摘要:
Glutathione (GSH) is an important intracellular antioxidant. It is closely related to the homeostasis of the intracellular redox state which can be changed through deoxynivalenol (DON) exposure. More importantly, GSH deficiency can directly lead to apoptosis and cellular metabolic disorders mediated by intracellular redox homeostasis. Therefore, real-time monitoring of dynamic changes of GSH levels in living cells is urgently needed for the evaluation of intracellular oxidative stress. Herein, for the first time, an aptamer-based recognition composite nanoprobe (Apt-AuNCs-MoS2) was successfully constructed. The nanoprobe was employed for real-time and in situ imaging of DON-induced intracellular GSH changes and evaluation of oxidative stress status. The results indicated that the aptasensor revealed excellent specificity and high sensitivity to GSH in the solution system, and the limit of detection was as low as 35 nM. The results about DON-mediated oxidative stress indicated that the oxidative stress in living cells tends to be saturated with more than 20 ppm DON concentration. Thus, the strategy provides a novel aptasensor for the determination of GSH, offers a promising platform for fluorescence imaging of intracellular GSH in living cells, and further enriches in situ analysis of the in vitro toxicology.

In this study, we investigated and compared the oil yield, physicochemical properties, fatty acid composition, nutrient content, and antioxidant ability of Xanthoceras sorbifolia Bunge (X. sorbifolia) kernel oils obtained by cold-pressing (CP), hexane extraction (HE), aqueous enzymatic extraction (AEE), and supercritical fluid extraction (SFE). The results indicated that X. sorbifolia oil contained a high percentage of monounsaturated fatty acids (49.31–50.38%), especially oleic acid (30.73–30.98%) and nervonic acid (2.73–3.09%) and that the extraction methods had little effect on the composition and content of fatty acids. X. sorbifolia oil is an excellent source of nervonic acid. Additionally, the HE method resulted in the highest oil yield (98.04%), oxidation stability index (9.20 h), tocopherol content (530.15 mg/kg) and sterol content (2104.07 mg/kg). The DPPH scavenging activity rates of the oil produced by SFE was the highest. Considering the health and nutritional value of oils, HE is a promising method for X. sorbifolia oil processing. According to multiple linear regression analysis, the antioxidant capacity of the oil was negatively correlated with sterol and stearic acid content and positively correlated with linoleic acid, arachidic acid and polyunsaturated fatty acid content. This information is important for improving the nutritional value and industrial production of X. sorbifolia.

Janus transitional metal dichalcogenides (TMDCs) have raised wide attraction because of its unique physical and chemical properties. This work focuses on the effect of single surface vacancy (Se and Te vacancy) on adsorption and sensing properties of Janus HfSeTe monolayer to several hazardous gases (CO, H2S, NH3, NO, NO2, and SO2). The adsorption structures, adsorption energy, charge transfer, electronic properties and work function are calculated using density functional theory. It is found that different surface vacancy (Se or Te vacancy) brings different adsorption behavior, and the introduction of vacancy significantly enhances the chemical interactions between Janus HfSeTe monolayer and adsorbed gases. The adsorption energy and charge transfer obviously increase for all these molecules compared to the adsorption on pristine HfSeTe monolayer. More notably, the adsorption of CO, NO, NO2 and SO2 brings new chemical bond formation, and the atomic density of states (DOS) suggests that there exist states overlapping between dangling Hf atom and atom in molecule. In addition, H2S and NH3 adsorptions make the work function decrease while other adsorptions lead to the risen of work function. Based on the distinguished adsorption properties and different degree of work function variation, Janus HfSeTe is a potential candidate in hazardous gas scavenger and sensing material.

Aging plays a significant role in the occurrence and development of idiopathic pulmonary fibrosis (IPF). In this study, we aimed to identify and verify potential aging-associated genes involved in IPF using bioinformatic analysis. The mRNA expression profile dataset GSE150910 available in the Gene Expression Omnibus (GEO) database and R software were used to identify the differentially expressed aging-related genes involved in IPF. Hub gene expression was validated by other GEO datasets. Gene ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed on differentially expressed aging-related genes. Subsequently, aging-related genes were further screened using three techniques (least absolute shrinkage and selection operator (LASSO) regression, support vector machine, and random forest), and the receiver operating characteristic curves were plotted based on screening results. Finally, real-time quantitative polymerase chain reaction (qRT-PCR) was performed to verify the RNA expression of the six differentially expressed aging-related genes using the blood samples of patients with IPF and healthy individuals. Sixteen differentially expressed aging-related genes were detected, of which the expression of 12 were upregulated and four were downregulated. GO and KEGG enrichment analyses indicated the presence of several enriched terms related to senescence and apoptotic mitochondrial changes. Further screening by LASSO regression, support vector machine, and random forest identified six genes (IGF1, RET, IGFBP2, CDKN2A, JUN, and TFAP2A) that could serve as potential diagnostic biomarkers for IPF. Furthermore, qRT-PCR analysis indicated that among the above-mentioned six aging-related genes, only the expression levels of IGF1, RET, and IGFBP2 in patients with IPF and healthy individuals were consistent with the results of bioinformatic analysis. In conclusion, bioinformatics analysis identified 16 potential aging-related genes associated with IPF, and clinical sample validation suggested that among these, IGF1, RET, and IGFBP2 might play a role in the incidence and prognosis of IPF. Our findings may help understand the pathogenesis of IPF.

The objective of the research was to investigate action mechanism of oxidative stress and cerebral injuries after subarachnoid hemorrhage (SAH) by Ghrelin and angiogenic factor G-patch and FHA domain 1 (Aggf1) and offer new research ideas to SAH clinical treatment and SAH-induced early cerebral injuries. SAH rat models were prepared by prechiasmatic anterior cistern injection. Specific Ghrelin and Aggf1 small interfering ribonucleic acid (siRNA) were designed and injected into silence Ghrelin or Aggf1 in rat left lateral ventricles. Rats were divided randomly into sham-operated (sham), SAH model, negative control siRNA, Ghrelin silence (Ghrelin((-/-))), and Aggf1 silence groups. Changes of rat neurological impairment, encephaledema, cerebral tissue phosphorylated protein kinase (p-Akt), and content changes of caspase-3 protein and oxidative stress indexes were observed, including glutathione (GSH) and oxidized glutathione (GSSG). Results showed scores of neurological impairment and water content in SAH model group were reduced compared with sham group, while p-Akt protein and GSH contents were enhanced. However, caspase-3 protein and GSSG contents were declined, showing statistically meaningful difference (P < 0.05). Compared with SAH model group, scores of neurological impairment, cerebral tissue water content, and caspase-3 protein and GSSG contents in silence Ghrelin and Aggf1 groups were increased, while p-Akt protein and GSH contents were decreased, demonstrating statistically meaningful difference (P < 0.05). To conclude, silence Ghrelin and Aggf1 aggravated early cerebral injuries after SAH, revealing that Ghrelin and Aggf1 could protect brains to some degree.

The effects of pyrolysis temperature on properties and adsorption performance of carbonized bacterial cellulose (CBC) produced from bacterial cellulose at 300, 400, 600 and 800 degrees C were investigated. As pyrolysis temperature increased, the BET surface area, C and ash contents of CBC increased while its mass yield and the contents of H, N and O decreased. Higher pyrolysis temperature resulted in CBC having more aromatic structure and less hydrophilic. The impacts of pyrolysis temperature, solution pH, contact time and initial concentration on the absorption of Cr(VI) onto CBC were systematically studied as well. The results showed that CBC400 prepared at 400 degrees C exhibited the highest Cr(VI) adsorption capacity for Cr(VI) up to 250.0 mg/g. The equilibrium adsorption and adsorption kinetics fitted the Langmuir isotherm and pseudo-second-order kinetic models well. The mechanisms of adsorption of Cr(VI) on CBC included electrostatic interaction, pi-pi interaction and functional groups complexation.

Nowadays, diabetes and obesity are two main health-threatening metabolic disorders in the world, which increase the risk for many chronic diseases. Apelin, a peptide hormone, exerts its effect by binding with angiotensin II protein J receptor (APJ) and is considered to be linked with diabetes and obesity. Apelin and its receptor are widely present in the body and are involved in many physiological processes, such as glucose and lipid metabolism, homeostasis, endocrine response to stress, and angiogenesis. In this review, we summarize the literatures on the role of the Apelin–APJ system in diabetes and obesity for a better understanding of the mechanism and function of apelin and its receptor in the pathophysiology of diseases that may contribute to the development of new therapies.

In this paper, we study the following quasilinear Schrodinger equation -Delta u + V(x)u - Delta(u(2))u = g(x, u), x is an element of R-2, where V(x) and g(x, u) are 1-periodic in x(i)(i = 1, 2). By using a change of variable and a version of Trudinger-Moser inequality, we obtain the existence of nontrivial solutions and infinitely many pairs of geometrically distinct solutions. Coal quality rating can help reduce greenhouse gas emissions, solving the global warming problem. It becomes more important as the carbon neutrality by the mid-21st century agreement is accepted by 195 countries, including China. In this paper, an improved fuzzy comprehensive evaluation method is introduced for coal quality rating. The data used in this work are of the Hostolgoi coalfield of the Xinjiang Province of China. Six industrial analysis indicators are determined as evaluation factors by taking the coal samples of different coal seam depths in the mining area. The super-standard multiple methods and the double-weight super-standard weighting method are combined to form a comprehensive weight. The results show that most of the coal samples of this coal mine are at grades I-II, and the overall coal is with good-quality stability. The evaluation results can improve the coal utilization efficiency and provide scientific guidance for evaluating and exploiting coal resources in coal geological exploration.

Proactive low-carbon consumption behaviors (PLCBs) are crucial to achieving carbon neutrality and identifying motivations for PLCBs is indispensable to changing individual consumption patterns. This study establishes a model by incorporating individual-group-level factors with psychological empowerment perception. The ordinary least-squares regression model was applied to identify the influencing factors of PLCBs with data collected from 1732 urban residents in eastern China. Results show that PLCBs are positively influenced by normative internalization, learning capacity, symbol concern, expertise level, and an environmentalism culture. In particular, the effect of learning capacity is the largest, with an influence coefficient of 0.271. A negative impact is observed between the consumerism culture and PLCBs. Moreover, psychological empowerment perception partly medicated the association between individual-group-level factors and PLCBs, and the maximum ratio of mediating effect to the full impact is 62.64%. The study sheds light on low-carbon-related behavioral management, and recommendations to promote PLCBs are further proposed.

In this work, the Chebyshev collocation scheme is extended for the Volterra integro-differential equations of pantograph type. First, we construct the operational matrices of pantograph and derivative based on Chebyshev polynomials. Also, the obtained operational matrices are utilized to approximate the derivatives of unknown functions. Furthermore, a detailed analysis of convergence is discussed in the weighted square norm. We conduct some numerical experiments to verify the high performance of the suggested numerical approach. The results show that the computational scheme is accurate.

In order to accurately judge the effective bearing area of forgings and detect the inner cavity of hot shaft forgings, a laser based detection method for inner cavity of hot shaft forgings is proposed. According to the characteristics of temperature field around the cavity, the laser technology is introduced to calculate the phase shift between two diffraction beams in any wavelength, normalized sensitivity, subtracted calculation of the two fringe patterns and detection of the size of the cavity. The experimental results show that the residual absolute value of the method is little different from the adaptive threshold, and the cavity location is accurate, which lays a foundation for improving the forging quality and yield.
