摘要:
在水产养殖产品升级与经营转型的大背景下,借我校水产养殖专业列入湖北省“荆楚卓越人才”协同育人计划的契机,针对水产养殖专业人才培养普遍存在的问题,结合我校水产专业人才培养的现状、目标定位与特色发展方向,笔者以教学一线教师的视角,从构建战略联盟长效机制的前提与基础、战略联盟的确定与遴选、人才培养知识体系的构建、专业技能及综合素质培养体系构建等方面,提出自己对“荆楚卓越水产人才”协同培养的一些思考与建议,以期为本校水产专业的学科建设和人才培养提供思路与探讨主题,并通过逐步教学实践探索,进而形成一套专业特色鲜明,能充分适应社会和行业需求的人才培养模式.

镧对稀有鮈鲫肝胰脏抗氧化酶活性及MDA含量的影响

摘要:
以氯化镧(LaCl_3·7H_2O)为试验毒物,以稀有鮈鲫(Gobiocypris rarus)为试验材料,采用21 d慢性毒性试验(La~(3+)浓度分别为0.00、0.04、0.08、0.16、0.32、0.80 mg/L),通过检测稀有鮈鲫肝胰脏中超氧物歧化酶(SOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量,研究了镧暴露胁迫对水生动物抗氧化系统的影响,以期从抗氧化生理响应的角度探讨稀土元素镧对水生动物的毒理机理。结果显示,与对照组相比,0.04~0.32 mg/L试验组肝胰脏SOD活性出现一定程度降低(P>0.05),但0.80 mg/L组SOD活性出现一定程度上升(P>0.05);0.04~ 0.32 mg/L试验组肝胰脏CAT活性较对照组出现一定程度降低(P>0.05),但0.80 mg/L组CAT活性出现一定程度上升(P>0.05),且0.80 mg/L组CAT活性显著高于0.04~0.32 mg/L组(P<0.05);0.04~0.16 mg/L试验组肝胰脏MDA含量较对照组出现一定程度降低(P>0.05),0.32 mg/L组MDA含量略微升高(P>0.05), 0.80 mg/L组MDA含量显著高于对照组(P<0.05),也显著高于其它La~(3+)暴露组(P<0.05)。研究表明,一定浓度的La~(3+)暴露胁迫会对环境中水生生物抗氧化系统产生不利影响。

饲料中添加稀土壳聚糖蝥合盐对鲫鱼肌肉生化组成的影响

摘要:
通过在日粮中添加不同剂量稀土壳聚糖螯合盐(rare earth-chitosan chelate, RECC),研究其对鲫鱼(Carassius auratus)肌肉品质的影响。试验分为4个组:空白对照组、0.08%组、0.16%组、0.24%组,各组日粮中稀土壳聚糖螯合盐添加水平分别为0.00%、0.08%、0.16%、0.24%。每组设5个重复,每个重复放养35尾试验鱼。采用生物化学分析和高效液相色谱法分析肌肉中营养组分。结果表明:在同一解冻时间时,各RECC添加组鲫鱼肌肉渗出性损失均较空白对照组有不同程度降低(P>0.05);各RECC添加组水分、粗脂肪、磷含量与空白对照组较为接近(P>0.05),粗蛋白质、钙含量均较空白对照组增加(P>0.05),灰分含量降低(P>0.05)。各RECC添加组鲫鱼肌肉中肌苷含量均高于或显著高于空白对照组,其中0.16%组显著高于空白对照组(P<0.05);肌苷酸含量也均高于空白对照组(P>0.05);胆固醇含量则均较空白对照组有不同程度下降(P>0.05)。以上结果表明,稀土壳聚糖螯合盐对鲫鱼肌肉成分不仅无不良影响,而且对其品质、风味有一定增强改善作用。

摘要:
This study was designed to evaluate the physiological and cytological responses in liver of GIFT tilapia (Oreochromis niloticus) to acute copper exposure. Fish were exposed to three Cu treatments at the following concentrations: 0 (control), 1.96 mg/L and 3.92 mg/L, and liver samples were collected after 96h of exposure. Observation of oil red O staining showed that acute copper exposure increased lipid accumulation in liver, which may due to changes of hepatic enzyme activities (malic enzyme, lipoprotein lipase, hepatic lipase, fatty acid synthetase, carnitine palmitoyltransferases l) and lesions of some organelles such as mitochondria and swollen rough endoplasmic reticulum of hepatocytes. This study indicated that acute copper exposure induced hepatic lipid metabolic disturbances and damaged hepatic organelle in GIFT tilapia. © 2018, Israeli Journal of Aquaculture - Bamidgeh. 摘要:
We evaluated the acute toxicity effects of lanthanum (La(III)) on gill and liver of rare minnow (Gobiocypris rarus). The median lethal concentration of La (III) at 96 h was 1.92 mg L–1. Rare minnow were reared in freshwater and exposed to 0.04, 0.08, 0.16, 0.32 and 0.80 mg L–1 La (III) for 21 d. Gill and liver samples were analyzed by light microscopy. The main histopathological changes induced by La (III) in gills were epithelial lifting, filamentary epithelial proliferation,edema, lamellar fusion, desquamation, and necrosis. Histopathological changes induced by La (III) in the liver included dilation of sinusoids, focal congestion, pyknotic nuclei, karyohexis and karyolysis, vacuolar degeneration, and numerous necrosis areas. Hypsometric analysis indicated significant changes in the measures of gill dimensions (average length, width, area), suggesting metabolic disturbance (gas exchange) upon La (III) exposure. The result showed that La (III) severely affects fish gill and liver. © 2017, Springer Science+Business Media, LLC.

A 42–day trial was conducted to evaluate the performance of juvenile (0.30 g initial weight) Pacific white shrimp Litopenaeus vannamei offered diets containing various levels of copper hydroxychloride (Cu2(OH)3Cl) containing 58.8% copper (Cu) or Cu sulfate (CuSO4) containing 25.5% Cu. The basal diet (360 g/kg protein, 80 g/kg lipid) was primarily composed of soybean meal, fish meal, corn protein isolate and whole wheat and was formulated to be deficient in Cu (approximately 10 mg Cu/kg). The basal diet (B10) was supplemented with 30, 90, 150, and 210 mg Cu/kg from each Cu source. With the exception of 1 treatment (H220, Cu supplemented diet with dietary copper 210 mg/kg from Cu hydroxychloride, n = 4), five replicate groups of 15 shrimp per tank (initial weight 0.30 g) were offered the test diets in slight excess. At the end of the trial, no significant differences were observed in final biomass (53.20–64.40 g), final mean weight (4.34–4.87 g), weight gain (1349–1544%), feed conversion ratio (FCR, 1.61–1.82), survival (81.3–96.0%), and thermal growth coefficient (0.082–0.087). The carapace, hepatopancreas, and whole shrimp Cu were significantly increased with dietary Cu levels from either source. No significant differences in hemolymph Cu contents were observed between the two sources. The Cu concentrations of hepatopancreas and whole shrimp were significantly lower in shrimp fed diets with Cu hydroxychloride than those in shrimp fed diets with Cu sulfate. The Cu contents of final whole body significantly increased with dietary Cu levels from either source and corresponded to a decreased trend in whole body Cu retention as dietary Cu increased regardless of source. Sequencing of intestinal and fecal microbiomes (16S V1–V3) captured distinct microbial communities based on source but not copper treatment group. However, the abundance of several specific family and genera of bacteria did appear to respond to different types and doses of dietary copper. Among these we observed a trend of decreased abundance of particular species of Vibrios, including V. harveyi, V. sinaloensis and V. orientalis. Results of this study confirm that both copper sources are utilized efficaciously by Pacific white shrimp, however, differential deposition patterns indicated the two sources of Cu are metabolized differently. Hence, additional studies to assess the specific metabolic pathways of various Cu sources as well as possible effects of high copper levels on resistance to infection should be undertaken in the future. Statement of relevance In this study, we assessed the efficacy of various levels of dietary copper supplementation with copper sulfate and copper hydroxychloride in Pacific white shrimp. Our finding shows that both copper sources are utilized efficaciously but metabolized differently by shrimp. This manuscript provides valuable information about effects of high copper levels from two copper sources in shrimp. © 2017 Elsevier B.V.

Two trials were conducted to evaluate the performance of Pacific white shrimp Litopenaeus vannamei offered diets containing various copper (Cu) levels from Cu hydroxychloride (Cu2(OH)3Cl) containing 58.81% copper in the clear water recirculating system. In both trials, the basal diet (360 g kg−1 protein, 80 g kg−1 lipid) containing approximately 10 mg Cu kg−1 was primarily comprised of fishmeal, soybean meal, corn protein concentrate and whole wheat. In trial 1, test diets were produced supplementing the basal diet with 5, 10, 20, 40 and 60 mg Cu kg−1 from Cu hydroxychloride. Four replicate groups of 15 shrimp per tank (initial weight 0.28 g) were offered diets in slight excess over 8 weeks. In trial 2, the basal diet was supplemented with 30, 90, 150, 210 and 270 mg Cu kg−1 from Cu hydroxychloride. Seven replicate groups of 15 shrimp per tank (initial weight 0.22 g) were offered feed in slight excess over 7 weeks. At the end of the two growth trials, no significant differences were observed in final biomass, final mean weight, percentage weight gain, feed conversion ratio (FCR) and survival. In trial 1, the Cu concentrations of the carapace, hepatopancreas and whole shrimp linearly increased with increasing dietary Cu supplements. In trial 2, polynomial regression of Cu concentrations of the carapace, hepatopancreas and whole shrimp against analysed dietary Cu content indicated that a plateau was reached at 215 mg analysed Cu kg−1. Results of this study indicate that there was no negative effect of high levels of Cu supplement with regard to growth and survival. Tissue levels generally increased up to around 200 mg Cu kg−1 diet and then decreased, possibly indicating a shift in physiology.

摘要:
Fish growth is an important index in aquaculture practice. However, the effect of growth under restricted feeding on quality of fish grown to the same size remains poorly unknown. In this study, yellow catfish were reared by feeding them at three different feeding rates such as low (FR-L group), medium (FR-M group) and high (apparent satiation) ration levels for 82, 58 and 34 days to reach the same size, respectively. After this period, the growth performance, whole-body amino acid composition and fatty acid profile of the fish were evaluated. Feeding restriction led to significant decreases in specific growth rate and feed conversion efficiency. Significant increases in dry matter, protein and energy contents were observed in fish under both restricted feeding treatments. FR-L-treated fish had significantly lower whole-body essential amino acid contents, and significantly higher Ala and non-essential amino acid contents compared to fish fed to satiation. Fish in restricted feeding treatments had significantly higher n-3 PUFA and n-3/n-6 ratio compared to the fish fed to satiation. The present results indicate that from a quality perspective, there are certain advantages in improvement of whole-body composition and enrichment of n-3 PUFA associated with the restricted feeding of yellow catfish at slow growth rate.

摘要:
This study was conducted to evaluate the effects of biofloc on the species and quantity of plankton in pond water. There were four treatments in the experiment: the control, no supplementation; treatment I, pond water supplemented with glucose; treatment II, pond water supplemented with both glucose and Bacillus subtilis; and treatment III, pond water supplemented with Bacillus subtilis only. Water quality, plankton, and biofloc formation were monitored every five days. Results showed that biofloc formation was enhanced by adding glucose to the pond water. The addition of glucose together with Bacillus subtilis shortened the formation time of mature bioflocs by about five days than the supplementation with glucose only. In ponds supplemented with glucose and those supplemented with glucose + bacillus, the ammonia nitrogen, and nitrite nitrogen content in the water column decreased, and the number of cyanobacteria and green algae were significantly inhibited, while the number of zooplankton such as Rotatoria increased during the experimental period. © 2017, Israeli Journal of Aquaculture - Bamidgeh. 摘要:
This study was designed to evaluate the effect of the replacement of fish oil (FO) by soybean oil (SO) on growth performance, liver lipid peroxidation, and biochemical composition in juvenile Chinese sucker, Myxocyprinus asiaticus. Fish (13.7±0.2g) in triplicate were fed five experimental diets in which 0% (FO as control), 40% (SO40), 60% (SO60), 80% (SO40), and 100% (SO100) FO were replaced by SO. The body weight gain of fish fed SO40, SO60, or SO80 diet was similar to FO group, but diets that have 100% soybean oil as dietary lipid significantly reduced fish growth (P<0.05). Although the level of SO resulted in increasing crude lipid content of the liver, the level of SO did not significantly alter the hepatosomatic index (HSI). Indicators of peroxidation, such as vitamin E (VE) and thiobarbituric acid-reactive substance (TBARS) contents, were changed as increasing dietary SO. It was shown that the inclusion of SO in the diets increased VE concentrations, but reduced TBARS in the liver and total cholesterol (T-CHO) in the plasma. Linoleic acid (LA) and linolenic acid (LNA) significantly increased in fish liver fed diets that contained SO, but eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and the ratio n-3/n-6 were significantly reduced by the inclusion of dietary SO (P<0.05). Our results indicated that the inclusion of SO increased the hepatic VE content and reduced lipid peroxidation in fish. However, diet containing 100% SO as dietary lipid could reduce growth performance. Thus, we recommended that 40–80% SO can be used as dietary lipid to replace FO for juvenile Chinese sucker. © 2017, Springer Science+Business Media Dordrecht.

Abstract There is growing interest in glutamate as a functional amino acid in nutrition and health. This study was conducted to determine whether glutamate precursor α-ketoglutarate (AKG) could alleviate lipopolysaccharide (LPS)-induced liver injury in young pigs. Twenty-four piglets were randomly assigned to the control, LPS, or LPS + AKG group. Piglets in the control and LPS groups were fed a basal diet, whereas piglets in the NAC group were fed the basal diet supplemented with 1 % AKG. On days 10, 12, 14, and 16 of the trial, piglets in the LPS and LPS + AKG groups received intraperitoneal administration of LPS (80 μg/kg BW), whereas piglets in the control group received the same volume of saline. On day 16 of the trial, blood samples were collected 3 h after LPS or saline injection. Twenty-four hours post-administration of LPS or saline (on day 17 of the trial), piglets were killed to obtain liver for analysis. Dietary AKG supplementation alleviated LPS-induced histomorphological abnormalities and mitigated LPS-induced increases in aspartate aminotransferase (AST) activity and AST/ALT ratio (P < 0.05). Compared with the LPS group, dietary supplementation with AKG decreased plasma glutamate concentration, while increasing hepatic concentrations of glutamate, glutamine, leucine, asparagine, lysine, alanine, serine, threonine, valine, and phenylalanine (P < 0.05). LPS challenge dramatically increased concentrations of malondialdehyde and decreased glutathione peroxidase activity in the liver. Additionally, LPS challenge enhanced concentrations of AMP and total protein, as well as RNA/DNA and total protein/DNA ratios, while decreasing hepatic ADP concentrations. These adverse effects of LPS challenge were ameliorated by AKG supplementation. Collectively, dietary AKG supplementation provides a new means to ameliorate LPS-induced liver injury by increasing anti-oxidative capacity and improving energy metabolism in young pigs. © 2015 Springer-Verlag Wien.

BACKGROUND: Genetically modified (GM) foods have caused much controversy. Construction of a food-grade delivery system is a desirable technique with presumptive impact on industrial applications from the perspective of bio-safety. The aim of this study was to construct a food-grade delivery system for Saccharomyces cerevisiae and to study the expression of monellin from the berries of the West African forest plant Dioscoreophyllum cumminsii in this system. RESULTS: A food-grade system for S. cerevisiae was constructed based on ribosomal DNA (rDNA)-mediated homologous recombination to enable high-copy-number integration of the expression cassette inserted into the rDNA locus. A copper resistance gene (CUP1) was used as the selection marker for yeast transformation. Because variants of transformants containing different copy numbers at the CUP1 locus can be readily selected after growth in the presence of elevated copper levels, we suggest that this system would prove useful in the generation of tandemly iterated gene clusters. Using this food-grade system, a single-chain monellin gene was heterologously expressed. The yield of monellin reached a maximum of 675 mg L-1. CONCLUSION: This system harbors exclusively S. cerevisiae DNA with no antibiotic resistance genes, and it should therefore be appropriate for safe use in the food industry. Monellin was shown to be expressed in this food-grade delivery system. To our knowledge, this is the first report so far on expression of monellin in a food-grade expression system in S. cerevisiae. © 2015 Society of Chemical Industry.

生物絮团是养殖水体中以好氧微生物为主体的有机体和无机物,经生物絮凝形成的团聚物,由细菌、浮游动植物、有机碎屑和一些无机物质相互絮凝组成。通过对生物絮团研究的发展过程、絮团的结构特征、絮凝机理、影响絮团的形成因素以及生物絮团技术在水产养殖应用中存在的问题进行了综述,为生物絮团技术在水产领域中的进一步研究和应用具有一定的指导意义。

试验在两种不同营养水平的饲料配方中分别添加0 mg/kg,50 mg/kg,100 mg/kg的酵母培养物益宁易SCP,制成6种试验饲料饲喂均重20.39±2.19g草鱼6周,评估了饲料中不同益宁易添加量对草鱼生长性能和肝、肠功能的影响.结果表明:在普通饲料中添加50 mg/kg的益宁易SCP可以促进草鱼的生长,保护草鱼的肝脏不受损害,并能够显著促进草鱼肠道的发育,增加其绒毛高度;而在降低营养标准的低价饲料中,添加量要需增加到100 mg/kg,效果才会更明显.

分别在饲料中添加不同含量的枯草(Bacillus subtilis)芽孢(A组实测芽孢数量为2.6×10~7 cfu/g,B组为4.4×10~6 cfu/g,空白组为0),饲喂静水水族箱中100 g左右的草鱼(Ctenopharyngodon idellus)10 d,测定饲喂枯草芽孢后草鱼粪便、养殖水体中芽孢数量和水质的变化。结果表明:(1)9 d内,水体中芽孢数量A组和B组在前6 d时不断增加,在6 d时达到稳定,其中A组和B组稳定时的含量分别为2.22×10~6 cfu/g和3.98×10~5 cfu/g。(2)10 d内,草鱼粪便干物质中的芽孢数量与饲料中的枯草芽孢含量相比,A组和B组粪便中的枯草芽孢损失率分别为89.76%~90.71%和83.83%~86.84%。(3)草鱼饲喂枯草芽孢后,A组和B组粪便中排出的枯草芽孢对水体中亚硝酸盐和氨氮有降低的趋势,但各组之间差异不显著(P>0.05),对化学需氧量(COD)没有影响。

采用A和B两种商品饲料加工工艺,分别在饲料配方中添加9.10×107cfu/g和1.05×107cfu/g的枯草芽孢,依次记为A1、A2、B1、B2,系统研究了不同加工工艺和芽孢添加量对饲料中芽孢数量的影响;并进一步研究了B1、B2在密闭和敞开两种不同保存方式下,饲料中芽孢数量的变化规律。研究结果表明:(1)在A和B生产工艺条件下,随着加工工段的先后,饲料芽孢的数量呈现递减趋势,A1和A2的成品芽孢存活率分别为37.47%和41.90%,B1和B2的成品存活率分别为28.35%和41.90%。(2)以低剂量饲料芽孢数量为横坐标,高剂量饲料芽孢数量为纵坐标,则在各工段中高剂量组与低剂量组呈现较好的线性回归关系,这表明枯草芽孢的存活率不受添加剂量的影响。(3)在100 d的保存期内,B1和B2饲料中芽孢数量与第1 d相比,芽孢的数量没有显著变化。

凡纳滨对虾饲料中菜籽粕替代鱼粉适宜比例的研究

以饲料中添加36%鱼粉作为对照组,用菜籽粕分别替代饲料配方中10%~50%的鱼粉,配成5种等氮等能试验饲料,饲喂凡纳滨对虾,研究不同替代比例对凡纳滨对虾生长性能和虾体氨基酸组成的影响.试验结果表明,饲养56d后,随着菜籽粕替代鱼粉比例的增加,对虾的质量增加率下降,但仅50%替代比例试验组的质量增加率为1542.74%,显著低于对照组(2066.07%,P<0.05);质量增加率(y)和菜籽粕实际添加量(x)的回归方程为y=-0.8674x2+12.917x +2031.7(r2=0.9081).通过拐点法计算得出的菜籽粕最适添加量为7.37%(换算成替代比例为12.70%),在99%的置信水平下,3.17%~22.48%的替代是完全可行的.菜籽粕替代鱼粉饲喂凡纳滨对虾,对其存活率、饵料系数、虾体氨基酸总量和鲜味氨基酸含量影响不大,当替代比例达到50%时,其存活率、虾体蛋氨酸和精氨酸含量才显著低于对照组,饵料系数显著高于对照组.综合凡纳滨对虾质量增加率、存活率、饵料系数、虾体氨基酸含量等各项指标,凡纳滨对虾饲料中用菜籽粕替代22.48%鱼粉完全可行.

摘要:
Alpha-ketoglutarate (AKG) is an intermediate of the Krebs cycle which bridges amino acid metabolism with glucose oxidation in animals. Of particular interest is the conversion of AKG into glutamate by mitochondrial glutamate dehydrogenase in the gastrointestinal tract where glutamate has multiple physiological functions (including regulation of cell function, neurotransmission, and gastric emptying). Additionally, AKG stimulates the initiation of catabolism of branched-chain amino acids (BCAA) via BCAA transaminase in enterocytes. Oxidation of AKG also provides large amounts of ATP and modulates cellular redox state in the small intestine. Translating the basic research into practice, results of recent studies indicate that dietary supplementation with AKG alleviates oxidative stress and injury in intestinal mucosal cells, while improving intestinal mucosal integrity and absorption of nutrients in endotoxin-challenged pigs. The beneficial effects of AKG are associated with increased activation of the mTOR signaling pathway and net protein synthesis. Thus, AKG is a novel and promising supplement in diets to improve intestinal health in animals and possibly humans.
