摘要:
探讨饲料中添加酵母细胞壁免疫多糖后对混养池塘中草鱼的免疫功能增强的影响,以商品混养饲料作为对照组,在此商品饲料中添加0.5%酵母细胞壁免疫多糖作为试验组。52天的试验结果表明:试验组抽样草鱼的肝体指数、脾脏指数与对照组相比均无显著性差异;试验组草鱼血清和肝脏匀浆液中谷丙转氨酶的活性分别为(4.50±0.71)、(1.535±0.568)U/mg,显著低于对照组;试验组草鱼脾脏匀浆液总蛋白质含量为(11.04±0.905)mg/ml,免疫球蛋白的含量为(0.42±0.028)mg/dl,均显著高于对照组,血清、肝脏匀浆液中酸性磷酸酶、碱性磷酸酶活性略高于对照组;而试验组草鱼血清和肝脏匀浆液谷草转氨酶略低于对照组,但均无显著差异。

摘要:
研究稀土壳聚糖螯合盐对鲫鱼非特异性免疫力的影响。结果表明:饲喂20天后,3个试验组血清溶菌酶活性较对照组均有不同程度提高,且0.24%组血清溶菌酶显著高于对照组;但在第40、60天时,3个试验组血清溶菌酶活性与对照组相比差异均不显著。在第40天时,3个试验组血清超氧化物歧化酶活性均高于对照组;在第20、60天时,各组血清超氧化物歧化酶活性无明显变化规律,差异均不显著。第20天时,0.24%组鲫鱼脾脏指数、头肾指数均显著高于对照组,0.08%组、0.16%组脾脏指数、头肾指数也均高于对照组;但在第40、60天时,各组脾脏指数、头肾指数无明显变化规律,差异均不显著。

摘要:
The expression of peroxisome proliferator-activated receptor gamma (PPAR gamma) was investigated in the hypothalamic-pituitary-adrenal (HPA) axis of weaned pigs after injection with 100 mu g/kg bodyweight Escherichia coli lipopolysaccharide (LPS) (n = 6) and control pigs injected with sterile saline (n = 6). LPS increased PPAR gamma mRNA and protein expression in the hypothalamus (23.8 and 3.1-fold relative to controls, respectively), pituitary gland (9.2 and 2.0-fold, respectively) and adrenal gland (3.5 and 2.3-fold, respectively) (P < 0.05). LPS also induced an increase in PPAR gamma immunohistochemical staining in the hypothalamus (1.3-fold), adenohypophysis (1.3-fold), adrenal cortex (1.4-fold) and adrenal medulla (1.6-fold) (P < 0.05). Concurrent with up-regulated expression of PPAR gamma, LPS increased the concentrations of plasma corticotrophin-releasing hormone (2.1-fold) and adrenocorticotrophin (1.4-fold) (P < 0.05). LPS also induced elevations of interleukin 6 and tumour necrosis factor alpha mRNA levels in the hypothalamus (4.0 and 3.2-fold, respectively), pituitary gland (20.7 and 5.1-fold, respectively) and adrenal gland (3.9 and 3.3-fold, respectively) (P < 0.05). PPAR gamma may play a role in the regulation of neuroendocrine responses associated with immunological stress in pigs. (C) 2009 Elsevier Ltd. All rights reserved.

谷氨酰胺是条件性必需氨基酸,也是三羧酸循环的中心物质,它在三羧酸循环中可以转化为谷氨酸,并能进一步形成谷氨酰胺。作为谷氨酰胺的前体物质,α-酮戊二酸是胃肠道细胞的主要能量来源,可以促进胃肠道细胞增殖,对维持胃肠道健康有很重要的作用,因此它能成为谷氨酰胺的最佳替代物。α-酮戊二酸将用于人类临床营养,同样可以应用于动物生产上,文章仅就α-酮戊二酸对动物机体肠道黏膜的作用进行了综述。

摘要:
对含非淀粉多糖酶小麦型饲料进行制粒加工,测定饲料还原糖及纤维素的含量,探讨了非淀粉多糖酶在饲料加工过程中对还原糖、纤维素、半纤维素降解的催化作用。试验结果表明,添加非淀粉多糖酶小麦型饲料在制粒过程中,还原糖增加17.76%(P〈0.01),半纤维素下降1.41个百分点(P〈0.01)、纤维素下降0.44个百分点(P〈0.05)。而无酶饲料在制粒过程中还原糖、纤维素及半纤维素均无明显变化。

摘要:
Neonates are at increased risk for inflammatory bowel disease, but effective prevention and treatments are currently limited. This study was conducted with the lipopolysaccharide (LPS)-challenged piglet model to determine the effects of dietary supplementation with α-ketoglutarate (AKG) on the intestinal morphology and function. Eighteen 24-day-old pigs (weaned at 21 days of age) were assigned randomly to control, LPS, and LPS + AKG groups. The piglets in the control and LPS groups were fed a corn-and soybean meal-based diet, whereas the LPS + AKG group was fed the basal diet supplemented with 1% AKG. On days 10, 12, 14, and 16, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16, D-xylose was orally administrated to all pigs at the dose of 0.1 g/kg BW, 2 h after LPS or saline injection, and blood samples were collected 3 h thereafter. Twenty-four hours post-administration of LPS or saline, pigs were killed to obtain intestinal mucosae for analysis. Compared with the control group, LPS challenge reduced (P < 0.05) protein levels, the ratio of villus height to crypt depth, and the ratio of phosphorylated mTOR to total mTOR in duodenal, jejunal, and ileal mucosa. These adverse effects of LPS were attenuated (P > 0.05) by AKG supplementation. Moreover, AKG prevented the LPS-induced increase in intestinal HSP70 expression. Collectively, these novel results indicate that dietary supplementation with 1% AKG activates the mTOR signaling, alleviates the mucosal damage, and improves the absorptive function of the small intestine in LPS-challenged piglets. The findings not only help understand the mode of AKGs actions in the neonatal gut but also have important implications for infant nutrition under inflammatory conditions. © Springer-Verlag 2010.

摘要:
L-type amino acid transporter-1 (LAT1) transports large, branched-chain, aromatic and neutral amino acids. About 64 Duroc Landrace Yorkshire pigs were used to study the effects of dietary crude protein (CP), energy and arginine on LAT1 expression in forebrain. The results showed that LAT1 expression in forebrain was sensitive to different levels of CP, energy and arginine. On the basis of Western blot analysis, a lower level of LAT1 presented in the brain tissues of pigs fed the low dietary CP diet (P < 0.05), a higher level were found in pigs fed the higher CP diet, with moderate to intense staining seen in pigs fed the diet plus 1% arginine. In contrast, pigs fed the control-energy diet had weak LAT1 expression, and those fed the diet supplemented with 1% arginine showed lowest LAT1 expression (P < 0.05). These results showed that LAT1 was highly expressed in the forebrain, and expression of LAT1 was affected by dietary protein, energy and arginine differently. Copyright © The Animal Consortium 2010.

摘要:
Ichthyophthirius multifiliis is an important freshwater teleost pathogen that often leads to significant economic losses to the aquaculture industry. The purpose of this study was to assess the acute toxicity of potassium ferrate(VI) to I. multifiliis theront and the concentration needed to prevent I. multifiliis infestation in goldfish, Carassius auratus. Five hundred theronts were exposed to concentrations of potassium ferrate(VI) in each well of a 96-well microtiter plate and observed for 4 h to determine the acute toxicity. Results showed that the exposure of I. multifiliis theronts to potassium ferrate(VI) at concentrations of 4.80 mg/L or more resulted in 100% mortality by 4 h; the LC50 value was estimated to be 1.71 mg/L Aqueous static renewal 96-h bioassays were carried out to determine the acute toxicity of potassium ferrate(VI) to goldfish. The LC50 value for potassium ferrate(VI) in goldfish was 42.51 mg/L. Goldfish were exposed to 4000 theronts/fish in aerated tap water (a dose previously shown to result in consistent infestation) and treated with a single dose of potassium ferrate(VI) after 30 min contact with theronts. Infection level and prevalence were recorded everyday after exposure. The results revealed that potassium ferrate(VI) at the 4.80 mg/L or more concentrations can significantly reduce not only the number of trophonts on the fin of goldfish on day 3 (P < 0.05), but also the prevalence of ichthyophthiriasis (P<0.05). Potassium ferrate(VI) at a concentration of 4.80 mg/L was considered to be the lowest effective dose to prevent infestation of I. multifillis in goldfish. (C) 2009 Published by Elsevier B.V.

试验以赤子爱胜蚯蚓为原料,对蚓激酶粗提取的工艺条件进行了研究,并对pH、硫酸铵的饱和度等基本工艺参数进行优化.结果表明,蚓激酶粗提取的最佳pH为7.8,第1次离心时,硫酸铵的饱和度为45%,第2次离心时,硫酸铵的饱和度为80%.

复合异位酸对山羊小肠氨基酸流通量和表观消化率的影响

试验研究了日粮添加复合异位酸对山羊小肠氨基酸流通量和表观消化率的影响.采用3×3拉丁方试验设计,选用3头安装有永久性瘤胃瘘管、十二指肠近端瘘管和回肠远端瘘管、平均体重为(25.4±0.75) kg的健康麻城黑山羊为试验动物.I组为空白对照,饲喂基础日粮.II组、III组分别饲喂含0.3%和0.6%复合异位酸(钠盐形式)的试验日粮.采用Co-EDTA标记法测定小肠氨基酸流通量和表观消化率.结果显示:III组缬氨酸和异亮氨酸十二指肠流通量极显著高于对照组(P<0.01),亮氨酸和必需氨基酸显著高于对照组(P<0.05);缬氨酸和丙氨酸的小肠表观消化率显著高于对照组(P<0.05).II组缬氨酸和异亮氨酸十二指肠流通量显著高于对照组(P<0.05).III组异亮氨酸十二指肠流通量显著高于II组(P<0.05).

本文旨在研究N-乙酰半胱氨酸(NAC)对脂多糖(LPS)应激仔猪生长性能及血浆生化指标的影响.选取来源一致、平均体重(11.58±0.26)kg的24头健康仔猪,随机分成2组,每组6个重复,每重复2头仔猪.试验分2个阶段,第1~9天对照组饲喂基础饲粮,试验组饲喂在基础饲粮中添加500mg/kg NAC.试验第10~20天分别将2个组同一重复猪中1头分出单独饲养,于第10天、第13天和第20天,1头仔猪腹膜下注射100mg/kg LPS,另一头仔猪注射等体积生理盐水.结果显示:1)饲粮中添加NAC可以缓解LPS刺激导致的日增重下降(P<0.05);2)饲粮中长时间添加NAC能极显著提高仔猪血浆谷胱甘肽水平(P<0.01),显著提高血浆超氧化物歧化酶活性(P<0.05),显著提高血浆表皮生长因子水平(P<0.05).由此可见,饲粮中添加NAC可缓解LPS应激导致的抑制仔猪生长.

试验研究了过氧化物酶体增殖物激活受体γ(PPARγ)的激动剂和抑制剂对脂多糖(LPS)应激断奶仔猪白细胞分类计数和血液生化指标的影响.选取24头健康断奶仔猪,分为4组,每组6个重复,每个重复1头猪.4个组分别为:对照组;LPS组;罗格列酮(ROS,PPARγ激动剂)+LPS组;双酚A型环氧树脂(BADGE,PPARγ抑制剂)+ROS+LPS组.注射LPS或生理盐水3 h后,前腔静脉采血待测.结果显示:1)ROS缓解了LPS导致的断奶仔猪血液白细胞总数和淋巴细胞数量的降低(P<0.05),BADGE阻断了ROS的缓解作用(P<0.05);2)ROS缓解了LPS导致的断奶仔猪血浆碱性磷酸酶活性的升高(P<0.05)及总蛋白、白蛋白和球蛋白含量的降低(P<0.10),BADGE阻断了ROS的缓解作用(P<0.10).结果表明,ROS在一定程度上缓解LPS应激导致的断奶仔猪白细胞总数和淋巴细胞数量的下降及部分血液生化指标的改变,而BADGE阻断了ROS的缓解作用.

The -93 SNP of MLH1 gene is associated with MLH1 gene methylation in endometrial and colorectal cancers. We undertook luciferase reporter assay and electrophoretic mobility shift assay (EMSA) to test whether the -93 SNP affects the MLH1 gene expression. The luciferase activity for -93A plasmid is significantly lower than -93G plasmid. In EMSA experiments, the -93A and -93G probes have different binding affinity to nuclear proteins of JEG3 cells. Our data indicate that -93 SNP affects MLH1 gene expression by altering protein binding to the promoter of MLH1 gene. © 2009 Elsevier Ltd. All rights reserved.

摘要:
针对近年来我国养殖鱼类体色变异越来越普遍的现象,总结了国内外有关鱼类体色方面的研究成果,分别从鱼体色素细胞的种类、色素体的合成与代谢、体色变化的内在生理调控机制(基因、神经和内分泌调节)以及营养、光照、水温、重金属、饵料等诸多外部因子方面,探讨了鱼类体色的成因以及上述因素对鱼体体色的影响,以期为后续对鱼类体色变异进行合理人工调控的研究和实践提供参考。

摘要:
试验研究罗格列酮(ROS)对脂多糖(LPS)刺激的断奶仔猪肠黏膜结构和抗氧化能力的影响.试验包括3个处理组,对照组注射生理盐水,LPS组注射100 μg/kg体重的LPS,ROS组在注射LPS之前的30 min注射3 mg/kg体重的RPS.注射LPS 3 h后取样.结果表明:ROS提高了十二指肠和回肠DNA的含量(p<O.05);ROS缓解了LPS导致的十二指肠、空肠和回肠隐窝深度的增加(p<0.10)和绒毛高度/隐窝深度的降低(p<0.01);ROS降低了空肠丙二醛含量(P<0.10),提高了十二指肠和空肠SOD活性(P<0.05).结果表明,ROS可能是通过提高肠道抗氧化功能而对LPS刺激的断奶仔猪小肠形态结构的损伤起到一定保护作用.

摘要:
Infected yellow catfish (Pelteobagrus fulvidraco) were sent from Niushan Lake Fishery, Hubei Province, China, to our laboratory for diagnosis. Macroscopic daffodil yellow mold was observed on the heads and fins of the fish and one Mucor species was isolated. Based on the morphological and molecular analysis, the species was identified as Mucor circinelloides. Its optimum growth temperature was 30 °C and it could not grow at 40 °C. The infectivity results showed wound infection could cause 100% cumulative mortalities at all experimental CFU (106, 107 and 108). The cumulative mortalities of the intraperitoneal infection increased along with the sporangiospore concentrations; the highest mortality was 90% with 108 CFU. Histopathological studies showed M. circinelloides could cause a series of pathological changes in the host tissues and they disseminated in different viscera, perhaps by the blood. This is the first report of M. circinelloides infection in yellow catfish. © 2009 Federation of European Microbiological Societies. Published by Blackwell Publishing Ltd.

Experiments were conducted to determine the effects of feeding different starch sources on piglets. Four diets were formulated with maize, brown rice, sticky rice and Hi-Maize 1043 as starch sources, with resistant starch (RS) contents of 2.3%, 0.9%, 0.0%, 20.6%, and amylose and amylopectin ratio of 0.23%, 0.21%, 0.18%, 0.06% respectively. Fifty-six pigs weaned at 28 days of age were randomly assigned to one of the four diets. In Exp. 1, six piglets in each group were fitted with an indwelling jugular catheter. After 25 days of feeding trial, venous blood samples were obtained at time zero and every 1 h for 4 h. In Exp. 2, the remaining piglets were used to determine the effects of different starch sources on the fractional synthesis rate (FSR).The results indicated that feeding the Hi-Maize 1043 diet decreased (p < 0.05) plasma contents of glucose, insulin, lactic acid and T 3 , while sticky rice increased plasma contents of glucose and insulin. The insulin contents in piglets fed the sticky rice diet was 69.2 μIU/ml at 1 h post-feeding which was highest among the starch diets. The FSR in the pancreas, spleen, duodenum, jejunum, ileum and colon in the corn group were much higher (p < 0.05) than that in the sticky rice group. These results suggest that RS is potentially beneficial for improving insulin sensitivity in young pigs and that the ratio of amylose and amylopectin have significantly effects on the FSR in splanchnic tissues in weaned piglets. Another finding of this study indicated maize with a ratio of amylose and amylopectin of 0.23 has the best starch sources for pig production. © 2009 The Authors.
