摘要:
This study was conducted to evaluate the effects of dietary protein levels on growth performance and digestive enzyme activities of juvenile hybrid culter (Culter alburnus female x Ancherythroculter nigrocauda male). Five isolipidic experimental diets were formulated to contain graded levels of protein ranging from 29.54-48.98%. These were fed to triplicate groups of fish (11.44 +/- 0.7 g/ fish) for 8 weeks. Results from the feeding trial indicated that growth performance, body composition, and digestive enzyme activities of the fish were significantly different (P<0.05) between the groups. Growth performance had a generally increasing trend with increasing dietary protein levels. Weight gain (160.76%) and specific growth rate (1.71) of fish in the group where feed contained 40.07% dietary protein was significantly higher than the group where feed contained 33.58% dietary protein (118.76% and 1.40, respectively), but there was no significant difference compared with 45.04% dietary protein. Based on the statistical significance, our results indicated that the dietary protein level should not be less than 40% for juvenile hybrid culter.

摘要:
The objective of this study was to isolate and identify yeast strains from the digestive tract of ducks, and evaluate in vitro their potential as probiotics in poultry. The yeast strains were isolated using malt extract agar medium, and identified through morphological, physiological, and biochemical tests as well as sequence homology analyses of 26S rDNA D1/D2 region. A total of 35 yeast strains were isolated from the guts of Cherry Valley meat ducks, including seven strains of Saccharomyces cerevisiae (S. cerevisiae). These seven strains of S. cerevisiae were further screened for their use as alternative yeast probiotics strains for poultry feed. The yeast strains were characterized for their cell surface hydrophobicity, autoaggregation ability, and resistance to high temperature (30°C, 37°C, and 42°C), low pH (2.0, 3.0, and 4.0), bile salts (0.3% and 0.6%), and nutrition starvation (2, 4, 6, 8, 10, and 12 days). The isolates of WHY-2 and WHY-7 had a higher survival percentage at 37°C, pH 2.0, 0.60% poultry bile salts, and 10 days of nutrition starvation, with higher cell surface hydrophobicity and autoaggregation, when compared with the other isolates, suggesting that the isolates WHY-2 and WHY-7, could be used as probiotic candidates. The data obtained in this study could help in selecting probiotic yeast candidates for use in poultry industry.

摘要:
为了研究日粮中添加凝结芽孢杆菌对仔猪生长性能、肠道菌群和血液生化指标的影响,试验选取21日龄健康断奶仔猪30头,随机分为3组(对照组、试验Ⅰ组、试验Ⅱ组),对照组饲喂基础日粮,试验Ⅰ组、试验Ⅱ组分别在基础日粮中添加凝结芽孢杆菌2.0× 106 cfu/g、2.0× 107 cfu/g.预试期3d,正试期21 d.于试验第21天采血,屠宰,测定其生长性能、肠道菌群和血液生化指标.结果表明:日粮中添加不同水平的凝结芽孢杆菌均显著降低了断奶仔猪的腹泻率(P<0.05);提高了仔猪回肠内容物中乳酸菌的数量(P<0.05),降低了回肠内容物中梭菌的数量(P<0.05);提高了血浆中磷酸丝氨酸、胱氨酸、酪氨酸等多种氨基酸的含量(P<0.05),降低了磷酰乙醇铵的含量(P<0.05).说明在日粮中添加凝结芽孢杆菌可以改善断奶仔猪的肠道菌群,降低仔猪的腹泻率.

摘要:
本试验旨在研究程序性坏死阻断剂Necrostatin-1 (Nec-1)对脂多糖(LPS)刺激断奶仔猪血细胞分类计数和血液生化指标的影响.选用25头断奶仔猪,分为对照组[10%二甲基亚砜(DMSO)+生理盐水]、LPS组(10%DMSO+LPS)、0.33Nec-1组[0.33 mg/kg体重(BW) Nec-1+LPS]、1.0Nec-1组(1.0 mg/kg BW Nec-1+LPS)和3.3Nec-1组(3.3 mg/kg BW Nec-1+LPS).注射10% DMSO或Nec-1 30 min后,对照组注射生理盐水,其他组注射100 μg/kg BW LPS,分别在注射LPS或生理盐水4h和24h后采血液样品待测.结果表明:注射LPS 4h后,0.33 mg/kg BW Nec-1、1.0 mg/kg BW Nec-1和3.3 mg/kg BW Nec-1显著缓解LPS刺激导致的血小板分布宽度升高(P<0.05);0.33 mg/kg BW Nec-1能显著缓解LPS刺激导致的血浆谷草转氨酶(AST)活性和AST/谷丙转氨酶(ALT)比例的升高(P<0.05),1.0 mg/kg BW Nec-1显著缓解血浆葡萄糖含量和肌酸激酶活性的降低(P<0.05);注射LPS 24 h后,0.33 mg/kg BW Nec-1、1.0 mg/kg BW Nec-1和3.3 mg/kg BW Nec-1可显著缓解LPS刺激导致的嗜酸性粒细胞比例的升高(P<0.05),并有缓解嗜酸性粒细胞绝对值升高的趋势(P<0.10);0.33 mg/kg BWNec-1缓解了肌酸激酶活性的降低并提高了血浆总胆固醇和甘油三酯含量(P<0.05),1.0 mg/kg BW Nec-1有缓解LPS刺激导致的血浆AST/ALT升高的趋势(P<0.10),3.3 mg/kg BW Nec-1能显著缓解AST/ALT比例的升高(P<0.05)和葡萄糖的降低(P<0.05),并有缓解AST升高和总胆固醇降低的趋势(P<0.10).以上结果表明,Nec-1对LPS刺激导致的免疫应激反应具有一定的缓解作用.

摘要:
为研究断奶仔猪日粮代谢葡萄糖的适宜水平,本试验选择体重为5.5 kg左右的21日龄断奶仔猪30头,随机分为5组,每组6个重复,每个重复1头猪。Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ组在断奶后1-14 d分别饲喂代谢葡萄糖水平为126.4、202.6、235.7、331.6、371.5 g/kg的5种日粮,在断奶后15-42 d分别饲喂代谢葡萄糖水平为37.6、132.5、300.0、354.3、412.5 g/kg的5种日粮,定期称重并记录采食量,颈静脉抽血检验。结果表明:Ⅱ组仔猪在2个阶段的日增重均显著高于Ⅰ、Ⅳ组(P〈0.05),Ⅱ组饲料转化效率均显著高于Ⅰ、Ⅳ、Ⅴ组(P〈0.05);断奶后1-14 d,Ⅰ组的总蛋白和白蛋白水平均显著高于Ⅲ组(P〈0.05);断奶后15-42 d,Ⅲ组的总蛋白含量显著高于Ⅰ、Ⅳ、Ⅴ组(P〈0.05),Ⅱ组的白蛋白含量显著高于日粮Ⅳ、Ⅴ组,Ⅴ组的尿酸含量显著高于Ⅰ、Ⅲ、Ⅳ组(P〈0.05)。本试验结果表明,日粮代谢葡萄糖水平会影响断奶仔猪生长性能以及血液生化指标,仔猪断奶后1-14 d,日粮代谢葡萄糖适宜水平为202.6-235.7 g/kg;仔猪断奶后15-42 d日粮代谢葡萄糖适宜水平为132.5-300.0 g/kg。

本试验旨在研究三丁酸甘油酯(TB)对乙酸(ACA)刺激仔猪小肠黏膜生长和肠道屏障功能的影响。试验选用18头体重接近、健康的28日龄断奶仔猪(杜×长×大),随机分为3个处理:对照组、乙酸组和TB组,每个处理6个重复,每个重复1头猪。试验期为21d。试验期内对照组和乙酸组饲喂基础日粮,TB组饲喂基础日粮+0.1%TB。于试验第15天清晨,乙酸组和TB组仔猪直肠灌注10mL 10%乙酸,对照组直肠灌注相应剂量的灭菌生理盐水。于试验第18、21天清晨前腔静脉采血,测定血浆二胺氧化酶(DAO)活力;第21天屠宰取样,取空肠、回肠等组织样品,测定仔猪肠道形态结构、肠道黏膜损伤相关基因mRNA水平等指标。结果表明:(1)未用乙酸刺激前,与对照组相比,日粮中添加0.1%TB对仔猪平均日增重有一定程度提高(P=0.089),且显著降低了仔猪料重比(P<0.05);(2)与对照组相比,乙酸组显著降低了空肠黏膜绒毛高度、回肠黏膜绒毛高度与隐窝深度比值及空肠绒毛表面积(P<0.05);(3)与乙酸组相比,TB组显著降低了血浆DAO活力(P<0.05),极显著降低了回肠AREG基因的相对表达量(P<0.01)。综合上述结果,日粮中添加0.1%TB能缓解乙酸诱导的仔猪肠黏膜生长及肠道屏障功能的损伤。

摘要:
This study was designed to evaluate the physiological and cytological responses in liver of GIFT tilapia (Oreochromis niloticus) to acute copper exposure. Fish were exposed to three Cu treatments at the following concentrations: 0 (control), 1.96 mg/L and 3.92 mg/L, and liver samples were collected after 96h of exposure. Observation of oil red O staining showed that acute copper exposure increased lipid accumulation in liver, which may due to changes of hepatic enzyme activities (malic enzyme, lipoprotein lipase, hepatic lipase, fatty acid synthetase, carnitine palmitoyltransferases l) and lesions of some organelles such as mitochondria and swollen rough endoplasmic reticulum of hepatocytes. This study indicated that acute copper exposure induced hepatic lipid metabolic disturbances and damaged hepatic organelle in GIFT tilapia.

摘要:
The extensive metabolism of amino acids (AA) as fuel is an important reason for the low use efficiency of protein in pigs. In this study, we investigated whether regulation of the pyruvate dehydrogenase kinase (PDK)/pyruvate dehydrogenase alpha 1 (PDHA1) pathway affected AA consumption by porcine intestinal epithelial (IPEC-J2) cells and intestinal bacteria in pigs. The effects of knockdown of PDHA1 and PDK1 with small interfering RNA (siRNA) on nutrient consumption by IPEC-J2 cells were evaluated. IPEC-J2 cells were then cultured with sodium dichloroacetate (DCA) to quantify AA and glucose consumption and nutrient oxidative metabolism. The results showed that knockdown of PDHA1 using siRNA decreased glucose consumption but increased total AA (TAA) and glutamate (Glu) consumption by IPEC-J2 cells (P <0.05). Opposite effects were observed using siRNA targeting PDK1 (P <0.05). Additionally, culturing IPEC-J2 cells in the presence of 5 mM DCA markedly increased the phosphorylation of PDHA1 and PDH phosphatase 1, but inhibited PDK1 phosphorylation (P <0.05). DCA treatment also reduced TAA and Glu consumption and increased glucose depletion (P <0.05). These results indicated that PDH was the regulatory target for shifting from AA metabolism to glucose metabolism and that culturing cells with DCA decreased the consumption of AAs by increasing the depletion of glucose through PDH activation.<br/> ©2018 American Chemical Society.

Objective: This experiment was conducted to investigate whether asparagine (Asn) could improve liver energy status in weaning pigs when challenged with lipopolysaccharide. Methods: Forty-eight weaned pigs (Duroc×Large White×Landrace, 8.12±0.56 kg) were assigned to four treatments: i) CTRL, piglets received a control diet and injected with sterile 0.9% NaCl solution; ii) lipopolysaccharide challenged control (LPSCC), piglets received the same control diet and injected with Escherichia coli LPS; iii) lipopolysaccharide (LPS)+0.5% Asn, piglets received a 0.5% Asn diet and injected with LPS; and iv) LPS+1.0% Asn, piglets received a 1.0% Asn diet and injected with LPS. All piglets were fed the experimental diets for 19 d. On d 20, the pigs were injected intraperitoneally with Escherichia coli LPS at 100 μg/kg body weights or the same volume of 0.9% NaCl solution based on the assigned treatments. Then the pigs were slaughtered at 4 h and 24 h after LPS or saline injection, and the liver samples were collected. Results: At 24 h after LPS challenge, dietary supplementation with 0.5% Asn increased ATP concentration (quadratic, p<0.05), and had a tendency to increase adenylate energy charges and reduce AMP/ATP ratio (quadratic, p<0.1) in liver. In addition, Asn increased the liver mRNA expression of pyruvate kinase, pyruvate dehydrogenase, citrate synthase, and isocitrate dehydrogenase β (linear, p<0.05; quadratic, p<0.05), and had a tendency to increase the mRNA expression of hexokinase 2 (linear, p<0.1). Moreover, Asn increased liver phosphorylated AMP-activated protein kinase (pAMPK)/total AMP-activated protein kinase (tAMPK) ratio (linear, p<0.05; quadratic, p<0.05). However, at 4 h after LPS challenge, Asn supplementation had no effect on these parameters. Conclusion: The present study indicated that Asn could improve the energy metabolism in injured liver at the late stage of LPS challenge. 为了解对三叶白术汤口服液的遗传毒性,为其安全性评价提供试验依据,进行了Ames试验(采用平板掺入法,每皿分别加入5 000、1 000、200、40、8μg)、中国仓鼠肺成纤维细胞(CHL)染色体畸变试验(试验药物浓度分别为5 mg/mL、1. 67 mg/mL、0. 56 mg/mL三个剂量组)和小鼠骨髓嗜多染红细胞微核试验(分别以2 000、1 000、500 mg/kg·bw的剂量给小鼠灌胃),3个遗传毒性试验均另设置阴性对照组和阳性对照组。结果显示,3项遗传毒性试验结果均为阴性。表明在此研究试验条件下,三叶白术汤口服液无遗传毒性。

摘要:
本试验旨在研究N-乙酰半胱氨酸(NAC)和冷应激对肉鸡肠道组织形态、能量状况和抗氧化能力的影响.试验将200只1日龄健康雄性科宝肉鸡随机分为4个处理组:对照组(基础日粮)、NAC组(基础日粮+0.1%NAC)、冷应激组(基础日粮)与冷应激+NAC组(基础日粮+0.1%NAC),每组5个重复,每个重复10只鸡.试验第1~2周按常规饲养方法控制温度,试验第3~6周冷应激组和冷应激+NAC组的温度控制在(10±2)℃,而对照组与NAC组维持在(26±2)℃.结果表明:冷应激处理使肉鸡平均日采食量、平均日增重、空肠低氧诱导因子1 (HIF-1) mRNA水平显著下降(P<0.05),饲料转化率、回肠一磷酸腺苷(AMP)含量和空肠隐窝深度极显著升高(P<0.01);日粮添加0.1% NAC显著提高了冷应激肉鸡肠道绒毛高度(VH)、十二指肠与空肠绒毛高度与隐窝深度比值(VH/CD)、空肠三磷酸腺苷(ATP)和腺苷酸池(TAN)含量、回肠ATP和二磷酸腺苷(ADP)及能荷(AEC)含量、十二指肠超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活力、回肠谷胱甘肽过氧化物酶(GSH-Px)活力、空肠血红素加氧酶(HMOX) mRNA水平(P<0.05).因此,日粮中添加0.1%的N-乙酰半胱氨酸可改善肠道组织形态和能量状况,提高冷应激肉鸡肠道抗氧化能力.

摘要:
Morphological studies of Sicuophora (Syn. Wichtermania) multigranularis Xiao et al., 2002, from the rectum of the frog, Quasipaa spinosa, performed using silver impregnation and scanning electron microscopy, confirmed the following newly recognized features: (1) only one apical suture on the right surface; (2) two naked regions at the posterior end of both the left and the right side of the body. Phylogenetic analysis based on the SSU-rRNA gene showed that S. multigranularis is a sister to a clade comprising all other Clevelandellida, strongly supporting the validity of the genus Sicuophora. This is also the first molecular data obtained for the genus Sicuophora. Because of the lack of molecular data, it will be necessary to obtain more genetic data from the family Sicuophoridae to discuss the question of the taxonomic status of the genus Sicuophora.

本试验旨在研究中链脂肪酸对脂多糖(LPS)诱导的断奶仔猪肠黏膜免疫屏障损伤的保护作用。将24头(21±1)d、体重为(9.0±0.7)kg的杜×长×大三元杂交断奶仔猪随机分为4个处理组,每个处理组6个重复,每个重复1头猪。试验采用2×2因子设计,2个主因子分别为日粮处理(中链脂肪酸或玉米油)和免疫应激(LPS或NaCl)。预试7 d,试验期为21 d。试验第21天,每个日粮组3个重复注射100μg/kg体重(BW)的LPS,另外3个重复猪注射等量的生理盐水,4 h后屠宰取肠道样品待测。结果表明:中链脂肪酸显著提高了空肠绒毛高度(P<0.05),降低了空肠、回肠固有层细胞密度和回肠PP结细胞密度(P<0.05);中链脂肪酸还缓解了LPS刺激导致的空肠杯状细胞数的升高和上皮间淋巴细胞数的减少。综上,日粮中添加中链脂肪酸有保护仔猪肠黏膜免疫屏障的功能。

本试验旨在研究N-乙酰半胱氨酸(N-acetylcysteine,NAC)灌胃对超早期断奶仔猪血浆中游离氨基酸、小肠形态结构及功能的影响.将22头体重为(2.99±0.41) kg的7日龄断奶仔猪随机分为对照组和NAC组,每组1 1个重复,每个重复1头猪.在饲喂人工乳的基础上,NAC组每天每头按50 mg/kg体重进行NAC灌胃,对照组每天每头进行同体积的生理盐水灌胃.于试验第11天按0.1 g/kg体重的剂量给仔猪灌服D-木糖溶液,1h后前腔静脉采血,然后屠宰取小肠组织样,刮取肠黏膜及制作组织切片.结果表明:与对照组相比,NAC组显著提高了超早期断奶仔猪平均日增重(P<0.05);提高了超早期断奶仔猪血浆中谷氨酰胺、胱氨酸、γ-氨基丁酸水平(P<0.05),降低了血浆中尿素氮、半胱氨酸、色氨酸水平和二胺氧化酶活力(P<0.05):NAC灌胃提高了哺乳仔猪十二指肠黏膜的绒毛高度/隐窝深度(P<0.05),有降低十二指肠隐窝深度的趋势(P<0.10),有提高空肠绒毛宽度和回肠绒毛高度的趋势(P>0.10).在本试验条件下,NAC灌胃可改善小肠形态结构与屏障功能,提高氨基酸的利用率,调节血浆中部分氨基酸的含量.

摘要:
试验旨在研究复合植物精油(OCT)对脂多糖(LPS)刺激仔猪肝脏的保护作用.选取18头仔猪,随机分为对照组、LPS组和OCT+LPS组,每组6个重复.对照组与LPS组饲喂基础日粮,OCT+LPS组在基础日粮中添加50 mg/kg OCT.试验期21 d.于试验第21天,LPS组与OCT+LPS组仔猪腹腔注射LPS,对照组注射等量的生理盐水,3h后采血,6h后屠宰.结果表明:与对照组相比,LPS刺激提高了血浆谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、谷酰转肽酶(GGT)、肝脏诱导型一氧化氮合酶(i-NOS)的活力(P<0.05),肝脏热休克蛋白70 (HSP70)基因的相对表达量显著提高(P<0.05);仔猪肝脏表皮生长因子(EGF)、白细胞介素10 (IL-10)、胰岛素样因子(IGF-1)、丝氨酸蛋白激酶(mTOR)基因的相对表达量显著降低(P<0.05).在日粮中添加50 mg/kg的OCT,可缓解LPS导致的仔猪血浆ALT、AST、GGT活力的升高以及肝脏i-NOS活力的升高(P<0.05),且有缓解LPS导致的仔猪肝脏MPO活力升高的趋势(P<0.1),另外可缓解LPS导致的仔猪肝脏HSP70基因相对表达量的升高以及EGF、IL-10、IGF-1、mTOR等基因相对表达量的降低(P<0.05).综上可知,日粮中添加50 mg/kg的OCT可缓解LPS刺激引起的肝脏炎症反应,提高仔猪肝脏细胞的生长和增殖,进而缓解LPS刺激导致的仔猪肝脏氧化应激损伤.

天然淋巴细胞(Innate lymphoidc ells,ILCs)是新发现的一类重要淋巴细胞亚群,它们起源于普通淋巴祖细胞,不同于T、B淋巴细胞。ILCs不表达抗原受体,在微生物的刺激下可快速活化并进入靶器官参与免疫应答。ILCs位于黏膜表面,可增强免疫反应,维持黏膜的完整性,在黏膜屏障炎症性反应及组织损伤中发挥重要作用。本文简要阐述了ILCs的分类、分化和迁移及在肠黏膜免疫中的作用。

为研究N-乙酰半胱氨酸(NAC)对乙酸(ACA)刺激仔猪肠黏膜屏障功能以及肠道黏膜生长的影响,试验选取体重相似且体况健康的仔猪18头,随机分为对照组、ACA组和NAC组,其中前两组饲喂基础饲粮,NAC组在基础饲粮中添加0.05%NAC,每组6个重复,每个重复1头猪。试验第15天对ACA组和NAC组仔猪直肠灌注10 m L10%的乙酸,对照组灌注相应剂量的生理盐水。第21天灌服D-木糖溶液后采集前腔静脉血样,深度麻醉后屠宰,取小肠黏膜样品待测。结果表明:与对照组相比,ACA组仔猪血浆二胺氧化酶(DAO)活性、十二指肠和回肠的隐窝深度分别提高了94.80%、14.31%、8.14%(P<0.05);十二指肠DAO活性降低了17.95%(P<0.05),空肠绒毛高度、十二指肠和回肠绒毛高度/隐窝深度比值,十二指肠和空肠绒毛表面积,及回肠RNA/DNA比值和TP/DNA比值分别降低了14.03%、26.72%、14.97%、12.40%、37.11%、38.91%、26.35%(P<0.05)。饲粮中添加NAC可以缓解以上这些负面影响。此外,相较于ACA组,NAC血浆中D-木糖和表皮生长因子(EGF)含量以及回肠中EGF m RNA水平提高了17.29%、26.20%、34.03%(P<0.05)。综上所述,NAC能够改善ACA刺激造成的仔猪小肠黏膜屏障功能受损,NAC可能是通过提高EGF含量以及EGF m RNA水平,促进细胞生长,修复肠道损伤。

试验旨在研究复合植物精油(OC T)对脂多糖(L PS)诱导的断奶仔猪的免疫应激的影响.试验选取28日龄左右的杜×长×大三元杂交仔猪,随机分成3个处理组:对照组(基础日粮,注射生理盐水),L PS组(基础日粮,注射LPS),OCT组(基础日粮+50 mg/kg OCT,注射LPS).每组8个重复,每个重复1头猪.于试验第21天注射L PS(100μg/kg体重)或等体积的生理盐水,注射3 h后前腔静脉采血进行白细胞分类计数;6 h后屠宰,取胸腺、脾脏,-80℃保存,用于测量胸腺、脾脏免疫相关基因白介素-4(IL-4)、白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子(TNF-α)、核因子(NF-κB)以及转录相关基因转录激活因子3(STAT3)的基因相对表达量.结果表明:①与对照组相比,L PS刺激降低了仔猪血液中白细胞、中性粒细胞、淋巴细胞、单核细胞及嗜酸性粒细胞数量(P<0.05);与LPS组相比,日粮中添加OCT提高了白细胞的数量(P<0.05);②与对照组相比,LPS刺激上调了脾脏、胸腺IL-6、STAT3 mRNA水平(P<0.05),下调了脾脏IL-4、TNF-α及胸腺IL-4、TNF-α、NF-κB mRNA水平(P<0.05);与LPS组相比,添加OCT下调了脾脏、胸腺IL-6、STAT3 mRNA水平(P<0.05);上调了胸腺IL-4、IL-10、TNF-α、NF-κB的mRNA水平(P<0.05).综合上述结果,LPS诱导发生了仔猪免疫应激,OCT在一定程度具有缓解作用.

Haemophilus parasuis is the causative agent of Glässer’s disease in pigs. H. parasuis can cause vascular damage, although the mechanism remains unclear. In this study, we investigated the host cell responses involved in the molecular pathway interactions in porcine aortic vascular endothelial cells (PAVECs) induced by H. parasuis using RNA-Seq. The transcriptome results showed that when PAVECs were infected with H. parasuis for 24 h, 281 differentially expressed genes (DEGs) were identified; of which, 236 were upregulated and 45 downregulated. The 281 DEGs were involved in 136 KEGG signaling pathways that were organismal systems, environmental information processing, metabolism, cellular processes, and genetic information processing. The main pathways were the Rap1, FoxO, and PI3K/Akt signaling pathways, and the overexpressed genes were determined and verified by quantitative reverse transcription polymerase chain reaction. In addition, 252 genes were clustered into biological processes, molecular processes, and cellular components. Our study provides new insights for understanding the interaction between bacterial and host cells, and analyzed, in detail, the possible mechanisms that lead to vascular damage induced by H. parasuis. This may lead to development of novel therapeutic targets to control H. parasuis infection. © 2018 by the authors. Licensee MDPI, Basel, Switzerland.

Enterotoxigenic Escherichia coli (ETEC) is primary pathogenic bacteria of piglet diarrhea, over two thirds of piglets diarrhea caused by ETEC are resulted from STa-producing ETEC strains. This experiment was conducted to construct the recombinant E. coli expressing STa and study the injury and mechanism of recombinant E. coli expressing STa on 7 days old piglets colon. Twenty-four 7 days old piglets were allotted to four treatments: control group, STa group (2 × 10 9 CFU E. coli LMG194-STa), LMG194 group (2 × 10 9 CFU E. coli LMG194) and K88 group (2 × 10 9 CFU E. coli K88). The result showed that E. coli infection significantly increased diarrhea rates; changed DAO activity in plasma and colon; damaged colonic mucosal morphology including crypt depth, number of globet cells, density of lymphocytes and lamina propria cell density; substantially reduced antioxidant capacity by altering activities of GSH-Px, SOD, and TNOS and productions of MDA and H 2 O 2 ; obviously decreased AQP3, AQP4 and KCNJ13 protein expression levels; substantially altered the gene expression levels of inflammatory cytokines. Conclusively, STa group had the biggest effect on these indices in four treatment groups. These results suggested that the recombinant strain expressed STa can induce piglets diarrhea and colonic morphological and funtional damage by altering expression of proteins connect to transportation function and genes associated with intestinal injury and inflammatory cytokines. © 2018 The Japanese Society of Veterinary Science.
