摘要:
为了解鄂西北地区猪场猪圆环病毒2型(Porcine circovirus type 2,PCV-2)不同免疫水平下的流行病学特点,试验采用ELISA法和PCR法分别对2017—2019年鄂西北地区猪场送检的1 456份血样和134份临床疑似PCV-2感染的病料进行了PCV-2抗体和病原检测,并统计采样猪的PCV-2疫苗免疫情况。根据2017—2019年采样猪PCV-2疫苗免疫记录,分别统计不同年份PCV-2疫苗免疫密度,分析不同免疫密度下PCV-2抗体水平差异。再根据采样猪的PCV-2疫苗免疫记录将猪分为未免疫、1次免疫及≥2次免疫共3个不同免疫频次组,分析不同免疫频次下PCV-2抗体阳性率和PCV-2病原阳性率差异。结果表明:2017年、2018年和2019年PCV-2疫苗免疫密度分别为91.2%、49.3%和39.9%,PCV-2抗体阳性率分别为78.1%、39.1%和34.8%,PCV-2疫苗免疫密度下降导致PCV-2抗体阳性率显著下降(P<0.05);未免疫、1次免疫和≥2次免疫的PCV-2抗体阳性率分别为32.4%、66.0%和86.5%,PCV-2病原阳性率分别为73.7%、42.0%和18.5%,PCV-2疫苗免疫频次增加能显著提高PCV-2抗体阳性率(P<0.05),显著降低PCV-2病原阳性率(P<0.05)。说明鄂西北地区PCV-2防控形势严峻,增加免疫密度和加强免疫频次可提高PCV-2抗体水平,降低猪只感染PCV-2病原的风险。

摘要:
为研究黄芩苷对仔猪感染副猪嗜血杆菌(Glaesserella parasuis,GPS)后血细胞分类计数和血液生化指标的影响,选取48头\"杜×长×大\"健康断奶仔猪,随机分为6组,分别为:空白对照组、GPS感染模型组、对照药物氟尼辛葡甲胺组和不同剂量黄芩苷处理组(肌内注射,剂量分别为25、50和100 mg/kg)。饲养3 d后进行攻毒及药物处理,攻毒后48 h和96 h分别采集血样进行血细胞分类计数和血液生化指标检测。结果显示,攻毒后48 h,氟尼辛葡甲胺能缓解副猪嗜血杆菌引起的血小板、单核细胞数量的降低(P<0.01),缓解嗜碱性粒细胞数量的降低及谷丙转氨酶含量的升高(P<0.05);25、50及100 mg/kg黄芩苷能缓解血小板、单核细胞数量的降低(P<0.01);25、100 mg/kg黄芩苷能缓解谷丙转氨酶含量的升高(P<0.05);50 mg/kg黄芩苷能缓解红细胞数量的降低(P<0.05)。攻毒后96 h,氟尼辛葡甲胺能缓解副猪嗜血杆菌引起的白细胞、中性粒细胞数量的升高及淋巴细胞数量的降低(P<0.01);25、50及100 mg/kg黄芩苷能缓解白细胞、中性粒细胞数量的升高;50 mg/kg黄芩苷能缓解血小板数量的降低(P<0.05);100 mg/kg黄芩苷能缓解葡萄糖、磷含量的降低(P<0.05)。以上结果表明,黄芩苷能在一定程度上缓解仔猪因感染GPS引起的血细胞分类计数和血液生化指标的异常变化。

摘要:
Haemophilus parasuis can elicit serious inflammatory responses, which contribute to huge economic losses to the swine industry. However, the pathogenic mechanisms underlying inflammation-related damage induced by H. parasuis remain unclear. Accumulating evidence indicates that long non-coding RNAs (lncRNAs) have important functions in the regulation of autoimmune disorders. Baicalin has been shown to have anti-inflammatory, anti-microbial, and anti-oxidant activities. In this study, we investigated whether lncRNAs were involved in the vascular injury or inflammation triggered by H. parasuis and whether baicalin regulated the lncRNA profiles of porcine aortic vascular endothelial cells (PAVECs) infected with H. parasuis. The results showed that the lncRNA and mRNA expression profiles of PAVECs were changed by H. parasuis. Important functions of lncRNAs and mRNAs were predicted. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses demonstrated that the targets of differentially expressed lncRNAs of H. parasuis infected PAVECs were mainly involved in the tumor necrosis factor (TNF) signaling pathway, apoptosis, and N-glycan biosynthesis; whereas nicotinate and nicotinamide metabolism, the cytosolic DNA-sensing pathway, the TNF signaling pathway, and the nuclear factor (NF)-kappa B signaling pathway were enriched in PAVECs pretreated with baicalin. In addition, top hub genes and lncRNAs were identified and validated by quantitative polymerase chain reaction. CCL5, GBP1, and SAMHD1 were significantly upregulated after H. parasuis infection, whereas they were significantly downregulated with baicalin pretreatment. LncRNA ALDBSSCT0000001677, ALDBSSCT0000001353, MSTRG.10724.2, and ALDBSSCT0000010434 had the same expression pattern. Collectively, these data suggested that baicalin could modify changes to the lncRNAs profiles or regulate lncRNAs that participate in inflammation-related signaling pathways, thereby alleviating tissue damage or inflammatory responses induced by H. parasuis. To our best knowledge, this is the first article of H. parasuis stimulating changes to the lncRNA profiles of PAVECs and the capability of baicalin to regulate lncRNA changes in PAVECs infected with H. parasuis, which might provide a novel therapeutic target for the control of H. parasuis infection.

摘要:
为评价黄芩苷的抗炎作用,研究黄芩苷对脂多糖(LPS)刺激后仔猪血清中炎性因子表达的影响,试验选取36头健康状况良好、体重(11.0±1.2) kg的"杜×长×大"断奶仔猪,随机分为阴性对照组、LPS组、氟尼辛葡甲胺组和黄芩苷组(黄芩苷剂量分别为按体重25,50,100 mg/kg添加)6组。饲喂7 d后,氟尼辛葡甲胺组和黄芩苷组提前0.5 h肌肉注射给药,然后与LPS组一起按体重0.1 mg/kg腹腔注射LPS,阴性对照组腹腔注射等量生理盐水,注射LPS 6 h后再次给药1次。检测注射LPS后3,24小时时猪血清中炎性因子TNF-α、IL-1β、IL-6和IL-8的含量,并于注射LPS 24 h后检测血管和腹膜中炎性因子TNF-α、IL-1β、IL-6和IL-8的基因表达量。结果表明:LPS刺激仔猪3 h后,LPS组血清中TNF-α、IL-1β、IL-6和IL-8的含量显著或极显著增加(P<0.05或P<0.01),氟尼辛葡甲胺能显著抑制血清中IL-6含量的升高(P<0.05),黄芩苷能显著或极显著抑制血清中TNF-α、IL-1β、IL-6和IL-8含量的升高(P<0.05或P<0.01);LPS刺激仔猪24 h后,LPS组血清中TNF-α含量极显著增加(P<0.01),氟尼辛葡甲胺对血清中TNF-α、IL-1β、IL-6和IL-8含量升高无显著抑制作用(P>0.05),100 mg/kg黄芩苷能极显著抑制血清中TNF-α、IL-6和IL-8含量的升高(P<0.01),显著抑制IL-1β含量的升高(P<0.05)。LPS刺激24 h后,仔猪血管和腹膜中TNF-α、IL-1β、IL-6和IL-8基因的表达显著或极显著上调(P<0.05或P<0.01);氟尼辛葡甲胺和黄芩苷能显著或极显著抑制血管中TNF-α、IL-1β、IL-6和IL-8基因表达上调(P<0.05或P<0.01);氟尼辛葡甲胺和黄芩苷能显著抑制腹膜中TNF-α、IL-1β、IL-6基因表达上调(P<0.05)。说明黄芩苷能有效地抑制LPS刺激后仔猪血清中炎性因子的分泌及血管和腹膜组织中相关基因的表达。

摘要:
为了研究黄芩苷对脂多糖(LPS)诱导后仔猪临床症状及血液学指标的影响,试验选取36头健康仔猪,分为空白对照组、LPS模型组、氟尼辛葡甲胺组、黄芩苷低剂量(25 mg/kg)组、黄芩苷中剂量(50 mg/kg)组和黄芩苷高剂量(100 mg/kg)组。各组猪在注射LPS前0.5 h给药,黄芩苷各剂量组及模型组猪按体重0.1 mg/kg腹腔注射LPS,空白对照组猪注射等量生理盐水,LPS诱导6 h后再给药1次。于注射LPS 3 h后测量各组猪的体温,并统计呕吐、颤抖、嗜睡猪的头数;于注射LPS后第3,24小时对各组猪采血,检测血常规指标和血液生化指标。结果表明:LPS诱导后第3小时,LPS模型组猪出现体温升高、颤抖、呕吐和嗜睡等临床症状,黄芪苷各剂量组猪的体温较LPS模型组均显著降低(P<0.05),猪的呕吐和嗜睡症状减轻;LPS诱导后第3小时,与空白对照组比,LPS模型组白细胞数和血小板总数均极显著降低(P<0.01);与LPS模型组比,黄芩苷中剂量组白细胞数极显著升高(P<0.01)。LPS诱导后第24小时,与空白对照组比,LPS模型组白细胞数极显著升高(P<0.01),血小板总数极显著降低(P<0.01);与LPS模型组比,黄芩苷中剂量组血小板总数显著升高(P<0.05)。LPS诱导后第3小时,与空白对照组比,LPS模型组总胆红素、肌酐、总蛋白、白蛋白和血糖含量出现显著或极显著变化(P<0.05或P<0.01);与LPS模型组比,黄芩苷中剂量组肌酐含量显著降低(P<0.05);LPS诱导后第24小时,与空白对照组比,LPS模型组总胆红素含量、天门冬氨酸氨基转移酶活性、肌酐和血糖含量出现显著或极显著变化(P<0.05或P<0.01)。与LPS模型组比,黄芩苷低剂量组和中剂量组能极显著(P<0.01)和显著(P<0.05)降低总胆红素含量。说明50 mg/kg黄芩苷肌肉注射给药可以有效缓解LPS刺激导致的仔猪体温升高、白细胞数异常变化、血小板总数降低及肝脏和肾脏功能的损伤。

摘要:
Glaesserella parasuis (G. parasuis) causes porcine vascular inflammation and damage. Baicalin is reported to have antioxidant and anti-inflammatory functions. However, whether baicalin protects piglets against G. parasuis challenge and the potential protective mechanism have not been investigated. Therefore, in this study, we comprehensively examined the protective efficacy of baicalin in piglets challenged with G. parasuis and the possible protective mechanism. Our results show that baicalin attenuated the release of the inflammation-related cytokines interleukin (IL) 1β, IL6, IL8, IL10, and tumour necrosis factor α (TNF-α) and reduced high mobility group box 1 (HMGB1) production and cell apoptosis in piglets infected with G. parasuis. Baicalin also inhibited the activation of the mitogen-activated protein kinase (MAPK) signalling pathway and protected piglets against G. parasuis challenge. Taken together, our data suggest that baicalin could protect piglets from G. parasuis by reducing HMGB1 release, attenuating cell apoptosis, and inhibiting MAPK signalling activation, thereby alleviating the inflammatory response induced by the bacteria. Our results suggest that baicalin has utility as a novel therapeutic drug to control G. parasuis infection.

摘要:
建立兽用黄连解毒方剂UPLC指纹图谱及黄芩苷、小檗碱和栀子苷3种成分含量测定方法.采用UPLC-PDA法研究不同工艺下黄连解毒方剂指纹图谱,发现共有峰30个并指认了13个峰.用UPLC法对方剂中黄芩苷、小檗碱和栀子苷含量进行测定,结果显示黄芩苷(5.00-799.45 μg/mL)、小檗碱(5.64-360.67 μg/mL)和栀子苷(5.00-320.13 μg/mL)线性良好,精密度、稳定性、重复性和加样回收率试验均符合要求,并检验了3种工艺的黄连解毒方剂样品.本研究为兽用黄连解毒方剂的质量控制研究提供了参考依据.

Glässer’s disease, caused by Haemophilus parasuis (H. parasuis), is associated with vascular damage and vascular inflammation in pigs. Therefore, early assessment and treatment are essential to control the inflammatory disorder. MicroRNAs have been shown to be involved in the vascular pathology. Baicalin has important pharmacological functions, including anti-inflammatory, antimicrobial and antioxidant effects. In this study, we investigated the changes of microRNAs in porcine aortic vascular endothelial cells (PAVECs) induced by H. parasuis and the effect of baicalin in this model by utilizing high-throughput sequencing. The results showed that 155 novel microRNAs and 76 differentially expressed microRNAs were identified in all samples. Subsequently, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the target genes of the differentially expressed microRNAs demonstrated that regulation of actin cytoskeleton, focal adhesion, ECM-receptor interaction, bacterial invasion of epithelial cells, and adherens junction were the most interesting pathways after PAVECs were infected with H. parasuis. In addition, when the PAVECs were pretreated with baicalin, mismatch repair, peroxisome, oxidative phosphorylation, DNA replication, and ABC transporters were the most predominant signaling pathways. STRING analysis showed that most of the target genes of the differentially expressed microRNAs were associated with each other. The expression levels of the differentially expressed microRNAs were negatively co-regulated with their target genes’ mRNA following pretreatment with baicalin in the H. parasuis-induced PAVECs using co-expression networks analysis. This is the first report that microRNAs might have key roles in inflammatory damage of vascular tissue during H. parasuis infection. Baicalin regulated the microRNAs changes in the PAVECs following H. parasuis infection, which may represent useful novel targets to prevent or treat H. parasuis infection. © 2020, Springer Science+Business Media, LLC, part of Springer Nature.

The objective of this study was to survey the prevalence of aflatoxin M1 (AFM1) in raw, pasteurized, extended shelf life (ESL) and UHT milk from central-south China. A total of 734 milk samples, comprising raw milk (133 samples), pasteurized milk (410), ESL milk (93) and UHT milk (98), were obtained between Nov. 01, 2018 and Mar. 31, 2019. The AFM1 concentrations in milk samples were detected by ELISA and confirmed by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Five hundred and ninety samples (80.4%) were positive for AFM1; the mean value was 23.7 ng/L, and the range was 5.1–104.4 ng/L. The percentages of AFM1 positive raw, pasteurized, ESL and UHT milk samples were 75.2, 82.2, 81.7 and 78.6%, respectively. The AFM1 concentration in raw milk was significantly lower (p < 0.05) than that in pasteurized and ESL milk, and the AFM1 level in UHT milk tended to be lower (p < 0.10) than that in pasteurized and ESL milk. The AFM1 levels exceeded the EU legal limit of 50 ng/L in 0, 11.5, 18.3 and 3.1% of raw, pasteurized, ESL and UHT milk samples, respectively. All milk samples tested had levels far below China's legal limit of 500 ng/L. Therefore, the AFM1 content in raw milk and milk products in central-south China does not pose a public health threat, but close attention should nonetheless be paid to milk AFM1, given the high positive rate of 80.4%. © 2019

“新农科”背景下卓越动物科学专业“晋级式”实践教学模式的研究与实践

摘要:
近年来,国内养殖行业发展迅速,对高技能人才需求极大,而动物科学专业学生由于对行业认知不够,实践动手能力较差,进入企业需要连续培训,融入行业时间变长。课题组在“新农科”建设背景下,首次提出了“晋级式”实践教学模式,即采用企业人事制度,设置3个等级:技术员(初级)、技术经理(中级)和技术总经理(高级),并设定相应技术考核要求,学员进行初级实践学习并达到所有技术要求,才能晋升中级、高级开展实践学习和考核,逐级晋升考核。课题组对该模式的目标、原则和实施思路进行了剖析,并以复合应用人才的实践教学培养为例,研究“晋级式”实践教学模式在动物科学专业人才培养中的应用评价,实施效果较好。

Diosmetin, as a methoxyflavone compound, was investigated for its interaction and mechanism with xanthine oxidase (XOD), which is a key enzyme catalyzing xanthine to uric acid and finally causing hyperuricemia. Using a kinetic analysis method, diosmetin was shown to possess a strong inhibitory activity for XOD in a reversibly competitive manner with an IC50value of (5.08 ± 0.48) × 10−6mol L−1. The results of fluorescence titrations showed that the combination of diosmetin with XOD gave spontaneous, exothermic static quenching, and hydrogen bonding and van der Waals forces were the main driving forces. It was suggested that diosmetin increased the polarity of the tyrosine and tryptophan microenvironment of XOD by synchronous fluorescence analysis. The results of ultraviolet absorption spectroscopy, circular dichroism and molecular docking showed that diosmetin could bind to the active site of XOD to form a new diosmetin-XOD complex. In these experiments, diosmetin reduced the content of α-helices, β-sheets and β-turns of XOD, which led to the instability of the XOD structure and the decrease of its catalytic function. Furthermore, the 4′-methoxy group of diosmetin had a higher affinity for XOD than the 4′-hydroxyl group of luteolin. To the best of our knowledge, this is the first comprehensive report to present the inhibitory activities of diosmetin against XOD. © The Royal Society of Chemistry and the Centre National de la Recherche Scientifique 2020.

<jats:title>Abstract</jats:title><jats:p>Dinuclear Ni(II) macrocyclic complex <jats:bold>A</jats:bold> ([Ni<jats:sub>2</jats:sub>L<jats:sup>1</jats:sup>OAc]ClO<jats:sub>4</jats:sub>⋅CH<jats:sub>3</jats:sub>OH⋅H<jats:sub>2</jats:sub>O) and dinuclear Ni(II) acyclic complex <jats:bold>B</jats:bold> ([Ni<jats:sub>2</jats:sub>L<jats:sup>2</jats:sup>](ClO<jats:sub>4</jats:sub>)<jats:sub>2</jats:sub>) were synthesized by condensation between H<jats:sub>2</jats:sub>L (3,3′‐(ethane‐1,2‐diylbis(benzylazanediyl))bis(methylene)bis(2‐hydroxy‐5‐methyl‐benzaldehyde)) and different molar equivalent 1,3‐propanediamine in the presence of Ni(II) ions. The structures of complexes were characterized by IR spectroscopy, elemental analysis, electrospray mass spectra and X‐ray single crystal diffraction. In complex <jats:bold>A</jats:bold>, the Ni(II)‐Ni(II) distance was 2.9413(7) Å, bridged by two phenolic oxygens and an acetate anion, while in complex <jats:bold>B</jats:bold>, it was 3.0540(1) Å, only bridged by two phenolic oxygens. The calf thymus DNA (CT‐DNA) binding ability was studied through spectroscopic and electrochemical methods, the corresponding binding constant for complexes <jats:bold>A</jats:bold> and <jats:bold>B</jats:bold> were 6.45×10<jats:sup>4</jats:sup> M<jats:sup>−1</jats:sup> and 8.65×10<jats:sup>4</jats:sup> M<jats:sup>−1</jats:sup>, respectively. The MS revealed that <jats:bold>B</jats:bold> can transform to <jats:bold>A</jats:bold> by adding more H<jats:sub>2</jats:sub>L, and <jats:bold>A</jats:bold> cannot transform to <jats:bold>B</jats:bold> in the presence of excess 1,3‐propanediamine.</jats:p>

HPLC法测定兽用除虫脲原料药含量及有关杂质

摘要:
建立了高效液相色谱法(HPLC)测定兽用除虫脲原料药含量及有关杂质的检测方法。采用C18色谱柱(250 mm×4.6 mm,5μm),以甲醇为流动相A,0.01 mol/L乙酸铵溶液为流动相B,进行梯度洗脱,流速1.0 mL/min,检测波长254 nm,进样量10μL,柱温25℃。除虫脲的线性范围为10~1000μg/mL(R 2=0.999),回收率97.01%~100.83%(RSD<1.0%,n=3),精密度(RSD=0.13%,n=9)、重复性(RSD=0.45%,n=6)良好,主成分与各杂质峰分离良好。以主成分自身对照法计算,各杂质峰面积的和不得大于对照溶液主峰面积的0.1倍(0.1%)。该方法简便灵敏,结果准确可靠,可作兽用除虫脲原料药含量及有关杂质测定。

摘要:
[目的]以脂多糖(LPS)诱导建立仔猪免疫损伤模型,研究黄芩苷对LPS诱导的仔猪血管紧密连接蛋白表达量和MLCK-MLC信号通路变化的影响,探讨黄芩苷对LPS诱导紧密连接蛋白表达量的影响及其保护机制。[方法]选取42头健康仔猪,设空白对照组、LPS模型组、LPS+丙酮酸乙酯处理组、LPS+氟尼辛葡甲胺处理组和LPS+黄芩苷处理组(剂量分别为25、50和100mg/kg)。饲喂7d后,提前0.5 h进行药物处理,处理组及模型组腹腔注射LPS,空白对照组注射等量生理盐水,LPS诱导6 h后再次给药。试验1d后,采取血管样品,用免疫组化及免疫荧光观察血管中紧密连接蛋白(ZO-1、Claudin-1和Occludin)的蛋白表达变化;Western blot法测定血管MLCK-MLC信号通路相关蛋白(MLCK、MLC2和p-MLC2)的表达量。[结果]LPS诱导仔猪后,血管中的紧密连接(ZO-1、Claudin-1和Occludin)蛋白表达量极显著降低(P<0.01),黄芩苷处理组能明显的提高仔猪血管紧密连接蛋白的表达量。LPS模型组MLCK-MLC信号通路各相关蛋白(MLCK、MLC2和p-MLC2)的表达量均极显著升高(P<0.01),黄芩苷能显著下调MLCK-MLC信号通路各相关蛋白(MLCK和p-MLC2)表达量(P<0.05)。[结论]LPS能引起仔猪的血管损伤,激活MLCK-MLC信号通路,影响紧密连接蛋白表达量及功能。黄芩苷对LPS诱导的仔猪血管紧密连接损伤具有保护作用。本试验结果为黄芩苷用于治疗猪的炎性血管损伤提供了科学依据。

[目的]体外分离培养猪腹膜间皮细胞,建立副猪嗜血杆菌感染猪腹膜间皮细胞的体外模型,研究副猪嗜血杆菌感染猪腹膜间皮细胞后引起的细胞炎症反应,筛选副猪嗜血杆菌感染猪腹膜间皮细胞的最佳感染时间和最佳感染比。[方法]从猪腹膜中分离猪腹膜间皮细胞,进行体外培养和鉴定。间皮细胞经传代培养,使用梯度浓度的副猪嗜血杆菌感染猪腹膜间皮细胞,于不同时间点收集细胞上清液,提取间皮细胞的RNA。采用ELISA法测定细胞培养上清液中炎性因子(IL-6、TNF-α和IL-1β)的含量;Q-RT PCR法测定细胞炎性因子(IL-6、TNF-α和IL-1β)的mRNA表达情况。[结果]体外培养的猪腹膜间皮细胞,经免疫组化鉴定确认抗波形蛋白抗体(Vim)、抗细胞角蛋白抗体(PCK)呈阳性,抗白细胞CD45抗体(CD45)、抗第Ⅷ因子相关抗体(F8)呈阴性,确认为猪腹膜间皮细胞。副猪嗜血杆菌感染能极显著上调炎性因子的mRNA表达水平,不同感染时间与不同感染比对猪腹膜间皮细胞的刺激反应不同,在6-20h之间随时间和感染比增加呈上升趋势。[结论]确定了副猪嗜血杆菌感染猪腹膜间皮细胞的最佳感染时间和最佳感染比,为研究副猪嗜血杆菌致腹膜炎症的作用机制提供了模型。

摘要:
Glässer’s disease, caused by Haemophilus parasuis, is a chronic disease related to an inflammatory immune response. Baicalin exerts important biological functions. In this study, we explored the protective efficacy of treatment with baicalin and the potential mechanism of activation of the MAPK signaling pathway in porcine aortic vascular endothelial cells (PAVECs) induced by H. parasuis. H. parasuis stimulated expression of receptor for advanced glycation end products, induced a significant increase in the level of protein kinase-α and protein kinase-δ phosphorylation, and significantly up-regulated ERK, c-Jun N-terminal kinase, and p38 phosphorylation in PAVECs. H. parasuis also up-regulated the levels of apoptotic genes (Bax, C-myc, and Fasl) and the expression levels of c-Jun and c-Fos, and induced S-phase arrest in PAVECs. However, treatment with baicalin inhibited expression of RAGE, suppressed H. parasuis-induced protein kinase-α and protein kinase-δ phosphorylation, reduced ERK, c-Jun N-terminal kinase, and p38 phosphorylation, down-regulated apoptotic genes (Bax, C-myc, and Fasl), attenuated phospho-c-Jun production from the extracellular to the nuclei, and reversed S-phase arrest in PAVECs. In conclusion, baicalin treatment inhibited the MAPK signaling pathway, thereby achieving its anti-inflammatory responses, which provides a new strategy to control H. parasuis infection. © The Author(s) 2019.

2,4-dinitrotoluene (2,4-DNT) is a common environmental pollutant, and was classified as a group 2B human carcinogenic compound by the International Agency for Research on Cancer. This study determined the toxic effects of 2,4-DNT exposure on zebrafish at the embryo-larvae stage, in terms of organ morphogenesis and the expression pattern of selected target genes related to lipid metabolism and oxygen transportation. The results showed that the 120-h post-fertilization LC50 of 2,4-DNT was 9.59 mg/L with a 95% confidence interval of 8.89–10.44 mg/L. The larvae treated with 2,4-DNT showed toxic symptoms including smaller body, less skin pigment production, yolk malabsorption, and disordered liver development. Further studies on the expression of genes related to lipid transport and metabolism, and respiration indicated that they were significantly affected by 2,4-DNT. It is concluded that 2,4-DNT exposure perturbed liver development and yolk absorption in early-life zebrafish, and disturbed the lipid metabolism /oxygen transport gene expression. © 2019 by the authors. [目的]产肠毒素性大肠杆菌(ETEC)可通过毒力因子引起仔猪肠道的分泌性腹泻,本实验拟通过研究黄芩苷铝在体外对ETEC毒力因子的影响,探讨黄芩苷铝治疗仔猪腹泻的作用机制。[方法]设置空白对照组以及4个不同浓度的黄芩苷铝组(125、250、500、1000μg/mL),分别处理6 h后收集上清液,采用ELISA法测定药物作用后上清液中耐热肠毒素(STa、STb)和不耐热肠毒素(LT)的释放情况;设置空白对照组以及3个不同浓度的黄芩苷铝组(250、500、750μg/mL),在4.5h和6h收集菌体并提取细菌RNA,运用Q-RT PCR测定黄芩苷铝对ETEC肠毒素基因(estA、estB、elt-Ⅰ)、定居因子基因(faeD、faeG)及鞭毛蛋白基因(filC) mRNA表达的影响。[结果]ELISA测定结果表明,500、1000μg/mL的黄芩苷铝均能够显著抑制ETEC中STa、STb和LT肠毒素的释放(p<0.05);Q-RT PCR测定结果表明在4.5 h,500、750μg/mL的黄芩苷铝均能极显著抑制ETEC中estA、elt-Ⅰ、faeD、faeG、fliC的mRNA表达(p<0.01),显著抑制estB的mRNA表达(p<0.05);6 h时250、500和750μg/mL的黄芩苷铝能显著抑制ETEC中estA、elt-Ⅰ的mRNA表达(p<0.01)。[结论]黄芩苷铝能抑制ETEC肠毒素的释放和毒力因子的表达,提示可能用于治疗仔猪腹泻。

本研究利用脂多糖(LPS)诱导建立仔猪免疫损伤模型,研究黄芩苷对LPS诱导的仔猪血管紧密连接蛋白表达量和M LCK-M LC信号通路变化的影响,并探讨其保护机制.试验选取42头健康仔猪,分为空白对照组(A组)、LPS模型组(B组)、LPS+丙酮酸乙酯处理组(C组)、LPS+氟尼辛葡甲胺处理组(D)和LPS+黄芩苷处理组(剂量分别为25、50和100 mg/kg,E、F、G组).饲喂7d后,攻毒前0.5h进行药物处理,处理组及模型组腹腔注射LPS,空白对照组注射等量生理盐水,LPS诱导6h后再次给药.试验1d后,采集主动脉血管样品2份,通过免疫组化及免疫荧光观察血管中紧密连接蛋白(闭锁小带蛋白-1 (ZO-1)、密封蛋白-1(Claudin-1)和闭合蛋白(Occludin))的表达变化;Western blotting法测定血管MLCK-MLC信号通路相关蛋白(肌球蛋白轻链激酶(MLCK)、肌球蛋白轻链2(MLC2)和磷酸化肌球蛋白轻链2(p-M LC2))的表达量.结果 表明,LPS诱导后,仔猪血管中ZO-1、Claudin-1和Occludin蛋白表达量均极显著降低(P<0.01),黄芩苷处理能明显提高仔猪血管紧密连接蛋白的表达量.LPS模型组MLCK和p-MLC2蛋白表达量均极显著升高(P<0.01),黄芩苷能显著下调MLCK和p-MLC2表达量(P<0.05),各组间MLC2表达量差异不显著(P>0.05).综上所述,LPS能引起仔猪的血管损伤,激活M LCK-M LC信号通路,影响紧密连接蛋白表达量及功能,黄芩苷对LPS诱导的仔猪血管紧密连接损伤具有保护作用.本试验结果为黄芩苷用于治疗猪的炎性血管损伤提供了科学依据.

BACKGROUND: Haemophilus parasuis (HPS) is the causative agent of Glasser's disease, characterized by arthritis, fibrinous polyserositis and meningitis, and resulting in worldwide economic losses in the swine industry. Baicalin (BA), a commonly used traditional Chinese medication, has been shown to possess a series of activities, such as anti-bacterial, anti-viral, anti-tumor, anti-oxidant and anti-inflammatory activities. However, whether BA has anti-apoptotic effects following HPS infection is unclear. Here, we investigated the anti-apoptotic effects and mechanisms of BA in HPS-induced apoptosis via the protein kinase C (PKC)-mitogen-activated protein kinase (MAPK) pathway in piglet's mononuclear phagocytes (PMNP). RESULTS: Our data demonstrated that HPS could induce reactive oxygen species (ROS) production, arrest the cell cycle and promote apoptosis via the PKC-MAPK signaling pathway in PMNP. Moreover, when BA was administered, we observed a reduction in ROS production, suppression of cleavage of caspase-3 in inducing apoptosis, and inhibition of activation of the PKC-MAPK signaling pathway for down-regulating p-JNK, p-p38, p-ERK, p-PKC-alpha and PKC-delta in PMNP triggered by HPS. CONCLUSIONS: Our data strongly suggest that BA can reverse the apoptosis initiated by HPS through regulating the PKC-MAPK signaling pathway, which represents a promising therapeutic agent in the treatment of HPS infection.
