现阶段果园土鸡的养殖是一种具有生态性的生态养殖方式,在果园中养鸡不仅可以对果园中闲置空间与资源进行充分的利用,同时也可以将鸡和果树量两种资源进行结合,互相发挥作用,实现互惠互利,进而达到鸡与果实的双丰收.但是在实际土鸡养殖的过程中,依然存在一些关于养殖的问题,并对果园土鸡养殖的效率造成一定的影响.为了解决其中存在的问题,文章中对果园土鸡养殖的新技术进行了探讨,以提升土鸡养殖的效率,并为广大同仁提供养殖方面的信息.

摘要:
The aim of the study was to determine the effects of solid-state fermentation of cottonseed meal (FCSM) with Candida utilis on broiler growth performance and toxicity in comparison to unfermented cottonseed meal (CSM) or soyabean meal (SBM, control). In total 150 one-day-old male Cobb 400 broilers were randomly assigned to 3 dietary treatments with 5 replications of 10 birds each. Animals were fed control diet with SBM, diet with CSM : SBM (1:1) and diet with FCSM : SBM (1:1) until 21 day of life. Fermentation decreased free gossypol content in CSM from 583.40 to 191.70 mg . kg(-1), and degraded total gossypol from 5830.19 to 3882.91 mg . kg(-1) CSM. In comparison to the control diet, FCSM : SBM (1:1) diet did not affect (P > 0.05) the average daily growth and feed conversion ratio, while CSM : SBM (1:1) diet decreased (P < 0.05) these parameters. However, FCSM : SBM (1:1) diet, similarly to CSM : SBM (1:1) diet, increased (P < 0.05) the relative weight of liver and the serum activity of alanine aminotransferase, and decreased (P < 0.05) the relative weight of thymus in comparison to the control diet. In conclusion, residual gossypol or/and degradation products of gossypol in FCSM may still be hepatotoxic and immunotoxic for broilers when SBM is half-replaced by FCSM in diet (155 g . kg(-1)), although the contents of free gossypol and total gossypol have been markedly reduced and no adverse effect on growth performance was observed.

摘要:
This paper gave detailed description of the fine structure of Apiosoma piscicola Blanchard 1885. These parasites were collected from the fry of Tachysurus fulvidraco during parasite surveys of fish at Jiangxia Fish Hatchery in Wuhan, Hubei province in May 2011. Its pellicle, peristomial apparatus, nuclei, trochal band and scopula were described, respectively. The relationship between these peritrich ciliates and their hosts was well discussed herein. The scopula of A. piscicola did not adhere directly to the surface of the host epithelial cells, but by means of a secreted adhesive pad of material according to the present study. Besides, no ultrastructural damage to host epithelial cells could be detected in our work. We concluded that these sessilids should be ectocommensals rather than ectoparasites, but not always harmless. In addition, their great variability of body shape and non-strict host specificity were also discussed.

摘要:
Milk is synthesized by mammary epithelial cells of lactating mammals. The synthetic capacity of the mammary gland depends largely on the number and efficiency of functional mammary epithelial cells. Structural development of the mammary gland occurs during fetal growth, prepubertal and post-pubertal periods, pregnancy, and lactation under the control of various hormones (particularly estrogen, growth hormone, insulin-like growth factor-I, progesterone, placental lactogen, and prolactin) in a species- and stage-dependent manner. Milk is essential for the growth, development, and health of neonates. Amino acids (AA), present in both free and peptide-bound forms, are the most abundant organic nutrients in the milk of farm animals. Uptake of AA from the arterial blood of the lactating dam is the ultimate source of proteins (primarily β-casein and α-lactalbumin) and bioactive nitrogenous metabolites in milk. Results of recent studies indicate extensive catabolism of branched-chain AA (leucine, isoleucine and valine) and arginine to synthesize glutamate, glutamine, alanine, aspartate, asparagine, proline, and polyamines. The formation of polypeptides from AA is regulated not only by hormones (e.g., prolactin, insulin and glucocorticoids) and the rate of blood flow across the lactating mammary gland, but also by concentrations of AA, lipids, glucose, vitamins and minerals in the maternal plasma, as well as the activation of the mechanistic (mammalian) target rapamycin signaling by certain AA (e.g., arginine, branched-chain AA, and glutamine). Knowledge of AA utilization (including metabolism) by mammary epithelial cells will enhance our fundamental understanding of lactation biology and has important implications for improving the efficiency of livestock production worldwide.

摘要:
本试验旨在研究亚麻籽油对脂多糖(LPS)刺激仔猪肝脏Toll样受体4(TLR4)和核苷酸结合寡聚化结构域(NOD)信号通路关键基因表达的影响。选取24头断奶仔猪,按体重相近原则随机分为4个组,分别为对照组、LPS组、2.5%亚麻籽油组(2.5%亚麻籽油+LPS)、5.0%亚麻籽油组(5.0%亚麻籽油+LPS),每组6个重复,每个重复1头猪,试验期21 d。试验组注射100 μg/kg体重的LPS,对照组注射等量的生理盐水。注射LPS或生理盐水4 h后屠宰仔猪,取肝脏,测定TLR4和NOD信号通路关键基因及相关炎性介质的mRNA表达水平。结果表明:1)LPS刺激显著提高了肝脏肿瘤坏死因子-α(TNF-α)、环氧酶2(COX2)、热休克蛋白70(HSP70)的mRNA相对表达量(P<0.05),2.5%亚麻籽油可显著降低COX2、TNF-α的mRNA相对表达量(P<0.05),5.0%亚麻籽油可显著降低TNF-α的mRNA相对表达量(P<0.05)。2)LPS刺激显著提高了肝脏TLR4、髓样分化因子88(MyD88)、白细胞介素-1受体相关激酶1(IRAK1)、NOD1、NOD2、受体互作蛋白2(RIPK2)、核因子-κB(NF-κB)的mRNA相对表达量(P<0.05);2.5%亚麻籽油可显著降低NOD1、NOD2的mRNA相对表达量(P<0.05),有降低RIPK2 mRNA相对表达量的趋势(0.05≤P<0.10);5.0%亚麻籽油可显著降低NOD2的mRNA相对表达量(P<0.05)。这表明LPS刺激导致仔猪发生炎症反应,亚麻籽油可能通过抑制NOD信号通路进而缓解肝脏炎症反应。

摘要:
The complete mitochondrial DNA sequence of Triplophysa venusta was determined using a PCR-based method in our present study. The genome is totally 16 574 bp in length and contains 13 protein-coding genes, 2 ribosomal RNA, 22 transfer RNA genes, and 1 non-coding control region. The overall base composition of T. venusta is A 27.8%, T 26.9%, C 26.9%, and G 18.4%, with an A + T bias of 54.7%. This complete mitogenome of T. venusta provides a basic data for studies on species identification, molecular systematics, and conservation genetics. The phylogenetic trees consistently showed that the Family Cobitidae was divided into three major clades: the subfamily Botiidae, the subfamily Nemaeheilidae, and the subfamily Cobitinae. Cobitinae and Nomaeheilidae formed a main Loach clade, with Botiidae clustered at the basal. © 2015 Taylor & Francis.

摘要:
In this study, the anti-inflammatory effects and mechanisms of baicalin on LPS-induced NLRP3 inflammatory pathway were investigated in piglet mononuclear phagocytes (control, LPS stimulation, LPS stimulation + 12.5 μg/ml baicalin, LPS stimulation + 25 μg/ml baicalin, LPS stimulation + 50 μg/ml baicalin and LPS stimulation + 100 μg/ml baicalin). The levels of reactive oxygen species (ROS), the secretion levels of IL-1β, IL-18 and TNF-α, mRNA expression levels of IL-1β, IL-18, TNF-α and NLRP3, as well as the protein levels of cleaved caspase-1 p20 were significantly increased after LPS-challenge in vitro. However, LPS stimulation did not influence apoptosis-associated speck-like protein and caspase-1 mRNA levels, which are also components of the NLRP3 inflammasome. Baicalin at 50 μg/ml and 100 μg/ml could inhibit the production of ROS, TNF-α, IL-1β and IL-18, and down-regulate mRNA expression of IL-1β, IL-18, TNF-α and NLRP3, as well as expression of cleaved caspase-1 p20. These results showed that the anti-inflammatory effects of baicalin occurred via the regulation of the release of ROS and mRNA expression of NLRP3. The anti-inflammatory activity of baicalin could be related to the suppression of NLRP3 inflammasome pathway under LPS stimulation. © 2016 The Author(s).

摘要:
为了探讨中药渣在母猪饲粮中添加的可行性,试验选用2~4胎次、预产期相近的健康二元母猪60头,随机分为对照组、中药渣组和发酵中药渣组,比较研究发酵前和发酵后中药渣对母猪繁殖性能和仔猪发育的影响。结果表明:与对照组相比,发酵中药渣组产仔数、产活仔数和断奶平均日增重均有所增加,断奶窝增重和哺乳仔猪腹泻率显著增加,母猪背膘损失有所降低;中药渣组木乃伊数略有增加,仔猪腹泻率明显增加。由上可见,饲粮中添加发酵中药渣制剂虽然在短期内造成仔猪一定程度的腹泻,但可提高母猪繁殖性能和仔猪生长性能。

摘要:
Dietary supplementation with N-acetylcysteine (NAC) has been reported to improve intestinal health and treat gastrointestinal diseases. However, the underlying mechanisms are not fully understood. According to previous reports, NAC was thought to exert its effect through glutathione synthesis. This study tested the hypothesis that NAC enhances enterocyte growth and protein synthesis independently of cellular glutathione synthesis. Intestinal porcine epithelial cells were cultured for 3 days in Dulbecco's modified Eagle medium containing 0 or 100 μM NAC. To determine a possible role for GSH (the reduced form of glutathione) in mediating the effect of NAC on cell growth and protein synthesis, additional experiments were conducted using culture medium containing 100 μM GSH, 100 μM GSH ethyl ester (GSHee), diethylmaleate (a GSH-depletion agent; 10 μM), or a GSH-synthesis inhibitor (buthionine sulfoximine, BSO; 20 μM). NAC increased cell proliferation, GSH concentration, and protein synthesis, while inhibiting proteolysis. GSHee enhanced cell proliferation and GSH concentration without affecting protein synthesis but inhibited proteolysis. Conversely, BSO or diethylmaleate reduced cell proliferation and GSH concentration without affecting protein synthesis, while promoting protein degradation. At the signaling level, NAC augmented the protein abundance of total mTOR, phosphorylated mTOR, and phosphorylated 70S6 kinase as well as mRNA levels for mTOR and p70S6 kinase in IPEC-1 cells. Collectively, these results indicate that NAC upregulates expression of mTOR signaling proteins to stimulate protein synthesis in enterocytes independently of GSH generation. Our findings provide a hitherto unrecognized biochemical mechanism for beneficial effects of NAC in intestinal cells. © 2015 Springer-Verlag Wien.

Lymphocyte count is an important phenotypic metric that has been reported to be related to the individual antiviral capacity of pigs and other mammals. To date, aside from information regarding several genes and pathways, little is known about the mechanism by which gene expression affects variation in lymphocyte count. In this work, we investigated the lymphocyte count variation after poly I:C stimulation and compared the transcriptomes of pigs with large and small differences of lymphocyte counts before and after poly I:C stimulation. Pigs with large and small differences of lymphocyte counts were designated as extreme response (ER) and moderate response (MR) pigs respectively. Lymphocyte counts in all animals were observed to decline after poly I:C stimulation. Transcriptomic analysis identified 1121 transcripts (981 differentially expressed genes) in MR pigs and 1045 transcripts (904 differentially expressed genes) in ER pigs. We found that the majority of the differentially expressed genes were involved in both innate and adaptive immune responses. However, the innate immune response of ER pigs was more rapid than that of MR pigs. Results indicated that the activation of signaling pathways associated with cell death, cytotoxicity and apoptosis may contribute to the poly I:C-induced decrease of lymphocyte counts in the periphery. Moreover, the differential expression patterns of chemokines and FAS either totally or partially provided an interpretation for the different degrees of decrease in the lymphocyte counts between MR and ER pigs. Overall, our study will provide further understanding of the molecular basis for the antiviral capacity of pigs and other mammals. © 2015 Stichting International Foundation for Animal Genetics.

摘要:
The objective of this study was to investigate the effects of microencapsulated Enterococcus fecalis (MEF) and the extract of Camellia oleifera seed (ECOS) on growth performance, morphological parameters and gene expressions related to barrier functions of the intestinal mucosa in broiler chickens. 160-day-old Arbor Acres male broilers were randomly allotted to 4 treatments with 8 replicates of 5 chicks in a 2. ×. 2 factorial arrangement, evaluating two levels of MEF (0 and 10. g/kg), and two levels of ECOS (0 and 300. mg/kg). The results showed that there was no significant interaction effect of MEF and ECOS on growth performance, morphometric parameters of jejunum and ileum, and mRNA expression of MUC2, occludin and claudin-1 in jejunum and ileum mucosa. Compared with other groups, diet supplemented with MEF significantly improved FCR from days 21 to 42, and significantly improved ADG from days 0 to 42. Dietary MEF significantly increased villus height and villus height to crypt depth ratio in jejunum and ileum, the concentrations of sIgA, IL-2, IL-6, TNF-α, and significantly up-regulated the gene expression of sIgA, MUC2, occludin and claudin-1 in jejunum and ileum mucosa. The interaction effect of MEF and ECOS was significant on the concentrations of sIgA, IL-2, IL-6, TNF-α in jejunum mucosa on day 42 and on the concentrations of sIgA, IL-2, TNF-α in ileum mucosa on day 42. These results indicated that MEF can improve growth performance and enhance intestinal barrier functions by improving its morphological development, increasing cytokine level, and up-regulating the gene expressions of sIgA, MUC2, occludin and claudin-1. But ECOS had slightly beneficial effects on growth performance and intestinal barrier functions. © 2015.
