湿法纺丝制备胶原纤维丝及紫外辐照对其性能的调控

摘要:
为提高胶原基生物材料的热稳定性,同时保持胶原自身良好的生物相容性,首先以安全无毒的磷酸盐缓冲液(PBS)为固定浴,通过湿法纺丝制备胶原纤维丝材料,以增强热稳定性和抗拉伸性能为目标,优化了制备条件。在此基础上,进一步通过紫外辐照,提高了胶原纤维丝的热稳定性和耐酶降解性。最后,通过体外酶降解实验和细胞毒性实验探索了胶原纤维丝的耐酶降解性能和促细胞增殖性能。研究结果表明湿法纺丝制备的胶原纤维丝具备自组装性能,最佳制备条件为20 mg/mL的胶原溶液和30×PBS的固定浴。紫外辐照能够提高胶原纤维丝的热稳定性并增强其抗拉伸强度和耐酶降解性能,但30 min以上的紫外辐照会大幅降低其抗拉伸强度。紫外辐照后的胶原纤维丝仍具有良好的促细胞增殖能力,可为胶原材料的性能提升提供一种新途径。

Collagen-polymer conjugates are being developed rapidly based on advancements in both polymer and protein science. The attachment of polymers to proteins can be achieved with the "grafting to" method or the "grafting from" method; the latter is more attractive. Although polymer-based collagen engineering provides an attractive strategy to regulate the native functions of collagen, there are still two important unresolved issues: the harsh modification conditions and the loss of fibrillogenesis property. Herein, a collagen-PNIPAAm conjugate (PCol) was prepared by the "grafting from" method to avoid harsh conditions. The amino groups of collagen were modified by a water-soluble NHS-functionalized ATRP initiator before in situ polymerization of NIPAAm on the collagen surface. On the other hand, the fibrillogenesis of PCol was realized by the induction of natural collagen, inspired by the xenogeneic collagen hybrid fibrils in organisms. Furthermore, the structure, fibrillogenesis, thermosensitivity, and biocompatibility of PCol were explored. This work may be useful in synthesizing optimized formulate collagen-polymer conjugates and helpful for the design of the novel collagen-based hybrid materials.

Self-assembly is one of the most vital molecular behaviors of natural collagen, which plays a prominent role in the design and fabrication of collagen-based biomaterials. However, an efficient and sensitive assay for monitoring collagen self-assembly is still lacking. Herein, we developed a quantum dot (QD)-based fluorescence sensing platform and applied it to monitor collagen self-assembly. Before collagen self-assembly, we observed a strong fluorescent signal when ZnCdSe/ZnS QDs were introduced to grass carp skin collagen (GCSC). After self-assembly, the formation of collagen fibril networks induced the aggregation of QDs, resulting in the fluorescence quenching of QDs and a significant decrease in the intensity of fluorescence. Moreover, the quenching efficiency was found to be linearly dependent with GCSC concentration in the range of 0.1-1 mg mL-1, with a detection limit of 0.1 mg mL-1. In addition, the proposed strategy displayed excellent specificity and wide applicability. These results provide guidance for the design of collagen-based biomaterials and help to deeply understand the mechanism behind collagen self-assembly. Shear treatment is always employed in the extraction, preparation and research of collagen materials. This study is aimed at investigating the effect of the pre-shearing treatment of neutral bovine tendon collagen solution on the molecular structure, assembly behavior and gel properties of collagen. Rheological and assembly degree (AD) tests showed that with the increase in the shear rate or the extension of shear time during the pre-shearing treatment, the equilibrium moduli and AD of collagen gels that assemble from the pretreated samples gradually decreased. Scanning electron microscopy (SEM) results indicated that with the deepening of the pre-shearing treatment (increased shear rate or shear time), the diameter of the resultant collagen fibers gradually decreased, and their pore size and porosity increased as a whole. The viscosity test of the collagen solution obtained after pre-shearing revealed that the viscosities of the samples were lower than that of the sample devoid of pre-shearing. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and circular dichroism (CD) analysis manifested that the molecular weights and CD characteristic peaks of the collagen molecules remained unchanged in the sheared solution. However, with the deepening of the pre-shearing treatment, the proportion of α chains in the SDS-PAGE image increased, while the total content of β and γ chains decreased; the triple-helix structure of collagen in the CD spectrum decreased, and the random coiled structure increased. Moreover, transmission electron microscopy (TEM) displayed that the D-periods of the fibers assembled from the sheared samples were also decreased. A mechanism for the effect of the pre-shearing treatment of the collagen solution on the molecular structure, assembly behavior and gel properties of collagen has been proposed. The findings in this work may provide new understanding and theory on the physical modification of proteins and the construction of new materials from polymer systems that are aggregated or assembled based on weak intermolecular interactions. © The Royal Society of Chemistry and the Centre National de la Recherche Scientifique 2020.

Pure collagen materials are expensive with poor mechanical properties, which need modifications in most cases. As the degradation product of collagen, gelatin is cheap, degradable and biocompatible, but few literatures have reported the research about gelatin-collagen composite materials. This is because gelatin and collagen have different soluble temperatures—gelatin is soluble in hot water (≥30 °C) and swells in cold water. However, a low temperature (2–10 °C) is required to prepare and store collagen solution, and neutral collagen solution denatures quickly above the room temperature. In this study, gelatin was ground into powders and swelled in neutral bovine tendon pepsin-soluble collagen solution (BPSC) to form a homogeneous gelatin-collagen mixture, in light of the swelling characteristics of gelatin in cold water. The assembly properties and gel properties of this composite material were further studied. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) test results showed that the bovine tendon collagen had typical type-I collagen structural characterizations with two α chains of about 100 kDa and one β chain of about 200 kDa; while the SDS-PAGE pattern of gelatin displayed bands continuously distributed from 30 to 200 kDa. Amino acid composition analysis test indicated that the content of polar amino acids and the sum of acidic and base amino acids for gelatin were higher than that of BPSC. Studies on gel properties demonstrated that gelatin-collagen mixed solution had collagen-like assembly characteristics and assembly kinetics. The moduli of the assembled gel at 35 °C were equivalent to that of pure bovine tendon collagen system; moreover, the system moduli didn’t change with time with elastic moduli (G′) of about 40 Pa. However, at 25 °C, the moduli of gelatin-collagen composite hydrogel increased with the extension of time, its G′ increased about 18 times within 8 h, and the ratio of elastic modulus to viscous modulus (G″) increased 4.6 times, showing a significant aging effect of structural strength. Meanwhile, the mechanical strength of the composite hydrogel was also regulated by temperature—the gel was highly elastic (G′≈3,000 Pa, G′>>G″) at a low temperature (5 °C); as the temperature rose, the system moduli gradually decreased and the elastic gel transformed into waterlike fluid at 50 °C little by little. What’s more, gelatin-collagen composite hydrogel also had reversible sol-gel performances and self-healing capability similar to the gelatin hydrogel. This novel preparation method for preparing composite materials and the resultant composite hydrogel are expected to be used in the fields of natural food gels, injectable hydrogels, cell scaffolds, drug sustained-release materials and so on, and improve and promote the processing performances, price and large-scale production of collagen-based materials.[Figure not available: see fulltext.]. © 2020, The Polymer Society of Korea and Springer.

摘要:
Thermal denaturation is the primary reason for the loss of excellent performance for collagen-based materials. It is particularly urgent to explore an effective structure restoration strategy for thermally denatured collagen. In this study, three different degrees of collagen thermal denaturation models were constructed; the effects of ultrahigh pressure treatments of varying intensities on the structure and properties of partially thermal denatured collagen were investigated. Those results showed that ultrahigh pressure partially restores the triple helix conformation and the α helix of low thermally denatured collagen molecules and moderately restores collagen thermal stability. However, this recovery is not a 'perfect' structural restoration; it cannot effectively restore the fibrotic ability of collagen which leads to a decrease in the fiber diameter of the product and an increase in the storage modulus of the product gel. As the degree of thermal denaturation of collagen increases, the ultrahigh pressure recovery effect is gradually lost. For moderate thermally denatured collagen, ultrahigh pressure treatments only recover a small amount of its triple helical conformation. For high thermally denatured collagen, ultrahigh pressure treatments exacerbate the destruction of the three-dimensional structure of collagen molecules, thereby losing the order of molecular structure and the capability of fibrosis self-assembly. © 2019, Springer Science+Business Media, LLC, part of Springer Nature.

Xenogeneic collagen co-assembly (XCCA) is a novel and efficient strategy for improving and regulating the performance of collagen assembled products. However, the effect of collagen molecular structures on XCCA has long been neglected and remains unclear. Herein, we investigated the telopeptide effect of collagen on XCCA by employing grass carp skin collagen (GCSC) with two different structures (tropocollagen and atelocollagen) and porcine skin collagen (PSC). Kinetic analysis showed that the co-assembly rate of PSC and grass carp skin atelocollagen (AC-GCSC) was faster than that of PSC and grass carp skin tropocollagen (TC-GCSC). Based on this observation, we further obtained different concentration dependences of XCCA by the alteration of telopeptide. Compared with PSC/TC-GCSC co-assembly, PSC/AC-GCSC co-assembly exhibited a higher sensitivity and a lower detection limit. Furthermore, telopeptide promoted the fabrication of thicker fibrils and improved the mobility of gels.

Heterotypic collagen fibril has long been found in the tissues of organisms, which plays an important role in the formation and function of complex structures of organisms. Inspiring by the phenomenon, scholars tried to incubate collagens from different sources into novel collagen materials in vitro, and the forming of heterogenic reconstituted collagen fibrils (RF) was often demonstrated by differential scanning calorimetry (DSC) and fluorescence quenching analysis. In this work, we used two type-I collagens from different species (bovine tendon and grasscarp fish skin) to co-assemble in vitro, and verified the formation of RF from a new rheological perspective. In addition, we also investigated the assembly behavior, surface hydrophilicity and hydrophobicity, microscopic morphology and cell proliferation ability of the RF. The results showed that the assembly rate, surface properties, fibril size, viscoelastic properties of RF can be delicately regulated by the method of heterogenic collagen reconstitution. This study provides new experimental evidence for the reconstitution of heterogenic collagens, and also offers a new means for the regulation of collagen gel performance, which would help to expand the application range of collagen gel materials. 摘要:
桂花富含天然抗氧化成分,在天然抗氧化剂领域具有良好的开发应用潜力。本研究以制取香料浸膏后的桂花残渣为原料,以乙醇为溶剂提取桂花中的抗氧化物质,得到桂花乙醇粗提物,并对该粗提物开展溶解性能、pH稳定性和热稳定性等理化性质分析。随后我们运用硅胶柱层析技术,对该粗提物进行极性分级,并对不同分级产物开展体外抗氧化活性的比较分析。在此基础上,进一步对具有良好抗氧化活性的分级产物开展了初步的结构鉴定,结果表明:桂花乙醇萃取物在溶解度参数大于10的溶剂中有较大的溶解度;其在中性、酸性和温度低于60℃的条件下较为稳定;此乙醇粗提物经硅胶柱层析分离可以得到3个分级产物。体外抗氧化活性评价结果表明,分级产物中组分1(Frac1)具有良好的1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除和还原能力,而组分3(Frac3)抑制亚油酸氧化的能力最强。高效液相色谱-质谱联用分析结果表明,Frac1中活性成分主要为黄酮甙类物质,而Frac3中的主要活性组分为双黄酮类物质。

摘要:
Side chain modification of collagen provides an attractive way to enhance their structure and functions, which is highly desirable for the development of promising biomaterials. However, the impact of structural change of side chains on the intrinsic self-assembly property of collagen was always ignored. Here, a series of acrylic acid-grafted-collagen (AA-g-Col) with different grafting density were prepared to explore the impact of side chain structural variation on the self-assembly of collagen. The results showed that excessive grafting density would weaken or even disappear the self-assembly property of AA-g-Col, but only affects the triple helix to a minor extent. Compared to pristine collagen, the mechanical property and cytocompatibility of AA-g-Col based matrices also deteriorated, along with the increase of grafting density. Therefore, this work contributed a new insight into the importance of grafting density for the study of modified collagen, which would be helpful for the design of optimized formulate collagen-based hybrid materials with both additional novel functions and tissue-mimicking fibrillary structures.

摘要:
A sensitive and robust electrochemical aptasensor for determining dopamine (DA) was developed using a grass carp skin collagen-graphene oxide (GCSC-GO) composite as a transducer and a label-free aptamer as a biological recognition element for the first time. In order to fabricate this sensor, the GCSC-GO composite was firstly prepared by ultra-sonication method and characterized by atomic force microscope, infrared spectroscopy, Raman spectroscopy, and electrochemical impedance spectroscopy. Subsequently, a label-free DA-binding aptamer was immobilized through strong interaction between collagen and aptamer. The fabricated electrochemical aptasensor was used to determine DA by differential pulse voltammetry. The results indicated that the peak current changes of the developed aptasensor was linear relationship with the DA concentrations from 1 to 1000 nM, and the detection limit was 0.75 nM (S/N = 3). Moreover, the fabricated aptasensor showed high selectivity for DA More importantly, the obtained aptasensor exhibited satisfactory recovery toward DA in human serum specimens with excellent stability. Abstract: Composite bacterial cellulose (BC) membranes containing collagen fibrils or collagen monomers (named BC/Col-fibre or BC/Col, respectively) were fabricated by immersing macroporous BC in a 3.0mg/mL collagen solution for 50h at 4°C with or without collagen fibrillogenesis before lyophilization. The structure of the membranes was characterized by scanning electron microscope, nitrogen adsorption–desorption experiment, and X-ray diffraction. The structure became much more compact upon the introduction of collagen but the crystal structure of BC did not change. The thermal stability and the cytocompatibility of the membranes were evaluated by thermogravimetric analysis and a cell adhesion assay, respectively. The thermal stability of BC was enhanced by collagen incorporation and by fibrillogenesis. The adhesive ability and the proliferation of NIH/3 T3 fibroblast cells in BC/Col-fibre were better than that in BC/Col, which was still superior to that in BC. The collagen amount was improved to 12% by immersing the macroporous BC in a 3.0mg/mL collagen solution for 50h at 4°C. Phytic acid, containing a myoinositol ring coupled with six phosphate groups, can react with the amino groups of collagen to regulate their self-assembly behavior. The aim of this research is to evaluate the effects of phytic acid on the selfassembly behavior of collagen, the structures and properties of the resulting fibrils and hydrogels. Turbidity and chloramine T assay suggested that phytic acid could improve the self-assembly kinetics and degree of collagen, and the optimal ratio of phytic acid/collagen was 1/1 (w/w). Scanning electron microscopy (SEM) analysis indicated that co-fibrils of collagen with phytic acid are more slender than that of pure collagen, and transmission electron microscopy (TEM) reveals that the characteristic D-periodicity of collagen fibrils is not affected by phytic acid. Besides, differential scanning calorimetry (DSC) and rheology revealed that the thermal stability of collagen fibrils and the viscoelasticity of collagen hydrogels could be improved by phytic acid and the optimal ratio of phytic acid/collagen is 1/1 (w/w). Ultra-high pressure technology has attracted a great deal of attention in recent years, and has been widely used in food science, medicine, and other fields. This study aimed to determine the effect of ultra-high pressure on the structure and properties of collagen. Native collagen extracted from bullfrog skin was processed under different ultra-high pressure treatment conditions (300, 400, and 500 MPa). Then systematic analysis of the molecular structures and properties of the samples after ultra-high pressure treatment were performed. It was found that the conformation of collagen molecules could be adjusted by ultra-high pressure treatment, and this regulation was closely related to the level of treatment pressure. A possible mechanism of the impact of ultra-high pressure on the collagen molecular structures was speculated according to the experimental results. At low pressure levels (300-400 MPa), the pressure perpendicular to collagen axis dominates and leads to a tightening of the triple helix structure of collagen, while the pressure parallel to collagen axis is dominant and the triple helix tends to dissociate like a zipper at high pressure levels (> 400 MPa). These structural changes would simultaneously result in various changes to thermal stability, self-assembly properties, and antigenicity of collagen. (C) 2018 Elsevier B.V. All rights reserved.

体外纤维重组是天然胶原的重要分子行为,直接影响胶原的性能和应用。本研究以草鱼胶原为研究对象,系统观察了体系温度、pH、离子强度和时间等环境因素对重组胶原纤维形貌和粒径分布的影响。在此基础上,进一步比较了不同环境因子调控下,胶原重组纤维产物的耐酶降解性能和热稳定性能。结果表明,重组胶原纤维粒径和粒径分布受环境因子的调控。中性pH环境下胶原纤维粒径大但分布不均一,体系温度越高、离子强度越大,重组纤维粒径越小,延长纤维重组时间有利于纤维粒径的增大。纤维产物性能分析结果表明,纤维粒径越大,则胶原重组纤维产物的耐酶降解性能和热稳定性能越好。

Self-assembled environment of collagen is one of the important factors for improving and regulating the properties of collagen-based biomaterials. This study aimed to investigate the effect of ionic liquids (ILs) on the fibril-formation and gel properties of grass carp (Ctenopharyngodon idellus) skin collagen. Fibrillogenic kinetics analysis showed that the collagen self-assembly can be suppressed by the introduction of ILs, and the inhibitory effect is influenced by concentration and types of ILs. Scanning electron microscopy test indicated that the assembled collagen fibrils in the presence of ILs had bigger diameters than that in the conventional buffer. Differential scanning calorimetry analysis revealed that the thermal stability of collagen fibrils can be significantly increased when self-assembly is performed in the presence of ILs. Moreover, the introduction of ILs enhanced the mechanical strength of collagen gels. Finding from this work provides a new idea for improving the performance of fish-sourced collagen biomaterials. © 2018, The Polymer Society of Korea and Springer Science+Business Media B.V., part of Springer Nature.

摘要:
In this study, the effects of collagen self-assembly, drying methods, and reswelling on collagen gel structure and biological properties were determined. The results showed that the self-assembly of collagen improved the regularity and pore size of the collagen gel and promoted collagen gel thermostability and cell proliferation. After drying and reswelling, the main structure of the self-assembly gel was composed of lamellar gel walls, but the gel structure shrank to some extent and pore size was reduced. The structure of the self-assembled gel after drying and reswelling was closely related to that of the drying method. © 2017 Taylor & Francis.
