Association between sensation, perception, negative socio-psychological factors and cognitive impairment

摘要:
Background: Evidence has suggested that sensation and socio-psychological factors may be associated with cognitive impairment separately in older adults. However, the association between those risk factors and cognitive impairment is still unknown.Objective: To investigate the association between sensation, perception, negative sociopsychological factors, and cognitive impairment in institutionalized older adults.Methods: From two public aged care facilities, 215 participants were investigated. The Minimental State Examination was applied to assess cognitive function. The sensory function was bifurcated into auditory and somatosensory realms which were evaluated using pure tone audiometry and Nottingham Sensory Assessment, respectively. Albert's test, left and right resolution, and visuospatial distribution were used to evaluate perception. Depression and social isolation were selected as negative socio-psychological factors and were evaluated by the Geriatric Depression Scale and the Lubben Social Network Scale. The multivariate analysis was performed utilizing binary logistic regression.Results: Participants with moderately severe or severe hearing loss exhibited significant cognitive impairment compared to those with mild hearing loss. It was observed that perceptual dysfunction and depression were independently related to cognitive impairment. However, there was no significant association between somatosensory function, social isolation, and cognitive impairment in the institutionalized older adults.Conclusion: More profound hearing loss, abnormal perception, and depression are associated with cognitive impairment in older adults. Subsequent research endeavors should delve into the causal mechanisms underpinning these associations and explore whether combined interventions have the potential to postpone the onset of cognitive impairment.

Purpose: To explore the relationship between Mycoplasma hyorhinis infection and tyrosine kinase inhibitor (TKI) resistance in lung adenocarcinoma patients. Methods: Mycoplasma hyorhinis infection can be verified with the monoclonal antibody PD4, which specifically recognizes a distinct protein of M. hyorhinis. Immunohistochemistry (IHC), using PD4 to detect M. hyorhinis, was performed on paraffin-embedded lung adenocarcinoma tissues of patients who had epidermal growth factor (EGFR) mutations and had received oral TKI. The number of patients enrolled in our study was 101. Assessments following TKI treatment were performed until objective disease progression or stable disease at the cutoff date was reached. In all of the patients, the primary endpoint was investigator-assessed progression-free survival (PFS). Results: Immunohistochemistry revealed that 61 of 101 cases (60.4%) of lung adenocarcinoma were positive for M. hyorhinis, which comprised of 31 low-positive cases and 30 high-positive cases; the remaining 40 cases (39.6%) were negative. The median PFS was significantly longer in the negative group [18months (95% CI 14.15–21.85)] than in the low-positive group [10months (95% CI 7.70–12.30); hazard ratio (HR) 4.095, 95% CI 2.254–7.438; p < 0.001] and in the high-positive group [4months (95% CI 2.85–5.15); HR 31.703, 95% CI 14.425–69.678; p < 0.001]. The results of the subgroup analysis were satisfactory. The PFS benefit with negative M. hyorhinis infection was consistent across subgroups. Conclusions: In this retrospective, exploratory analysis, M. hyorhinis infection significantly reduced PFS. With increased levels of M. hyorhinis infection, the progression of the disease was more advanced, likely due to the hydrolysis of TKI by M. hyorhinis. A strong correlation was found between M. hyorhinis infection and TKI resistance in lung adenocarcinoma. This study provides potent evidence that M. hyorhinis hydrolyses TKI and will assist in the research of related mechanisms in the future. Implications for cancer survivors: It provides an option to improve the efficacy of TKI, including strategies to decrease M. hyorhinis infection, thereby reducing long-term distress in TKI resistance patients with EGFR mutations. © 2021, The Author(s), under exclusive licence to Springer-Verlag GmbH, DE part of Springer Nature.

去泛素化是蛋白质翻译后修饰的重要形式之一,它影响了靶蛋白在细胞内的定位、稳定性和功能,以及相应的细胞周期、分化、凋亡调节、内吞、自噬和损伤后DNA修复等细胞生理活动。综合国内外有关去泛素化酶的研究文献进行分析和总结,重在阐述去泛素化酶的复杂功能。去泛素化酶可与多个靶蛋白相互作用,而细胞内多个其他蛋白又可以修饰或影响同一个去泛素化酶。去泛素化酶之间、去泛素化酶与靶蛋白之间、细胞内其他蛋白与去泛素化酶之间形成了一个复杂的相互作用网络。论文可以为去泛素化酶的功能研究提供参考。

摘要:
目的:深入了解胃癌患者术后主要照顾者的照顾需求,为医护人员实施健康教育提供理论依据,从而提高照顾者的照顾能力,减轻患者痛苦,提高其生存质量。方法:研究采用深度访谈法收集15位主要照顾者的资料,进行现场录音和笔录,将获得的资料根据Colaizzi内容分析法进行整理和分析后提炼出主题。结果:共归纳出3个主题:(1)照顾者对照护知识的态度和认知是被动的、浅表的;(2)照顾者的照护需求内容具有多样化特点;(3)照顾者对情感支持需求大。结论:胃癌患者术后主要照顾者对照护知识需求和情感支持需求大,医护人员应该关注照顾者,提供有针对性和可操作性的照护知识指导,共同提高患者和照顾者的生活质量。

Background: Quinolines have been characterized as a class of potential antitumor agents, and a large number of natural and synthetic quinolines acting as antitumor agents were reported. Methods: A series of 7-chloro-4(1H)-quinolone derivatives were synthesized. The antiproliferative effect of these compounds was evaluated by MTT assay against five human tumor cell lines. The mechanism of action of the selected compound 7h was also investigated. Results and Discussion: Most of the compounds had more potent antiproliferative activities than the lead compound 7-chloro-4(1H)-quinolone 6b. Compound 7h was found to be the most potent antiproliferative agent against human tumor cell lines. Further investigation demonstrated that compound 7h triggered ATG5-dependent autophagy of colorectal cancer cells by promoting the functions of LC3 proteins. Conclusion: These results were useful for designing and discovering more potent novel antitumor agents endowed with better pharmacological profiles. © 2020 Bentham Science Publishers.

[目的]对哥本哈根倦怠问卷(the Copenhagen Burnout Inventory,CBI)进行修订并验证其信效度。[方法]采取便利抽样法,选取陕西省、四川省、湖北省10余所三级甲等医院487名医护人员,采用修订后CBI对其进行调查。[结果]CBI修订版内容效度指数为0.791。探索性因子分析提取3个公因子,累积方差贡献率为67.98%,因子载荷为0.515~0.830。验证性因子分析结果显示拟合指数理想。Cronbach’s α系数为0.936,各维度Cronbach’s α系数分别为0.853,0.932,0.907。条目与总分相关系数为0.530~0.852,条目间相关系数为0.430~0.653。重测Cronbach’s α系数为0.914。[结论]CBI修订版信效度良好,可以作为测量医护人员职业倦怠的评估工具。

回顾游离肿瘤细胞术后灭活的相关研究,总结蒸馏水、生理盐水、聚维酮碘溶液和乙醇灭活游离肿瘤细胞的原理,归纳腹膜细胞学检查、流式细胞仪检测等游离肿瘤细胞检测常用手段的特点和优劣,整理肿瘤切除术后冲洗液发挥灭活游离肿瘤细胞所需的温度、浓度、冲洗次数、冲洗体积、时间等客观条件,探究冲洗液发挥最大灭瘤效果的最适宜条件范围。肿瘤切除术后游离肿瘤细胞灭活受多种因素影响,检测也会干扰冲洗液实际灭瘤效果的判断,因此本文综述游离肿瘤细胞灭活的研究状况并指出目前研究可能存在的不足,为将来游离肿瘤细胞灭活的进一步研究提供参考。

摘要:
目的 描述患者性别角色和满意度分布现状,探讨不同性别角色与护理服务和满意程度的相关关系.方法 采用便利抽样方法,选择武汉某三甲医院172名患者为研究对象.本研究采用问卷调查方法,以现场发放和回收问卷为主,问卷有患者一般资料问卷、贝姆性别角色量表(BSRI)、纽卡斯尔护理满意度量表(NNSS).采用SPSS23.0软件对相关数据进行统计分析.结果 ①BSRI:172名患者中,双性化角色占比2.9%,男性化角色占比15.1%,女性化角色占比16.9%,未分化角色占比65.1%.②NNSS:不满意率患者占比在36.1% ~44.8%之间,满意率患者占比在55.2%~63.9%之间.(③性别角色与NNSS条目2~6、8、11、14、16的相关性均具有统计学意义(均P <0.05);双性化与NNSS条目4、8,男性化与NNSS条目2~5、8,女性化与NNSS条目5、16的相关性均具有统计学意义(均P<0.05);NNSS条目10、12、14、17与双性化、女性化的相关性均具有统计学意义(均P<0.05),NNSS条目5、7、13、17与男性化、未分化的相关性均具有统计学意义(均P<0.05).结论 172名患者中,性别角色分布差异明显,以性别角色未分化最多,性别角色双性化最少;护理服务整体满意率较高,评分2.92~3.17分,以对患者需求了解和回应程度的满足评分最高,倾听烦恼及回应、隐私保护评分最低;性别角色男性化对满意度条目最多,未分化角色最少,双性化与不满意的程度有相关性,女性化/未分化与满意的程度有相关性.因此,护理人员应当避免对患者进行过多刚性化服务,尤其针对未分化者,应当采用柔性间接的管理,进而满足患者需求,提高护理服务满意度.

摘要:
不孕不育是一个遍及全球的生殖健康问题,近年不孕症患者呈逐年增长趋势,而辅助生殖技术(ART)是目前治疗不孕不育最有效的方法。由于不孕不育患者在接受ART治疗的不同阶段均面临不同的心理压力,且负性情绪会导致妊娠率与受孕率降低。因而,笔者在本研究中以ART为视点,分析辅助生殖技术(ART)女性患者与其负性情绪的关系。通过分析接受助孕手段女性患者的心理健康状况,并采用Zung焦虑自评量表(SAS)和抑郁自评量表(SDS)对进入治疗周期前的患者进行测试,将该患者的焦虑和抑郁情况进行分类,采取有针对性的措施和手段。以提高辅助生殖临床胚胎着床率和妊娠率,为临床护士开展行体外受精-胚胎移植助孕女性患者行心理护理提供了参考。

辅助生殖技术是治疗女性不孕的一种手段,但是焦虑、抑郁等负性情绪在患者中普遍存在,笔者旨在了解体外受精-胚胎移植助孕患者的焦虑、抑郁现状,并分析其影响因素.随机抽取300例接受体外受精-胚胎移植助孕的患者,在治疗周期前采用一般情况调查问卷、焦虑自评量表、抑郁自评量表进行横断面调查,以分析相关影响因素.结果 表明,在治疗周期前,进行体外受精-胚胎移植助孕患者的焦虑、抑郁人数比例分别为39.3%和62.3%.其中经济因素、不孕不育因素以及家庭因素是体外受精-胚胎移植女性患者焦虑、抑郁情绪的主要影响因素(p<0.05).焦虑、抑郁情绪普遍存在于体外受精-胚胎移植助孕的女性患者中,还受到诸如文化程度等因素影响.因此,建议医务人员帮助患者及家属树立治疗的信心,并强调家庭支持系统的重要性,让患者可以随时感受到家庭的温暖,从而减少焦虑、抑郁等负性情绪的发生.

<jats:title>Abstract</jats:title><jats:p>The p53 tumor suppressor is a critical factor in the DNA damage response (DDR), and regulation of p53 stability has a key role in this process. In our study, we identified USP49 as a novel deubiquitinase (DUB) for p53 from a library consisting of 80 DUBs and found that USP49 has a positive effect on p53 transcriptional activity and protein stability. Investigation of the mechanism revealed that USP49 interacts with the N terminus of p53 and suppresses several types of p53 ubiquitination. Furthermore, USP49 rendered HCT116 cells more sensitive to etoposide (Eto)-induced DNA damage and was upregulated in response to several types of cell stress, including DNA damage. Remarkably, USP49 expression was regulated by p53 and USP49 in knockout mice, which are more susceptible to azoxymethane/dextran sulfate sodium (AOM/DSS)-induced colon tumors. These findings suggest that USP49 has an important role in DDR and may act as a potential tumor suppressor by forming a positive feedback loop with p53.</jats:p>

Deubiquitination is a posttranslational protein modification prevalent in mammalian cells. Deubiquitinases regulate the functions of the target protein by removing its ubiquitin chain. In this study, the effects of the deubiquitinase USP38’s functions on the LSD1 protein and on cell physiology were investigated. Western blotting, real-time quantitative PCR, immunoprecipitation, denaturing immunoprecipitation and luciferase reporter assays were used to analyze the protein stability, protein interactions and changes in the ubiquitin chain. Cell proliferation assays, colony formation assays, drug treatments and western blotting were used to explore the functions of USP38 in cells. The deubiquitinase USP38 stabilizes protein LSD1 in cells by binding LSD1 and cleaving its ubiquitin chain to prevent the degradation of LSD1 by the intracellular proteasome. USP38 enhances the ability of LSD1 to activate signaling pathways and hence promotes cellular abilities of proliferation and colony formation through interacting with LSD1. Furthermore, USP38 enhances the drug tolerance of human colon cancer cells. USP38 is an LSD1-specific deubiquitinase that affects cellular physiology through interacting with LSD1.

组蛋白去甲基化酶LSD1是一个调控基因表达,影响机体发育和肿瘤发生的重要蛋白,它在细胞内的含量是受到精确调控的,其中LSD1蛋白的泛素化和去泛素化就是重要的调控方式,因此,筛选并研究LSD1的去泛素化修饰酶就显得非常重要。通过蛋白稳定性实验,实时定量PCR,免疫共沉淀,泛素蛋白链检测和信号通路激活实验来鉴定蛋白。通过生长曲线测定来研究基因对细胞增殖的影响。结果显示,USP17能够直接结合LSD1并切除其上的多聚泛素链而稳定LSD1在细胞内的蛋白水平,同时增强LSD1激活的STAT3信号通路,并且促进肿瘤细胞的增殖。因此,USP17是一个LSD1去泛素化酶。

HER2蛋白质的胞外区影响乳腺癌细胞的功能

HER2蛋白高表达是部分乳腺癌病人的肿瘤转移的高风险因素之一,针对该蛋白的靶向治疗卓有成效,但也有明显的副作用,因此,非常有必要进一步深入研究HER2蛋白的致病机理,以便开发更多的靶向药物。使用PCR技术将HER2基因N端的151个核苷酸进行扩增并将可能的糖基化位点进行突变,使用HA标签标记小肽。将重组了HER2基因突变体的核苷酸序列转染乳腺癌细胞SKBR3后,进行生长曲线、细胞黏附及抗药性研究及western blot分析。结果表明,HER2蛋白的N端突变体抑制乳腺癌细胞的黏附和生长,其中的糖基化位点突变后进一步降低了细胞的黏附和生长能力。通过激活抗凋亡蛋白BAK和抑制促凋亡蛋白BAD、BID和BIM的功能,HER2蛋白的N端小肽促进了乳腺癌细胞的抗药性。对开发新型的针对HER2蛋白高表达乳腺癌的靶向药物有一定的借鉴意义。

Mycoplasma fermentans is a pathogenic bacterium that infects humans and has potential pathogenic roles in respiratory, genital and rheumatoid diseases. NAD+-dependent deacetylase is involved in a wide range of pathophysiological processes and our studies have demonstrated that expression of mycoplasmal deacetylase in mammalian cells inhibits proliferation but promotes anti-starvation stress tolerance. Furthermore, mycoplasmal deacetylase is involved in cellular anti-oxidation, which correlates with changes in the proapoptotic proteins BIK, p21 and BIM. Mycoplasmal deacetylase binds to and deacetylates the FOXO3 protein, similar with mammalian SIRT2, and affects expression of the FOXO3 target gene BIM, resulting in inhibition of cell proliferation. Mycoplasmal deacetylase also alters the performance of cells under drug stress. This study expands our understanding of the potential molecular and cellular mechanisms of interaction between mycoplasmas and mammalian cells. © 2017 Elsevier GmbH

泛素化和去泛素化是两种广泛存在的蛋白质翻译后修饰方式。阐述了其影响了目标蛋白在细胞内的定位、稳定性和功能。其中,去泛素化过程涉及组蛋白的修饰,细胞周期,细胞分化,细胞凋亡,内吞和自噬的调控及DNA损伤修复,也参与机体的肿瘤发生,肌肉萎缩,组织发育及抗病毒感染。

Mycoplasmas are a group of microbes that can cause human diseases. The mycoplasmal lipoprotein p37 promotes cancer metastasis, at least in part, by interacting with EGFR. In this study, we show that the p37 lipoprotein binds another member of the EGFR family, HER2, through the HER2 extracellular domain. The binding of p37-HER2 promotes phosphorylation of HER2 and activates the downstream signaling molecule Erk1/2. Because the HER2 signaling pathway contributes to breast tumor metastasis, our results imply that the mycoplasmal lipoprotein p37 may also be involved in breast cancer metastasis. This study contributes to our understanding of mycoplasmal lipoprotein p37 function and its potential involvement in tumorigenesis. © 2016

建立一个高效的分离和分化小鼠成肌细胞的方案,以便探讨MAML1在转基因小鼠中肌管的形成和肌肉发生中的作用。使用细胞分化方法鉴定MAML1在肌肉发生中的作用。使用Western blot检测肌肉发生的标志蛋白肌球蛋白Myosin的表达。研究发现,传代次数少于5次及卫星细胞对成肌细胞在体外形成正确的肌管非常重要。不传代直接进行诱导分化,能促使成肌细胞形成更多的肌管。在MAML1转基因小鼠中,MAML1通过促进形成更多、更粗壮而长的肌管,提高肌球蛋白的蛋白水平来促进肌肉发生。这些MAML1转基因的肌管能够持续存在超过5天,然后才会降解。

肿瘤坏死因子(TNF-α)是一种炎症相关的细胞因子。通过基因组PCR对Maml1基因敲除和MAML1转基因小鼠进行了鉴定,通过去除血清和增加TNF-α来检测小鼠骨骼肌成肌细胞的抗饥饿能力。通过分化培养基诱导、TNF或staurosporine处理来研究成肌细胞肌管形成能力。结果表明,TNF-α促进了成肌细胞抗饥饿的能力,促进成肌细胞分化形成更多、更长、更持久的肌管。同样,MAML1基因也能够促进肌管形成。对进一步了解TNF-α对骨骼肌成肌细胞生理功能的影响提供了借鉴。
