摘要:
为研究日粮中添加羧甲基纤维素钠(Sodium Carboxymethyl Cellulose,CMC)对脂多糖(Lipopoly saccharide,LPS)刺激仔猪肝脏损伤的保护作用,本试验选用 24 头(8.09±0.23)kg杜×长×大断奶仔猪,随机分为 4 组,即对照组(基础日粮)、CMC组(基础日粮+0.2%CMC)、LPS组(基础日粮+LPS)、LPS+CMC组(基础日粮+0.2%CMC+LPS).在试验第 17 天,LPS组和LPS+CMC组仔猪腹腔注射LPS,对照组和CMC组注射等量生理盐水.4 h后采血,屠宰取肝脏样品待测.结果表明:日粮添加CMC降低了血浆谷草转氨酶(AST)、碱性磷酸酶(ALP)活性(P<0.05),缓解了LPS导致的肝脏病理损伤;日粮添加CMC缓解了LPS刺激导致肝脏过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性的降低及环氧合酶 2(COX2)mRNA表达量的升高(P<0.05);日粮添加CMC降低了肝脏TNF-α和IL-6 水平及髓样分化因子88(MyD88)、核苷酸结合寡聚域受体1(NOD1)、白细胞介素-6(IL-6)(P<0.05)及肿瘤坏死因子-α(TNF-α)(P<0.001)mRNA表达量.由此可见,CMC通过提高肝脏抗氧化能力,降低炎性因子水平,缓解LPS刺激导致的肝脏损伤.

本试验旨在研究日粮中添加甘氨酸(Gly)对脂多糖(LPS)应激断奶仔猪肠黏膜屏障损伤的保护作用。选用24头21日龄断奶仔猪,随机分为4组,即对照组(基础日粮)、LPS组(基础日粮+LPS)、1.0%Gly组(基础日粮+1.0%Gly+LPS)、2.0%Gly组(基础日粮+2.0%Gly+LPS)。预饲期7 d,正试期28 d,在试验第28天,LPS组、1.0%Gly组和2.0%Gly组仔猪腹腔注射LPS(100μg/kg BW),对照组注射无菌生理盐水。注射4 h后,仔猪屠宰,取脾脏、肝脏、肠系膜淋巴结及空肠、回肠样品待测。结果表明:(1)各处理组的平均日增重、平均日采食量及耗料增重比无显著差异。(2)与对照组相比,LPS应激升高了回肠丙二醛(MDA)水平(P<0.05),降低空肠、回肠谷胱甘肽(GSH)水平(P<0.05);与LPS组相比,日粮中添加Gly降低回肠MDA水平、升高GSH水平(P<0.05)。(3)与对照组相比,LPS应激增加肝脏移位细菌数(P<0.05);与LPS组相比,日粮中添加Gly降低肝脏移位细菌数(P<0.05)。(4)与对照组相比,LPS应激降低空肠绒毛高度/隐窝深度比值(P<0.05),且有降低空肠绒毛高度趋势(P<0.1);与LPS组相比,日粮中添加Gly对空肠、回肠的绒毛高度、隐窝深度及绒毛高度/隐窝深度比值没有显著影响。(5)与对照组相比,LPS应激增加回肠肥大细胞密度(P<0.05);与LPS组相比,日粮中添加Gly降低空肠肥大细胞密度(P<0.05)。(6)与对照组相比,LPS应激升高空肠、回肠CRF(P<0.05)、CRFR1(P<0.001)和GR(P<0.05)的mRNA相对表达量;与LPS相比,日粮添加Gly降低了空肠、回肠CRFR1(P<0.05)及空肠GR(P<0.001)的mRNA相对表达量。结果提示,日粮添加Gly可缓解LPS应激导致的肠黏膜屏障功能障碍。

<jats:p><jats:bold>Background:</jats:bold> Holly (<jats:italic>Ilex latifolia</jats:italic> Thunb.) polyphenols extracts (HPE) contain high amounts of polyphenols, including phenolic acids, triterpenoids, tannic acids, and so on, which have strong antioxidant function. This experiment was aimed to explore the protective effect and mechanism of HPE against hepatic injury induced by diquat.</jats:p><jats:p><jats:bold>Methods:</jats:bold> Thirty-two weaned piglets were allotted by a 2 × 2 factorial experiment design with diet type (basal diet vs. HPE diet) and diquat challenge (saline vs. diquat). On the 21st day, piglets were injected with diquat or saline. One week later, blood samples were collected. Then all piglets were slaughtered and hepatic samples were collected.</jats:p><jats:p><jats:bold>Results:</jats:bold> Dietary HPE supplementation improves hepatic morphology, the activities of plasma aspartate aminotransferase, alanine aminotransferase, and glutamyl transpeptidase, and enhances hepatic anti-oxidative capacity, while it regulates the expression of ferroptosis mediators (transferrin receptor protein 1, heat shock protein beta 1, solute carrier family 7 member 11, and glutathione peroxidase 4) in diquat-challenged piglets.</jats:p><jats:p><jats:bold>Conclusion:</jats:bold> These results indicate that dietary HPE supplementation enhances hepatic morphology and function, which is involved in modulating antioxidant capacity and ferroptosis.</jats:p>

<jats:title>Abstract</jats:title><jats:p>Flaxseed oil is rich in α-linolenic acid (ALA), which is the metabolic precursor of EPA and DHA. The present study investigated the effect of flaxseed oil supplementation on lipopolysaccharide (LPS)-induced muscle atrophy and carbohydrate oxidation impairment in a piglet model. Twenty-four weaned pigs were used in a 2 × 2 factorial experiment including dietary treatment (5 % maize oil<jats:italic>v</jats:italic>. 5 % flaxseed oil) and LPS challenge (saline<jats:italic>v</jats:italic>. LPS). On day 21 of treatment, the pigs were injected intraperitoneally with 100 μg/kg body weight LPS or sterile saline. At 4 h after injection, blood, gastrocnemius muscle and longissimus dorsi muscle were collected. Flaxseed oil supplementation increased ALA, EPA, total<jats:italic>n</jats:italic>-3 PUFA contents, protein:DNA ratio and pyruvate dehydrogenase complex quantity in muscles (<jats:italic>P</jats:italic>< 0·05). In addition, flaxseed oil reduced mRNA expression of toll-like receptor (TLR) 4 and nucleotide-binding oligomerisation domain protein (NOD) 2 and their downstream signalling molecules in muscles and decreased plasma concentrations of TNF-<jats:italic>α</jats:italic>, IL-6 and IL-8, and mRNA expression of TNF-<jats:italic>α</jats:italic>, IL-1<jats:italic>β</jats:italic>and IL-6 (<jats:italic>P</jats:italic>< 0·05). Moreover, flaxseed oil inclusion increased the ratios of phosphorylated protein kinase B (Akt) 1:total Akt1 and phosphorylated Forkhead box O (FOXO) 1:total FOXO1 and reduced mRNA expression of FOXO1, muscle RING finger (MuRF) 1 and pyruvate dehydrogenase kinase 4 in muscles (<jats:italic>P</jats:italic>< 0·05). These results suggest that flaxseed oil might have a positive effect on alleviating muscle protein loss and carbohydrates oxidation impairment induced by LPS challenge through regulation of the TLR4/NOD and Akt/FOXO signalling pathways.</jats:p>

This study was conducted to elucidate the biological effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on cell proliferation, differentiation and gene expression in C2C12 myoblasts. C2C12 were treated with various concentrations of EPA or DHA under proliferation and differentiation conditions. Cell viability was analyzed using cell counting kit-8 assays (CCK-8). The Edu assays were performed to analyze cell proliferation. To analyze cell differentiation, the expressions of myogenic marker genes were determined at the transcriptional and translational levels by qRT-PCR, immunoblotting and immunofluorescence. Global gene expression patterns were characterized using RNA-sequencing. Phosphorylation levels of ERK and Akt were examined by immunoblotting. Cell viability and proliferation was significantly inhibited after incubation with EPA (50 and 100 muM) or DHA (100 muM). Both EPA and DHA suppressed C2C12 myoblasts differentiation. RNA-sequencing analysis revealed that some muscle-related genes were significantly downregulated following EPA or DHA (50 muM) treatment, including insulin-like growth factor 2 (IGF-2), troponin T3 (Tnnt3), myoglobin (Mb), myosin light chain phosphorylatable fast skeletal muscle (Mylpf) and myosin heavy polypeptide 3 (Myh3). IGF-2 was crucial for the growth and differentiation of skeletal muscle and could activate the PI3K/Akt and the MAPK/ERK cascade. We found that EPA and DHA (50 muM) decreased the phosphorylation levels of ERK1/2 and Akt in C2C12 myoblasts. Thus, this study suggested that EPA and DHA exerted an inhibitory effect on myoblast proliferation and differentiation and downregulated muscle-related genes expression.

本试验旨在研究脂多糖(LPS)刺激后不同时间断奶仔猪脾脏Toll样受体4( TLR4)和核苷酸结合寡聚域受体( NOD)炎症信号通路关键基因表达的变化.选择42 头体重为( 7.1 ± 0.9) kg的杜×长×大断奶仔猪,按注射LPS之前( 0 h)和注射LPS后不同时间点( 1、2、4、8、12、24 h)随机分为7个处理,每个处理6头猪.预饲14 d后,腹腔注射100 μg/kg BW的LPS,分别于注射LPS之前和注射LPS后不同时间点,将仔猪麻醉屠宰,取血液和脾脏样品,测定血细胞分类计数和脾脏炎症信号通路关键基因及炎症相关基因的mRNA表达量.结果表明:LPS刺激导致部分血细胞分类计数发生显著的变化( P<0.05) ;使 TLR4 和 NOD 炎症信号通路关键基因mRNA表达量显著上调( P<0.05) ,且在2~8 h达到峰值;也导致肿瘤坏死因子-α、白细胞介素-1β、白细胞介素-6、环氧合酶2、热休克蛋白70 mRNA表达量显著升高( P<0.05),且在1~4 h达到峰值. LPS刺激8~24 h后,这些炎症相关基因均逐渐恢复至正常水平或略低于正常水平.总体来看,LPS刺激激活了炎症信号通路,促进炎性细胞因子的表达,并随着时间的延长,这些基因呈现先上升、达到高峰后逐渐下降的变化.

The intestinal microbiota plays a vital role in metabolism, pathogen resistance, and immune development in host cells, and is modifiable by dietary change. Lentinan (LNT), a type of mushroom polysaccharide, is known to ameliorate intestinal inflammation with the potential of therapeutic effect on digestive diseases. We hypothesized that LNT could alleviate Escherichia coli lipopolysaccharide (LPS)-induced intestinal injury via regulating the composition and metabolites of intestinal microbiota in a piglet model. Twenty-four weaned piglets were used in a 2 × 2 factorial design, and the main factors included a dietary treatment (basal or LNT diet) and immunological challenge (LPS or saline). After feeding basal or LNT diet for 21 days, pigs were injected with LPS or saline. At 4 h post-injection, pigs were killed and jejunum, ileum and cecal digesta were collected. LNT improved intestinal morphology and barrier function. LNT also inhibited inflammatory signaling pathways (toll-like receptor 4 and nucleotide binding oligomerization domain protein) and pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-1β and interleukin-6) expression, as well as up-regulated the heat shock protein 70 expression in small intestine. In addition, LNT enhanced the concentrations of propionate, butyrate, isobutyrate and isovalerate in cecal digesta, resulting in a significant increase in histone acetylation without affecting the protein level of G protein-coupled receptor 41 (GPR41), a short chain fatty acid receptor. Bacterial 16S rRNA gene pyrosequencing showed that LNT had a great impact on gut microbiota composition at different taxonomic levels. Moreover, the correlation analysis revealed some potential relationships between cecal metabolites and certain intestinal microbiota. These results indicate that LNT promotes intestinal health, in part, through altering intestinal microbiota composition and increasing the short chain fatty acid synthesis, which subsequently lead to a reduction in inflammation and hyper-acetylation of histones. © 2019 The Royal Society of Chemistry.

LPS can induce an inflammatory immune response in the intestine, and long non-coding RNA (lncRNA) is involved in the process of inflammatory disease. However, the biological role of lncRNA in the intestinal inflammation of piglets remains unclear. In this study, the lncRNA expression profile of the ileal mucosa of piglets challenged by LPS was analysed using lncRNA sequencing. In total, 112 novel lncRNAs were predicted, of which 58 were up-regulated and 54 down-regulated following LPS challenge. Expression of 15 selected lncRNAs was validated by quantitative PCR. We further investigated the target genes of lncRNA that were enriched in the signalling pathways involved in the inflammatory immune response by utilising Gene Ontology and Kyoto Encyclopaedia of Genes and Genomes analysis, with cell adhesion molecules and mTOR signalling pathway identified. In addition, the co-expression networks between the differentially expressed lncRNAs and the target mRNAs were constructed, with seven core lncRNAs identified, which also demonstrated that the relationship between lncRNAs and the target genes was highly correlated. Our study offers important information about the lncRNAs of the mucosal immune system in piglets and provides new insights into the inflammatory mechanism of LPS challenge, which might serve as a novel target to control intestinal inflammation.

猪场生物安全体系是指企业以'杀灭传染源、切断传播途径、保护易染猪群'为原则,准确评估生产所面临的疫病风险,并制定一系列有效预防和控制措施,应对所产生危害或潜在风险的综合性防控体系,它将排除疫病威胁,保护动物健康的各种方法集成化。在规模化的养殖生产中,对疫病的防治仍然是养猪业发展的关键所在。由此可见,选取科学严谨的生物安全体系以及系统化的防治举措,对于应对诸如非洲猪瘟等疫病暴发至关重要。本文以猪场非洲猪瘟疫病的防疫为例,从提高预防措施、加强饲养管理、规范防治程序方面对构建生物安全体系的方法进行介绍。

本试验旨在研究脂多糖(LPS)刺激后不同时间断奶仔猪肌肉炎症和肌肉蛋白质降解相关基因表达的变化规律.选择42头(7.1±0.9) kg杜×长×大三元杂断奶仔猪,按注射LPS之前(0 h)和注射LPS后1、2、4、8、12、24 h随机分为7个处理,每个处理6头猪.预试14d后,腹腔注射100 tg/kg体重的LPS.按以上时间点将仔猪屠宰,取背最长肌样品待测.结果 表明:背最长肌炎性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、IL-6的mRNA表达量在注射LPS 1~2 h后达到峰值;Toll样受体4信号通路关键基因Toll样受体4(TLR4)、骨髓分化因子88 (MyD88),核苷酸结合寡聚域信号通路关键基因核苷酸结合寡聚域2 (NOD2)、受体互作蛋白激酶2(RIPK2)的mRNA表达量在注射LPS 2~4 h后达到峰值;肌肉蛋白质降解相关基因叉头转录因子-1 (FOXO-1)、FOXO-4、肌肉环指蛋白1(MuRF1)、肌萎缩F-box (MAFbx)的mRNA表达量在注射LPS 12 h后达到峰值.可见,LPS刺激诱导肌肉释放大量炎性细胞因子,使TLR4和NOD炎症信号通路关键基因及肌肉蛋白质降解相关基因mRNA显著表达.

Here, the potential mechanisms of the protective effects of fish oil against LPS-induced liver injury in a piglet model were investigated by using RNA sequencing. Twenty-four piglets were used in a 2 × 2 factorial design, and the main factors included diet (5% corn oil or 5% fish oil) and immunological challenge (LPS or saline, on d 19). All piglets were slaughtered at 4 h after challenge, and liver samples were collected. Fish oil improved liver morphology and reduced TNF-α, IL-1β and IL-6 productions after LPS challenge. RNA sequencing analysis showed fish oil had significant effect on the expressions of genes involved in immune response during LPS-induced inflammation. Selected gene expression changes were validated using quantitative RT-PCR. Fish oil reduced the expressions of pro-inflammatory genes IL1R1, IL1RAP, CEBPB and CRP, and increased that of anti-inflammatory genes IL-18BP, NFKBIA, IFIT1, IFIT2 and ATF3. Moreover, fish oil restored the expressions of some lipid metabolism-related genes, such as ACAA1, ACACA, ACADS and ACADM, which were only decreased in pigs fed a corn oil diet after LPS challenge. Our RNA sequencing reveals novel gene-nutrient interactions following fish oil supplementation and evoked inflammation, which add to the current understanding of the benefits of n-3 polyunsaturated fatty acids against liver injury. © The Author(s) 2018.

Antimicrobial growth promoters (AGPs) are commonly used in the livestock industry at subtherapeutic levels to improve production efficiency, which is achieved mainly through modulation of the intestinal microbiota. However, how different classes of AGPs, particularly ionophores, regulate the gut microbiota remains unclear. In this study, male Cobb broiler chickens were supplemented for 14 days with or without one of five commonly used AGPs including three classical antibiotics (bacitracin methylene disalicylate, tylosin, and virginiamycin) and two ionophores (monensin and salinomycin) that differ in antimicrobial spectrum and mechanisms. Deep sequencing of the V3-V4 region of the bacterial 16S rRNA gene revealed that two ionophores drastically reduced a number of rare bacteria resulting in a significant decrease in richness and a concomitant increase in evenness of the cecal microbiota, whereas three antibiotics had no obvious impact. Although each AGP modulated the gut microbiota differently, the closer the antibacterial spectrum of AGPs, the more similarly the microbiota was regulated. Importantly, all AGPs had a strong tendency to enrich butyrate-and lactic acid-producing bacteria, while reducing bile salt hydrolase-producing bacteria, suggestive of enhanced metabolism and utilization of dietary carbohydrates and lipids and improved energy harvest, which may collectively be responsible for the growth-promoting effect of AGPs. © MDPI AG. 本试验旨在研究亚麻籽油对脂多糖(LPS)刺激断奶仔猪肠黏膜结构和免疫细胞的影响。选择24头(9.15±0.90) kg杜×长×大断奶仔猪,随机分为4组,每组6个重复,每个重复1头猪。试验采用2×2双因子设计,主因子包括:1)饲粮处理(5%玉米油或5%亚麻籽油) ;2)免疫应激(注射生理盐水或LPS) 。饲粮中添加5%玉米油或5%亚麻籽油饲喂21 d,在试验第21天,每组1 / 2的猪腹膜注射100 μg / kg BW的LPS,另外1 / 2的猪腹膜注射等量生理盐水。注射LPS或生理盐水4 h后,将仔猪麻醉屠宰,取肠道样品待测。结果表明:1) LPS刺激显著降低了空肠、回肠绒毛高度和空肠隐窝深度(P<0.05) ,显著提高空肠固有层细胞密度和嗜中性粒细胞数目(P<0.05) ,有降低空肠上皮间淋巴细胞数目(P = 0.080)和回肠杯状细胞数目(P = 0.059)的趋势;2) LPS刺激对亚麻籽油组仔猪空肠、回肠绒毛高度无显著影响(P>0.05) ;饲粮添加5%亚麻籽油有提高回肠派氏结细胞密度的趋势(P = 0.064) ,且在LPS刺激下,显著降低空肠固有层细胞密度(P<0.05) ,并有增加空肠上皮间淋巴细胞数目的趋势(P = 0.069) 。结果提示,5%亚麻籽油在一定程度上可维护LPS刺激断奶仔猪的小肠黏膜结构和免疫功能。

This study was conducted to envaluate whether glycine could alleviate Escherichia coli lipopolysaccharide (LPS)-induced intestinal injury by regulating intestinal epithelial energy status, protein synthesis, and inflammatory response via AMPK, mTOR, TLR4, and NOD signaling pathways. A total of 24 weanling piglets were randomly allotted to 1 of 4 treatments: (1) non-challenged control; (2) LPS-challenged control; (3) LPS + 1% glycine; (4) LPS + 2% glycine. After 28 days feeding, piglets were injected intraperitoneally with saline or LPS. The pigs were slaughtered and intestinal samples were collected at 4 h postinjection. The mRNA expression of key genes in these signaling pathways was measured by real-time PCR. The protein abundance was measured by Western blot analysis. Supplementation with glycine increased jejunal villus height/crypt depth ratio. Glycine also increased the jejunal and ileal protein content, RNA/DNA ratio, and jejunal protein/DNA ratio. The activities of citroyl synthetase in ileum, and α-ketoglutarate dehydrogenase complex in jejunum, were increased in the piglets fed diets supplemented with glycine. In addition, glycine decreased the jejunal and ileal phosphorylation of AMPKα, and increased ileal phosphorylation of mTOR. Furthermore, glycine downregulated the mRNA expression of key genes in inflammatory signaling. Meanwhile, glycine increased the mRNA expression of negative regulators of inflammatory signaling. These results indicate that glycine supplementation could improve energy status and protein synthesis by regulating AMPK and mTOR signaling pathways, and relieve inflammation by inhibiting of TLR4 and NOD signaling pathways to alleviate intestinal injury in LPS-challenged piglets. © 2018 by the authors. Scope: Flaxseed oil is a rich source of α-linolenic acid (ALA), which is the precursor of the long-chain n-3 polyunsaturated fatty acids (PUFAs), including docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). This study investigates the protective effect of flaxseed oil against intestinal injury induced by lipopolysaccharide (LPS). Materials and results: Twenty-four weaned pigs were used in a 2 × 2 factorial experiment with dietary treatment (5% corn oil vs 5% flaxseed oil) and LPS challenge (saline vs LPS). On day 21 of the experiment, pigs were administrated with LPS or saline. At 2 h and 4 h post-administration, blood samples were collected. After the blood harvest at 4 h, all piglets were slaughtered and intestinal samples were collected. Flaxseed oil supplementation led to the enrichment of ALA, EPA, and total n-3 PUFAs in intestine. Flaxseed oil improved intestinal morphology, jejunal lactase activity, and claudin-1 protein expression. Flaxseed oil downregulated the mRNA expression of intestinal necroptotic signals. Flaxseed oil also downregulated the mRNA expression of intestinal toll-like receptors 4 (TLR4) and its downstream signals myeloid differentiation factor 88 (MyD88), nuclear factor kappa B (NF−κB), and nucleotide-binding oligomerization domain proteins 1, 2 (NOD1, NOD2) and its adapter molecule, receptor-interacting protein kinase 2 (RIPK2). Conclusion: These results suggest that dietary addition of flaxseed oil enhances intestinal integrity and barrier function, which is involved in modulating necroptosis and TLR4/NOD signaling pathways. © 2018 WILEY-VCH Verlag GmbH & Co. <jats:title>Abstract</jats:title><jats:p>This experiment aimed to explore whether glutamate (Glu) had beneficial effects on intestinal injury caused by <jats:italic>Escherichia coli</jats:italic> LPS challenge via regulating mTOR, TLRs, as well as NODs signaling pathways. Twenty-four piglets were allotted to 4 treatments including: (1) control group; (2) LPS group; (3) LPS + 1.0% Glu group; (4) LPS + 2.0% Glu group. Supplementation with Glu increased jejunal villus height/crypt depth ratio, ileal activities of lactase, maltase and sucrase, and RNA/DNA ratio and protein abundance of claudin-1 in jejunum and ileum. In addition, the piglets fed Glu diets had higher phosphorylated mTOR (Ser<jats:sup>2448</jats:sup>)/total mTOR ratio in jejunum and ileum. Moreover, Glu decreased TNF-α concentration in plasma. Supplementation with Glu also decreased mRNA abundance of jejunal TLR4, MyD88, IRAK1, TRAF6, NOD2 and increased mRNA abundance of ileal Tollip. These results indicate that Glu supplementation may be closely related to maintaining mTOR and inhibiting TLR4 and NOD signaling pathways, and concomitant improvement of intestinal integrity under an inflammatory condition.</jats:p>

The hepatoprotective effects of flaxseed oil (FO) were tested in a pig model of lipopolysaccharide (LPS)-induced liver injury. Twenty-four piglets were used in a 2 × 2 factorial design including diet and an LPS challenge. After 3 weeks of feeding with 5% FO or 5% corn oil, pigs were challenged with LPS or saline. FO alleviated LPS-induced morphological liver damage, reduced serum alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase activities, and increased claudin-1 protein expression. FO decreased tumor necrosis factor-α interleukin-6 and cyclooxygenase 2 mRNA expression, and heat shock protein 70 mRNA and protein expression. FO downregulated receptor interacting protein kinase (RIP)1, RIP3, and mixed-lineage kinase domain-like protein (MLKL) mRNA expression, RIP1 and RIP3 protein expression, and MLKL phosphorylation. FO downregulated mRNA expression of toll-like receptor 4, nucleotide-binding oligomerization domain protein 1 and multiple downstream signaling molecules, and decreased p38 and extracellular signal-regulated kinase 1/2 phosphorylation. These results indicate that FO inhibits necroptotic and inflammatory signaling pathways to protect the liver from inflammation-related injury. © 2018 Elsevier Ltd

This study was conducted to investigate whether medium-chain triglycerides (MCTs) attenuated lipopolysaccharide (LPS)-induced liver injury by down-regulating necroptotic and inflammatory signaling pathways. A total of 24 pigs were randomly allotted to four treatments in a 2 × 2 factorial design including diet (0 and 4% MCTs) and immunological challenge (saline and LPS). After three weeks of feeding with or without 4% MCTs, pigs were challenged with saline or LPS. MCTs led to a significant increase in eicosapentaenoic acid, docosahexaenoic acid and total (n-3) polyunsaturated fatty acid concentrations. MCTs attenuated LPS-induced liver injury as indicated by an improvement in liver histomorphology and ultrastructural morphology of hepatocytes, a reduction in serum alanine aminotransferase and alkaline phosphatase activities as well as an increase in claudin-1 protein expression. In addition, MCTs also reduced serum tumor necrosis factor-α (TNF-α), interleukin (IL)-1β and IL-6 concentrations, liver TNF-α and IL-1β mRNA expression and protein concentrations and enhanced liver heat shock protein 70 protein expression in LPS-challenged pigs. Moreover, MCTs decreased mRNA expression of receptor-interacting serine/threonine-protein kinase (RIP) 3, mixed-lineage kinase domain-like protein (MLKL) and phosphoglycerate mutase 5 and inhibited MLKL phosphorylation in the liver. Finally, MCTs decreased liver mRNA expression of toll-like receptor (TLR) 4, nucleotide-binding oligomerization domain protein (NOD) 1 and multiple downstream signaling molecules. MCTs also suppressed LPS-induced p38 mitogen-activated protein kinase (MAPK) phosphorylation and increased extracellular signal-related kinase 1/2 phosphorylation in the liver. These results indicated that MCTs are capable of attenuating LPS-induced liver damage by suppressing hepatic necroptotic (RIP1/RIP3/MLKL) and inflammatory (TLR4/NOD1/p38 MAPK) signaling pathways. © 2018 by the authors. 摘要:
天然淋巴细胞(Innate lymphoidc ells,ILCs)是新发现的一类重要淋巴细胞亚群,它们起源于普通淋巴祖细胞,不同于T、B淋巴细胞。ILCs不表达抗原受体,在微生物的刺激下可快速活化并进入靶器官参与免疫应答。ILCs位于黏膜表面,可增强免疫反应,维持黏膜的完整性,在黏膜屏障炎症性反应及组织损伤中发挥重要作用。本文简要阐述了ILCs的分类、分化和迁移及在肠黏膜免疫中的作用。

本试验旨在研究天冬酰胺( Asn)对脂多糖( LPS)刺激断奶仔猪生长性能和下丘脑-垂体-肾上腺( HPA)轴的Toll样受体4( TLR4)和核苷酸结合寡聚化结构域( NOD)信号通路关键基因mRNA表达的影响.试验选取24头健康的( 21 ± 2)日龄“杜×长×大”断奶仔猪[( 8. 12 ± 0.56) kg],按体重相近原则随机分为4组,分别为: 1)对照组(基础饲粮) ; 2) LPS组(基础饲粮+LPS) ; 3) LPS+0.5% Asn组(基础饲粮+0.5%Asn+LPS) ; 4) LPS +1.0% Asn组(基础饲粮+ 1.0% Asn+LPS) .每组6个重复,每个重复1头猪.试验第20天,LPS组、LPS+0.5% Asn组和 LPS+1.0% Asn组的试验猪注射100 μg /kg BW的LPS,对照组试验猪则注射等量的生理盐水.试验期为20 d.结果表明,与对照组相比,LPS刺激以及饲粮中添加Asn对LPS刺激断奶仔猪的生长性能没有显著影响( P>0.05) .LPS刺激可以显著提高HPA轴中TLR4、髓样分化蛋白88( MyD88) 、NOD2、受体相互作用丝氨酸/苏氨酸蛋白激酶2( RIP2)和核转录因子-κB( NF-κB)的mRNA表达量( P<0.05) .饲粮中添加Asn可以显著降低下丘脑中NF-κB的mRNA表达量(线性,P<0.05) ,并有降低肾上腺中TLR4和NOD2的mRNA表达量的趋势(线性,P<0.10) .此外,饲粮中添加Asn可以显著降低垂体中肿瘤坏死因子受体相关因子6( TRAF6)的mRNA表达量(二次,P<0.05) ,显著增加了肾上腺中RIP2的mRNA表达量(线性,P < 0. 05;二次,P < 0.05) .由此可见,Asn虽然对LPS刺激仔猪的生长性能没有现在影响,但其可通过降低下丘脑 NF-κB的mRNA表达量,而对下丘脑TLR4信号通路具有一定的抑制作用.
