摘要:
Tribonema biomass is considered promising biorefinery feedstock for the co-production of biodiesel and valuable bioproducts; however, the extraction of these useful compounds produces large amounts of algal residues, which produce increased environmental concerns. Herein, cellulose was extracted from the waste residue of T. utriculosum via alkalization and bleaching, followed by the production of high-value-added cellulose nanocrystals (CNCs) via acid hydrolysis. The hydrolysis was performed with 60% (wt%) H2SO4 at a yield of 13.31%, resulting in the generation of rod-shaped nanoparticles averaging 39.5 nm in diameter and 239.2 nm in length. The structural characterization analysis revealed that the prepared CNCs had high crystallinity (73.0%) due to the removal of non-cellulose components and amorphous regions by chemical treatment, as well as possessing good aqueous suspension stability (zeta potential = – 40.1 mV). Although the CNCs showed lower thermal stability than extracted cellulose, they spanned a broader temperature range due to two-stage degradation behaviour, with higher residue weight (16.7%). This work represents the first report on the preparation of a high-value-added industrial product, CNCs, from the filamentous microalga T. utriculosum, aiming to maximize benefits from waste algal residue reutilization. 摘要:
Some studies have revealed thyrotoxicity of phthalates; however, associations of phthalate exposure with papillary thyroid cancer (PTC) remain unclear. We conducted a pair-matching case-control study of 111 PTC cases and 111 age- and sex-matched non-PTC controls to examine associations between urinary concentrations of phthalate metabolites and PTC. Phthalate metabolites were determined in fasting urine specimens by ultra-performance liquid chromatography – tandem mass spectrometry (UPLC-MS/MS). After adjusting for potential confounders and other phthalate metabolites, the concentrations of the sum of di (2-ethylhexly) phthalate (DEHP) metabolites in urine were positively associated with PTC [odds ratio (OR) = 5.35; 95% confidence interval (CI): 1.61–17.83], suggesting the effect of phthalates exposure on PTC development. The findings require confirmation. © 2019 Elsevier Ltd

The binding mode of 5-((4-((4-chlorophenoxy)methyl)-5-iodo-1H-1,2,3-triazol-1-yl)methyl)-2-methylpyrimidin-4-amine (PA-1) was studied using a combination of X-ray crystallography, DFT calculation, and molecular docking approaches. The crystal structure of PA-1 primarily indicated the formation of intermolecular hydrogen bonding and halogen bonding between N-H center dot center dot center dot N and C-I center dot center dot center dot N. DFT calculation demonstrated that the geometric structure of PA-1 is in excellent agreement with the crystal structure. The frontier molecular orbital analysis indicated the important role of the 5-iodo-1,2,3-triazole and benzene ring in the biological activities of PA-1. Further molecular electrostatic potential surface analysis confirmed the possibility of iodine atom participating in the halogen bonding with the negatively charged center; this was validated by the molecular docking study, which showed that the iodine atom of PA-1 could form a halogen bonding with the O atom of Asp521 in the active site of E. coli. PDHc-E1. These findings demonstrate that halogen bonding interaction could be used for the further optimization of PA-1 to discover more potent PDHc-E1 inhibitors and antifungal compounds. Monascin, a specific type of monascus pigments, exhibits many bioactivities. In this study, the antioxidative activity of monascin was investigated by theoretical and experimental methods. First, the antioxidant potential of six monascus pigments was predicted by density functional theory (DFT) and time-dependent density functional theory (TDDFT) at the B3LYP/6-31+G (d, p) level, and monascus yellow pigments were predicted to have strong antioxidant capacity, as they can transfer hydrogen to free radicals and accept electrons from radicals. Then, the free radical-scavenging capacity of monascin for 2,2-diphenyl-1-picrylhydrazyl (DPPH), superoxide, and hydroxyl radicals was experimentally validated by electron spin resonance (ESR) measurement. Monascin exhibited a quenching effect on DPPH, superoxide, and hydroxyl radicals in a dose-dependent manner. Specifically, the scavenging activity of monascin for DPPH, superoxide, and hydroxyl radicals was 97.5%, 59.5%, and 68.6%, respectively, when 0.1 mg mL-1 monascin was present. Our study provides theoretical evidence for the strong antioxidative activity of monascin and offers a simple and reliable strategy to determine the antioxidative activity. © 2020 The Royal Society of Chemistry.

In order to further determine the interaction between polysaccharides and polyphenols, the complex of soluble dietary fiber (SDF) and two phenols were prepared. According to the results, at the conditions of pH = 4, temperature = 60 °C, concentration ratio of polysaccharide and phenol = 4:1, the maximum adsorption of catechin and gallic acid in the SDF complex was 155.74 and 134.05 mg/g, respectively. Due to UV–Vis and FT-IR analysis, it could be speculated that the interaction between SDF and phenols resulted in chemical combination. Furthermore, the monosaccharide composition and molecular weight distribution of SDF were significantly altered after conjugated with phenols. However, it showed no significant difference between two different SDF-phenol complexes, referring to GC and GPC data. The surface of SDF-phenol complex was loose with uniform arrangement, while the physical mixture surface was porous with irregular holes and cracks. Both SDF-CC and SDF-GA complex indicated higher thermal stability than the mixtures by TGA analysis. © 2020 Elsevier Ltd

Biomimetic dynamic membrane (BDM) has been employed as a promising membrane separation technology regarding water/wastewater treatment (Model pollutant is methylene blue). Given its catalytic function on micro-pollutant removal and fouling control, detailed mechanism for impacts of fabrication method, carriers (CNT and GO) and laccase on the construction of biomimetic layer and enzyme immobilization have not been clear so far. In this work, the BDM performance with various fabrication methods, carriers and laccase were investigated and verified. The BDM fabrication tests demonstrated that BDM with mixed filtration method had better filtration performance (up to 120 L m−2 h−1 flux and 80% removal rate) than BDM with stepwise filtration method. Moreover, the laccases immobilized on GO exhibited a stronger laccase activity than those on CNT. Increasing CNT or GO dosage strengthened removal rate, but lowered flux, meanwhile flux and removal rate exhibited a significant fluctuation with certain laccase dosage. At 25 g m−2 CNT or GO dosage and 50 g m−2 laccase dosage, the optimized flux and removal rate values were obtained. Further study investigated the surface morphology and property of BDM, showing that BDM with mixed filtration method turned out to be the optimized enzyme immobilization mechanism and fabrication method. In addition, during multiple filtration cycles, with the optimized conditions, the removal rate, flux and laccase activity of BDM could maintain at high levels. On account of the finding of the present study, selecting a suitable fabrication method, appropriate CNT or GO dosage and laccase dosage can indeed optimize the structure of biomimetic layer and enzyme immobilization, expanding its possibility on sustainable operation. © 2019 Elsevier Ltd

The co-existence of polysaccharides and enzymes in the food matrix could form complexes that directly influence the catalytic efficacy of enzymes. This work investigated the self-assembly behaviors of α-amylase and charged polysaccharides and fabricated the α-amylase/polysaccharides complex coacervates. The results showed that the linear charge density of polysaccharides had a critical impact on the complex formation, structure, and enzyme protection under acidic conditions. At low pH, α-amylase formed compact and tight coacervates with the λ-carrageenan. However, α-amylase/pectin coacervates dissociated when the pH was lower than 3.0. The optimized binding ratio of α-amylase/λ-carrageenan was 12:1, and α-amylase/pectin was 4:1. Finally, the α-amylase/λ-carrageenan complex coacervates effectively immobilized the enzyme and almost 70% of enzyme activity remained in coacervates after exposure to pH3.0 for 1 h. This study demonstrates that the change in the linear charge density of polysaccharides could regulate the enzyme-catalyzed process in food processing by a simple and fine-controlled method. © 2020 Elsevier Ltd

The preparation mainly composed of extraction, pre-purification and dehydration is essential for the research and development of natural polysaccharides. The methods or conditions used in the three procedures had significant effects on the composition, structure and function of the polysaccharides obtained. Temperature, pH, enzyme, ultrasound and microwave were the important factors associated with their physicochemical changes. Molecular degradation and intermolecular interaction were two of the main mechanisms responsible for the changes. The degradations of polysaccharides responding to hydrothermal and ultrasonic conditions could be partly descripted by multiple linear regression model, implying the possibility for the prediction and control of polysaccharide degradation. Moreover, the interactions between polysaccharide and other compounds, forming complexes natively or conditionally, could be selectively triggered or eliminated to obtain polysaccharides under certain functions. This work shows new insights into the preparation of polysaccharides, which could benefit the efficient utilization of their natural and modified properties.

Deoxynivalenol (DON) in wheat (Emai596) was degraded by five ways of ozone treatments, including ozone-gas treated wheat flour, wheat kernels before and after tempering, and tempering and washing with ozonated-water. For lab-scale milling, the effect on flour quality was investigated. The DON was quantified via high-performance liquid chromatography after extraction with immunoaffinity columns. The measure indicated all treatments could decrease the DON content of flour below 1.00 mg/kg. Ozone-gas treated wheat kernels after tempering had the highest DON degradation rate 33.33%. Ozonation produced no significant difference in gluten qualities, dynamic viscoelasticity, pasting and mixing properties, when wheat kernels were treated with ozone-gas or ozonated-water. Although ozone-gas treated flour had the highest whiteness (85.56), but it had negative effect on the rest of quality indicators. The results suggested the treatment of Ozone-gas treated tempered wheat kernels could not only degrade DON most efficiently but also ensure quality of flour. © 2020 The Author(s). 摘要:
The interfacial tension is one of the most important fundamental properties and presents crucial impacts throughout vegetable oil production, application and digestion. In this study, composition of vegetable oil including tocopherols, phytosterols, phenolic compounds, phospholipids, fatty acid composition and other constituents were determined. Furthermore, interfacial tension and its relationship with vegetable oil compositions were analyzed. Distribution and profile of composition of vegetable oil were remarkably different. The interfacial tension results showed physical refined vegetable oil exhibited an obviously lower interfacial tension than chemical refined oil attributed to abundant minor compositions. Moreover, the correlation analysis results indicated that phenolic compounds demonstrated the greatest influence on interfacial tension of vegetable oil against water with r = − 0.671, p = 0.009, followed by free fatty acid value, linoleic acid of triglyceride and phospholipids with r = − 0.639, 0.626, − 0.576 and p = 0.014, 0.017 and 0.031, respectively. No significant correlation was found between interfacial tension and other minor compositions. These results contribute to regulating lipid metabolism and evaluating oil quality more scientifically. © 2019, Springer Science+Business Media, LLC, part of Springer Nature.

Tribonema minus was cultivated in different concentrations of sugarcane wastewater (SW) diluted with mBG-11 medium to produce biomass for biodiesel, bioproduct, and biomaterial production. The results showed that T. minus grew mixotrophically in 50%SW, with the highest biomass accumulation (7.86 g/L) and nutrient removal efficiency (84.85% of nitrogen, 62.57% of phosphorus, and 44.72% of COD). Excluding 100%SW, the chrysolaminarin and cellulose contents increased with increasing SW concentration; the highest contents of 8.11% and 25.69% dry weight were reached in 75%SW, respectively. Although fewer lipids and palmitoleic acid accumulated at higher SW concentrations, their productivities were significantly higher than those in the control due to the higher contribution of biomass. Moreover, the fatty acid profiles produced at the tested concentrations showed superior biodiesel properties. These findings suggested that the addition of mBG-11 medium to SW might be an effective strategy for valuable biomass production in T. minus and SW bioremediation. © 2020 Elsevier Ltd

摘要:
Developing a rapid and stable method for analyzing the quality parameters of rice is important. Near-infrared (NIR) spectroscopy combined with chemometric techniques have been used to predict the critical contents of rice and shown its accuracy and stability. To further improve the predictive ability, we combine the derivative method of fractional order Savitzky–Golay derivation (FOSGD) with the wavelength selection method of competitive adaptive reweighted sampling (CARS). Compared with the traditional integer order Savitzky–Golay derivation (IOSGD), the FOSGD could improve the resolution ratio of the raw spectra more effectively. The wavelength selection method, CARS, could further extract the informative variables from the processed spectra. Four key contents of rice samples, including moisture, amylose, chalkiness degree, and gel consistency, were utilized to validate this method. The prediction results indicated that partial least squares (PLS) models optimized with FOSGD-CARS own higher accuracy and stability with smaller the root mean squared error of cross validations (RMSECVs) and root mean squared error of predictions (RMSEPs). The proposed method is convenient and provides a practical alternative for rice analysis. © The Author(s) 2020.

Effects of inulin on protein in frozen dough during frozen storage were investigated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM). The strength of electrophoretic bands in A1 (32-57 kDa) and A2 (20-25 kDa) regions and the content of freezable water and sulfhydryl in frozen dough with inulin were lower than those of the blank under the same storage time. The gluten structure of frozen dough with 2.5 wt% long-chain inulin was more dense and compact than that of the sample with 5.0 wt% short-chain inulin after 2 weeks. Moreover, 4 weeks later, the change of the a-helix and ß-sheet with 2.5 wt% long-chain inulin was lowest. These characteristics suggested that long-chain inulin exhibited a better protection effect on protein in frozen dough and showed a promising prospect for application in the food industry. © 2020 The Royal Society of Chemistry.

Automatic evisceration is a very important production technology in the chicken industry. In this study, an automatic chicken eviscerating robot system was developed and evaluated based on machine vision for locating the chicken carcasses and anal incision. This system consisted of chicken carcass conveying device, robotic manipulator, end-effector, control system and machine vision system. Based on machine vision, this system employed color space conversion, threshold segmentation, morphological operation method and active contour algorithm to obtain the chicken carcass images of the removed wings and legs. Subsequently, anal incision contour was extracted and its position was detected by the maximum inscribed circle algorithm. All the algorithms were implemented in the system for recognizing the individual carcass and automatically locating the targets of different sizes, which guided the manipulator to complete the evisceration. Present results indicated that the maximum location deviation of contour centroid of chicken carcass and anal incision was 5 and 4 pixels, respectively, while the average time of image processing was 954 ms. Then the eviscerating manipulator could accurately grasp the viscera, the residual rate and the breakage rate of viscera were 6.2% and 15%, respectively. The high accuracy obtained from the evaluation results shows that the machine vision system can be applied successfully to robotic evisceration in the processing of chicken. Hence, the development of the method for the eviscerating robot is necessary and significant, in particular for providing great foundation for robotic evisceration in poultry industry. © 2020 Institution of Chemical Engineers

The integrity of the intestinal mucosal barrier is important for the health of the host. In this study, longan pulp polysaccharides (LP) prevented the intestinal mucosal injury by increasing the expression of mucin 2, tight junction proteins zonulae occludens-1 (ZO-1), claudin-1, claudin-4, and adherens junction E-cadherin in cyclophosphamide-treated mice. To further identify the principle bioactive component of LP, four acidic polysaccharides (LPIa, LPIIa, LPIIIa, and LPIVa) were puri fi ed, and their intestinal protection activity in vitro was compared. LPIa, LPIIa, and LPIIIa displayed an ability to increase mRNA expression of ZO-1, claudin-1, occludin, and E-cadherin in differentiated Caco-2 cells, especially LPIa. LPIa has specific structure characteristics: porous surface structure, a high molecular weight (1.47 x 10(5) Da), and two specific glycosidic linkages of alpha-Araf-(1--> and -->5)-alpha-Araf-(1-->. These structure characteristics might primarily contribute to greater intestinal barrier protective effect of LPIa.

摘要:
Thermal denaturation is the primary reason for the loss of excellent performance for collagen-based materials. It is particularly urgent to explore an effective structure restoration strategy for thermally denatured collagen. In this study, three different degrees of collagen thermal denaturation models were constructed; the effects of ultrahigh pressure treatments of varying intensities on the structure and properties of partially thermal denatured collagen were investigated. Those results showed that ultrahigh pressure partially restores the triple helix conformation and the α helix of low thermally denatured collagen molecules and moderately restores collagen thermal stability. However, this recovery is not a 'perfect' structural restoration; it cannot effectively restore the fibrotic ability of collagen which leads to a decrease in the fiber diameter of the product and an increase in the storage modulus of the product gel. As the degree of thermal denaturation of collagen increases, the ultrahigh pressure recovery effect is gradually lost. For moderate thermally denatured collagen, ultrahigh pressure treatments only recover a small amount of its triple helical conformation. For high thermally denatured collagen, ultrahigh pressure treatments exacerbate the destruction of the three-dimensional structure of collagen molecules, thereby losing the order of molecular structure and the capability of fibrosis self-assembly. © 2019, Springer Science+Business Media, LLC, part of Springer Nature.

Background: There is growing public awareness regarding the health effects of indoor nanoscale particulate matter (INPM) since people spend the majority of their time indoors. INPM could have a direct entry route into the brain via the axons of the olfactory nerve and migrating across the blood-brain barrier (BBB). Using animals to explore this possibility is not a reliable method to fully demonstrate human physiological responses. We, therefore, set out to develop a human 3D functional blood-brain barrier model to examine the potential effects of INPM on the cerebral nervous system. Methods: Human astrocytes were co-cultured and human umbilical vein endothelial cells in 3D within a microfluidic chip to simulate the micro-complex physiological structure of the human BBB. This 3D human organotypic model has then been made to investigate any INPM-induced BBB dysfunction linked to potential cellular responses. Results: A 3D human functional blood-brain barrier was constructed in this study. We observed the translocation of INPM across the blood-brain barrier. The 3D human organotypic chip initially reflected damage to the nervous system with abnormal astrocyte proliferation and a decline in cell viability. We also looked at the behavior of oxidative stress-related biomarkers (ROS, GSH-Px, and MDA). INPM was implicated in aggravating inflammation via reactive oxygen species (ROS). The Keap1-Nrf2-ARE pathway is a key mechanism in cellular resistance to oxidative stress by mediating and activating a variety of antioxidant and detoxification enzymes. Following ROS accumulation, INPM induced abnormal expression of nuclear transcription factor Nrf2. This behavior disturbed the expression of, γ-glutamate synthase (γ-GCS) and heme oxygenase (HO-1), which further exacerbated the imbalance of the antioxidant system. Conclusions: This functional 3D human organotypic chip effectively mimics the physiological response of the human BBB. The chip provides a micro-complex structure to simulate the internal environment of the human blood-brain barrier, and partially simulates the physiological responses of the BBB to INPM exposure. Based on this model, INPM was shown to affect the blood-brain barrier biofunction by disrupting the Keap1-Nrf2-ARE pathways. © 2020 The Authors

采用电子舌测定鲜味,超纳滤、反相高效液相色谱从风干武昌鱼中分离纯化出纯度均一的鲜味肽,基质辅助激光解析电离-飞行时间质谱仪、氨基酸自动分析仪、傅里叶变换红外光谱测定鲜味肽的分子质量、氨基酸组成和二级结构,探讨鲜味肽结构与风味的关系。结果显示:从风干武昌鱼中纯化得到了一种鲜味较强的肽P2a-2,其分子质量为1 304.59 Da。P2a-2的鲜味氨基酸谷氨酸(21.97%)、甘氨酸(10.67%)和丙氨酸(9.51%)比例较高,且助味氨基酸组氨酸的比例最高(27.54%),β-转角为主要的二级结构(90.16%),推断P2a-2呈现较佳鲜味与其高含量的鲜味氨基酸和β-转角相关。

A 3D functional human microvascular network in a microfluidic device was developed in this study. The established model enables endotheilal cells to form vessel-like microtissues and behave physiological functions which are closer to cells in human blood vessels. It provides a possible bridge for how the research outcomes translate to humans. The ɑ-MnO2 nanowires, which were successfully synthesized using the hydrothermal approach. The vascular toxicity was evaluated based on this organotypic 3D microvessel model. Our results demonstrated that ɑ-MnO2 nanowires can induce cytotoxicity in dose-dependently. Because the accumulation of reactive oxygen species (ROS) and depletion of glutathione (GSH) are observed in the nanowire-treated cells. In addition, intravascular accumulation of ɑ-MnO2 nanowires could induce endothelial dysfunction by oxidative stress. This is expressed in disorder of NO expression and IL-6 up-regulation. Based on these results, we concluded that a mechanism for ɑ-MnO2 nanowire-induced vascular toxicity, which involves the accumulation of ROS, up-regulation of oxidative stress and inflammatory factors. This investigation may provide a fundamental insight to understand the nanotoxicity of wire-shaped nanomaterials. © 2019, © 2019 Taylor & Francis Group, LLC.
