微波水解-液相色谱-原子荧光光谱法测定硒蛋白中的硒代氨基酸

摘要:
针对酶解法测定硒代氨基酸操作复杂,时间长,水解不完全且水解产物不稳定等问题,建立一种微波水解-液相色谱-原子荧光光谱仪测定硒蛋白中硒代氨基酸的方法.样品经HCl(6 mol/L)微波水解后,调节pH值至7.5,采用阴离子色谱柱,pH=6.0的(NH4)2 HP04(40 mmol/L)溶液为流动相,流速0.6 mL/min,等度洗脱12 min的条件对硒代胱氨酸(SeCys2)、甲基-硒代半胱氨酸(MeSeCys)和硒代蛋氨酸(SeMet)进行分离,用原子荧光光谱仪测定其含量.微波水解条件采用星点设计-响应面法进行优化,优选最佳水解条件为:称样量20 mg,水解温度150℃,水解时间40 min.SeCys2、MeSeCys和SeMet均在0~100 ng浓度范围内呈线性关系,相关系数均大于0.999,检出限分别为0.07、0.03和0.14 mg/kg,RSD分别为5.6%、4.6%、3.7%(n=6).用来测定硒代氨基酸,耗时短,成本低,水解完全,操作简单,水解产物稳定,能够科学评价硒蛋白中硒代氨基酸的组成,对硒蛋白产品的质量控制和溯源提供了科学方法,值得推广应用.

摘要:
植物油用途广泛,因其较高的营养价值和良好的润滑性可作为食用油和工业润滑油。但是植物油含有不饱和烃、含氧化合物等化学成分导致氧化稳定性差,而氧化反应产生的毒性物质会影响食用油的安全性,而工业润滑油的使用寿命又与氧化稳定性密切相关,所以抗氧化性能是食用油和工业润滑油非常重要的性能指标。文章从食用油和工业润滑油两方面综述了植物油抗氧化性能的检测方法,讨论比较检测方法的适用性、优缺点以及植物油在不同行业中抗氧化性能评价指标和改善方式的差异,旨在为评价不同使用功能的植物油抗氧化性能提供参考。

摘要:
Bacterial magnetosomes, consisting of magnetite (Fe3O4) or greigite (Fe3S4) and enveloped with biomembrane, are a type magnetic nano-particle with a diameter of 30~120 nm. The exoperidium of the bacterial magnetosomes has the vesicle of the active groups with abundant oxhydryl and imino, providing a basis for bio-modification/chemical modification. In the present study, the magnetic response and the feature of being easily modified were researched. Glutaric dialdehyde was used as bi-functional reagent to combine the magnetosomes and nucleic acid aptamer for enrichment and purification of ochratoxin A in the samples. The results suggested that the adsorption rate of this method for OTA was 99%, and the recovery rate was 96%. In conclusion, this method presented a simple, efficient, as well as organic solvent reduction methods for OTA enrichment and purification, which provided a new idea in bacterial magnetosomes application and OTA purification. © 2022, Dept. of JCCOA. 摘要:
对白云边酒酿造酒醅中分离得到的不同种属的酵母菌进行发酵性能比较,从中筛选一株耐温性好、产酒力强、产正丙醇低的酵母Schizosaccharomyces pombe Scc7.与S.pombe标准菌株1056相比,Scc7在高粱糖化液中发酵酒精度提高了8.7%,正丙醇含量降低了9.9%,乙酸乙酯含量提高了22.2%,并产生3-羟基丁酮.以麸皮为培养基发酵制备Scc7菌剂模拟固态白酒发酵后发现,与储存4个月的大曲相比,将Scc7菌剂以30%比例同1个月大曲混合发酵可使酒精度提高20.8%,正丙醇降低57.78%.本研究表明,Scc7制备的功能菌剂在白酒酿造中具有良好的应用前景.

摘要:
Proteus mirabilis is an opportunistic pathogen associated with nosocomial infections and foodborne diseases. The resistance and biofilm formation of P. mirabilis have been a great concern. In this study a multidrug-resistant P. mirabilis strain 012 was exposed to a lethal dose of ampicillin (10 mg/mL, 2.5-fold minimal bactericidal concentration) for 24 h at 37°C. After resuscitation and isolation, five variant isolates were selected and subjected to ampicillin induction by repeatedly streaking on ampicillin-containing plates (10 mg/mL) for at least three times. In biofilm formation assays by using crystal violet staining, we found that the variant strains had enhanced biofilm-forming abilities. (-)-epigallocatechin-3-gallate (EGCG) at a minimum inhibitory concentration (MIC) (256 μg/mL) significantly reduced the biofilm formation of all variant strains and the wild-type strain (p < 0.01). Sub-MIC of EGCG (128 μg/mL) suppressed the biofilms of wild-type and two variants. However, it stimulated the biofilms of the other three variants. The antibiofilm effects of EGCG against the wild-type strain were further confirmed by confocal laser scanning microscopy. Scanning electron microscopy revealed that EGCG induced variants to form more fibrous structures. Our results revealed that a lethal dose of antibiotic exposure increased antibiotic resistance and biofilm formation of P. mirabilis. EGCG may be used as a promising antibiofilm agent to prevent the P. mirabilis biofilm formation in the food industry. However, the sub-MIC of EGCG is not effective and will not be applied. © 2022 Mary Ann Liebert, Inc., publishers.

为了降低米酒的高级醇含量,提高米酒品质,以异常威克汉姆酵母(Wickerhamomyces anomalus)、扣囊复膜酵母(Saccharomyces fibuligera)和酿酒酵母(Saccharomyces cerevisiae)为菌株混合发酵制备米酒,对其挥发性风味物质及感官品质进行分析,并采用单因素试验及响应面试验对其发酵条件进行优化。结果表明,W. anomalus、S. fibuligera、S. cerevisiae按1∶1∶1比例制备米酒品质较好,米酒高级醇含量为164.95 mg/L,感官评分为87.3分。米酒最优发酵工艺条件为:接种量3×106CFU/m L、后发酵温度21℃、后发酵时间2.5 d。在此优化条件下,米酒感官评分为90.1分,高级醇含量为153.1 mg/L,与优化前相比,其高级醇含量下降了7.2%,感官品质有所提高。

Trapa bispinosa Roxb. is a traditional Chinese food which is well known for its medicinal properties. The shell of Trapa bispinosa has anticancer activity, maybe due to its high content of polyphenols. There are few studies on the chemical composition of Trapa bispinosa shells, then we isolated the active components from Trapa bispinosa shell and clarified the mechanism of its anticancer activity. One monomer compound was separated from the ethanol extract of the Trapa bispinosa shell by fractional extraction, silica gel, Sephadex LH-20 gel column chromatography and liquid phase separation. The structure, identified by NMR was 1,2,3,6-tetra-O-galloyl-β-D-glucose. The results of the CCK-8 assay showed that 1,2,3,6-tetra-O-galloyl-β-D-glucose could significantly inhibit the proliferation of gastric cancer SGC7901 cells, and the effect was close to that of 5-fluorouracil. Here, 1,2,3,6-tetra-O-galloyl-β-D-glucose could affect the cell cycle of SGC7901 cells. At the dose of 200 μg/mL and an incubation time of 48 h, SGC7901 cells remained in the G1 phase, apoptosis occurred, the intracellular calcium ion concentration increased and the mitochondrial membrane potential decreased. Transcriptome sequencing analysis showed that the differentially expressed genes were mainly enriched in the P53 signalling pathway associated with apoptosis. The results of qPCR and Western blot showed that 1,2,3,6-tetra-O-galloyl-β-D-glucose could induce apoptosis of SGC7901 cells by up-regulating the expression levels of P21, PUMA, PERP and IGF-BP3 genes, down-regulating the CyclinD gene, increasing the expression levels of cytochrome C, caspase-3, caspase-9 protein and decreasing that of the protein BCL-2.

Background: Mannans are the main components of hemicellulose in nature and serve as the major storage polysaccharide in legume seeds. To mine new mannanase genes and identify their functional characteristics are an important basis for mannan biotechnological applications.<&wdkj&>Objective: In this study, a putative mannanase gene (ManBs31) from the genome of the marine bacterium Alteromonadaceae Bs31 was characterized.<&wdkj&>Methods: Amino acid sequence analysis and protein structural modeling were used to reveal the molecular features of ManBs31. The catalytic domain of ManBs31 was recombinantly produced using Escherichia coli and Pichia pastoris expression systems. The biochemical properties of the enzymes were determined by reducing sugar assay and thin-layer chromatography.<&wdkj&>Results: Sequence analysis revealed that ManBs31 was a multidomain protein, consisting of a catalytic domain belonging to glycoside hydrolase family 5 (GH5) and two cellulose-binding domains. Recombinant ManBs31-GH5 exhibited the maximum hydrolytic performance at 70 ºC and pH 6. It showed the best hydrolysis capacity toward konjac glucomannan (specific enzyme activity up to 1070.84 U/mg) and poor hydrolysis ability toward galactomannan with high side-chain modifications (with a specific activity of 344.97 U/mg and 93.84 U/mg to locust bean gum and ivory nut mannan, respectively). The hydrolysis products of ManBs31-GH5 were mannooligosaccharides, and no monosaccharide was generated. Structural analysis suggested that ManBs31-GH5 had a noncanonical +2 subsite compared with other GH5 mannanases.<&wdkj&>Conclusion: ManBs31 was a novel thermophilic endo-mannanase and it provided a new alternative for the biodegradation of mannans, especially for preparation of probiotic mannooligosaccharides.

The study aimed to investigate the effect and mechanism of eugenol treatment on fresh-peeled Chinese water chestnuts (CWCs). The results found that eugenol treatment maintained the appearance of fresh-peeled CWCs, accompanied by higher L* value, total solids and O2 contents, as well as lower browning degree, weight loss rate, CO2 content, a* and b* values. In addition, eugenol treatment significantly reduced the activities of peroxidase, phenylalanine ammonia-lyase, and polyphenol oxidase, as well as the total content of soluble quinone in fresh-peeled CWCs. Meanwhile, fresh-peeled CWCs treated with eugenol showed markedly lower content of total flavonoids, which may be related to yellowing. Furthermore, eugenol treatment suppressed the rates of O2·- and OH·- production as well as the contents of H2O2 and malondialdehyde in fresh-peeled CWCs. During the storage, eugenol treatment not only increased the activities of catalase, superoxide dismutase, ascorbate peroxidase and glutathione reductase as well as the DPPH free radical scavenging rate, but also increased the total phenolics, ascorbic acid and glutathione contents. In summary, eugenol treatment delayed the surface discoloration of fresh-peeled CWCs by improving the antioxidant capacity, inhibiting the phenolic compound metabolism and scavenging ROS, thus effectively maintaining the quality of fresh-peeled CWCs while extending their shelf life. 摘要:
本实验旨在优化菱角壳黄酮提取工艺,并考察其对人宫颈癌Hela细胞增殖的抑制作用。采用单因素实验考察了提取时间、料液比、乙醇浓度以及提取温度对菱角壳总黄酮得率的影响,并以菱角壳总黄酮得率为响应值进行响应面分析,得出菱角壳黄酮提取的最佳提取工艺。同时,采用CCK-8法测定菱角壳黄酮对人宫颈癌Hela细胞增殖的影响,并测定其IC50值,考察菱角壳黄酮的抗肿瘤活性。实验结果表明,菱角壳黄酮最佳提取工艺为:提取时间28 min、料液比1:33 g/mL、乙醇浓度53%。在该条件下菱角壳粗提物总黄酮的提取率可达3.455%±0.16%。5、100、200、400、800 μg/mL浓度的菱角壳黄酮粗提物、纯化物溶液孵育均对Hela细胞增殖均具有较好的抑制作用,且存在时间和剂量依赖性。以作用时间48h为例,菱角壳黄酮粗提物、菱角壳黄酮纯化物、5-Fu的IC50值分别为271.46、268.16、152.09 μg/mL。因此,同等时间下,菱角壳黄酮提取物的对于Hela细胞的增殖抑制作用弱于阳性对照5-Fu。

Background: Gastric cancer (GC) is ranked as the third leading cause of cancer-related mortality worldwide. 1,2,3,4,6-Pentagalloyl-β-D-glucose (β-PGG) has various pharmacological activities and has been shown to suppress cancer development. However, the mechanism by which β-PGG inhibits gastric cancer has not been elucidated. Objective: This study explored the potential targets and mechanism of β-PGG in GC using the network pharmacology approach combined with in-vitro experiments. Methods: The PharmMapper software was used to predict the potential targets of β-PGG, and GC-related genes were identified on the GeneCards database. PPI analysis of common genes was performed using the STRING database. The potential regulatory mechanism of β-PGG in GC was explored through Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses. The binding ability of key genes and target proteins was verified by molecular docking. The effects of β-PGG on genes and proteins were evaluated using the CCK-8 assay, cell cycle analysis, apoptosis assay, real-time fluorescence quantification polymerase chain reaction (qRT-PCR), and Western blotting. Results: Eight hub genes involved in cell cycle progression and apoptosis were identified. Cancer-related signaling pathways were identified using the Cytoscape tool. Some of those genes were significantly enriched in the p53 signaling pathway. The CCK-8 assay showed that β-PGG inhibited the proliferation of GC cells. Cell cycle and apoptosis experiments revealed that β-PGG induced cell cycle arrest and apoptosis of gastric cancer cells. qRT-PCR and Western blot analysis showed that β-PGG inhibited β-PGG cells by modulating the p53 signaling pathway. Conclusion: In the present study, the targets and mechanism of β-PGG in gastric cancer were explored. The results indicate that β-PGG can be used to develop treatments for GC. 目的 优化酶法提取鲟鱼鱼油工艺,探讨其对非酒精性脂肪肝病(non-alcoholicfattyliverdisease,NAFLD)的干预作用。方法 以鲟鱼下脚料为原料,用酶法提取鱼油,在单因素实验的基础上,采用正交实验得到提取鲟鱼鱼油的最佳工艺。利用气相色谱-质谱法检测鱼油的脂肪酸组成及含量。采用高脂饲喂方法建立NAFLD小鼠模型,探究鲟鱼鱼油对小鼠NAFLD的改善作用。结果 鲟鱼鱼油最佳提取条件为:1500U/g木瓜蛋白酶、料液比1:1 (mL/g)、pH 7、45℃酶解时间2 h。此条件鱼油的提取率为82.94%,所得鱼油为淡黄色、澄清透明,酸价为0.1mgKOH/g、过氧化值为0.49mmol/kg,符合一级鱼油国家标准。提取的鲟鱼鱼油饱和脂肪酸为23.56%、不饱和脂肪酸为74.71%。动物实验结果显示,正常饲料中添加鲟鱼鱼油时,小鼠各项指标均无显著性变化;高脂组小鼠血清中血清总胆固醇、甘油三酯、低密度脂蛋白、丙氨酸氨基转移酶、天冬氨酸氨基转移酶显著升高,高密度脂蛋白显著降低,而高脂组中添加深海鱼油和鲟鱼鱼油可显著改善血脂异常;添加鲟鱼鱼油可明显改善由高脂饲喂引起的小鼠肝组织脂肪变性及炎性浸润;添加鲟鱼油可显著改善小鼠肝组织炎性细胞因子基因IL-1β、TNF-α、IL-6、MCP1,脂代谢有关基因CD36、SREBF1及胆固醇代谢相关基因HMGCR、CYP7A1的表达。结论 本研究优化了鲟鱼鱼油提取工艺,获得的鲟鱼鱼油中ω-3不饱和脂肪酸含量较高,证明了鲟鱼鱼油对NAFLD的改善作用,可作为深海鱼油的替代产品。

摘要:
首次根据Genbank中已知的7237条羊肚菌序列,对所有编码转录因子的序列进行了功能分类,并重点对羊肚菌中的锌指蛋白(ZFP)编码序列进行了生物信息学分析。结果表明,Genbank中所有7237条羊肚菌来源的核苷酸序列,有167条是编码转录因子的序列,其中88条序列编码了锌指蛋白。这些序列都含有相应的保守基序,编码蛋白质的二级结构以α-螺旋为主,三级结构极为相似,功能上以亲水性蛋白为主。

鱼油对高脂小鼠血脂的改善作用

研究鱼油对高脂模型小鼠血脂的改善作用。将6—7周龄雄性C57BL/6J小鼠分为三组,饲喂正常饮食、高脂饮食或高脂补充鱼油18周后,测定体重、食物摄入量、肝脏指数和空腹血糖,并用酶法检测血清中甘油三酯(Triglyceride, TG)、总胆固醇(total cholesterol, TC)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol, HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol, LDL-C)浓度及游离脂肪酸(nonestesterified fatty acid, NEFA)含量。通过使用苏木精—伊红染色法及油红O染色法评价了肝组织形态学改变。结果表明,在高脂饮食小鼠中,体重增加,肝脏甘油三酯(TG)水平较正常小鼠高出约38%(P为0.002 8);与高脂饮食小鼠相比,高脂补充鱼油的小鼠TG降低了6.80%(P为0.673 8)、LDL-C降低了33.23%(P为0.002 3),HDL-C水平显著性提升了51.25%,此外,高脂补充鱼油组小鼠表现出更大的代谢变化,脂肪堆积减少33%(P为0.1207)。这些结果表明鱼油补充对高脂饮食诱导脂代谢的有益影响,鱼油可能通过调节脂肪代谢来改善高脂饮食引起的肥胖,血脂异常的潜力,这一结果可以应用于预防肥胖的个性化营养。

伊曲茶碱在两种混合溶剂中的固液相平衡

伊曲茶碱是一种口服有效的抗帕金森药物,通过固液相平衡实验为伊曲茶碱结晶动力学的研究及结晶工艺的设计、优化提供参考数据。采用动态法,在283.15—323.15 K、常压下,测定了伊曲茶碱在2种二元混合溶剂(乙醇+乙酸乙酯、甲基乙基酮)中的溶解度;用Apelblat方程、λh方程对溶解度数据进行拟合;通过Van′t Hoff方程计算2种溶剂体系的溶解熵、溶解焓以及吉布斯自由能。结果表明:伊曲茶碱的溶解度与温度以及乙醇的摩尔分数有关。伊曲茶碱在不同摩尔分数混合溶剂中的溶解度随着温度的升高而逐渐增加;同一温度下,其在所处的溶剂体系中的溶解度随着乙醇摩尔分数的增加而减小。Apelblat方程关联结果优于λh方程,能更好地拟合溶解度数据。伊曲茶碱溶解过程是吸热的,熵增的,溶解焓是溶解过程中溶解吉布斯自由能的主要贡献者。

六氯丁二烯降解菌BJ-04的筛选鉴定及降解特性

六氯丁二烯是一种新型的持久性有机污染物,利用微生物降解环境中六氯丁二烯是一种无二次污染、非常绿色高效的方法。该研究从武汉东西湖区马池桥附近污泥中采样,通过连续传代驯化,分离筛选到一株能够以六氯丁二烯为唯一碳源生长的菌株,具有高效降解六氯丁二烯(HCBD)能力。16S rRNA基因测序鉴定菌株;通过气相色谱法和气相色谱-质谱联用法来测定底物降解及产物生成情况,并对降解菌株的抗生素抗性和底物广谱性进行探究。结果表明,该菌的16S r RNA序列与根癌农杆菌(Agrobacterium tumefaciens)具有99%的一致性,构建系统发育树显示该菌株与Agrobacterium tumefaciens聚为一支,将其命名为Agrobacterium sp. BJ-04,降解菌在72 h内可将1 mmol/L的HCBD降解至0.1 mmol/L左右,但是使用氯离子电极法未检测到Cl-浓度变化,气相色谱-质谱联用法检测到可能的中间产物正丁醇生成,此外该菌对氨苄青霉素、氯霉素、卡那霉素、链霉素、庆大霉素和萘啶酸均具有抗生素抗性,能够利用正己烷、环己烷、苯、正丁醇为碳源进行生长。对菌株BJ-04系统研究为将来用于处理废水及土壤中污染物六氯丁二烯提供理论和技术支持,也为充分利用微生物代谢解决环境污染问题提供菌株资源和技术储备。

BACKGROUND: The incidence of metabolic syndrome (MetS) is increasing, and n-3 polyunsaturated fatty acids (PUFAs) in salmon (Oncorhynchus) phospholipids can effectively reduce the risk of MetS. RESULTS: Under the intervention of 4% salmon phospholipid, the levels of fasting blood glucose (FBG), insulin, monocyte chemoattractant protein-1 (MCP-1), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were significantly reduced in the plasma of MetS mice, whereas adiponectin was significantly increased. By screening, we found that the 18 differential metabolites, consisting of seven triglycerides (TGs), six diglycerides (DGs), one phosphatidylethanolamine (PE), three sphingomyelins (SMs) and one eicosanoid, could be the key differential metabolites, and two metabolic pathways were significantly affected: glycerolipid metabolism and glycerophospholipid metabolism. CONCLUSION: 4% salmon phospholipids could affect MetS by inhibiting insulin resistance, reducing inflammatory factors and promoting the synthesis of PE, yet the mechanism required further study. Scutellarin related drugs have superior therapeutic effects on cerebrovascular and cardiovascular diseases.Here,an optimal biosynthetic pathway for scutellarin was constructed in Yarrowia lipolytica platform due to its excellent metabolic potential.By integrating multi-copies of core genes from different species,the production of scutellarin was increased from 15.11 mg/L to 94.79 mg/L and the ratio of scutellarin to the main by-product was improved about 110-fold in flask condition.Finally,the production of scutellarin was improved 23-fold and reached to 346 mg/L in fed-batch bioreactor,which was the highest reported titer for de novo production of scutellarin in microbes.Our results represent a solid basis for further production of natural products on unconventional yeasts and have a potential of industrial implementation.
