摘要:
Simple Summary Summer heat stress (HS) seriously affects the reproductive and lactation performance of sows and the long-term development of their offspring. Mannose oligosaccharide (MOS) is widely used as an ingredient in animal feed as it can limit the colonization of enteric pathogens. However, most animal experiments that use MOS to alleviate the effects of HS are often performed with broilers. The effect of MOS in sows affected by HS is unclear. Based on our results, dietary supplementation with MOS tended to alleviate HS in sows compared with the control. However, no significant interactive effects were found. The aim of this study was to determine the effects of dietary supplementation with mannose oligosaccharide (MOS) on the condition of the body and the reproductive and lactation performances of sows. Eighty pregnant sows were randomly assigned to four groups with a 2 x 2 factorial design: with or without MOS (1 g/kg) and with or without heat stress (HS) challenge. The temperature in the HS groups (HS and HM group) was controlled at 31.56 +/- 1.22 degrees C, while the temperature in the active cooling (AC) groups (AC and AM group) was controlled at 23.49 +/- 0.72 degrees C. The weight loss of sows in the AC group was significantly lower than that of sows in the HS group (p < 0.01). The weight and backfat thickness loss of sows supplemented with MOS displayed a downward trend. The average birth weight of the litter significantly increased in the HM group (basic diet + MOS) compared with the HS group (p < 0.05). The milk protein of sows significantly decreased under the HS condition at 2 and 12 h after delivery (p < 0.05). However, the milk immunoglobin G (IgG) of sows in the HS group increased significantly compared with that of sows in the HM group (p < 0.05) at 12 and 24 h after delivery. The levels of serum urea nitrogen (UREA) and glucose (GLU) decreased significantly under the HS condition (p < 0.05), while the level of interleukin-6 (IL-6) increased significantly under the HS condition (p < 0.05). Dietary supplementation with MOS also significantly reduced TNF-alpha under the AC conditions (p < 0.05). In conclusion, HS significantly affected the body condition, lactation performances and their offspring of sows. However, dietary supplementation with 1 g/kg MOS did not result in statistically significant changes.

肠道不仅是所有营养物质消化吸收的最终场所,也是动物体内最大的免疫器官,是机体防御体系的第一道屏障,维护动物肠道健康对保障养猪生产至关重要。肠道是机体应激反应的中心器官,各种应激因素致使肠道结构损伤与功能紊乱,从而导致仔猪健康不良与生长受阻。因此,建立稳定可靠的肠道损伤模型是研究仔猪肠道损伤机制及营养干预的关键,本文主要介绍了本实验室成功建立的几种仔猪肠道损伤模型,筛选了几种肠道功能紊乱的标志性分子,为猪和人类的肠道功能障碍研究及营养干预提供参考。

摘要:
本试验旨在研究表达Ⅰ型耐热肠毒素(STa)的重组大肠杆菌诱发7日龄仔猪小肠炎症反应.选取24头7日龄健康仔猪,随机分为4个处理组:对照组(人工乳)、STa组(人工乳+2×109 CFU重组菌LMG194-pBAD-STa)、LMG194组(人工乳+2×109 CFU大肠杆菌LMG194)和K88组(人工乳+2×109 CFU大肠杆菌K88),每组6头.试验第5天进行攻毒,第7天屠宰取样,观察空肠组织形态学变化,并检测血清中炎性细胞因子含量及空肠免疫相关基因的相对表达量.结果显示,与对照组相比,S T a与K88组仔猪空肠绒毛明显萎缩变短,有明显损伤甚至脱落;STa、LMG194及K88组血清中IL-10、IL-8和IL-6浓度均显著升高(P<0.05),LMG194和K88组血液和肠道iFABP浓度均显著降低(P<0.05),STa和K88组血清中TNF-α浓度显著降低、NF-κB浓度显著升高,LMG194组NF-κB浓度显著降低、TNF-α浓度显著升高(P<0.05);STa和LMG194组CCL2和CXCL9基因表达水平显著上调(P<0.05),V N N1基因表达水平显著下调(P<0.05),STa和LMG194组IFN-γ组基因水平分别显著上升和下降(P<0.05);K88组CXCL9和IFN-γ基因表达水平显著下调(P<0.05),VNN1基因表达水平显著上调(P>0.05),CCL2基因表达水平无明显差异(P>0.05).以上结果表明,表达Ⅰ型耐热肠毒素STa的重组大肠杆菌几乎具有与K 88一样的毒性,可导致7日龄仔猪小肠黏膜受损,诱发严重的炎症反应,导致仔猪腹泻,可作为仔猪腹泻模型的候选菌株.

试验旨在研究表达Ⅰ型耐热肠毒素(S T a)的重组大肠杆菌对7日龄仔猪小肠功能的影响.选取24头7日龄仔猪,随机分为4个处理组,分别为对照组、STa组、LMG194组和K88组.整个试验期间均以人工乳饲喂,预饲期4 d,试验第5天进行攻毒,STa组、LMG194组及K88组分别在早晚两次口服灌喂2×109 CFU大肠杆菌LMG194-pBAD-STa、LMG194和K88,对照组灌服等量生理盐水,试验第7天早上各组灌服D-木糖并采血,屠宰并取空肠、回肠组织样品,测量仔猪血常规指标、血浆D-木糖含量及二胺氧化酶(DAO)活力,以及吸收与转运相关基因与蛋白表达量.结果显示,与对照组相比,STa组仔猪血液中中性粒细胞、中性粒细胞比率显著降低(P<0.05),淋巴细胞比率显著升高(P<0.05),LMG194组与K88组仔猪血液中中性粒细胞比率显著降低(P<0.05),淋巴细胞比率显著升高(P<0.05);STa组与K88组仔猪血浆D-木糖含量显著降低(P<0.05);STa组、LMG194组及K88组血浆DAO活力显著升高(P<0.05);STa组与LMG194组空肠AQP8、AQP10、KCN J13和b0,+A T基因表达量均显著降低(P<0.05),K88组空肠AQP8、AQP10基因表达量显著升高(P<0.05);STa组回肠AQP8基因表达量显著降低(P<0.05),AQP10、SGLT-1基因表达量显著升高(P<0.05),LMG194组、K88组AQP8基因表达量显著升高(P<0.05),K88组AQP10、KCNJ13、b0,+AT、SGLT-1基因表达量显著降低(P<0.05);STa组、LMG194组及K88组空肠HSP70蛋白的表达量显著升高(P<0.05).以上结果表明,表达Ⅰ型耐热肠毒素的重组大肠杆菌可导致7日龄仔猪小肠产生炎症,吸收与转运能力下降.

摘要:
The aim of the present study was to construct a recombinant strain by using the probiotics Lactobacillus acidophilus expressing the epidermal growth factor (EGF), which can improve intestinal morphology, and to test its effect on the intestinal histology in LPS (lipopolysaccharide)-challenged mice. The EGF gene was cloned into the shuttle vector pSET4s and then transformed into the L. acidophilus MG6243 strain by electroporation. Two recombinant strains were confirmed by SDS-PAGE and named ΔMG6243-1 and ΔMG6243-2. Sixty mice were randomly assigned to six treatments: 1) non-challenged control, 2) LPS-challenged control, 3) LPS-challenged + 200 μl MG6243, 4) LPS-challenged + 200 μl ΔMG6243-1, 5) LPS-challenged + 200 μl ΔMG6243-1 + inducer IPTG, and 6) LPS-challenged + 200 μl ΔMG6243-2. There were ten replicates for each treatment, and each replicate had one mouse. On d 15, the LPS-challenged mice were injected intraperitoneally with LPS at 10 mg/kg BW, while the non-challenged mice were injected with 0.9% NaCl solution. Three hours after the challenge, all mice were killed. Results showed that both ΔMG6243-1 and ΔMG6243-2 could increase villus height in the duodenum, ileum and jejunum (P < 0.05). In addition, the recombinant L. acidophilus decreased the crypt depth in the duodenum and ileum (P < 0.05). These results indicated that the recombinant L. acidophilus could effectively improve the intestinal histology in LPS-challenged mice. © 2018 Chulalongkorn University Printing House.

摘要:
Trihexanoin is a short-chain triglyceride (SCT). Many studies have reported that SCTs play important roles in the maintenance of intestinal epithelial structure and function. The present work was to investigate the effects of trihexanoin on growth performance, carbohydrate and fat metabolism, as well as intestinal morphology and function in weaned piglets. Twenty weaned piglets (21 ± 2 d) were randomly allocated to one of two treatment groups: The control group (basal diet supplemented with 0.5% soya oil); the TH group (basal diet supplemented with 0.5% trihexanoin). Dietary trihexanoin supplementation significantly reduced diarrhea rate; increased the concentrations of LDL, HDL and total protein in plasma; decreased cholesterol concentrations and glutamyl transpeptidase activity in plasma; improved intestinal morphologic structure; altered the mRNA levels and abundances of proteins related to glycogen and fat metabolism, mucosal barrier function, antioxidant capacity and water transport capacity; and altered the community of intestinal microflora. These results indicate that dietary trihexanoin supplementation could reduce diarrhea, regulate carbohydrate and fat metabolism, exert beneficial effects on the intestinal mucosal barrier, protect the intestinal mucosa from injuries, improve intestinal transport and absorption, and enhance antioxidant capacity. In conclusion, dietary supplementation with 0.5% trihexanoin improves the intestinal function and health of weaned piglets. © 2018 by the authors. Licensee MDPI, Basel, Switzerland.

A useful animal model of intestinal injury is pivotal for studying its pathogenesis and developing nutritional interventions (e.g., amino acid supplementation). Here, we propose the use of indomethacin (IDMT) to induce intestinal inflammation in neonatal pigs. Fourteen-day-old piglets fed a milk replacer diet receive intraperitoneal administration of IDMT (5 mg/kg body weight) for 3 consecutive days. On day 4, blood and intestinal samples are obtained for physiological and biochemical analyses. IDMT increases blood DAO activity, I-FABP concentration, neutrophil and eosinophil numbers; intestinal MMP3 mRNA levels, MPO activity, and MDA concentration; but reduces the plasma concentration of citrulline (synthesized exclusively by enterocytes of the small intestine), intestinal GSH-Px activity, and mRNA levels for villin, I-FABP, TRPV6, AQP10, and KCNJ13. Moreover, extensive hemorrhagic spots, thinned intestinal wall, and ulcers in the distal jejunum and ileum are observed in IDMT-challenged piglets. Furthermore, IDMT decreases intestinal villus height and villus surface area in the piglet jejunum. Collectively, this work establishes a porcine model of intestinal injury for designing novel nutritional means to improve gut function in pigs and humans. © 2018 Frontiers in Bioscience. 幼龄仔猪PEDV感染肠道损伤模型的建立

试验旨在建立猪流行性腹泻病毒(PEDV)感染幼龄仔猪肠道损伤模型。试验选用16头7日龄健康幼龄仔猪(杜×长×大),随机分为两个处理组:对照组和PEDV组,每个处理8个重复,每个重复1头猪。试验期为10d,试验期间两个处理组饲喂相同的基础日粮。于试验第7天晚上对PEDV组仔猪口腔灌服PEDV病毒(剂量为104.5 TCID50),对照组灌服等量的生理盐水。于试验第10天早上空腹灌服D-木糖(0.1g/kg体重),1h后前腔静脉采血,屠宰取样,取十二指肠、空肠、回肠等组织样品,测定平均日增重(ADG)、腹泻率(DR)、肠道形态结构、肠道黏膜损伤相关基因mRNA水平。试验结果表明:(1)PEDV感染显著降低了仔猪ADG(P<0.05),极显著提高了仔猪腹泻率(P<0.01);(2)PEDV感染极显著降低了血浆D-木糖含量、空肠和回肠绒毛高度、十二指肠、空肠和回肠绒毛高度与隐窝深度的比值(P<0.01),显著提高了十二指肠和空肠隐窝深度(P<0.05);(3)PEDV感染极显著提高了仔猪空肠黏膜PEDV M基因的相对表达量(P<0.01),极显著降低了肠绒毛蛋白(villin)和肠型脂肪酸结合蛋白(i-FABP)基因的相对表达量(P<0.01)。以上结果表明,口腔灌服PEDV可以成功诱导建立幼龄仔猪肠道损伤模型。

摘要:
The traditionally classified nutritionally non-essential amino acids are now known to be insufficiently synthesized for maximal growth and optimal health in piglets. This study determined the effects of dietary supplementation with an amino acid blend (AAB; glutamate:glutamine:glycine:arginine:N-acetylcysteine = 5:2:2:1:0.5) on piglet growth performance and intestinal functions. Sixteen piglets (24-day-old) were randomly assigned to a corn and soybean meal-based diet supplemented with 0.99% alanine (isonitrogenous control) or 1% AAB. On day 20 of the trial, blood and intestinal tissue samples were obtained from piglets. Compared with the control, AAB supplementation reduced (P < 0.05) diarrhoea incidence; plasma alanine aminotransferase and diamine oxidase activities; intestinal concentrations of hydrogen peroxide, malondialdehyde, and heat shock protein-70, and intestinal mRNA levels for interleukin-1β, interferon-γ, and chemokine (C–X–C motif) ligand-9; and the numbers of Enterobacterium family, Enterococcus genus and Clostridium coccoides in the colon digesta. Furthermore, AAB supplementation enhanced (P < 0.05): the plasma concentrations of serine, aspartate, glutamate, cysteine, tyrosine, phenylalanine, tryptophan, lysine, arginine, citrulline, ornithine, taurine, and γ-aminobutyric acid; intestinal villus height and surface area, villus height/crypt depth ratio, antioxidative enzyme activities, and mRNA levels for porcine β-defensin-1, sodium-independent amino acid transporters (b 0,+ AT and y + LAT1), aquaporin (AQP) 3, AQP8, AQP10, nuclear factor erythroid 2-related factor 2 and glutathione S-transferase omega-2, and protein abundances of AQP3, AQP4, claudin-1, occludin and myxovirus resistance 1; and the numbers of Bifidobacterium genus and Lactobacillus genus in the colon digesta. Collectively, these comprehensive results indicate that dietary AAB supplementation plays an important role in improving piglet growth and intestinal function. © 2018, Springer-Verlag GmbH Austria, part of Springer Nature.

Enterotoxigenic Escherichia coli (ETEC) is primary pathogenic bacteria of piglet diarrhea, over two thirds of piglets diarrhea caused by ETEC are resulted from STa-producing ETEC strains. This experiment was conducted to construct the recombinant E. coli expressing STa and study the injury and mechanism of recombinant E. coli expressing STa on 7 days old piglets colon. Twenty-four 7 days old piglets were allotted to four treatments: control group, STa group (2 × 109 CFU E. coli LMG194-STa), LMG194 group (2 × 109 CFU E. coli LMG194) and K88 group (2 × 109 CFU E. coli K88). The result showed that E. coli infection significantly increased diarrhea rates; changed DAO activity in plasma and colon; damaged colonic mucosal morphology including crypt depth, number of globet cells, density of lymphocytes and lamina propria cell density; substantially reduced antioxidant capacity by altering activities of GSH-Px, SOD, and TNOS and productions of MDA and H2O2; obviously decreased AQP3, AQP4 and KCNJ13 protein expression levels; substantially altered the gene expression levels of inflammatory cytokines. Conclusively, STa group had the biggest effect on these indices in four treatment groups. These results suggested that the recombinant strain expressed STa can induce piglets diarrhea and colonic morphological and funtional damage by altering expression of proteins connect to transportation function and genes associated with intestinal injury and inflammatory cytokines. © 2018 The Japanese Society of Veterinary Science.

Enterotoxigenic Escherichia coli strains induce human and animal intestinal dysfunction and injury, and cause diarrhea in weanling pigs. A recombinant E.coli strain LMG194-STa which expressed single toxin STa of ETEC was constructed by directly inserting the STa gene of ETEC into the expression vector pBAD202 and then transferring the recombinant plasmid pBAD-STa into the E.coli LMG19 host strain. Diarrhea and intestinal injury in piglets were induced by oral administration of recombinant strain LMG194-Sta. In vitro, the recombinant strain LMG194-Sta had the same toxicity to the IPEC cell as wild type strain K88, and higher toxicity than the host strain LMG194. In vivo, LMG194-STa caused severe diarrhea in piglets as K88, the diarrhea rate of LMG194-STa and K88 groups was higher than that in the LMG194 and control groups. Both K88 and LMG194-STa induced intestinal inflammation, injury, and atrophy, while adversely affecting the expression of genes for cytokines, transporters and ion channels, and nutrient metabolism. Thus, we established a porcine model of recombinant E. coli strain LMG194-STa-induced diarrhea for future nutritional and mechanistic studies of intestinal dysfunction. © 2018 Frontiers in Bioscience. The present study was to determine the efficacy of dietary supplementation with oleum cinnamomi (OCM) on growth performance and intestinal functions in piglets. Sixteen piglets (24-day-old) were randomly assigned to the control or OCM groups. Piglets in the control group were fed a basal diet, whereas piglets in the OCM group were fed the basal diet supplemented with 50 mg/kg OCM. On day 20 of the trial, blood samples and intestinal tissues were obtained from piglets. Compared with the control group, dietary OCM supplementation increased (p < 0.05) average daily feed intake, plasma insulin levels, villus width and villous surface area in the duodenum and jejunum, DNA levels and RNA/DNA ratios in the ileum, the abundance of Enterococcus genus and Lactobacillus genus in caecum digesta, mRNA levels for epithelial growth factor receptor (EGFR), Ras, extracellular signal-regulated kinase 1/2 (Erk1/2), b-cell lymphoma-extra large (Bcl-xL), villin, junctional adhesion molecule A (JAM-A), myxovirus resistance (MX) 1, MX2 and regenerating islet-derived protein 3 gamma (REG3G), and protein abundances of Ras and claudin-1, but decreased (p < 0.05) diarrhoea incidence; the abundances of Enterobacteriaceae family, Enterococcus genus, Lactobacillus genus, Bifidobacterium genus, and Clostrium coccoides in the colon digesta, and AMP-activated protein kinase (AMPK) mRNA levels and caspase-3 protein abundance in the jejunal mucosa of piglets. Taken together, these data indicate that dietary OCM supplementation modulates intestinal microbiota and improves intestinal function in weanling pigs. OCM is an effective feed additive and alternative to feed antibiotics for improving intestinal health in swine. © 2018 by the authors. Licensee MDPI, Basel, Switzerland.

This study aims to determine whether Lactobacillus casei (L. casei) could relieve liver injury in piglets challenged with lipopolysaccharide (LPS). Piglets were randomly allocated into one of the three groups: control, LPS, and L. casei. The control and LPS groups were fed a corn- and soybean meal-based diet, whereas the L. casei group was fed the basal diet supplemented with 6 × 106 cfu/g L. casei. On Day 31 of the trial, piglets in the LPS and L. casei groups receivedintraperitoneal administration of LPS (100 μg/kg body weight), while the control group received the same volume of saline. Blood and liver samples were collected for analysis. Results showed that L. casei supplementation decreased the feed/gain ratio (p = 0.027) and diarrhea incidence (p < 0.001), and attenuated LPS-induced liver histomorphological abnormalities. Compared with the control group, LPS challenge dramatically increased glutamyl transpeptidase activity (p = 0.001) in plasma as well as the concentrations of Interleukin 6 (IL-6) (p = 0.048), Tumor necrosis factor-alpha (TNF-α) (p = 0.041), and Malondialdehyde (MDA) (p = 0.001) in the liver, while decreasing the hepatic SOD activity. LPS also increased (p < 0.05) the mRNA levels for IL-6, IL-8, TNF-α, Toll-like receptors 4 (TLR4), Nuclear factorkB (NF-kB) and Heat shock protein 70 (HSP70) in the liver. The adverse effects of LPS challenge were ameliorated by L. casei supplementation. In conclusion, dietary L. casei alleviates LPS-induced liver injury via reducing pro-inflammatory cytokines and increasing anti-oxidative capacity. © 2017 by the authors. Porcine epidemic diarrhea virus (PEDV) infects the intestine of young pigs, but effective measures for prevention and treatment are lacking. N-Acetylcysteine (NAC) has been shown to reduce endotoxin-induced intestinal dysfunction. This study was conducted with the PEDV-infected neonatal piglet model to determine the effect of NAC supplementation on intestinal function. Thirty-two 7-day-old piglets were randomly allocated to one of four treatments in a 2×2 factorial design consisting of two liquid diets (0 or 50mg/kg BW NAC supplementation) and oral administration of 0 or 104.5 TCID50 (50% tissue culture infectious dose) PEDV. On day 7 of the trial, half of the pigs (n=8) in each dietary treatment received either sterile saline or PEDV (Yunnan province strain) solution at 104.5 TCID50 per pig. On day 10 of the trial, d-xylose (0.1g/kg BW) was orally administrated to all pigs. One hour later, jugular vein blood samples were collected, and then all pigs were killed to obtain the small intestine. PEDV infection increased diarrhea incidence, while reducing ADG. PEDV infection also decreased plasma d-xylose concentration, small intestinal villus height, mucosal I-FABP and villin mRNA levels but increased mucosal MX1 and GCNT3 mRNA levels (P<0.05). Dietary NAC supplementation ameliorated the PEDV-induced abnormal changes in all the measured variables. Moreover, NAC reduced oxidative stress, as indicated by decreases in plasma and mucosal H2O2 levels. Collectively, these novel results indicate that dietary supplementation with NAC alleviates intestinal mucosal damage and improves the absorptive function of the small intestine in PEDV-infected piglets. © 2017, Springer-Verlag Wien.

Haemophilus parasuis can cause a severe membrane inflammation disorder. It has been documented that superoxide dismutase (SOD) is a potential target to treat systemic inflammatory diseases. Therefore, we constructed an experimental H. parasuis subunit vaccine SOD and determined the protective efficacy of SOD using a lethal dose challenge against H. parasuis serovar 4 strain MD0322 and serovar 5 strain SH0165 in a mouse model. The results demonstrated that SOD could induce a strong humoral immune response in mice and provide significant immunoprotection efficacy against a lethal dose of H. parasuis serovar 4 strain MD0322 or serovar 5 strain SH0165 challenge. IgG subtype analysis indicated SOD protein could trigger a bias toward a Th1-type immune response and induce the proliferation of splenocytes and secretion of IL-2 and IFN-γ of splenocytes. In addition, serum in mice from the SOD-immunized group could inhibit the growth of strain MD0322 and strain SH0165 in the whole-blood killing bacteria assay. This is the first report that immunization of mice with SOD protein could provide protective effect against a lethal dose of H. parasuis serovar 4 and serovar 5 challenge in mice, which may provide a novel approach against heterogeneous serovar infection of H. parasuis in future. © 2017, Canadian Science Publishing. 试验旨在研究表达耐热肠毒素(STa)的重组大肠杆菌对7日龄仔猪肠道形态结构及抗氧化功能的影响。选取24头7日龄仔猪,随机分在4个日粮处理组,分别为对照组(人工乳),STa组(人工乳+2×10^9 CFU重组菌LMG194-pBAD-STa),LMG194组(人工乳+2×10^9 CFU大肠杆菌LMG194),K88组(人工乳+2×10^9 CFU大肠杆菌K88)。试验第5天进行攻毒,第7天屠宰取样,测量小肠黏膜的绒毛高度和隐窝深度,检测空肠、回肠、结肠及血清中的过氧化氢酶(CAT)、总一氧化氮合成酶(TNOS)与诱导型一氧化氮合酶(iNOS)的活力及血清中丙二醛(MDA)与过氧化氢(H_2O_2)的含量。试验结果表明,与对照组相比,STa组仔猪各肠段绒毛高度均显著降低(P〈0.05),十二指肠隐窝深度及绒毛高度与隐窝深度的比值显著降低(P〈0.05),十二指肠、空肠、回肠绒毛表面积显著降低(P〈0.05);同时,STa组回肠、结肠CAT活力显著降低,血清、空肠、回肠和结肠中的TNOS活力显著降低(P〈0.05),STa组结肠iNOS活力显著降低(P〈0.05),STa组血清中的MDA与H_2O_2含量显著提高(P〈0.05)。结果显示,表达STa的重组大肠杆菌可导致7日龄仔猪肠道结构损伤和抗氧化能力下降。

<jats:named-content content-type="genus-species">Clostridium perfringens</jats:named-content>
encodes at least two different quorum sensing (QS) systems, the Agr-like and LuxS, and recent studies have highlighted their importance in the regulation of toxin production and virulence. The role of QS in the pathogenesis of necrotic enteritis (NE) in poultry and the regulation of NetB, the key toxin involved, has not yet been investigated. We have generated isogenic
<jats:italic>agrB</jats:italic>
-null and complemented strains from parent strain CP1 and demonstrated that the virulence of the
<jats:italic>agrB</jats:italic>
-null mutant was strongly attenuated in a chicken NE model system and restored by complementation. The production of NetB, a key NE-associated toxin, was dramatically reduced in the
<jats:italic>agrB</jats:italic>
mutant at both the transcriptional and protein levels, though not in a
<jats:italic>luxS</jats:italic>
mutant. Transwell assays confirmed that the Agr-like QS system controls NetB production through a diffusible signal. Global gene expression analysis of the
<jats:italic>agrB</jats:italic>
mutant identified additional genes modulated by Agr-like QS, including operons related to phospholipid metabolism and adherence, which may also play a role in NE pathogenesis. This study provides the first evidence that the Agr-like QS system is critical for NE pathogenesis and identifies a number of Agr-regulated genes, most notably
<jats:italic>netB</jats:italic>
, that are potentially involved in mediating its effects. The Agr-like QS system thus may serve as a target for developing novel interventions to prevent NE in chickens.

摘要:
This study determined whether N-acetylcysteine (NAC) could improve intestinal function through phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR), epithelial growth factor receptor (EGFR), toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-κB), adenosine 5′-monophosphate-activated protein kinase (AMPK), and type I interferon (IFN) signaling pathways in a piglet model of lipopolysaccharides (LPS) challenge. Thirty-two piglets (24-day-old) were randomly allocated to one of four treatments, with eight replicates per treatment and one piglet per replicate. The experiment consisted of four treatments in a 2×2 factorial arrangement with two diets (supplemented with 0 or 500mg NAC/kg diet) and saline or LPS administration. On day 20 of the trial, piglets in the LPS and LPS+NAC groups were intraperitoneally injected with 0 (saline) or 100μg LPS/kg BW. Blood samples were obtained at 3h and intestinal mucosae were collected at 6h post LPS or saline injection. The growth performance was not affected by dietary NAC. LPS induced intestinal dysfunction, as indicated by: (1) reductions in the small-intestinal glutathione concentrations and plasma d-xylose levels; (2) elevations in plasma diamine oxidase activity, mucosal MMP3 mRNA levels and caspase-3 protein abundance; (3) reduced the activities of the small-intestinal mucosal maltase, sucrase and lactase. The adverse effects of LPS on porcine intestinal function and redox status were mitigated by NAC supplementation through the activation of multiple signaling pathways involving PI3K/Akt/mTOR, EGFR, TLR4/NF-κB, AMPK, and type I IFN. Our findings provide novel mechanisms for beneficial effects of NAC in protecting the intestine from inflammation in animals. © 2017, Springer-Verlag Wien.

大肠杆菌是导致仔猪腹泻的主要致病菌之一,耐热肠毒素A(STa)是其毒性最强的致病因子。为更好研究STa导致腹泻的机理,本试验尝试建立单一肠毒素STa作用下的仔猪腹泻模型。试验选取7日龄左右健康且大小匀称的杜×长×大三元杂交哺乳仔猪24头(未做大肠杆菌相关疾病免疫),按单因子设计随机分为4个组,每组设置6个重复,每个重复1头仔猪。4个处理组分别为空白对照组、大肠杆菌LMG194-pBAD-STa组(
