摘要:
The condition of the motor oil in civilian cars is difficult to monitor; hence, we propose a method to evaluate the degree of degradation of motor oil using an on-board diagnostic (OBD) system. Three civilian cars and four motor oils (containing mineral oils and synthetic oils) were subjected to five groups of road tests under urban traffic and high-way conditions. The operation information, oil service time, mileage, engine operation time, idle time of the engine, and number of start-ups of the engine were obtained using the proposed OBD system. Physiochemical properties and changes in the components of motor oils during road tests were analyzed in laboratory. The theoretical model of the comprehensive indicators of driving parameters and oil properties were established. The proposed method was successfully applied to different cars, motor oils, and operating conditions in road tests. All the theoretical models had high accuracy and precision. Herein, we provide a method to monitor the oil condition with real-time driving parameters and provide a reference for end users to change their motor oil reasonably.

摘要:
In this study, a multi-phenol antioxidant 2,4,6-tris(3,5-di-tert-butyl-4-hydroxyphenylthio)-1,3,5-triazine (THA) was synthesized. The thermal stability of THA and its oxidation resistance in triisodecyl trimellitate (TDTM) ester oil were evaluated compared with several commercial phenol antioxidants. Further investigations on the oxidation resistance and tribological performance of THA with dinonyl diphenylamine (Am) and ZDDP composites were conducted to determine the additive compatibility in TDTM. The results indicate that the THA possesses higher thermal stability and more effectiveness of oxidation resistance than the commercial phenol antioxidants. The synergistic anti-oxidative effect is confirmed for the THA/Am mixtures in TDTM ester oil. However, the addition of THA alone or THA/Am mixtures shows no improvement for the tribological performance of TDTM. Excitingly, the THA/Am/ZDDP composite additives not only have the superior oxidation resistance, but exhibit the effective friction and wear reductions. It suggests the excellent compatibility of the synthesized THA antioxidant with Am and ZDDP additives in TDTM ester oil. © 2019, Springer Science+Business Media, LLC, part of Springer Nature.

摘要:
In this study, it is tried to establish a theoretical model based on oil oxidation stability to evaluate the motor oil life. Five civilian cars and five motor oils were taken to eight road tests. The road tests contained two experiments taken in two test cars with the same car model, synthetic motor oil and test time, and three experiments taken in one test car with the same mineral oil to study the reliability and repeatability of the proposed method. The change characteristics of oxidation onset temperature, total acid number, and kinematic viscosity of test oils were analyzed. A theoretical model was established with the oxidation onset temperature and column chromatography method. The results showed the established theoretical model has the potential to quantitatively determine the remaining service time and operation mileage of the test oils with high accuracy and reliability. The test synthetic oils are able to operate approximately 11,000–13,000 km in 534–686 days in the typical urban traffic condition of China. The test mineral oils are capable of operating approximately 8200 km with 40 km/day or capable of operating 5723 km in 410 days. The proposed approach can provide some reference for end-users to drain their motor oil reasonably.<br/> ©IMechE 2018.

Due to the lack of a Panax japonicus var. major reference genome, we assembled a reference transcriptome from P. japonicus C. A. Mey transcriptome sequencing data, and 203 283 unigenes were obtained. In this study, with the assistance from the Trinity, Bowtie2 and SAMtools softwares, 218 m465 single nucleotide polymorphisms (SNPs) were identified by mapping the Illumina sequences to the reference transcriptome. The SNP forms included 126 262 transformations and 92 203 transversions. A large number of SNP loci were associated with triterpenoid saponin synthesis: 54 SNPs were associated with cytochrome P450, one with glycosyl transferase and 94 with the biosynthesis of the triterpenoid saponin backbone. © 2019 The Royal Society of Chemistry.

GroEL, a member of the Hsp60 class molecular chaperones, has been extensively studied by general molecular biological method not only on its ATPase activities but also on refolding mechanism. Here, a C80 isothermal calorimeter was applied to understand the molecular mechanism of GroEL-assisted refolding. The power–time curves were obtained from different reaction systems, and the integral heat was calculated and analyzed. The ATPase activity of HSP60 was proven to be very stable at high temperature and had a high temperature dependence. In ATP hydrolysis experiments, we found that ATP hydrolysis catalyzed by GroEL was an exothermic reaction, in which divalent cations especially magnesium cation played an important role. And the ATP concentration also influences the ATP hydrolysis rate. Moreover, GroEL can assist in refolding thermodenatured xylanase with the presence of ATP and co-chaperone GroES, and the refolding process was demonstrated to be endothermic reaction, indicating that non-native proteins interacted with hydrophobic apical domains of GroEL, mainly hydrophobic interaction. The thermodynamic parameters presented in this study illustrated the work mechanism of GroEL by microcalorimetry and have important implications for further study of other interaction mechanism. © 2018, Akadémiai Kiadó, Budapest, Hungary.

Although the reason is not clear, mileage is commonly recommended as an indicator to evaluate the oil drain interval of a car. This paper tries to investigate whether the mileage is the most suitable indicator to evaluate the oil drain interval based on scientific experiments and data analysis method. Three civilian cars and motor oils (contained mineral oil, semisynthetic oil, and synthetic oil) were subjected to road tests under urban traffic condition. The real-time driving parameters comprising speed, mileage, idle time, engine operation time, and number of starts were obtained by on-board diagnostic system. Synchronous oil samples were collected about every 30 days. The physicochemical properties and component changes of experimental oils were analyzed in laboratory. The effects of driving parameters on oil properties were studied with the improved grey relational analysis method. The results show that the influencing factors of driving parameters on motor oil degradation were different in different experimental cars. The influence of mileage on oil degradation was not obvious. The idle time of engine, engine operation time, and number of starts were the three most significantly influential factors on oil deterioration after comprehensive analysis of the three experimental cars.

摘要:
The 5′ untranslated region (5′UTR) plays a key role in post-transcriptional regulation, but interaction between nucleotides and directed evolution of 5′UTRs as synthetic regulatory elements remain unclear. By constructing a library of synthesized random 5′UTRs of 24 nucleotides in Saccharomyces cerevisiae, we observed strong epistatic interactions among bases from different positions in the 5′UTR. Taking into account these base interactions, we constructed a mathematical model to predict protein abundance with a precision of R 2 = 0.60. On the basis of this model, we developed an approach to engineer 5′UTRs according to nucleotide sequence activity relationships (NuSAR), in which 5′UTRs were engineered stepwise through repeated cycles of backbone design, directed screening, and model reconstruction. After three rounds of NuSAR, the predictive accuracy of our model was improved to R 2 = 0.71, and a strong 5′UTR was obtained with 5-fold higher protein abundance than the starting 5′UTR. Our findings provide new insights into the mechanism of 5′UTR regulation and contribute to a new translational elements engineering approach in synthetic biology. © 2018 American Chemical Society.

As oxidation stability of engine oil was hard to evaluate in field, this paper tried to propose a field test method to determine the oxidation onset temperature (OOT) of gasoline engine oil. A theoretical model of grey average value of oil spot image and OOT was established by using spot test method, image processing, and Pressure Differential Scanning Calorimetery method. The oil sample unknown OOT was taken to spot test and image processing in field to obtain the grey average value. The OOT can be calculated by bringing the grey average value to the established theoretical model. This approach was successfully used for OOT determination of 4 different kinds of engine oils with good accuracy and precision. The absolute error of mineral oils was less than 4.52°C, and synthetic oils were less than 1.2°C. This convenient, low-cost, and reliable method can achieve OOT of engine oil in field determination and provide reference for engine oil drain interval evaluation. 摘要:
Magnetic ZnFe<inf>2</inf>O<inf>4</inf>/ZnO (ZFO/ZnO) multi-porous nanotubes have been first fabricated via a facile electrospinning and subsequent calcination process. A series of ZFO/ZnO photocatalysts with different ZFO molar content and morphologies are also obtained by varying the molar ratio of Zn/Fe metal salt and its dosage. The morphology, composition, crystal structure and specific surface area of achieved photocatalysts are systematically examined. TEM images demonstrate ZFO/ZnO-3 multi-porous nanotubes possess perfect 1D nanotube profile with hierarchical pores. HRTEM images confirm the formation of ZFO/ZnO heterojunctions. DRS spectra show that ZFO/ZnO-3 multi-porous nanotubes exhibit an enhanced absorption both in UV and visible-light region. PL spectra and photocurrent responses of ZFO/ZnO-3 multi-porous nanotube demonstrated that the photogenerated electrons and holes are effectively separated. Above all, ZFO/ZnO-3 multi-porous nanotubes photocatalysts with a larger specific surface area of 57.79 m<sup>2</sup> g<sup>−1</sup>exhibit the best photocatalytic efficiency of 99% after 150 min under the solar irradiation for the decolorization of RhB. Moreover, ZFO/ZnO photocatalysts not only possess magnetic separation property, but also keep a relatively high photocatalytic efficiency even after four cycles, which is beneficial for practical application. In addition, both the formation and potential photocatalytic mechanisms of ZFO/ZnO-3 multi-porous nanotubes are proposed in detail. ©2016 Elsevier B.V.

Maintaining an appropriate concentration of dissolved oxygen in aqueous solution is critical for efficient operation of a bioreactor, requiring sophisticated engineering design and a system of regulation to maximize oxygen transfer from the injected air bubbles to the cells. Bacterial hemoglobins are oxygen-binding proteins that transfer oxygen from the environment to metabolic processes and allow bacteria to grow even under microaerophilic conditions. To improve the oxygen utilization efficiency of cells and overcome the oxygen shortage in bioreactors, the gene coding for the Campylobacter jejuni single domain hemoglobin (CHb) gene was artificially synthesized and functionally expressed under the control of inducible expression promoters P-T7 and P-vgh in Escherichia coli. The effects of the recombinants P-T7-CHb and P-vgh-CHb on cell growth were evaluated in aerobic shake flasks, anaerobic capped bottles and a 5-L bioreactor, and a pronounced improvement in cell biomass was observed for CHb-expressing cells. To determine the growth curves, CHb gene expression, and CHb oxygen-binding capacity of specific recombinants with different promoters, we determined the time course of CHb gene expression in the two recombinants by semi-quantitative RT-PCR and CO differential spectrum assays. Based on the growth patterns of the two recombinants in the bioreactor, we proposed different recombinant types with optimal performance under specific culture conditions.

Bacteria hemoglobin could bind to the oxygen, transfer it from the intracellular microenvironment to the respiration process and sustain the energy for the metabolism and reproduction of cells. Heterologous expression of bacteria hemoglobin gene could improve the capacity of the host on oxygen-capturing and allow it to grow even under microaerophilic condition. To develop a system based on hemoglobin to help bacteria cells overcome the oxygen shortage in fermentation, in this study, Campylobacter jejuni truncated hemoglobin (CtrHb) gene was synthesized and expressed under the control of constitutive expression promoters P2 and PSPO1-II in Escherichia coli. As showed by the growth curves of the two recombinants P2-CtrHb and PSPO1-II-CtrHb, constitutive expression of CtrHb improved cell growth under aerobic shaking-flasks, anaerobic capped-bottles and bioreactor conditions. According to the NMR analysis, this improvement might come from the expression of hemoglobin which could boost the metabolism of cells by supplying more oxygen to the respiratory chain processes. Through semi-quantitative RT-PCR and CO differential spectrum assays, we further discussed the connection between the growth patterns of the recombinants, the expression level of CtrHb and oxygen binding capacity of CtrHb in cells. Based on the growth patterns of these recombinants in bioreactor, a possible choice on different type of recombinants under specific fermentation conditions was also suggested in this study. © 2015 Elsevier Inc.

摘要:
The contents of three main functional components including amino acids, microelements, and polysaccharides were determined in submerged-fermentation mycelium and compared with data of fruit body in Grifola frondosa. The results showed that the contents of three main functional components in mycelium were higher than or as same as the fruit body. This indicated that submerged-fermentation mycelium was not only better in output and period than traditional culture method, but also better in nutrient components than fruit body. So the process of submerged-fermentation has very bright future in mycelium production of G. frondosa. © Springer-Verlag Berlin Heidelberg 2014.

A selective, sensitive and accurate high-performance liquid chromatographic- tandem mass spectrometry (HPLC-MS/MS) method for simultaneous determination of iloperidone and its two active metabolites, P88 and P95, in human plasma has been first developed and validated. The analytes and internal standard (IS), pioglitazone hydrochloride, were extracted from human plasma via liquid-liquid extraction with ethyl acetate and separated on a CAPCELL PAK C18 MG IIIcolumn (150 mm × 2.0 mm, 5 μm) set at 40. °C. The mobile phase was acetonitrile: 5 mM ammonium formate containing 0.3% formic acid (pH 4.8) (25:75, v/v), with a flow rate of 0.35. mL/min. The mass spectrometer was operated in multiple reaction monitoring (MRM) mode using the transitions m/ z 427.2. →. 261.2 for iloperidone, m/. z 429.1. → 261.1 for P88 and P95, and m/. z 357.1. →. 133.7 for the I.S. (pioglitazone hydrochloride). The method was validated to be linear over the concentration range of 10-10,000. pg/mL for iloperidone and P88, 50-15,000. pg/mL for P95. The mean recoveries were more than 78.88%, and the intra- and inter-day precisions were less than 10.24% and accuracy was -5.78 to 5.40%, which indicated that the quantitative method was reliable and accurate. The validated method has been successfully applied to a human pharmacokinetic study of iloperidone and two active metabolites, P88 and P95, after oral administration of 4. mg iloperidone tablets in 12 healthy Chinese volunteers. © 2013 Elsevier B.V.

Denitrification is a major biological process to reduce nitrate to molecular nitrogen (N2). In shallow eutrophic lakes, this process can remove the largest portion of fixed nitrogen and plays an important role in self-purification of this ecosystem. To understand the structure of denitrifying communities in a shallow eutrophic lake, denitrifier communities in four sub-lakes of East Lake in Wuhan, China, were explored by restriction fragment length polymorphisms (RFLP) analysis and sequencing of nirS gene clone libraries. nirS is a functional marker gene for denitrification encoding cytochrome cd 1-containing nitrite reductase, which catalyzes the reduction of nitrite to nitric oxide. Both RFLP fingerprints clustering analysis and phylogeny analysis based on the amino acid sequences of NirS revealed that NirS-type communities in East Lake sediment could be roughly divided into three clusters. Cluster I accounted for 74-82 % of clones from the moderately eutrophic sub-lakes Tuan, Tang Ling, and Guo Zheng. Cluster II accounted for 76 % of the communities in hypertrophic sub-lake Miao Lake and cluster III as a minor group (7 % of the total), mainly presented in Miao Lake. Phylogenetic analysis revealed that cluster I was related to the reference clones from a broad range of ecological environments, and clusters II and III were more phylogenetically related to the reference clones from entrophic environments. Canonical correspondence analysis indicated that total nitrogen, total phosphate, total organic carbon, and NH4-N and NO2-N were important environmental factors affecting the dispersion of NirS-type denitrifier in the sediments. Cluster I showed a weak relationship with the nutrient content, while cluster II and III were positively related with the nutrient content. Principal coordinates analysis indicated that NirS-type communities from Tuan Lake, Tang Ling Lake, and Guo Zheng Lake sediments were divergent from those found in river, estuary sediment, and forest soil but similar to communities in constructed wetland sediment despite large geographic distances. The communities from the hypertrophic sub-lake Miao Lake deviated from other sub-lakes and the reference communities and clustered independently. Our results support the argument that environmental factors regulate the composition and distribution of the functional bacterial groups. © 2013 Springer Science+Business Media New York.

SaPIN2a, the proteinase inhibitor of nightshade (Solanum americanum), has been proposed to regulate proteolysis in phloem development. In this study, we expressed and characterized recombinant SaPIN2a in Escherichia coli. Purified recombinant SaPIN2a (rSaPIN2a) had a strong inhibitory effect on serine proteinase chymotrypsin (IC50 36.1 nmol/L), but its inhibitory activities toward trypsin (IC50 398.6 nmol/L) and especially toward subtilisin (IC50 5004 nmol/L) were low. It did not inhibit cysteine proteinase papain and aspartic proteinase cathepsin D. rSaPIN2a was a competitive inhibitor of trypsin, and a noncompetitive inhibitor of chymotrypsin and subtilisin.

摘要:
In this paper, the influence of different strains and mediums on the yield of submerged-fermentation mycelium of Grifola frondosa was studied. Five strains of Grifola frondosa, including Gr1, Gr2, Gr3, Gr4 and Gr5, were submerged-fermentation cultured and the growing situation of mycelium was measured and analyzed. The results showed that the growth of Gr1 and Gr5 were better than other 3 strains and corn powder medium was more useful to the growth of Grifola frondosa than PDA medium and basic medium.

The biological effect of CdTe quantum dots (QDs) on Halobacterium halobium R1 (H. halobium R1) growth was analyzed by a microcalorimetric technique. By using a TAM air eight channels microcalorimeter, the thermogenic curves of H. halobium R1 growth were obtained at 37 °C. To analyze the results, the maximum heat power (Pm) and the growth rate constants (k) were determined, which showed that they were correlated to the concentration of QDs. The addition of quantum dots caused a gradual increase of Pm and k at low concentrations of QDs, and a conspicuous decrease at high concentrations. For confirmation, the turbidity (OD600) and respiratory rate at different concentrations of QDs were studied. The morphology of H. halobium R1 cells both in the absence and presence of QDs was examined by transmission electron microscopy (TEM). The results of these studies were corroborated with ones derived from microcalorimetry. In this work, the mechanism of cytotoxicity of QDs was explored through fluorescence spectroscopy, inductively coupled plasma mass spectrometry (ICP-MS) and microcalorimetry. It was clear that metabolic mechanism of H. halobium R1 growth was changed by the addition of QDs. To the best of our knowledge, the thermokinetics and toxicology of CdTe QDs against H. halobium R1 were obtained for the first time by microcalorimetry. © 2010 IOP Publishing Ltd.
