发酵木薯酒精糟对生产猪生产性能及营养物质消化的影响

摘要:
试验研究不同水平的发酵木薯酒精糟对生长猪生产性能的影响.选择200头初始体重约(26.35±0.43) kg的生长猪,随机分为5个处理组,每组4个重复,每个重复10头猪.5个处理组分别为:A组为对照组,饲喂基础日粮;B组、C组和D组分别为发酵木薯酒精糟用量为2%、6%和15%的配合日粮组,E组则为用发酵木薯酒精糟直接替代6%基础日粮的混合日粮组.结果表明,B组和C组的日增重较A组分别提高了6.45%(P>0.05)和4.84%(P>0.05),各试验组料肉比均低于对照组,分别降低了7.48%、9.06%、5.12%和0.39%.日粮中添加发酵木薯酒精糟后有效改善了营养物质消化率,B组、C组、D组及E组干物质消化率较对照组分别提高了8.62%(P<0.05)、7.50%(P>0.05)、4.75%(P>0.05)和5.26%(P>0.05);粗蛋白消化率较对照组分别提高了11.43%(P<0.05)、9.50%(P<0.05)、9.61%(P<0.05)和10.61%(P<0.05);粗脂肪消化率较对照组分别提高了15.66%(P<0.05)、15.86%(P<0.05)、12.98%(P<0.05)和14.10% (P<0.05);钙的消化率则较对照组分别提高了10.46%(P<0.05)、6.50%(P>0.05)、6.93%(P>0.05)和10.60%(P<0.05).B组、C组、D组及E组较A组的养殖效益均有所提高,每千克饲料的增重成本分别降低了0.683、0.833、0.523元和0.238元,其中使用处理C组饲料的效益最好.本试验结果说明,生长猪日粮中发酵木薯酒精糟的适宜添加量为2%-6%,能有效提高猪的生产性能和养殖效益.

胸膜肺炎放线杆菌(Actinobacillus pleuropneumoniae,APP)是一种能够引起猪传染性胸膜肺炎的细菌病原体,它给养猪业带来巨大的经济损失。外膜蛋白W(Omp W)在细菌的生命活动中起着重要作用,大量研究表明Omp W与细菌的感染、耐药性以及分子调节等都有着紧密联系,因而阐明ompW基因对胸膜肺炎放线杆菌调节机制对于该疾病的防控具有重要意义。以胸膜肺炎放线杆菌ompW基因为研究对象,成功构建自杀性质粒p EM-Omp W,为构建胸膜肺炎放线杆菌Omp W突变株以及探究ompW基因对胸膜肺炎放线杆菌分子调节机制提供了基础。

摘要:
microRNA分子在调节宿主细胞应对细菌感染的过程中起重要作用。为研究miR-194b-3p对马链球菌兽疫亚种(SeZ)粘附猪肾上皮细胞(PK15)能力的影响,本实验将miR-194b-3pagomir和antagomir分别转染PK15细胞,检测SeZ对PK15细胞粘附能力的变化。结果表明:在PK15细胞中转染miR-194b-3p agomir,SeZ C55138粘附率显著下降;转染miR-194b-3p antagomir, SeZ C55138粘附率显著上升,确定了miR-194b-3p能够显著抑制SeZC55138对PK15细胞的粘附。同时对其机制进行研究,显示PK15细胞表面CD44是miR-194b-3p的一个调控靶基因, miR-194b-3p通过调节CD44的表达参与影响了上述粘附过程。本实验为深入理解SeZ与PK15细胞的互作机制提供了线索。

摘要:
Clostridium perfringens is the causative pathogen of avian necrotic enteritis. Lactobacillus spp. are well-characterized probiotics with anti-microbial and immune-modulatory activities. In the present study, we investigated the effects of L. acidophilus and L. fermentum on the growth, α-toxin production and inflammatory responses of C. perfringens. In in vitro culture experiments, both lactobacilli inhibited the growth of C. perfringens (P < 0.01), accompanied with a decrease in pH (P < 0.01). Supernatants from lactobacilli cultures also suppressed the growth of C. perfringens during 24 h of incubation (P < 0.01), but this inhibitory effect disappeared after 48 h. Both lactobacilli decreased the α-toxin production of C. perfringens (P < 0.01) without influencing its biomass, and even degraded the established α-toxin (P < 0.01). Lower environmental pH reduced the α-toxin production as well (P < 0.01). Preincubation with L. acidophilus decreased the attachment of C. perfringens to cells (P < 0.01) with the cell cytotoxicity being unaffected. Both lactobacilli pretreatment reduced the up-regulation of proinflammatory factors, peptidoglycan (PGN) receptors and nuclear factor kappa B (NF-κB) p65 in C. perfringens-challenged chicken intestinal epithelial cells (P < 0.05). In conclusion, L. acidophilus and L. fermentum inhibited the pathological effects of C. perfringens in vitro conditions. © 2017 Guo, Liu, Zhang, Li, Li, Ding and Guo.

试验旨在研究包被肉桂油对肉鸡非特异性免疫功能的影响.选用360只1日龄科宝肉鸡,随机分为6个处理组,每组6个重复,每个重复10只鸡.对照组饲喂玉米-豆粕型基础日粮,正对照组在基础日粮中添加50 mg/kg的黄霉素,试验组分别添加50、100、200和300 mg/kg的包被肉桂油(以肉桂醛计),试验期为42 d.结果显示:与对照组相比,50和200 mg/kg试验组14日龄血液中单核细胞的比例上调(P<0.05);试验组21日龄血清中溶菌酶的含量增加且呈线性正相关(R2=0.824),试验组42日龄血清中溶茵酶的含量增加且呈线性正相关(R2=0.625);试验组42日龄外周血淋巴细胞的LPS刺激指数上调(P<0.001).与正对照组相比,50 mg/kg试验组14日龄血液中单核细胞的比例上调(P<0.05);试验组21日龄血清中溶菌酶的含量增加(P<0.05);试验组42日龄外周血淋巴细胞的LPS刺激指数上调(P<0.05).结果表明:日粮中添加包被肉桂油有利于促进肉鸡外周血B淋巴细胞的增殖,增强非特异性免疫功能.

Pork is one of the most economical sources of animal protein for human consumption. Meat quality is an important economic trait for the swine industry, which is primarily determined by prenatal muscle development and postnatal growth. Identification of the molecular mechanisms underlying skeletal muscle development is a key priority. MicroRNAs (miRNAs) are a class of small noncoding RNAs that have emerged as key regulators of skeletal muscle development. A number of muscle-related miRNAs have been identified by functional gain and loss experiments in mouse model. However, determining miRNA-mRNA interactions involved in pig skeletal muscle still remains a significant challenge. For a comprehensive understanding of miRNA-mediated mechanisms underlying muscle development, miRNAome analyses of pig skeletal muscle have been performed by deep sequencing. Additionally, porcine miRNA single nucleotide polymorphisms have been implicated in muscle fiber types and meat quality. The present review provides an overview of current knowledge on recently identified miRNAs involved in myogenesis, muscle fiber type and muscle protein metabolism. Undoubtedly, further systematic understanding of the functions of miRNAs in pig skeletal muscle development will be helpful to expand the knowledge of basic skeletal muscle biology and be beneficial for the genetic improvement of meat quality traits.

摘要:
Clostridium perfringens encodes at least two different quorum sensing (QS) systems, the Agr-like and LuxS, and recent studies have highlighted their importance in the regulation of toxin production and virulence. The role of QS in the pathogenesis of necrotic enteritis (NE) in poultry and the regulation of NetB, the key toxin involved, has not yet been investigated. We have generated isogenic agrB-null and complemented strains from parent strain CP1 and demonstrated that the virulence of the agrB-null mutant was strongly attenuated in a chicken NE model system and restored by complementation. The production of NetB, a key NE-associated toxin, was dramatically reduced in the agrB mutant at both the transcriptional and protein levels, though not in a luxS mutant. Transwell assays confirmed that the Agrlike QS system controls NetB production through a diffusible signal. Global gene expression analysis of the agrB mutant identified additional genes modulated by Agrlike QS, including operons related to phospholipid metabolism and adherence, which may also play a role in NE pathogenesis. This study provides the first evidence that the Agr-like QS system is critical for NE pathogenesis and identifies a number of Agr-regulated genes, most notably netB, that are potentially involved in mediating its effects. The Agr-like QS system thus may serve as a target for developing novel interventions to prevent NE in chickens. © 2017 American Society for Microbiology. All Rights Reserved.

This study determined whether N-acetylcysteine (NAC) could improve intestinal function through phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR), epithelial growth factor receptor (EGFR), toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-κB), adenosine 5′-monophosphate-activated protein kinase (AMPK), and type I interferon (IFN) signaling pathways in a piglet model of lipopolysaccharides (LPS) challenge. Thirty-two piglets (24-day-old) were randomly allocated to one of four treatments, with eight replicates per treatment and one piglet per replicate. The experiment consisted of four treatments in a 2 × 2 factorial arrangement with two diets (supplemented with 0 or 500 mg NAC/kg diet) and saline or LPS administration. On day 20 of the trial, piglets in the LPS and LPS + NAC groups were intraperitoneally injected with 0 (saline) or 100 μg LPS/kg BW. Blood samples were obtained at 3 h and intestinal mucosae were collected at 6 h post LPS or saline injection. The growth performance was not affected by dietary NAC. LPS induced intestinal dysfunction, as indicated by: (1) reductions in the small-intestinal glutathione concentrations and plasma d-xylose levels; (2) elevations in plasma diamine oxidase activity, mucosal MMP3 mRNA levels and caspase-3 protein abundance; (3) reduced the activities of the small-intestinal mucosal maltase, sucrase and lactase. The adverse effects of LPS on porcine intestinal function and redox status were mitigated by NAC supplementation through the activation of multiple signaling pathways involving PI3K/Akt/mTOR, EGFR, TLR4/NF-κB, AMPK, and type I IFN. Our findings provide novel mechanisms for beneficial effects of NAC in protecting the intestine from inflammation in animals. © 2017, Springer-Verlag Wien.

Supplementation of branched-chain amino acids (BCAA) has been demonstrated to promote skeletal muscle mass gain, but the mechanisms underlying this observation are still unknown. Since the regulation of muscle mass depends on a dynamic equilibrium (fasted losses-fed gains) in protein turnover, the aim of this study was to investigate the effects of BCAA supplementation on muscle protein synthesis and degradation in fed/fasted states and the related mechanisms. Fourteen 26-(Experiment 1) and 28-day-old (Experiment 2) piglets were fed reduced-protein diets without or with supplemental BCAA. After a four-week acclimation period, skeletal muscle mass and components of anabolic and catabolic signaling in muscle samples after overnight fasting were determined in Experiment 1. Pigs in Experiment 2 were implanted with carotid arterial, jugular venous, femoral arterial and venous catheters, and fed once hourly along with the intravenous infusion of NaH 13 CO 3 for 2 h, followed by a 6-h infusion of [1- 13 C]leucine. Muscle leucine kinetics were measured using arteriovenous difference technique. The mass of most muscles was increased by BCAA supplementation. During feeding, BCAA supplementation increased leucine uptake, protein synthesis, protein degradation and net transamination. The greater increase in protein synthesis than in protein degradation resulted in elevated protein deposition. Protein synthesis was strongly and positively correlated with the intramuscular net production of α-ketoisocaproate (KIC) and protein degradation. Moreover, BCAA supplementation enhanced the fasted-state phosphorylation of protein translation initiation factors and inhibited the protein-degradation signaling of ubiquitin-proteasome and autophagy-lysosome systems. In conclusion, supplementation of BCAA to reduced-protein diet increases fed-state protein synthesis and inhibits fasted-state protein degradation, both of which could contribute to the elevation of skeletal muscle mass in piglets. The effect of BCAA supplementation on muscle protein synthesis is associated with the increase in protein degradation and KIC production in the fed state. © 2016 by the author; licensee MDPI, Basel, Switzerland.

为了解鄂西北地区规模化猪场所用猪瘟疫苗的类别及免疫效果,对鄂西北地区38家规模化猪场猪瘟疫苗的使用情况进行调研,采集829份血清进行了猪瘟抗体水平检测.结果表明:38家规模猪场中有26家(68.4%)使用政府供应疫苗,12家(31.6%)使用商品疫苗;猪瘟细胞苗和脾淋苗在猪场的使用比例分别为71.1%和28.9%;猪场母猪猪瘟疫苗使用剂量高于推荐剂量的1.2 ~6倍,生长育肥猪使用的剂量为推荐剂量的1~4倍,猪场使用政府供应的疫苗时剂量高于商品疫苗;20家猪场群体免疫合格(52.6%),18家猪场群体免疫不合格(47.4%);受检的829份血清中,阴性血清158份(19.1%),阳性血清609份(73.5%).说明鄂西北地区猪瘟疫苗以政府供应猪瘟细胞苗为主,不同养殖场免疫效果差别大,需要规范免疫程序,加强免疫抗体水平检测,以降低该地区猪瘟疫病风险.

发酵杏鲍菇菌糠对育肥猪生长性能和表观消化率的影响

本试验旨在研究发酵杏鲍菇菌糠对育肥猪生长性能和表观消化率的影响,并确定发酵杏鲍菇菌糠在育肥猪日粮中的适宜添加量。选取20头平均体重为20 kg左右的杜长大三元杂交育肥猪,采用单因子设计随机分配到4个处理组:空白对照组、5%菌糠组、10%菌糠组和15%菌糠组。每组5个重复,每个重复1头育肥猪。将发酵后的杏鲍菇菌糠作为生长育肥猪的饲料原料,取代不同量的麸皮,按照NRC(2012)配制猪的基础日粮。试验

摘要:
代谢葡萄糖可准确地评定饲料的营养价值以及不同年龄和生理阶段的动物对葡萄糖的需要量.本研究旨在通过测定不同日粮外源代谢葡萄糖值,建立仔猪日粮外源代谢葡萄糖摄入量的估测方法.试验选择(25±2)日龄体重6~7 kg的杜×长×大三元杂交安装回肠末端“T”型瘘管断奶仔猪20头,随机分为4个处理,每个处理饲喂不同淀粉(或葡萄糖)含量的日粮.测定不同日粮的仔猪外源代谢葡萄糖水平,同时以日粮外源代谢葡萄糖含量为因变量Y,分别以日粮中淀粉、粗脂肪、粗蛋白和粗纤维含量为自变量,建立估测日粮外源代谢葡萄糖水平的多元回归公式.结果表明:4种断奶仔猪日粮的外源代谢葡萄糖水平分别为209.37、258.02、394.07 g/kg和420.19g/kg.估测日粮外源代谢葡萄糖水平的多元回归公式为Y=-39.84+9.09X1+10.61X2(R2=0.9999,X1和X2分别表示日粮中淀粉和粗纤维含量).

本试验旨在研究Toll样受体4(Toll-like receptor-4,TLR4)信号通路负调控因子在断奶仔猪不同组织的分布情况。试验选取12头杜×长×大断奶仔猪,屠宰,取脾脏、胸腺、下丘脑、垂体、肾上腺、空肠、回肠、肝脏、背最长肌和腓肠肌等组织。应用实时荧光定量PCR技术测定TLR4信号通路负调控因子在仔猪不同组织中的mRNA表达水平,包括单免疫球蛋白IL-1相关蛋白(Single immunoglobulin IL-1 R-related molecule,SIGIRR)、细胞因子信号传导抑制因子1(Suppressor ofcytokine signaling 1,SOCS1)、防辐射105 (Radioprotective 105,RP105)和Toll反应蛋白(Toll-interacting protein,Tollip)。结果表明:TLR4信号通路负调控因子在所测定的10个组织中均有一定程度的表达,但是组织分布规律存在着一定的差异。总体看来,主要在肝脏、回肠和脾脏中的表达量较高,在胸腺、垂体和空肠中的表达量居中,在其它组织中的表达量较低。综上所述,TLR4信号通路负调控因子在不同组织中表达差异较大,可能是负调控因子在TLR4信号传导通路中结合的传导因子与结合位点的不同,以及机体对负调控因子的适应机制不同,造成不同组织中负调控因子的mRNA表达量不同。

本试验旨在研究脂多糖(LPS)刺激对仔猪核苷酸结合寡聚化结构域蛋白(NODs)信号通路关键基因及其负调控因子m RNA表达的影响。选取12头杜×长×大断奶仔猪,分成2个处理组,每个处理组6个重复。试验组注射100μg/kg体重的LPS,对照组注射等量的生理盐水。注射LPS或生理盐水4 h后,屠宰仔猪,取空肠、回肠、肝脏、背最长肌和腓肠肌,应用实时荧光定量PCR技术测定NODs信号通路关键基因及其负调控因子m RNA表达水平,包括NOD1、NOD2、受体相互作用蛋白激酶2(RIPK2)、核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)、矢车菊苷β1(ACAP1)和Erbb2相互作用蛋白(ERBB2IP)m RNA表达水平。结果表明:与对照组相比,LPS刺激显著提高了肝脏NOD1、NOD2、RIPK2、NF-κB和TNF-α,背最长肌NOD2、RIPK2和TNF-α,腓肠肌NOD2、RIPK2、NF-κB和TNF-α,空肠RIPK2和TNF-α的m RNA表达量(P<0.05),LPS刺激有提高回肠RIPK2和NF-κB m RNA表达量的趋势(P<0.10);同时,LPS刺激显著降低了空肠ACAP1和ERBB2IP,回肠和肝脏ACAP1的m RNA的表达量(P<0.05)。这表明,LPS激活仔猪肠道、肝脏和肌肉组织中NODs信号通路关键基因的表达,而抑制其负调控因子ACAP1和ERBB2IP的表达。

摘要:
The metabolic roles of mitochondria go far beyond serving exclusively as the major producer of ATP in tissues and cells. Evidence has shown that mitochondria may function as a key regulator of skeletal muscle fiber types and overall well-being. Maintaining skeletal muscle mitochondrial content and function is important for sustaining health throughout the lifespan. Of great importance, β-hydroxy-β-methylbutyrate (HMB, a metabolite of L-leucine) has been proposed to enhance the protein deposition and efficiency of mitochondrial biogenesis in skeletal muscle, as well as muscle strength in both exercise and clinical settings. Specifically, dietary supplementation with HMB increases the gene expression of peroxisome proliferator-activated receptor gamma co-activator 1-alpha (PGC-1α), which represents an upstream inducer of genes of mitochondrial metabolism, coordinates the expression of both nuclear- and mitochondrion-encoded genes in mitochondrial biogenesis. Additionally, PGC-1α plays a key role in the transformation of skeletal muscle fiber type, leading to a shift toward type I muscle fibers that are rich in mitochondria and have a high capacity for oxidative metabolism. As a nitrogen-free metabolite, HMB holds great promise to improve skeletal muscle mass and function, as well as whole-body health and well-being of animals and humans. © 2015 Springer-Verlag.

摘要:
The brush-border membrane (BBM) of enterocytes is responsible for the digestion and absorption of nutrients and ions in the small intestine. To identify the BBM proteins involved in epithelial cell maturation along the crypt-villus axis, enterocytes were sequentially isolated from the villus tip to the crypt of the jejunum from 21-day-old suckling piglets. After preparation of BBM vesicles, we detected 194 proteins in the jejunal epithelial cells by isobaric tags using relative and absolute quantification (iTRAQ) techniques. Of these, 56 BBM proteins were differentially expressed along the crypt-villus axis. During differentiation, the expression of proteins related to digestion and absorption of nutrients was primarily downregulated at the upper, middle villus, or crypt compared to the villus tip, while expression of proteins related to structural and enzyme regulator proteins was largely upregulated. We verified the differences in Na+/K+-transporting ATPase, galectin-3, and an intestinal-type fatty acid binding protein by western blot or immunochemical analysis. Identification of BBM-associated proteins helps enhance our understanding of digestion and absorption in piglets and other mammals, including humans.

摘要:
Haemophilus parasuis (H. parasuis) is the causative agent of Glässer’s disease, a severe membrane inflammation disorder. Previously we showed that Baicalin (BA) possesses anti-inflammatory effects via the NLRP3 inflammatory pathway in an LPS-challenged piglet model. However, whether BA has anti-inflammatory effects upon H. parasuis infection is still unclear. This study investigated the anti-inflammatory effects and mechanisms of BA on H. parasuis-induced inflammatory responses via the NF-κB and NLRP3 inflammasome pathway in piglet mononuclear phagocytes (PMNP). Our data demonstrate that PMNP, when infected with H. parasuis, induced ROS (reactive oxygen species) production, promoted apoptosis, and initiated transcription expression of IL-6, IL-8, IL-10, PGE2, COX-2 and TNF-α via the NF-κB signaling pathway, and IL-1β and IL-18 via the NLRP3 inflammasome signaling pathway. Moreover, when BA was administrated, we observed a reduction in ROS production, suppression of apoptosis, and inhibition of the activation of NF-κB and NLRP3 inflammasome signaling pathway in PMNP treated with H. parasuis. To our best knowledge, this is the first example that uses piglet primary immune cells for an H. parasuis infection study. Our data strongly suggest that BA can reverse the inflammatory effect initiated by H. parasuis and possesses significant immunosuppression activity, which represents a promising therapeutic agent in the treatment of H. parasuis infection.

为解决直接添加植酸酶到水产饲料中不利于植酸酶充分发挥作用的问题。试验参考吉富罗非鱼饲料配方,以豆粕、棉粕和菜粕为原料,按比例为15??9??7混合,采用植酸酶进行体外水解,在单因素试验的基础上,以水解效率为指标,采用正交试验研究加酶量(A)、反应温度(B)、加水量(C)和酶解时间(D)4个不同因素对植酸酶体外水解植物性饲料原料效率的影响。结果表明,各因素对植酸酶体外水解植物性饲料原料效率的影响程度依次是A>B>C>D。其最佳酶解工艺条件为:加酶量为2000 U/kg底物,反应温度35℃,加水量50%,酶解时间为3 h。在此条件下得出的植酸酶水解效率为42.16%。

试验旨在研究凝结芽孢杆菌的发酵条件、发酵液的浓缩方式、载体吸附等工艺,并评价其产品性能.采用10 L全自动发酵罐发酵凝结芽孢杆菌,采用有机膜和矿物质吸附剂对发酵液进行浓缩、吸附.采用体外消化试验、体外药敏试验、耐高温试验和产品储存试验评价产品性能.结果表明:①凝结芽孢杆菌制剂的最佳工艺参数为搅拌速度200 r/min,通气量350 L/h,浓缩设备为有机陶瓷膜过滤设备,矿物质吸附剂与发酵液比例为1∶1.②产品在85℃处理10 min存活率为71%,经胃液消化后存活率为86.8%,过肠液后存活率为69%;产品在25 ℃和35℃储存30 d后仍保持50%作用的存活率,储存60 d后菌种数依旧可以到达108数量级.由此可见,此产品具有耐酸、耐胆盐、耐高温和易于储存等优良特性.
