摘要:
肥胖是一种代谢性炎症反应性疾病,肥胖动物体内脂肪组织巨噬细胞含量明显增高,且极化表型由抗炎的M2型转化为促炎的M1型。近年来,研究者发现在与脂肪组织相连的交感神经周围存在一种特殊的巨噬细胞,即交感神经相关性巨噬细胞,具有摄取和分解去甲肾上腺素的作用,导致脂肪分解障碍引起肥胖。我们对脂肪组织巨噬细胞、交感神经相关性巨噬细胞的生物学特性及其与肥胖的联系进行了综述。

摘要:
目的:分析发表在中国知网(China National Knowledge Infrastructure,CNKI)上的关于我国胃癌患者照顾者研究的发展动态,为今后的深入研究提供参考。方法:以中国知网(CNKI)为数据来源,选择知网下的文献数据库,在输入检索条件下的"主题"字段输入"胃癌"或者"照顾者",在"篇名"字段输入"照顾者"或者"家属","主题"与"篇名"之间用"并且"连接,选择"模糊"词频进行检索,对检索到的第一作者为中国大陆的文献进行分析。结果:共检索出65篇文献,选择符合纳入条件的21篇文献进行研究分析。结论:来自全国各地的研究者都越来越关注胃癌患者的照顾者,其中以描述性研究居多。在今后的研究中应该进一步拓展相关研究的广度与深度,探讨某种合适又高效的干预方式对照顾者进行干预后以达到提高患者的生活质量、促进医患关系和谐的目的。

摘要:
肌肉是维持人体生命活动的重要物质.它是由肌原纤维相互包裹形成肌束,通过肌膜固定支持并保持一定的形态,继而肌钙蛋白和肌球蛋白控制粗肌丝和细肌丝相互滑动,使肌小节缩短从而控制肌肉的收缩来维持人体的运动.肌肉损伤一般发生于外力作用下的挫伤或间接外力作用下的拉伤,但其他如生物、化学因素也可以导致肌肉损伤.肌肉损伤在一定程度上可以修复.目前,肌肉损伤与修复相关的研究已经向组织的超微结构、分子机制和基因水平拓展,主要致力于研究如何从细胞和基因水平上促进肌肉的再生,以及寻找安全有效的生物替代物来修复受伤的肌肉组织.本综述介绍了中外学者对肌肉结构、功能的认识,重点阐述了肌肉损伤后引发炎症反应,卫星细胞被激活促进肌纤维再生,以及这一系列过程中众多基因调控和蛋白的参与的分子机制,为后续研究提供参考.

呼吸道传染病的流行病学及预防控制工作一直是医学界关注的热点问题。2019年12月,我国湖北省武汉市发生不明原因的肺炎疫情,武汉市成为全球关注的焦点。目前新型冠状病毒感染的肺炎尚在疫情发病的初期,发病还存在较强的不确定性,尚无特异性治疗。目前人类已知的冠状病毒,根据病毒的血清学特点和核苷酸序列分类分为α、β、γ和δ属,引起人感染的冠状病毒有HCoV-229E、HCoV-OC43、SARS-CoV、HCoV-NL63、HCoV-HKU1和MERS-CoV 6种[1]。

摘要:
Acute respiratory distress syndrome (ARDS) is associated with a mortality of 45%. Our previous research indicated that anti-vascular endothelial growth factor (VEGF) could maintain the normal structure and function of the respiratory barrier. However, systemic application of VEGF antagonists would lead to animal death. This study attempts to study the targeted drug delivery for ARDS. In this study, we used soluble fms-like tyrosine kinase-1 (sFlt)-targeted ultrasound microbubbles to antagonize the effect of VEGF on lung tissue. Ninety male BALB/c mice were randomly assigned to 6 groups: phosphate buffer saline (PBS) group (PBS+PBS); blank group (PBS+empty microbubbles); lipopolysaccharide (LPS) group (LPS+PBS); ARDS group (LPS+empty microbubbles); control group (PBS+sFlt microbubbles); and treatment group (LPS+sFlt microbubbles). After administration of LPS or PBS in the corresponding groups, the sFlt-targeted microbubbles or empty microbubbles were injected into the blood circulation. Then the lungs were irradiated with ultrasound, which ruptured the drug-loaded microbubbles and helped release drugs to the lung tissues targeted. The lung injury score, lung wet/dry ratio (W/D), liver and kidney functions, and the mortality of the mice in all groups were investigated at the predetermined time point. The difference in mortality between groups was examined by Fisher test. Other data were analyzed by one-way analysis of variance (ANOVA). A value of P<0.05 indicates that the difference was significant. The results showed that the PaO2 levels were normal in the PBS group, the blank group, and the control group. The LPS group and ARDS group showed significant hypoxia. PaO2 was improved significantly in the treatment group. The lung injury score and W/D were normal in the PBS group, the blank group, and the control group. The lung injury score and W/D increased significantly in the LPS group and ARDS group and decreased in the treatment group (P<0.05). The mortality rate of the ARDS model was 60% (95% confidence interval 47.5%–72.5%), and that with sFlt-targeted microbubbles was significantly lower at only 40% (95% confidence interval 27.5%–52.5%, P<0.05). It was concluded that anti-VEGF with sFlt targeted ultrasound microbubbles attenuated the lung injury and ultimately reduced the 7-day mortality effectively. It might be a suitable therapeutic tool for the treatment of ARDS. © 2020, Huazhong University of Science and Technology.

强化一维持健康教育对冠心病患者自我管理影响的研究

目的 探讨强化-维持健康教育对社区冠心病患者疾病相关知识水平、自我管理、再入院率的影响.方法 2017年3-12月采用方便抽样法选取武汉市某社区冠心病患者60例为研究对象,对其实施强化-维持健康教育,以冠心病相关知识问卷评价患者知识水平,冠心病自我管理行为量表(CSMS)评估患者自我管理能力,监测患者再入院率.结果 强化-维持干预后患者疾病相关知识总分及各维度得分明显高于干预前(P<0.05),干预后冠心病相关知识问卷中诱发因素、药物知识2个维度得分低于干预前(P<0.05),其他维度比较差异无统计学意义(P>0.05);强化-维持干预后CSMS总分及各维度得分优于干预前(P<0.05);强化-维持干预后再入院率明显低于干预前(P<0.05).结论 强化-维持健康教育能有效提高并维持社区冠心病患者疾病相关知识水平,从而提升其自我管理能力,降低其再入院率.

[目的]分析60例肝硬化患者家属的疾病不确定感水平,探索治疗性沟通系统对肝硬化患者家属疾病不确定感的干预效果。[方法]采用自行设计的问卷和疾病不确定感家属量表对60例肝硬化患者家属进行调查;根据患者住院号尾数的奇数、偶数分为对照组、观察组各30例。对照组以常规健康教育方式,观察组以治疗性沟通系统方式。[结果]肝硬化患者家属疾病不确定感总分为78.00±10.69。观察组疾病不确定感、不明确性维度、缺乏澄清维度得分均低于对照组,2组比较差异有统计学意义;观察组干预后疾病不确定感及各维度得分均降低,2组比较差异有统计学意义。[结论]肝硬化患者家属疾病不确定感处于中等水平,治疗性沟通系统有助于降低其疾病不确定感水平。

目的 探讨瑞舒伐他汀预处理对大鼠局灶性脑缺血再灌注后大脑中动脉血管平滑肌细胞(VSMCs)中炎性细胞因子白细胞介素(IL)-1β、IL-6、肿瘤坏死因子α(TNF-α)表达的影响以及可能的机制.方法 36只健康成年SD大鼠,雌雄不限,随机分为假手术组,局灶性脑缺血再灌注组,瑞舒伐他汀预处理组,每组12只.大脑中动脉缺血2h再灌注24 h后,Real-time PCR及Western blotting法检测大鼠大脑中动脉VSMCs中IL-1β、IL-6和TNF-α以及核因子κB(NF-κB) mRNA和蛋白的表达.结果 再灌注24 h后,模型组VSMCs IL-1β、IL-6以及TNF-αmRNA和蛋白的表达明显增加,而给予瑞舒伐他汀预处理后,可明显抑制大脑中动脉VSMCs中IL-1β、IL-6和TNF-αmRNA和蛋白的高表达,同时也可发现,VSMCs中NF-κB mRNA和蛋白的表达也明显减少.结论 瑞舒伐他汀预处理可有效抑制大脑中动脉VSMCs中IL-1β、IL-6和TNF-α mRNA和蛋白的表达,从而减轻缺血再灌注后炎症反应对脑组织的进一步损伤作用.瑞舒伐他汀预处理对IL-1β、IL-6及TNF-α的作用可能与其减少VSMCs中NF-κB的表达有关.

摘要:
Pregnant women, a vulnerable population, can be exposed to a variety of environmental metals that may adversely affect their health at elevated concentrations. Moreover, environmental exposure and risk disparities are associated with several factors such as sociodemographic characteristics, pregnancy complications, and nutritional supplementation indicators. Our aim was to investigate whether and how the urinary metal concentrations vary according to these factors in a large pregnancy and birth cohort. Urinary levels of seven metals including manganese, zinc, selenium, arsenic, cadmium, thallium, and lead were assessed in 7359 participants across Wuhan City in China. In all, 7359 urine samples from the third trimester were analyzed and 15 sociodemographic characteristics, pregnancy complications, and nutritional supplementation indicators were assessed: maternal age, household income, multivitamin supplements during pregnancy, etc. Concentrations of Zn, As, and Cd were detected in all urine samples; Mn, Se, Tl, and Pb were detected in 95.3%, 99.8%, 99.8%, and 99.9% urine samples, respectively. The geometric mean (GM) of creatinine-adjusted urinary Mn, Zn, Se, As, Cd, Tl, and Pb concentrations were 1.42, 505.44, 18.24, 30.49, 0.64, 0.55, and 3.69 mug/g. Factors that associated with environmental metals concentration were as follows: (1) Urinary Zn and Cd concentrations increased with maternal age. (2) Pregnant women with higher education had lower Cd concentrations. (3) Pregnant women with folic acid supplementation had lower Mn and with multivitamin supplementation had higher Se and lower Tl. Our results demonstrated that the maternal age, education level, folic acid supplementation, and multivitamin supplementation were significantly associated with environmental metal concentrations. Furthermore, studies are recommended to explore the influence of diet on biological metals concentrations in more detail.

Farnesoid X receptor (FXR) is an important regulator of glucose and lipid homeostasis. However, the exact role of FXR in diabetes remains to be fully elucidated. The present study examined the effects of chenodeoxycholic acid (CDCA), an agonist of FXR, on metabolism profile in a rat model of type 2 diabetes mellitus (T2DM). Male Wistar rats (8-week-old; n=40) were randomized into the following four groups (n=10): Untreated control, CDCA-treated, T2DM, and CDCA-treated T2DM. To establish the T2DM model, the rats were fed a high-fat diet (HFD) for 4 weeks and received a single low-dose intraperitoneal injection of streptozotocin (30 mg/kg), followed by an additional 4 weeks of HFD feeding. CDCA was administrated (10 mg/kg/d) intraperitoneally for 10 days. Reverse transcription-quantitative polymerase chain reaction and western blotting assays were performed to determine the RNA and protein expression of FXR, phosphoenolpyruvate carboxykinase, G6Pase, proliferator-activated receptor-γ coactivator-1 and short heterodimer partner in rat liver tissue. The results revealed that FXR activation by CDCA did not reduce body weight, but it lowered the plasma levels of fasting glucose, insulin and triglycerides in the T2DM rats. CDCA administration reversed the downregulation of the mRNA and protein expression of FXR in the T2DM rat liver tissue samples. Furthermore, treatment with CDCA reduced the mRNA and protein expression levels of phosphoenolpyruvate carboxykinase, glucose 6-phosphatase and peroxisome proliferator-activated receptor-γ coactivator-1 in the liver tissue samples of the T2DM rats. By contrast, CDCA treatment increased the mRNA and protein expression levels of short heterodimer partner in the liver tissue samples of the T2DM rats. In conclusion, FXR agonist treatment induces beneficial effects on metabolism in the rat T2DM model. In conclusion, the present study indicated that the FXR agonist may be useful for the treatment of T2DM and hypertriglyceridemia.

Mycoplasmas are a group of microbes that can cause human diseases. The mycoplasmal lipoprotein p37 promotes cancer metastasis, at least in part, by interacting with EGFR. In this study, we show that the p37 lipoprotein binds another member of the EGFR family, HER2, through the HER2 extracellular domain. The binding of p37-HER2 promotes phosphorylation of HER2 and activates the downstream signaling molecule Erk1/2. Because the HER2 signaling pathway contributes to breast tumor metastasis, our results imply that the mycoplasmal lipoprotein p37 may also be involved in breast cancer metastasis. This study contributes to our understanding of mycoplasmal lipoprotein p37 function and its potential involvement in tumorigenesis. © 2016
