic>α</jats:italic> and IL-6 in LPS-challenged piglets. Moreover, HP up-regulated mRNA expression of intestinal tight junction proteins such as claudin-1 and occludin. In addition, bacterial 16S rRNA gene sequencing showed that HP altered hindgut microbiota composition by enriching <jats:italic>Prevotella</jats:italic> and enhancing SCFA production following LPS challenge. These results collectively suggest that HP is capable of alleviating LPS-triggered intestinal injury by improving intestinal disaccharidase activities, barrier function and SCFA production, while reducing intestinal inflammation.</jats:p>

试验旨在研究谷氨酰胺(Gln)对灌服大豆分离蛋白(SPI)仔猪肝脏和肌肉抗氧化功能及肝脏形态结构的影响.将24头7日龄健康仔猪(杜×长×大)随机分为3组:对照组、SPI组、SPI+Gln组,每组8个重复,每个重复1头猪.试验第1~5 d各组仔猪均饲喂人工乳,试验第6~7 d SPI组和SPI+Gln组每头仔猪饲喂含大豆分离蛋白(10 g/kg BW)的人工乳,对照组饲喂含酪蛋白(7.8 g/kg BW)的人工乳.试验第8~10 d SPI + Gln组灌服Gln(250 mg/kg BW),其余2组灌服丙氨酸(305 mg/kg BW).试验第11 d屠宰仔猪,取肝脏、腓肠肌、背最长肌组织样品,检测抗氧化酶活性和氧化产物的含量,并制作苏木素-伊红染色肝脏切片.结果表明,SPI可诱导肝脏脂肪变性、细胞空泡化和细胞排列无序,并提高了仔猪肝脏MDA(P<0.05)、腓肠肌H2O2及背最长肌MDA的含量,但显著降低了腓肠肌、背最长肌CAT和GSH-Px活性.Gln处理缓解了SPI仔猪肝细胞形态的异常与脂肪变性,提高了肝脏GSH-Px活性,并显著降低了SPI仔猪肝脏MDA和腓肠肌的H2O2的含量(P<0.05).因此,SPI诱导仔猪肝脏形态结构异常、肝脏和肌肉抗氧化能力下降,而Gln可有效缓解肝细胞损伤,提高仔猪肝脏与腓肠肌抗氧化能力.

本试验旨在研究复合氨基酸对断奶仔猪背最长肌生长的影响。选取16头体重(5.49±0.35)kg、健康的24日龄杜×长×大断奶仔猪随机分成对照组和复合氨基酸组,每组8个重复,每个重复1头猪。对照组饲喂基础日粮+0.99%的丙氨酸(等氮控制),复合氨基酸组饲喂基础日粮+1.0%的复合氨基酸(谷氨酸:谷氨酰胺:甘氨酸:精氨酸:N-乙酰半胱氨酸=5:2:2:1:0.5)。试验期为21 d,于试验第21天麻醉、屠宰取样。结果表明:与对照组相比,复合氨基酸组提高了仔猪背最长肌蛋白质含量和蛋白质/DNA(P<0.05),降低了RNA/DNA(P<0.05);提高了背最长肌缬氨酸、组氨酸、3-甲基组氨酸和精氨酸的含量(P<0.05),降低了背最长肌苏氨酸、谷氨酰胺、β-丙氨酸、赖氨酸和氧化型谷胱甘肽(GSSG)的含量(P<0.05);提高了背最长肌PepT1和b0,+AT基因的相对表达量(P<0.05),降低了MSTN基因的相对表达量(P<0.05)。结果表明添加复合氨基酸对断奶仔猪背最长肌的生长有一定的促进作用。

为了研究肉桂油对断奶仔猪肝脏的激素水平、抗氧化指标和相关基因相对表达量的影响,试验选取16头仔猪(杜×长×大)随机分为对照组和肉桂油组,每组8头,对照组饲喂基础日粮,肉桂油组在基础日粮中添加50 mg/kg的肉桂油,于试验第21天屠宰取样,测定和分析仔猪肝脏部分激素[类胰岛素生长因子1(IGF-1)、前列腺素E2(PGE2)]、炎性细胞因子[白介素1β(IL-13)]、抗氧化指标[谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、髓过氧化物酶(MPO)和总超氧化物歧化酶(T-SOD)]的含量和mTOR、EGFR信号通路相关基因[哺乳动物雷帕霉素靶蛋白(mTOR)、起始因子4E结合蛋白(4E-BP1)、70 ku核糖体蛋白S6激酶(P70S6K)、细胞外信号调节激酶(ERK)、丝氨酸/苏氨酸蛋白激酶(Akt)]、炎性和免疫相关基因[趋化因子9(CXCL9)、趋化因子11(CXCL11)、热休克105/110蛋白1(HSPH1)、干扰素α(IFN-α)、鼠黏液/流感病毒抵抗基因1(MX1)、鼠黏液/流感病毒抵抗基因2(MX2)、再生胰岛衍生蛋白3γ (REG3G)]及细胞凋亡相关基因[促凋亡基因(Bax)、低氧诱导因子-1(HIF-1)]的mRNA表达水平.结果 表明:与对照组比较,肉桂油组显著提高了断奶仔猪肝脏中IL-1β水平(P<0.05),显著降低了肝脏PGE2水平(P<0.05);显著提高了仔猪肝脏GSH-Px活性(P<0.05),显著降低了MDA含量(P<0.05);显著上调了仔猪肝脏mTOR、4 E-BP1、ERK、Bax、CXCL9、MX1、REG3G基因的mRNA相对表达量(P<0.05),显著下调了Akt、CXCL11、HSPH1、IFN-α、MX2和HIF-1基因的mRNA相对表达量(P<0.05).说明日粮中添加肉桂油可以调节仔猪肝脏PGE2和IL-1β水平,提高肝脏抗氧化能力,对断奶仔猪肝脏相关基因相对表达量有一定的调控作用.

Porcine epidemic diarrhea virus (PEDV) has caused enormous economic losses to the swine industry worldwide in recent years. Puerarin (PR), a major isoflavonoid isolated from the Chinese herb Gegen, possesses many pharmacological activities, including anti-inflammatory, and anti-viral activities. This study was conducted with both PEDV-infected African green monkey kidney cells (Vero) and neonatal pigs to determine the effect of PR on PEDV infection and to elucidate the underlying mechanisms by using proteomic analyses. Twenty-four piglets fed a milk replacer were randomly allocated into one of three groups (Control, PEDV, and PEDV + PR). After a 5-day period of adaption, piglets (n = 8/group) in the PEDV + PR were orally administered with PR (0.5 mg/kg body weight) between days 5 and 9, whereas piglets in the other two groups received the same volume of liquid milk replacer. On day 9, piglets were orally administered with either sterile saline or PEDV (Yunnan province strain) at 104.5 TCID50 (50% tissue culture infectious dose) per pig. On day 12 of the trial, jugular vein blood and intestinal samples were collected. In addition, Vero cells were assigned randomly into three groups (Control, PEDV, PEDV + PR). Cells in the PEDV and PEDV + PR groups were infected with PEDV at a multiplicity of infection of 0.01, while cells in the control group were treated with the same volume of sterile saline. One hour later, cells in the Control and PEDV groups were cultured in serum-free DMEM, while cells in the PEDV + PR group were supplemented with PR. After 36 h of culture, cells were harvested. PR attenuated the reductions in cell proliferation in vitro and growth performance in PEDV-infected piglets, and inhibited PEDV replication and the expression of several cytokines (including IL-8) both in vitro and in vivo. Proteomic analyses identified that the abundances of 29 proteins in the ileum were altered by PEDV infection and restored to the control level by PR. Pathway analyses revealed that PR restored the expression of several interferon-stimulated genes and selectively upregulated the expression of guanylate-binding proteins. Western blot analyses showed that PR supplementation inhibited the PEDV-induced NF-κB activation. Collectively, these results indicate that PR could exert antiviral and anti-inflammatory effects in piglets infected with PEDV and have the potential to be an effective antiviral feed additive. © Copyright © 2020 Wu, Zhang, Yi, Wu, Chen, Guo, Li, Ji, Wang, Zhao, Hou and Wu.

摘要:
为研究复合植物精油(OCT)对脂多糖(LPS)刺激断奶仔猪肠道结构和抗氧化能力的影响,本试验选取28日龄左右健康的杜×长×大三元杂交仔猪24头,按照体重相近原则随机分为3个处理组:对照组、LPS组和OCT组.对照组和LPS组仔猪饲喂基础日粮,OCT组仔猪饲喂基础日粮+50 mg/kg OCT.试验期21 d.于试验第21天,LPS组和OCT组仔猪注射LPS(100μg/(kg·BW)),对照组仔猪注射等量生理盐水.LPS或生理盐水注射后3 h采血;6 h后,屠宰全部仔猪取肠道样品测定有关指标.结果表明,与对照组相比,LPS组仔猪空肠绒毛高度/隐窝深度有降低趋势(P>0.05),回肠绒毛高度及绒毛高度/隐窝深度显著降低(P<0.05);血浆过氧化氢酶(CAT)活性、血浆和空肠超氧化物歧化酶(SOD)活性显著降低(P<0.05),空肠诱导型一氧化氮合酶(iNOS)活性、血浆和回肠H 2 O2含量显著上升(P<0.05).与LPS组相比,OCT组仔猪空肠绒毛高度、空肠绒毛高度/隐窝深度和回肠绒毛高度显著提高(P<0.05),回肠绒毛高度/隐窝深度有提高趋势(P>0.05);血浆CAT、空肠SOD活性显著提高(P<0.05),血浆和回肠H 2 O2含量显著下降(P<0.05).以上结果表明,日粮中添加50 mg/kg OCT可以在一定程度上缓解由LPS刺激引起的仔猪氧化应激,改善仔猪肠道结构.

本试验旨在研究表达Ⅰ型耐热肠毒素(STa)的重组大肠杆菌诱发7日龄仔猪小肠炎症反应.选取24头7日龄健康仔猪,随机分为4个处理组:对照组(人工乳)、STa组(人工乳+2×109 CFU重组菌LMG194-pBAD-STa)、LMG194组(人工乳+2×109 CFU大肠杆菌LMG194)和K88组(人工乳+2×109 CFU大肠杆菌K88),每组6头.试验第5天进行攻毒,第7天屠宰取样,观察空肠组织形态学变化,并检测血清中炎性细胞因子含量及空肠免疫相关基因的相对表达量.结果显示,与对照组相比,S T a与K88组仔猪空肠绒毛明显萎缩变短,有明显损伤甚至脱落;STa、LMG194及K88组血清中IL-10、IL-8和IL-6浓度均显著升高(P<0.05),LMG194和K88组血液和肠道iFABP浓度均显著降低(P<0.05),STa和K88组血清中TNF-α浓度显著降低、NF-κB浓度显著升高,LMG194组NF-κB浓度显著降低、TNF-α浓度显著升高(P<0.05);STa和LMG194组CCL2和CXCL9基因表达水平显著上调(P<0.05),V N N1基因表达水平显著下调(P<0.05),STa和LMG194组IFN-γ组基因水平分别显著上升和下降(P<0.05);K88组CXCL9和IFN-γ基因表达水平显著下调(P<0.05),VNN1基因表达水平显著上调(P>0.05),CCL2基因表达水平无明显差异(P>0.05).以上结果表明,表达Ⅰ型耐热肠毒素STa的重组大肠杆菌几乎具有与K 88一样的毒性,可导致7日龄仔猪小肠黏膜受损,诱发严重的炎症反应,导致仔猪腹泻,可作为仔猪腹泻模型的候选菌株.

肠道不仅是所有营养物质消化吸收的最终场所,也是动物体内最大的免疫器官,是机体防御体系的第一道屏障,维护动物肠道健康对保障养猪生产至关重要。肠道是机体应激反应的中心器官,各种应激因素致使肠道结构损伤与功能紊乱,从而导致仔猪健康不良与生长受阻。因此,建立稳定可靠的肠道损伤模型是研究仔猪肠道损伤机制及营养干预的关键,本文主要介绍了本实验室成功建立的几种仔猪肠道损伤模型,筛选了几种肠道功能紊乱的标志性分子,为猪和人类的肠道功能障碍研究及营养干预提供参考。

摘要:
试验研究了日粮中添加凝结芽孢杆菌对仔猪血浆抗氧化指标和肠黏膜生长的影响。选取30头体重相近的21日龄健康断奶仔猪随机分为3组(对照组、试验Ⅰ组和试验Ⅱ组),每组10个重复,每个重复1头猪。对照组饲喂基础日粮,试验Ⅰ、Ⅱ组在基础日粮的基础上分别添加2.0×10^6、2.0×10^7 CFU/g的凝结芽孢杆菌。正试期21 d,试验结束时前腔静脉采血后屠宰取样。结果显示:与对照组相比,试验Ⅰ和Ⅱ组断奶仔猪血浆超氧化物歧化酶(SOD)活性、空肠和回肠绒毛高度与隐窝深度的比值、回肠总蛋白(TP)含量及RNA与DNA的比值均显著提高(P<0.05);试验Ⅱ组仔猪血浆丙二醛(MDA)含量显著降低(P<0.05);试验Ⅰ组空肠TP含量及TP/DNA显著提高(P<0.05)。在本试验条件下,日粮中添加凝结芽孢杆菌可以提高断奶仔猪的抗氧化能力,促进仔猪肠道黏膜细胞的增殖与更新,从而改善了肠道黏膜形态结构。

为了研究日粮中添加凝结芽抱杆菌对断奶仔猪肠道抗氧化和抗病毒能力的影响,试验选取24头 21 日龄的健康断奶仔猪(杜×长×大),随机分为 3组(对照组、BC6 组、BC7 组),每组 8个重复,对照组饲喂基础日粮,BC6组饲喂基础日粮+凝结芽抱杆菌(2×106 cfu/g饲料),BC7组饲喂基础日粮+凝结芽抱杆菌(2×107 cfu/g饲料),于试验第 21天屠宰取样,检测空肠都膜组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)的活力和丙二酶(MDA)、过氧化氢(H202)含量,以及空肠炎性抗病毒和免疫调节相关基因[热休龙蛋白 1(HSPH1)、趋化因子(CCL2)、干扰素-α(IFN-α)、干扰素-?(IFN-?)、白细胞介素-4(IL-4)、抗氧化应激转录因子 2(Nrf-2)基因]及相关蛋白 [2'-5'寡聚腺普酸合成酶 1(OAS1)、抗黏液病毒 1(MX1)]的相对表达量.结果表明:与对照组相比,BC6组空肠MDA、H202的含量显著降低(P<0.05),HSPH1.,IFN-?基因的相对表达量显著下调(P<0.05),0ASl蛋白相对表达量显著升高(P<0.05);BC7组空肠 H202的含量显著降低(P<0.05),SOD和 CAT活力显著升高(P<0.05),IL-4、HSPHl和 Nrf-2基因的相对表达量显著下调(P<0.05),IFN-?基因的相对表达量显著上调(P<0.05),MX1蛋白相对表达量显著升高(P<0.05).说明日粮中添加凝结芽抱杆菌可以提高断奶仔猪肠道抗氧化能力和抗病毒能力,促进仔猪肠道吸收,从而维护肠道稳态,且日粮中添加 2×107cfu/g的凝结芽抱杆菌效果更好.

本研究旨在探究日粮中添加凝结芽孢杆菌(BC)对断奶仔猪小肠功能的影响。试验选取平均体重为(6.5±0.21) kg的21日龄三元杂交(杜×长×大)断奶仔猪24头,随机分为3组,每组8个重复,每个重复1头猪。对照组饲喂基础日粮,BCⅠ、BCⅡ组分别在基础日粮中添加2×106、2×107 CFU/g凝结芽孢杆菌。于试验第21天,所有仔猪灌服D-木糖(0.1 g/(kg·BW)),1 h后采血,屠宰,取回肠黏膜样品及回肠、结肠食糜。结果表明:与对照组相比,BCⅠ、BCⅡ组血浆二胺氧化酶(DAO)活力显著降低(P<0.05),BCⅠ组血浆D-木糖含量显著增加(P<0.05);BCⅠ、BCⅡ组回肠谷胱甘肽过氧化物酶(GSH-Px)的活力显著增加(P<0.05),BCⅡ组回肠谷胱甘肽(GSH)含量增加(P>0.05),BCⅠ组回肠丙二醛(MDA)含量显著降低(P<0.05);BCⅠ组回肠黏膜水通道蛋白3(AQP3)、碱性氨基酸转运载体(b0,+AT)、钠葡萄糖共转运载体1(SGLT-1)的mRNA表达量均显著增加(P<0.05),BCⅡ组回肠黏膜AQP3、AQP10、内向整流钾离子通道13(KCNJ13)、b0,+AT、SGLT-1的mRNA表达量均显著增加(P<0.05);BCⅠ组回肠及结肠肠球菌、双歧杆菌数量均显著升高(P<0.05),BCⅡ组回肠肠球菌及结肠肠球菌及双歧杆菌数量均显著升高(P<0.05)。综合试验结果,日粮中添加凝结芽孢杆菌可提高断奶仔猪小肠道吸收转运能力及抗氧化能力,改善断奶仔猪小肠内菌群结构。

The intestinal microbiota plays a vital role in metabolism, pathogen resistance, and immune development in host cells, and is modifiable by dietary change. Lentinan (LNT), a type of mushroom polysaccharide, is known to ameliorate intestinal inflammation with the potential of therapeutic effect on digestive diseases. We hypothesized that LNT could alleviate Escherichia coli lipopolysaccharide (LPS)-induced intestinal injury via regulating the composition and metabolites of intestinal microbiota in a piglet model. Twenty-four weaned piglets were used in a 2 × 2 factorial design, and the main factors included a dietary treatment (basal or LNT diet) and immunological challenge (LPS or saline). After feeding basal or LNT diet for 21 days, pigs were injected with LPS or saline. At 4 h post-injection, pigs were killed and jejunum, ileum and cecal digesta were collected. LNT improved intestinal morphology and barrier function. LNT also inhibited inflammatory signaling pathways (toll-like receptor 4 and nucleotide binding oligomerization domain protein) and pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-1β and interleukin-6) expression, as well as up-regulated the heat shock protein 70 expression in small intestine. In addition, LNT enhanced the concentrations of propionate, butyrate, isobutyrate and isovalerate in cecal digesta, resulting in a significant increase in histone acetylation without affecting the protein level of G protein-coupled receptor 41 (GPR41), a short chain fatty acid receptor. Bacterial 16S rRNA gene pyrosequencing showed that LNT had a great impact on gut microbiota composition at different taxonomic levels. Moreover, the correlation analysis revealed some potential relationships between cecal metabolites and certain intestinal microbiota. These results indicate that LNT promotes intestinal health, in part, through altering intestinal microbiota composition and increasing the short chain fatty acid synthesis, which subsequently lead to a reduction in inflammation and hyper-acetylation of histones. © 2019 The Royal Society of Chemistry.

Trihexanoin is a short-chain triglyceride (SCT). Many studies have reported that SCTs play important roles in the maintenance of intestinal epithelial structure and function. The present work was to investigate the effects of trihexanoin on growth performance, carbohydrate and fat metabolism, as well as intestinal morphology and function in weaned piglets. Twenty weaned piglets (21 ± 2 d) were randomly allocated to one of two treatment groups: The control group (basal diet supplemented with 0.5% soya oil); the TH group (basal diet supplemented with 0.5% trihexanoin). Dietary trihexanoin supplementation significantly reduced diarrhea rate; increased the concentrations of LDL, HDL and total protein in plasma; decreased cholesterol concentrations and glutamyl transpeptidase activity in plasma; improved intestinal morphologic structure; altered the mRNA levels and abundances of proteins related to glycogen and fat metabolism, mucosal barrier function, antioxidant capacity and water transport capacity; and altered the community of intestinal microflora. These results indicate that dietary trihexanoin supplementation could reduce diarrhea, regulate carbohydrate and fat metabolism, exert beneficial effects on the intestinal mucosal barrier, protect the intestinal mucosa from injuries, improve intestinal transport and absorption, and enhance antioxidant capacity. The aim of the present study was to construct a recombinant strain by using the probiotics Lactobacillus acidophilus expressing the epidermal growth factor (EGF), which can improve intestinal morphology, and to test its effect on the intestinal histology in LPS (lipopolysaccharide)-challenged mice. The EGF gene was cloned into the shuttle vector pSET4s and then transformed into the L. acidophilus MG6243 strain by electroporation. Two recombinant strains were confirmed by SDS-PAGE and named ΔMG6243-1 and ΔMG6243-2. Sixty mice were randomly assigned to six treatments: 1) non-challenged control, 2) LPS-challenged control, 3) LPS-challenged + 200 μl MG6243, 4) LPS-challenged + 200 μl ΔMG6243-1, 5) LPS-challenged + 200 μl ΔMG6243-1 + inducer IPTG, and 6) LPS-challenged + 200 μl ΔMG6243-2. There were ten replicates for each treatment, and each replicate had one mouse. On d 15, the LPS-challenged mice were injected intraperitoneally with LPS at 10 mg/kg BW, while the non-challenged mice were injected with 0.9% NaCl solution. Three hours after the challenge, all mice were killed. Results showed that both ΔMG6243-1 and ΔMG6243-2 could increase villus height in the duodenum, ileum and jejunum (P < 0.05). In addition, the recombinant L. acidophilus decreased the crypt depth in the duodenum and ileum (P < 0.05). These results indicated that the recombinant L. acidophilus could effectively improve the intestinal histology in LPS-challenged mice. © 2018 Chulalongkorn University Printing House.

This experiment aimed to investigate the effects of a symbiotic, consisting of microencapsulated Lactobacillus plantarum (MLP) and fructooligosaccharide (FOS) on growth performance, blood immune parameters, and intestinal microbiota in weaned piglets. Ninety weaned piglets were assigned to three dietary groups (CON: basal diet; ANT: basal diet + aureomycin; SYN: basal diet + MLP and FOS) for a four-week trial. Compared to CON, pigs in the SYN group had higher weight gain and feed intake, and lower diarrhea rate (P < .05). Also, pigs in the SYN group had improved plasma IgA and IgG concentrations, and increased (P < .05) numbers of lactic acid bacteria in the colon compared to CON. In conclusion, feeding a synbiotic based on MLP and FOS had beneficial effects on growth performance, plasma immune parameters, and intestinal microbiota, indicating the potential of it to serve as an alternative to feed antibiotics in weaned pig diets. © 2017 The Author(s).
