Vibrio parahaemolyticus usually occurs in coastal areas and is generally recognized as a marine bacterium. It has become the leading cause of gastroenteritis worldwide. In the present study, 96 V. parahaemolyticus isolates were obtained from freshwater crayfish (Procambarus clarkii) and classified by multilocus sequence typing. Fifty-three sequence types (STs) were identified among the 96 isolates analyzed, 38 of which were novel STs. These isolates fell into six groups and 42 singletons, suggesting a high level of genetic diversity. Screening for 9 virulence and virulence-related genes in the isolates revealed that 40 isolates contained more than two genes with possible roles in pathogenicity. The virulence of the representative isolates VP66 (trh(+), ureC(+), T3SS1(+), T3SS2 beta(+), T6SS2(+)) and VP80 (T3SS1(+), T6SS1(+), T6SS2(+)) were further assessed in zebrafish and mouse infection model in vivo, and the tested isolates were shown to be lethal to both zebrafish and mice. These results suggest that crayfish may serve as a carrier of V. parahaemolyticus in freshwater, and that some isolates may have the potential to cause foodborne disease in humans. The effects of n3 polyunsaturated fatty acids (PUFA) on cardiovascular disease are controversial. We currently explored the effects of various ratios of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) on high-fat-induced atherosclerosis. In model apoE(-/-) mice, high-fat diets (HFD) were partially replaced with fish and algal oils (DHA/EPA 2:1, 1:1 and 1:2) and/or plant oils enriched in linoleic and alpha-linolenic acids with an n6/n3 ratio of 4:1. PUFA supplementation significantly reduced the atherosclerotic plaque area, serum lipid profile, inflammatory response, aortic ROS production, proinflammatory factors and scavenger receptor expression as compared to those in the HFD group. However, plant oils did not have a significant effect on the following: serum HDL-C level; aortic ABCA1, ABCG1 and LAL mRNA expression; and CD36 and LOX-1 protein expression. Compared to the plant-oil-treated group, the DHA/EPA 1:1 group had a smaller atherosclerotic plaque area, higher serum HDL-C levels and lesser CD36 and MSR-1 mRNA expression; the DHA/EPA 2:1 group had lower serum TC, LDL-C and TNF-a levels and lower aortic ROS levels. Our study suggested that n3 PUFA from animals had more potent atheroprotective effects than that from plants. Supplementation involving higher DHA/EPA ratios and an n6/n3 ratio of 4:1 was beneficial for reducing serum "bad cholesterol" and a 1:1 DHA/EPA ratio with an n6/n3 ratio of 4:1 was beneficial for improving serum "good cholesterol" and inhibiting ox-LDL uptake. Our results suggest that achieving an n6/n3 ratio of 4:1 in the diet is also important in addition to having an optimal DHA/EPA ratio. (C) 2016 Elsevier Inc. All rights reserved.

A dual extraction based on solid phase extraction (SPE) and solidified floating organic drop microextraction (SFODME) was developed for As species in tea leaves and tea infusion by electrothermal vaporization inductively coupled plasma mass spectrometry, including total, suspended, soluble, organic and inorganic As as well as As(III) and As(V). In SPE step, titanium dioxide nanotubes were used for preconcentration of analytes and removal of sample matrix. Elution solution from SPE was employed for further preconcentration and separation of analytes with SFODME. Under optimal conditions, detection limits of this method were 0.046 and 0.072 pg mL-1 with relative standard deviations of 6.3% and 5.8% for As(III) and As(V) (n = 9, c = 1.0 ng mL-1), respectively. A preconcentration factor of 500-fold was achieved for As(III) and As(V). This method was successfully applied for analysis of speciation of arsenic and its distribution in tea leaves, tea infusion and certified reference material of tea leaves. © 2016 Elsevier Ltd.

Food-grade nanoemulsions are potential vehicles of labile lipophilic compounds such as β-carotene, but much work is needed to improve physical and chemical stabilities. The objective of this work was to study impacts of eugenol on physical and chemical stabilities of β-carotene-loaded nanoemulsions prepared with whey protein and lecithin. The combination of whey protein and lecithin resulted in stable nanoemulsions with eugenol added at 10% mass of soybean oil. Nanoemulsions, especially with eugenol, drastically reduced the degradation of β-carotene during ambient storage, heating at 60 and 80 °C, and UV radiation at 254, 302, and 365 nm. The droplet diameter of the nanoemulsion without eugenol increased from 153.6 to 227.3 nm after 30-day ambient storage, contrasting with no significant changes of nanoemulsions with eugenol. Heating or UV radiation up to 8 h did not significantly change the droplet diameter. Therefore, eugenol can be used to improve the stability of nanoemulsion delivery systems. © 2015 Published by Elsevier Ltd.

Chitosan/whey protein isolate film incorporated with sodium laurate-modified TiO<inf>2</inf>nanoparticles was developed. The nanocomposite film was characterized by scanning electron microscopy, X-ray diffraction and differential scanning calorimetry, and investigated in physicochemical properties as color, tensile strength, elongation at break, water vapor permeability and water adsorption isotherm. Our results showed that the nanoparticles improved the compatibility of whey protein isolate and chitosan. Addition of nanoparticles increased the whiteness of chitosan/whey protein isolate film, but decreased its transparency. Compared with binary film, the tensile strength and elongation at break of nanocomposite film were increased by 11.51% and 12.01%, respectively, and water vapor permeability was decreased by 7.60%. The equilibrium moisture of nanocomposite film was lower than binary film, and its water sorption isotherm of the nanocomposite film fitted well to Guggenheim-Anderson-deBoer model. Lotus root attracts increasing attention mainly because of its phenolic compounds known as natural antioxidants. Its thirteen varieties were systematically analyzed on the content, distribution, composition and antioxidant activity of phenolic compounds for a better understanding of this aquatic vegetable. The respective mean contents of total phenolics in their flesh, peel and nodes were 1.81, 4.30 and 7.35 mg gallic acid equivalents (GAE)/g fresh weight (FW), and those of total flavonoids were 3.35, 7.69 and 15.58 mg rutin equivalents/g FW. The phenolic composition determined by a high-performance liquid chromatography method varied significantly among varieties and parts. The phenolics of flesh were mainly composed of gallocatechin and catechin; those of peel and node were mainly composed of gallocatechin, gallic acid, catechin and epicatechin. The antioxidant activities of phenolic extracts in increasing order were flesh, peel and node; their mean concentrations for 50% inhibition of 2,2-diphenyl-1-picrylhydrazyl radical were 46.00, 26.43 and 21.72 μg GAE/mL, and their mean values representing ferric reducing antioxidant power were 75.91, 87.66 and 100.43 μg Trolox equivalents/100 μg GAE, respectively. "Zoumayang", "Baheou", "No. 5 elian" and "Guixi Fuou" were the hierarchically clustered varieties with relatively higher phenolic content and stronger antioxidant activity as compared with the others. Especially, their nodes and peels are promising sources of antioxidants for human nutrition. © 2016 by the authors; licensee MDPI, Basel, Switzerland.

摘要:
This study is based on the development of bacterial count quantification generated by medium dielectric variations and consequent polarization material release. The proposed approach is an action method for the fast detection of bacterial concentration in orange juice. The sensing method relies on bacterial attachment up to biofilm formation. Furthermore, different media provide more oxygen group content to enhance capacitance and self-discharge. The test took only 30 min and it provided the means for rapid bacterial detection in the juice industry. © 2015 Elsevier Ltd. All rights reserved.

Three water-soluble hetero-polysaccharides, designated LP1–3, were isolated from litchi pulp. Their structures, solution properties and immunomodulatory activities were evaluated. LP1 contained (1 → 4,6)-β-D-Glc and (1 → 4)-α-L-Gal, while LP2 contained (1 → 3)-α-L-Ara and (l → 2)-β-D-Gal, and LP3 contained α-L-Ara and (l → 4)-β-Rha. Their molecular weights ranged from 105,880 to 986,470 g/mol. LP1 had a spherical conformation with hyper-branched structure and LP2 was semi-flexible chain, while the polysaccharide chains of LP3 were cross linked to form network-like conformation in solution. In addition, all fractions strongly stimulated mesenteric lymph node cell proliferation, IFN-γand IL-6 secretion in the dose range of 25–100 μg/mL compared with untreated control group (p < 0.05). LP1 exhibited the strongest stimulation of mesenteric lymph node cell proliferation and cytokine secretion, which may be attributed to its unique chemical structure and chain conformation. This is the first report on the solution properties and intestinal immunity activities of polysaccharides from litchi pulp.<br/> ©2016 Elsevier Ltd

In this paper, 13 4-N-nitrophenyl substituted amino-4H-1,2,4-triazole derivatives were synthesized and their aromatase inhibitory activities were measured. The results show that the substitution of the groups on benzyl group can further improve their bioactivity and the compound with Cl on the para position of benzyl has the highest bioactivity (IC50= 9.02 nM). A QSAR model was constructed from the 13 compounds with genetic function approximation using DS 2.1 package. This model can explain 90.09% of the variance (R2Adj), while it can predict 84.95% of the variance (R2cv) with the confidence interval of 95%. © 2016 Elsevier Ltd

This work provides an evaluation of an ultrasound-assisted, combined extraction, centrifugation and ultrafiltration process for the optimal recovery of polyphenols. A purple sweet potato (PSP) extract has been obtained using ultrasonic circulating extraction equipment at a power of 840 W, a frequency of 59 kHz and using water as solvent. Extract ultrafiltration, using polyethersulfone (PES), was carried out for the recovery of polyphenol, protein and anthocyanin. Pre-treatment, via the centrifugation of purple sweet potato extract at 2500 rpm over 6 min, led to better polyphenol recovery, with satisfactory protein removal (reused for future purposes), than PSP extract filtration without centrifugation. Results showed that anthocyanin was efficiently recovered (99%) from permeate. The exponential model fit well with the experimental ultrafiltration data and led to the calculation of the membrane's fouling coefficient. The optimization of centrifugation conditions showed that, at a centrifugation speed of 4000 rpm (1195x g) and duration of 7.74 min, the optimized polyphenol recovery and fouling coefficient were 34.5% and 29.5 m -1 , respectively. The removal of proteins in the centrifugation process means that most of the anthocyanin content (90%) remained after filtration. No significant differences in the intensities of the HPLC-DAD-ESI-MS 2 peaks were found in the samples taken before and after centrifugation for the main anthocyanins; peonidin-3-feruloylsophoroside-5-glucoside, peonidin-3-caffeoyl-p-hydroxybenzoylsophoroside-5-glucoside, and peonidin-3-caffeoyl-feruloyl sophoroside-5-glucoside. This proves that centrifugation is an efficient method for protein removal without anthocyanin loss. This study considers this process an ultrasound-assisted extraction-centrifugation-ultrafiltration for purple sweet potato valorization in "green" technology. © 2016 by the authors; licensee MDPI.

摘要:
In recent decades, the polysaccharides from the medicinal plants have attracted a lot of attention due to their significant bioactivities, such as anti-tumor activity, antioxidant activity, anticoagulant activity, antidiabetic activity, radioprotection effect, anti-viral activity, hypolipidemic and immunomodulatory activities, which make them suitable for medicinal applications. Previous studies have also shown that medicinal plant polysaccharides are non-toxic and show no side effects. Based on these encouraging observations, most researches have been focusing on the isolation and identification of polysaccharides, as well as their bioactivities. A large number of bioactive polysaccharides with different structural features and biological effects from medicinal plants have been purified and characterized. This review provides a comprehensive summary of the most recent developments in physiochemical, structural features and biological activities of bioactive polysaccharides from a number of important medicinal plants, such as polysaccharides from Astragalus membranaceus, Dendrobium plants, Bupleurum, Cactus fruits, Acanthopanax senticosus, Angelica sinensis (Oliv.) Diels, Aloe barbadensis Miller, and Dimocarpus longan Lour. Moreover, the paper has also been focused on the applications of bioactive polysaccharides for medicinal applications. Recent studies have provided evidence that polysaccharides from medicinal plants can play a vital role in bioactivities. The contents and data will serve as a useful reference material for further investigation, production, and application of these polysaccharides in functional foods and therapeutic agents. © Taylor & Francis Group, LLC.

In this study, the influence of tea extract (TE) on the growth of probiotics in skim milk was examined. Lactobacillus plantarum ST‐III, Bifidobacterium bifidum Bb02, Lactobacillus acidophilus NCFM, and Lactobacillus rhamnosus GG were used in this study. The introduction of TE in milk significantly stimulated the propagation and acidification of L. rhamnosus GG and L. acidophilus NCFM. The antioxidant capacities and the total free amino acid contents of all fermented milk products were enhanced by the addition of TE; however, there were different antioxidant properties and free amino acid contents of fermented milk samples fermented by different bacteria. With a 9% (w/w) level, the fermentation with L. rhamnosus GG and L. acidophilus NCFM showed larger numbers of viable cells and faster acidifying rates, as well as excellent antioxidant capacity and abundant free amino acids. The stimulative effects of TE on probiotics can be considered for industrial purposes and has practical implications for commercial applications. Portuguese This study investigated the influence of tea extract on growth of 4 probiotics in skim milk. The results showed TE contributed to improving the propagation and production of metabolites for probiotics, shortening fermentation time, and enhancing the nutritional quality of the product.

In this study, fibrillogenesis and thermal dissociation of pepsin-soluble collagen (PSC), extracted from snakehead (Channa argus) skin, were monitored by fluorescence method based on thioflavin T (Th-T), where the accuracy and sensitivity were evaluated and compared with those of turbidity assay. The fluorescence method revealed the fibrillogenesis dynamics of collagen with better sensitivity, especially at nucleation and plateau stages. The melting temperature (T-m) of PSC was estimated to be 47 degrees C by circular dichroism spectroscopy; below this temperature, the triple-helical structure should be intact. After that, the dynamic process of collagen dissociation was explored by the fluorescence method, and verified by morphological analysis of the fibrils and the proportion of retained fibrils. The thermal dissociation critical temperature (TDCT) of PSC fibrils was confirmed to be 39 degrees C. The fluorescence intensity of fibril-incorporated Th-T gradually decreases in the dissociation process, and the decrease rate can be accelerated by increasing temperature. Finally, the thermal stability of triple-helical structures of free-, assembled- and dissociated-PSC was compared. Thus, we demonstrated the formation and thermal dissociation of collagen fibrils in vitro by a fluorescence method based on Th-T. This approach may advance the understanding of fibril formation and inverse dissociation of fish-sourced collagen in vitro. (C) 2016 Elsevier B.V. All rights reserved.

Folate is a vital vitamin for the human being and its deficiency can lead to a variety of clinical abnormalities ranging from neural tube defects to cancers. Benzo(a)pyrene (BaP), a strong mutagen and carcinogen, is considered one of the common contaminants in food. The aim of this study was to investigate the positive effect of folate on cancer prevention at a fundamental level. In the present study, we investigated the impact of folic acid on BaP-induced mutagenicity and genotoxicity by means of in vitro and in vivo experiments. The reformed Ames test was applied to study the antimutagenicity of folic acid against BaP. The protective effect of folic acid on cytotoxicity caused by BaP in human liver cell line LO2 was evaluated by MTT assay. In addition, the effect of folic acid on the BaP-induced genotoxicity in vivo was assessed by mouse bone marrow micronucleus assay. The results indicated that folic acid significantly inhibited the reverse mutation of Salmonella typhimurium strains TA98 and TA100, and protected the viability of human liver cells against BaP (p<0.01). The micronucleus test showed that all doses of folic acid had a remarkable protective effect for the female mice (p<0.01). In conclusion, folic acid was found to reduce the mutagenicity and genotoxicity induced by BaP.

Three litchi polysaccharide fractions (LPFs), LP-4, LP-6 and LP-8, were obtained by fractional precipitation using 40%, 60% and 80% ethanol, respectively. The physicochemical properties, chemical antioxidant, and cellular antioxidant and antiproliferative activities of the three polysaccharide fractions were compared. LP-6 contained the highest contents of uronic acid and binding protein among the three fractions, whereas LP-8 contained the least. Amino acid composition analysis of the binding protein revealed that LP-6 contained the most acidic and aromatic amino acids. However, LP-8 contained more galactose and mannose than LP-4 and LP-6. LP-6 exhibited the highest chemical antioxidant activities, with an oxygen radical absorbance capacity of 28.14 μmol TE per g DW. LP-8 exhibited higher cellular antioxidant activity and a greater inhibitory effect on the proliferation of A549, HepG2 and MGC-803 cells at a concentration of 100-800 μg mL-1 than LP-4 and LP-6. In summary, the different LPFs exhibited different antioxidant and antiproliferative activities with differential physicochemical properties. © The Royal Society of Chemistry.

NAC genes have been characterized in numerous plants, where they are involved in responses to biotic and abiotic stress, including low oxygen stress. High concentration of CO2 is one of the most effective treatments to remove astringency of persimmon fruit owing to the action of the accumulated anoxia metabolite acetaldehyde. In model plants, NAC genes have been identified as being responsive to low oxygen. However, the possible relationship between NAC transcription factors and persimmon astringency removal remains unexplored. In the present research, treatment with a high concentration of CO2 (95%) effectively removed astringency of “Mopan” persimmon fruit by causing decreases in soluble tannin. Acetaldehyde content increased in response to CO2 treatment concomitantly with astringency removal. Using RNA-seq and Rapid amplification of cDNA ends (RACE), six DkNAC genes were isolated and studied. Transcriptional analysis indicated DkNAC genes responded differentially to CO2 treatment; DkNAC1, DkNAC3, DkNAC5 and DkNAC6 were transiently up-regulated, DkNAC2 was abundantly expressed 3 days after treatment, while the DkNAC4 was suppressed during astringency removal. It is proposed that DkNAC1/3/5/6 could be important candidates as regulators of persimmon astringency removal and the roles of other member are also discussed. © 2015 by the authors; licensee MDPI, Basel, Switzerland.

A high performance size exclusion-fluorescence detection (HPSEC-FD) method combined with fluorescein isothiocyanate (FITC) prelabeling was established for the microanalysis of polysaccharide–protein complexes from longan pulp (LPP). FITC-labeled LPP (LPPF) was fractionated by gel filtration chromatography. The weight-average molecular weight and FITC substitution degree of LPPF were 39.01 kDa and 0.20%, respectively. The HPSEC-FD calibration curves linear over the range of 1–200 μg/mL in mouse plasma, spleen and lung samples with correlation coefficients greater than 0.995. The inter-day and intra-day precisions of the method were not more than 6.9%, and the relative recovery ranged from 93.7% to 106.4%. The concentration–time curve of LPPF in plasma following intravenous (i.v.) administration at 40 mg/kg body weight well fitted to a two-compartment model. LPPF rapidly eliminated from plasma according to the short half-lives (t1/2α = 2.23 min, t1/2β = 39.11 min) and mean retention times (MRT0–t = 1.15 h, MRT0–∞ = 1.39 h). After administration over 5 to 360 min, the concentration of LPPF in spleen homogenate decreased from 7.41 to 3.68 μg/mL; the concentration in lung homogenate decreased from 9.08 to 3.40 μg/mL. This study investigated the protective properties of lotus seedpod oligomeric procyanidins (LSOPC) against nonalcoholic fatty liver disease (NAFLD) and its underlying mechanism. Sprague-Dawley (SD) male rats were fed a basic diet, a high-fat diet (HFD), or HFD plus 0.2 or 0.5% (w/w) LSOPC for 12 weeks. Administration of LSOPC markedly reduced serum and hepatic biochemical parameters and protein expression of advanced glycation endproducts (AGEs). Additionally, 0.5% (w/w) LSOPC treatment remarkably reversed the increasing tendency of receptor of advanced glycation endproduct (RAGE) to normal level. Furthermore, dietary LSOPC significantly decreased the protein levels of mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-κB) and down-regulated genes involved in pro-inflammatory cytokines and adhesion molecules. Taken together, these findings demonstrate that LSOPC may protect obese rats with long-term HFD-induced NAFLD against RAGE-MAPK-NF-κB signaling suppression.<br/> ©2015 American Chemical Society.

摘要:
The effect of media milling on lipid-lowering and antioxidant activities of chitosan was studied in rats fed high-fat diets. Results showed that media-milled chitosan was more effective than chitosan in reducing body weight gain and liver fat accumulation of rats. Compared with chitosan, the reducing effects of media-milled chitosan on serum triacylglycerol (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were increased by 10.1, 7.5 and 10.2%, and liver TG and TC-reducing effects were increased by 16.2 and 14.6%, respectively. Rats fed media-milled chitosan showed decreased levels of free fatty acid (FFA) and thiobarbituric acid-reactive substances (TBARS) and increased superoxide dismutase (SOD) activity. The cholesterol and fat in feces of rats fed media-milled chitosan were higher than those of rats fed chitosan. These results suggested media milling enhanced the lipid-lowering and antioxidant activities of chitosan, and the reason might be partly due to its effect on strengthening the ability of chitosan in promoting fecal lipid excretions. A growing body of evidence has indicated that high-fat diet-induced nonalcoholic fatty liver disease is usually accompanied by oxidized low-density lipoprotein (ox-LDL) deposited in the liver. The current study aimed to investigate the effect of quercetin on high-fat diet-induced ox-LDL accumulation in the liver and to explore the potential underlying mechanisms. The results demonstrate that quercetin supplementation for 24 weeks significantly alleviated high-fat diet-induced liver damage and reduced hepatic cholesterol and ox-LDL level. Quercetin notably inhibited both mRNA and protein expression of CD36 (reduced by 53% and 71%, resp.) and MSR1 (reduced by 25% and 45%, resp.), which were upregulated by high-fat diet. The expression of LC3II was upregulated by 2.4 times whereas that of p62 and mTOR was downregulated by 57% and 63% by quercetin treatment. Therefore, the significantly improved autophagy lysosomal degradation capacity for ox-LDL may be implicated in the hepatoprotective effect of quercetin; scavenger receptors mediated ox-LDL uptake might also be involved. © 2015 Liang Liu et al.
