乳酸菌抗菌性能评价及协同作用研究

摘要:
目的 提高乳酸菌抗菌性能,拓宽其抗菌谱。方法 对可共生的7株益生菌的体外生长性能、产酸性、抗氧化性能以及抑菌性能进行了比较,筛选出综合性能较好的菌株,并进行复配探究最佳复合比例。结果 筛选出鼠李糖乳杆菌HN001、干酪乳杆菌C11、R5和植物乳杆菌R2综合性能较好。通过抑菌谱测定,筛选出对细菌抑制作用最强的鼠李糖乳杆菌HN001和对霉菌抑制作用最高的植物乳杆菌R2。对两株菌以不同比例进行复合,得到最佳抑菌比例HN001:R2为5:1。结论 鼠李糖乳杆菌HN001和植物乳杆菌R2的组合较单一菌株具有更强的抑菌性能和更广的抑菌谱,具有协同抗菌作用,在防治食源性致病菌方面具有一定的应用潜力。

酶法快速测定植物油中游离甾醇

摘要:
目的 建立固相萃取(solid phase extraction,SPE)结合酶法测定植物油中游离甾醇含量的快速检测方法。方法 采用SPE结合酶法40 min内实现植物油中的游离甾醇含量的快速测定,并应用于植物油中游离甾醇含量测定。结果 方法学验证结果显示,植物甾醇含量在浓度范围0.3~7.5 mg/m L之间线性良好,检出限为0.018 mg/g,日内精密度为7.94%,日间精密度为6.88%,回收率在84.75%~99.50%之间。实际样品的检测结果显示,酶法与气相色谱-质谱法(gas chromatography-mass spectrometry,GC-MS)的相对误差在±20%以内,且酶法的检测效率比GC-MS提高3倍。常见的几种植物油中,游离甾醇含量较高的是玉米油和菜籽油,分别为1.53~3.66 mg/g和1.98~3.02 mg/g。结论 本研究建立的SPE结合酶法检测植物油中游离甾醇的快速检测方法稳定性、重现性好、灵敏可靠,可满足植物油中游离甾醇的现场批量快速检测。

摘要:
A variety of mycotoxins from different sources frequently contaminate farm products, presenting a potential toxicological concern for animals and human. Mycotoxin exposure has been the focus of attention for governments around the world. To date, biomarkers are used to monitor mycotoxin exposure and promote new understanding of their role in chronic diseases. The goal of this research was to develop and validate a sensitive UHPLC-MS/MS method using isotopically-labeled internal standards suitable for accurate determination of 18 mycotoxin biomarkers, including fumonisins, ochratoxins, Alternaria and emerging Fusarium mycotoxins (fumonisin B1, B2, and B3, hydrolyzed fumonisin B1 and B2, ochratoxin A, B, and alpha, alternariol, alternariol monomethyl ether, altenuene, tentoxin, tenuazonic acid, beauvericin, enniatin A, A1, B, and B1) in human urine. After enzymatic digestion with β-glucuronidase, human urine samples were cleaned up using HLB solid phase extraction cartridges prior to instrument analysis. The multi-mycotoxin and analyte-specific method was validated in-house, providing satisfactory results. The method provided good linearity in the tested concentration range (from LOQ up to 25–500 ng/mL for different analytes), with R2 from 0.997 to 0.999. The limits of quantitation varied from 0.0002 to 0.5 ng/mL for all analytes in urine. The recoveries for spiked samples were between 74.0% and 133%, with intra-day precision of 0.5%–8.7% and inter-day precision of 2.4%–13.4%. This method was applied to 60 urine samples collected from healthy volunteers in Beijing, and 10 biomarkers were found. At least one biomarker was found in all but one of the samples. The high sensitivity and accuracy of this method make it practical for human biomonitoring and mycotoxin exposure assessment. © 2020 by the authors.

摘要:
A 3D functional human microvascular network in a microfluidic device was developed in this study. The established model enables endotheilal cells to form vessel-like microtissues and behave physiological functions which are closer to cells in human blood vessels. It provides a possible bridge for how the research outcomes translate to humans. The ɑ-MnO2 nanowires, which were successfully synthesized using the hydrothermal approach. The vascular toxicity was evaluated based on this organotypic 3D microvessel model. Our results demonstrated that ɑ-MnO2 nanowires can induce cytotoxicity in dose-dependently. Because the accumulation of reactive oxygen species (ROS) and depletion of glutathione (GSH) are observed in the nanowire-treated cells. In addition, intravascular accumulation of ɑ-MnO2 nanowires could induce endothelial dysfunction by oxidative stress. This is expressed in disorder of NO expression and IL-6 up-regulation. Based on these results, we concluded that a mechanism for ɑ-MnO2 nanowire-induced vascular toxicity, which involves the accumulation of ROS, up-regulation of oxidative stress and inflammatory factors. This investigation may provide a fundamental insight to understand the nanotoxicity of wire-shaped nanomaterials. © 2019, © 2019 Taylor & Francis Group, LLC.

Folic acid is a necessary micronutrient for normal human growth and development. Benzo(a)pyrene (BaP) is a ubiquitously distributed environmental pollutant and its metabolite, benzo(a)pyrene-diol-epoxide, is known to exert a strong teratogenic and carcinogenic effect on the body’s tissues and cells. The aim of this study was to investigate the mechanism by which folic acid can inhibit the toxic effects of BaP both in vivo and in vitro. We measured changes in 16HBE cell activity affected by the intervention of folic acid on BaP using the cell counting kit-8 assay and that of cell cycle distribution by flow cytometry. At the same time, we assessed the xeroderma pigmentosum group A, xeroderma pigmentosum group C, excision repair cross complementation group 1, cyclinD1, and CKD4 mRNAs, and their related protein expression both in mouse lung tissue and in 16HBE cells. In conclusion, the mechanisms by which this effect is mediated were not entirely elucidated by our study, possibly because folic acid antagonizes the toxic effects of BaP by upregulating the levels of excision repair cross complementation group 1, xeroderma pigmentosum group A, and xeroderma pigmentosum group C gene expression to improve the rate of DNA repair, in turn accelerating the speed of repair for DNA damage caused by BaP. Meanwhile, folic acid could restrain BaP-induced cyclinD1 protein expression, which could help cells return to their normal cell cycle.

摘要:
This paper described an improved method for high-throughput and sensitive determination of zearalenone and its five metabolites (zearalanone, α-zearalenol, β-zearalenol, α-zearalanol and β-zearalanol) in human serum. Serum samples were measured both before and after enzyme hydrolysis to assess the free and total amount of each compound by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) in multi reaction monitoring (MRM) mode following off-line 96-well μElution solid-phase extraction (SPE). All the analytes were completely separated on a C18 column within 6 min. It enabled multi-sample preparation at the same time eliminating tedious evaporation and reconstitution steps, allowing 96 (one plate) samples to be processed and analyzed within 24 h. Using an isotope labelled internal standard ( 13 C-ZEN), high recoveries were achieved for all the compounds in the range 91.6%-119.5%, with intra-day and inter-day relative standard deviations (RSDs) of less than 8%. The limits of detection (LOD) and the limits of quantification (LOQ) were 0.02-0.06 ng mL -1 (0.6-2 fmol) and 0.1-0.2 ng mL -1 (3-6 fmol), respectively, demonstrating a notable enhancement in sensitivity compared to the existing methods. The validated method was applied to the analysis of paired urine and serum samples collected from 125 healthy individuals in Henan Province, locating in the middle area of China. ZEN metabolites in human serum were significantly lower than those in urine. Only one serum sample was positive for ZEN after enzyme digestion, whereas at least one of ZEN biomarkers was detected in 75.2% of the paired urine samples. Some comparison and discussion were also included in this paper. © 2019 by the authors.

水浸法削减大米粉中镉的工艺优化及对其品质影响

摘要:
采用响应面法优化水浸法削减镉污染的大米粉中镉的工艺条件,在最优条件下测定水浸法对大米粉营养成分的影响,并用扫描电镜、差示扫描量热仪、X射线衍射、红外光谱等方法分别研究水浸法对大米粉形态、热力学特征、淀粉晶体结构及官能团的影响。结果表明:水浸法削减大米粉中镉的最佳条件为水浸温度42℃、料液比1∶3(g/m L)、水浸时间47 h,此时镉含量为(0.459±0.006)mg/kg,大米粉中的镉去除率达到(61.75±4.31)%,处理后大米粉中镉含量为(0.176±0.003)mg/kg。在最佳工艺条件下,大米粉中的蛋白质含量显著降低(P<0.05),脂肪、淀粉、灰分也略有下降(P<0.05)。大米粉水浸后糊化起始温度降低,而峰值温度变化很小,说明大米粉晶体片层被破坏,稳定性降低,大米粉中直链淀粉含量可能升高,但水浸处理并没有破坏米粉内淀粉粒晶体成分。水浸大米粉面间距有略微的降低,说明水浸可能导致大米粉产品较原料产品的硬度、咀嚼度、回弹性、黏聚性下降。大米粉在水浸过程中并没有产生新的化学键或基团。

羧甲基赖氨酸(CML)是食品加工中经美拉德反应由蛋白质与糖类通过非酶催化反应产生的一种晚期糖基化终末产物,是一种新型食品潜在有害物质。本文通过研究外源游离性CML对秀丽隐杆线虫寿命、子代数量、热环境中线虫存活时间的影响,探讨其可能的毒性机制。结果表明,外源游离性CML导致线虫产卵期缩短,寿命试验以及热刺激下存活的线虫数量呈现出剂量-反应关系。影响了TOR信号通路的Skn-1基因和生殖系统信号通路中的hsf-1基因,提高线虫的热环境阻力的抵抗力并延长其寿命。高浓度的CML降低了线虫的中DAF-16的表达水平从而影响其寿命,抑制线虫体内Folt-1基因的表达,从而减少其子代线虫的数量。

Nε-(carboxymethyl)lysine (CML), which is a compound produced when food is processed, has aroused concern in recent years because of its potentially dangerous effects. This study aimed to investigate the mechanism of free CML-induced toxic injury in mice. The inflammatory cytokine tumor necrosis factor-α, transforming growth factor-β, vascular cell adhesion molecule-1 mRNA expression levels of CML-infected mice liver and kidney tissues significantly increased. While CML receptor—receptor for advanced glycation end products (RAGE) protein expression in male mice liver tissue had a more significant change than the control group, there was no significant difference in other dose groups compared with the control group. In conclusion, the foodborne free CML can be induced by oxidative stress and immune response to liver and kidney tissue injury in mice. Additionally, the free CML may also bind to RAGE, which activates the downstream inflammatory pathway. © 2019 Wiley Periodicals, Inc.

摘要:
3-Monochloropropane-1,2-diol (3-MCPD) is a common food processing contaminant and a simple, rapid, sensitive and low cost monitoring technology is needed due to its potential carcinogenic nature. Carbon dots directly intercepted on filter paper provide high fluorescence intensity and can be adapted for use as a sensor. We synthesized a carbon dot-filter paper in combination with a molecularly imprinted polymeric film to extract 3-MCPD from samples. This grafted paper-based sensor exhibited a high adsorption capacity (68.97mgg(-1)), an excellent selectivity (imprinting factor=4.5) and a low detection limit (0.6ngmL(-1)). Recoveries ranged from 97.2% to 105.3% with relative standard deviations <5.6%. The results obtained using this method were linearly correlated to those of the classic GC-MS method (r=0.998). Based on the Chinese National Standard, this study provides a novel and powerful platform for the simple, rapid, sensitive and on-site analysis of 3-MCPD in soy sauce.

摘要:
建立了QuEChERS-高效液相色谱-串联质谱(HPLC-MS/MS)同时快速测定蔬菜中29种农药残留的分析方法。样品以乙腈提取,氯化钠盐析,以十八烷基硅烷键合硅胶(C18)和N-丙基乙二胺(PSA)净化,以WatersXBridgeC18色谱柱分离,电喷雾正离子扫描,多反应监测(MRM)模式检测,基质匹配标准溶液外标法定量。在此优化条件下,29种农药分子在不同加标水平下的平均回收率为64.62%~123.55%,相对标准偏差(RSD)为0.7%~14.3%,方法定量限(LOQ)为0.40~8.00μg/kg。该方法简便快速、灵敏度高、重现性好,能满足蔬菜中农药残留的快速检测要求。

Background: A growing body of evidence shows that indoor concentrations of airborne particles are often higher than is typically encountered outdoors. Since exposure to indoor PM2.5 is thought to be associated with cardiovascular disease, the health impacts of indoor air pollution need to be explored. Based on animal models, ambient particulate matter has been proved to promote coagulation which is very likely involved in the pathogenic development of cardiovascular disease. However, animal models are insufficient to predict what will happen with any certainty in humans. For this reason, the precise pathogenic mechanisms behind the development of cardiovascular disease in humans have not yet been determined. Results: We generated a 3D functional human microvascular network in a microfluidic device. This model enables human vascular endothelial cells to form tissue-like microvessels that behave very similarly to human blood vessels. The perfusable microvasculature allows the delivery of particles introduced into these generated human-like microvessels to follow the fluid flow. This exposure path effectively simulates the dynamic movement of airborne nanoscale particles (ANPs) within human vessels. In this study, we first identified the existence of ANPs in indoor air pollution. We then showed that ANPs could activate endothelial cells via ROS induced inflammation, and further resulted in abnormal expression of the coagulation factors (TF, TM and t-PA) involved in coagulation cascades. In addition, we found that a protein could cover ANPs, and this biointeraction could interfere with heparan sulfate (HS). Human organotypic 3D microvessel models provide a bridge for how research outcomes can translate to humans. Conclusions: The 3D human microvessel model was used to determine the physiological responses of human vessels to ANP stimulation. Based on the obtained data, we concluded that ANPs not only disrupts normal coagulation functions, but also act directly on anticoagulant factors in human vessels. These experimental observations provide a potential biological explanation for the epidemiologically established link between ANPs and coagulation abnormality. This organ-on-chip model may provide a bridge from in vitro results to human responses. © 2019 The Author(s).

Phytosterols have been extensively studied because it plays essential roles in the physiology of plants and can be used as nutritional supplement to promote human health. We use a rapid method by coupling near-infrared spectroscopy (NIRS) and chemometric techniques to quickly and efficiently determine three essential phytosterols (beta-sitosterol, campesterol and stigmasterol) in vegetable oils. Continuous wavelet transform (CWT) method was adopted to remove the baseline shift in the spectra. The quantitative analysis models were constructed by partial least squares (PLS) regression and randomization test (RT) method was used to further improve the models. The optimized models were used to calculate the phytosterol contents in prediction set in order to evaluate their predictability. We have found that the phytosterol contents obtained by the optimized models and Gas Chromatography/Mass Spectrometry (GC/MS) analysis are almost consistent. The root mean square error of prediction (RMSEP) and ratio of prediction to deviation (RPD) for the three phytosterols are 525.7590, 212.2245, 65.1611 and 4.0060, 4.7195 and 3.5441, respectively. The results have proved the feasibility of the proposed method for rapid and non-destructive analysis of phytosterols in edible oils.

In this study, we performed a 3D organotypic culture of human lung fibroblast (HLF) in a microfluidic device. This 3D HLF model could partly simulate the biological responses of human lung. ɑ-MnO2 nanowires were successfully synthesized using the hydrothermal approach. Material characterization identified the existence of ɑ-MnO2 nanostructure. Based on this 3D organotypic HLF model, our results demonstrated that ɑ-MnO2 nanowires can induce cytotoxicity in dose-dependently. Because the accumulation of reactive oxygen species (ROS) and depletion of glutathione (GSH) are observed in the nanowire-treated cells. In addition, ROS generation induced intracellular increase of malondialdehyde (MDA), DNA damage and activated alpha-smooth muscle actin expression. Based on these results, we concluded that a mechanism for ɑ-MnO2 nanowire-induced pulmary toxicity, which involves the accumulation of ROS, up-regulation of oxidative stress and inflammatory factors. This investigation may provide a fundamental insight to understand the nanotoxicity of wire-shaped nanomaterials. © 2019, © 2019 Taylor & Francis Group, LLC.

笔者在本研究中提出了用正交法优化微生物发酵法削减大米粉中镉污染的工艺条件,在最优条件下测定了微生物发酵对两种污染程度不同的大米的主要化学成分的影响,并用差示扫描量热法(differential scanning calorimetry,DSC)、X射线衍射(X ray diffraction,XRD)、红外光谱等方法分别研究了微生物发酵大米热力学特征、淀粉晶体结构及官能团的影响。结果表明,最佳的发酵菌种为植物乳杆菌,发酵法削减大米中镉的最佳条件为接种量3.5%,发酵时间为48h,料液比为1:3。在此条件下,镉含量为0.459±0.006mg/kg米中的镉去除率达到80.02±1.10%,可以得到镉含量为0.092±0.003 mg/kg的大米;镉含量为0.873±0.031mg/kg大米的镉去除率达到77.25±4.47%,可以得到镉含量为0.199±0.039mg/kg的大米。在最佳工艺条件下,两种大米中的水分和蛋白质、淀粉含量显著降低(p<0.05),脂肪含量也有略微下降。两种大米发酵后糊化起始温度降低,污染较小的大米峰值温度降低,而污染较严重的大米峰值温度变化不大。两种大米的结晶度都有一定下降,特别是污染程度较低的大米下降明显。但两种大米在发酵过程中都没有发生剧烈的化学变化而产生新的化学键或基团。

Calibration transfer is indispensable for practical applications of near infrared (NIR) spectroscopy due to the need for precise and consistent measurements across different spectrometers. In this work, a method for multi-spectrometer calibration transfer is described based on independent component analysis (ICA). A spectral matrix is first obtained by aligning the spectra measured on different spectrometers. Then, by using independent component analysis, the aligned spectral matrix is decomposed into the mixing matrix and the independent components of different spectrometers. These differing measurements between spectrometers can then be standardized by correcting the coefficients within the independent components. Two NIR datasets of corn and edible oil samples measured with three and four spectrometers, respectively, were used to test the reliability of this method. The results of both datasets reveal that spectra measurements across different spectrometers can be transferred simultaneously and that the partial least squares (PLS) models built with the measurements on one spectrometer can predict that the spectra can be transferred correctly on another.

Diets containing various docosahexaenoic acid (DHA)/eicosapentaenoic acid (EPA) ratios protect against liver damage in mice fed with a high-fat diet (HFD). However, it is unclear whether these beneficial roles of DHA and EPA are associated with alterations of fatty acid (FA) composition in the liver. This study evaluated the positive impacts of n-6/n-3 polyunsaturated fatty acids (PUFAs) containing different DHA/EPA ratios on HFD-induced liver disease and alterations of the hepatic FA composition. ApoE−/− mice were fed with HFDs with various ratios of DHA/EPA (2: 1, 1: 1, and 1: 2) and an n-6/n-3 ratio of 4: 1 for 12 weeks. After treatment, the serum and hepatic FA compositions, serum biochemical parameters, liver injury, and hepatic lipid metabolism-related gene expression were determined. Our results demonstrated that dietary DHA/EPA changed serum and hepatic FA composition by increasing contents of n-6 and n-3 PUFAs and decreasing amounts of monounsaturated fatty acids (MUFAs) and the n-6/n-3 ratio. Among the three DHA/EPA groups, the DHA/EPA 2: 1 group tended to raise n-3 PUFAs concentration and lower the n-6/n-3 ratio in the liver, whereas DHA/EPA 1: 2 tended to raise n-6 PUFAs concentration and improve the n-6/n-3 ratio. DHA/EPA supplementation reduced the hepatic impairment of lipid homeostasis, oxidative stress, and the inflammatory responses in HFD-fed mice. The DHA/EPA 2: 1 group had lower serum levels of total cholesterol, triglycerides, and low-density lipoprotein cholesterol and higher levels of adiponectin than HFD group. The DHA/EPA 1: 2 group had elevated serum levels of aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase, without significant change the expression of genes for inflammation or hepatic lipid metabolism among the three DHA/EPA groups. The results suggest that DHA/EPA-enriched diet with an n-6/n-3 ratio of 4: 1 may reverse HFD-induced nonalcoholic fatty liver disease to some extent by increasing n-6 and n-3 PUFAs and decreasing the amount of MUFAs and the n-6/n-3 ratio.
